1.Use of Traditional Chinese Medicine Injections in Our Hospital during 2003~2006

OBJECTIVE:To evaluate the use of traditional Chinese medicine injections in our hospital for references of clinical rational administration.METHODS:The consumption sum,DDDs,daily drug costs(DDC) of traditional Chinese medicine injections in our hospital during 2003~2006 were analyzed statistically.RESULTS:Over the 4 years,the incremental rate of the consumption sum of traditional Chinese medicine injections in our hospital experienced a big change,with annual incremental rates of 187%,95%,and —0.4%,respectively.Dominating the first places over the 4 years in respect of DDDs has been the category for cardio-cerebral vessels.The antitumor traditional Chinese medicine injections took the lead in consumption sum and DDC.CONCLUSION:The consumption quantity of traditional Chinese medicine injections experienced a fast increase,suggesting its great market demand.But it has distinctive defects which limited its more broad use.
2.Studies on the relationship of PDCD 4 with the invasion of astrocytic gliomas

Objective To investigate the relationship of programmed cell death 4(PDCD4) with the invasion of astrocytic glio-mas .Methods Using the immunohistochemical method to detect the expression of PDCD4 in astrocytic gliomas in different grades . Measuring the peritumoral low-density area on MRI scan ,then compared with the results of immunohistochemical expression .Re-sults The downregulation of PDCD4 was with the increasing of the malignant grade of astrocytic gliomas .The tumor grade malig-nancy was positively correlated with the grade of the peritumoral low-density area on MRI scan(P<0 .05) ,while the expression of PDCD4 was negatively correlated with the grade of astrocytic gliomas (P<0 .01) .Conclusion PDCD4 might serve as one of the in-dicators of invasion and malignant phenotype for astrocytic gliomas .
3.Effect of electroacupuncture on c-Jun amino terminal kinase signaling pathway and inflammatory cytokines interferon-γ in substantia nigra cells of rotenone-induced rats model of Parkinson's disease

Objective To observe the effects of electroacupuncture (EA) on c-Jun amino terminal kinase (JNK) signaling pathway and inflammatory cytokines interferon-γ (IFN-γ) in substantia nigra (SN) cells of rotenone-induced rats model of Parkinson's disease (PD),and explore the underlying mechanism of EA on PD.Methods A total of 32 male Sprague-Dawley mice were randomly and evenly divided into a normal group,a shamoperation group,a model group and an EA group.Model group and EA group were injected intradermally with rotenone (1 mg/kg,dissolved in DMSO and saline,concentration:O.25 mg/ml) on the nape of neck.Sham-operation group was injected the same dose of DMSO and saline.Normal group had no intervention.EA group was applied to Fengfu (DU16) and Taichong (LR3) acupoints after the establishment of PD model in rats.Behavioral assessment was conducted after the treatments,the rats were sacrificed for sampling substantia nigra tissue to detect the expressions of tyrosine hydroxylase (TH),p-c-Jun amino terminal kinase (p-c-Jun) and interferon-γ(IFN-γ) protein with Western blotting (WB).Results Model rats showed significant PD syndrome characteristics,comparing with normal group and sham group,the difference was not statistically significant (P > 0.05).The results of open box test showed that the scores of model group rats decreased significantly in terms of the horizontal movement [(19.12 ±2.34) points] and vertical locomotor activity [(5.27 ± 1.04) points] when compared with normal group and sham group,the difference was statistically significant (P < 0.05).After EA treatment,locomotor activity of rats increased significantly when compared with model group (P < 0.05),however,the normal group and sham group was not statistically and significantly different in locomotor activity (P > 0.05).Compared with normal group,the expression of TH protein in (0.183 ± 0.0213) reduced significantly and the expressions of p-c-Jun (0.388 ± 0.0283) and IFN-γ protein(0.453 ± 0.0332) increased significantly in model group,the difference was statistically significant (P <0.05).Compared with normal group,the expression of TH protein(0.324 ± 0.0538) reduced and the expressions of p-c-Jun(0.207 ± 0.0592) and IFN-γ protein (0.239 ± 0.0215) increased in EA group,the difference was not statistically significant (P > 0.05).Compared with model group,the expression of TH protein increased significantly in EA group(P < 0.05),the expressions of p-c-Jun and IFN-γ protein reduced significantly in EA group(P < 0.05).Conclusion EA therapy may reduce the expression of IFN-γ protein in SN of PD rats model by regulating the expression of JNK/mitogen-activated protein kinase (MAPK) pathway,which may delay the process of PD.
4.Localization of genetic susceptibility loci for bipolar disorder on chromosome 20 in Shandong peninsular

Objective To identify the genetic loci associated with bipolar disorder in Chinese population by the ge?nome scan on chromosome 20 in bipolar disorder patients and healthy people from Shandong peninsula. Method Geno?typing was conducted on DNA pool samples of 104 bipolar disorder patients and 1000 controls using thirteen microsatel?lite markers on chromosome 20 spaced at intervals of approximately 10 cM and GeneMapper 4.0 software. CLUMP soft?ware was used to analyze multiallelic markers. Result Significant difference of allele frequencies was found at marker D20S186 (20p12.2) between patients and controls (P=0.022). Conclusion The data suggest that the D20S186 locus (20p12.2) is significantly associated with susceptibility to bipolar disorder in a Chinese Han population in Shandong pen?insula. Future studies should focus on the fine mapping of this region and assessment of candidate genes.
5.Study of zinc element and ZnT3 mRNA expression in mouse brain

AIM:To explore the interaction and function between ZnT3 mRNA expression and zinc elemental distribution in brain slice of mouse.METHODS:Zinc distribution was determined by SRXRF and ZnT3 mRNA expression in tissue was examined by RT-PCR method.RESULTS:Zinc content in cerebral cortex and hippocampus was significantly higher than that in other positions.The highest expression of ZnT3 mRNA was observed in cerebrum,hippocampus and testis.However,the ZnT3 mRNA was not detected in heart,liver,lung,spleen,kidney,intestine,olfactory bulb and cerebellum.CONCLUSION:ZnT3 facilitates the accumulation of zinc in synaptic vesicles and may play important roles in structuring of vesicular zinc pool.
6.A screen for genetic loci associated with bipolar disorder on chromosome 4

Objective To find out association mapping of loci related to bipolar disorder on chromosome 4 with microsatellite markers in DNA pooling samples from bipolar disorder cases and normal controls in Shandong province. Methods A total of 22 microsatellite markers on chromosome 4 spaced at approximately 10 cM were selected and two separated DNA pooling samples consisting of 104 bipolar disorder cases and 1000 normal controls were genotyped respectively. Statistic analysis was performed by Chi-square method with CLUMP software to compare the difference in the ratio of each allele in these loci between the two pooling samples. Result Significant statistic differences were found at D4S1592 and D4S402 on chromosome 4 between cases and controls(P<0.01 ).( D4S1592:x2 = 15.968, P=0.006; D4S402:x2 =31.553, P=0.002). Conclusion The loci of D4S1592 and D4S402 on chromosome 4 are found to be associated with bipolar disorder patients in Shandong province, further screening of the susceptibility genes around these loci is needed.
7.miR-205-5p/E2F1 signal axis is involved in the regulation of radiosensitivity of glioma cells through suppressing the classical Wnt/β-catenin signaling pathway

Objective:To explore the mechanism of miR-205-5p/E2F1 signal axis in regulating the glioma U251, U87 radiotherapy resistance.Methods:X-ray gradual ascending and intermittent induction method was used to irradiate the glioma U251 cells to establish U251/TR, U87/TR radiation-resistant cell lines. Then, the morphology, migration, invasion and proliferation abilities of cells (U251/TR, U87/TR radiation-resistant cells and U251, U87 radiation-sensitive cells) were analyzed. Luciferase gene detection system and point mutation technique were employed to analyze the mechanism of miR-205-5p and E2F1 gene activity on U251 and U87 radiation-resistant cell lines.Results:Compared with the radiation-sensitive U251 cells, the radiation-resistant cells U251/TR, U87/TR showed increased proliferation activity, enhanced migration and invasion abilities and decreased apoptosis under X-ray irradiation. miR-205-5p mimics transfection could down-regulate the expression of E2F1 factor in U251/TR cells, inhibit cell proliferation, invasion and migration and increase the radiosensitivity of U251/TR cells. miR-205-5p mimics transfection combined with with E2F1 down-regulation exerted anti-tumor effect and decreased cell tolerance by suppressing the Wnt/β-catenin signaling pathway activity.Conclusions:The glioma radiation-resistant cell line U251/TR, U87/TR can be established by X-ray gradual ascending and intermittent induction method. The miR-205-5p/E2F1 signal axis exerts tumor-suppressing effect through the classical Wnt/β-catenin signaling pathway, which can be used as an therapeutic target to increase the radiosensitivity of glioma.

Objective:To investigate the knowledge, attitudes, and practices (KAP) of pulse oximetry among pediatric healthcare providers in China and analyze the factor influencing the KAP.Methods:A self-developed questionnaire was used for an online research on the KAP of 11 849 pediatric healthcare providers from 31 provinces, autonomous regions, and municipalities of China from March 11 to 14, 2022.The factors influencing the KAP of pulse oximetry among pediatric healthcare providers were examined by Logistic regression. Results:The scores of KAP, of pulse oximetry were 5.57±0.96, 11.24±1.25 and 11.19±4.54, respectively.The corresponding scoring rates were 69.61%, 74.95%, and 55.99%, respectively. Logistic regression results showed that the gender and working years of pediatric healthcare providers, the region they were located, and whether their medical institution was equipped with pulse oximeters were the main factors affecting the knowledge score (all P<0.05). Main factors influencing the attitude score of pediatric healthcare providers included their knowledge score, gender, educational background, working years, region, medical institution level, and whether the medical institution was equipped with pulse oximeters (all P<0.05). For the practice score, the main influencing factors were the knowledge score, gender, age, and whether the medi-cal institution was equipped with pulse oximeters (all P<0.05). Conclusions:Chinese pediatric healthcare providers need to further improve their knowledge about and attitudes towards pulse oximetry.Pulse oximeters are evidently under-used.It is urgent to formulate policies or guidelines, strengthen education and training, improve knowledge and attitudes, equip more institutions with pulse oximeters, and popularize their application in medical institutions.
