1.GANODERMA SPORE MAY PREVENT THE DECREASE OF CELL PROLIFERATION AND NEURONAL SURVIVAL IN THE HIPPOCAMPUS OF FETAL AND POSTNATAL RATS INDUCED WITH GESTATIONAL HYPERTENSION

Objective To investigate the intervening effects of Ganoderma spore on the decrease of cell proliferation and neuronal survival in the hippocampus of fetal and postnatal rats induced with gestational hypertension. Methods Fourty SD pregnant rats were divided into four groups including the control group,Nw-nitro-L-arginine methylester(L-NAME)+distilled water(DW) group,L-NAME+L-Arginine group and L-NAME+Ganoderma spore(GS) group.The hippocampal tissue of the brain was detected by immunohistochemistry,Western blotting,RT-PCR,flow cytometry and electron microscopy. Results After the application of L-NAME,the expressions of hypoxia inducing factor-1?(HIF-1?) and vascular endothelial growth factor(VEGF) were increased at the hippocampus of E21 brain and continued up to P30 brain.The microvessel density of E21 hippocampus was increased and the structural abnormalities of blood capillary at P30 hippocampus were showed.The cell proliferation was decreased at E21 hippocampus and so was the neuronal number at P30 hippocampus.With the administration of Ganoderma spore,HIF-1? and VEGF were down-regulated at E21 hippocampus and were not detected at P30 hippocampus.The microvessel density of E21 hippocampus reached a normal level and the blood capillary ultrastructure of P30 hippocampus was restored.The cell proliferation of E21 hippocampus and neuronal number of P30 hippocampus were recuperatively increased.Conclusion Ganoderma spore may prevent the decrease of cell proliferation and neuronal survival in the hippocampus of fetal and postnatal rats induced with gestational hypertension.
2.A study of post-traumatic stress disorder symptoms and its drug intervention among middle age acute leukemia patients

Objective To investigate and understand the characteristics and drug intervention of post-traumatic stress symptoms(PTSD) among middle age acute leukemia patients.Methods The Essen Trauma-Inventory (ETI) was administrated to 100 middle age acute leukemia patients who were admitted in our department and study the PSTD symptoms.Results The positive rate of PTSD is 21.21% in sample of the patients.Total PTSD symptoms scores were (18.10 ± 9.42),there is no significant different between the 3 main PTSD symptoms in these patients(P > 0.05).Type of leukemia,sexuality,levels of education and marital status are influencing factors of PSTD symptoms,there is significant different between the subgroups (P <0.01).Drug intervention can significantly relieve PTSD symptoms in these patients (Z =13.56,P < 0.01).Conclusions Middle age acute leukemia patients are generally found PTSD symptoms and with a high incidence rate of PTSD,so it needs timely diagnosis and treatment.
3.The clinical effect of leukocyte filter preventing nonhemolytic febrile transfusion reactions

Objective To investigate the clinical effect of using leukocyte filter to prevent nonhemolytic febrile transfusion reactions(NHFTR) in hematologic patients who were trarisfused erythrocyte suspension. Methods Select 720 inpatients need erythrocyte transfusion and have transfusion records. The patients were randomly divided into investigation group and control group. We made a routine examination and recorded the patients' body temperature, heart rate, blood pressure, respiration rate and transfusion adverse reaction before and after erythrocyte suspeusion transfusion. Results Before blood transfusion, the body temperature of patients between two groups were normal, but after blood transfusion, there were 35 patients with NHFTR in the control group, while only 5 cases in the investigation group which used leucocyte filter, the difference between two groups is significant( P<0.01 ). The change of the patients' body temperature were compared between two groups before and after transfusion, the changes in the control group was significantly higher than the investigation group( P<0.01 ). Compared the changes of serum total bilirubin level in all patients before and after transfusion,there was no difference in both groups. The incidence of NHFTR increased with increasing of the transfusion times, but compared with the investigation group,the incidence of the control group was higher significantly(P<0.01 ). Conclusion Leukocyte filter can prevent the occurring of NHFTR efficiently, enhance the quality of transfusion,guarantee the blood transfusion safety.
4.Ganoderma spores may regulate the levels of mitochondria-related molecular substances in hippocampus of young rats birthed by rats with gestational hypertension

OBJECTIVE: To investigate the effects of Ganoderma spores on mitochondria-related molecular substances in hippocampus of young rats birthed by rats with gestational hypertension. METHODS: Nitric oxide synthase (NOS) inhibitor Nw-nitro-L-arginine methylester (L-NAME) was intraperitoneally injected into pregnant rats to induce gestational hypertension, and Ganoderma spores were administered orally. The effects of Ganoderma spores on levels of mitochondria-related molecular substances in hippocampus of young rats birthed by the rats with gestational hypertension were evaluated with immunoradiometric assay of cAMP, RT-PCR analysis of related genes, and detection of enzyme activity. RESULTS: In hippocampus of the new-born rats birthed by rats with gestational hypertension, the cAMP level, mitochondrial DNA (mtDNA) level and adenosine triphosphatase (ATPase) activity were decreased, and the expression level of peroxisome proliferator activated receptor gamma coactivator 1 alpha (pgc1 alpha) was unchanged compared to the normal control group. The cAMP level, mtDNA level, ATPase activity and pgc1 alpha expression level in hippocampus of 30-day post-natal rats were lower than those of the rats in normal control group. After oral administration of Ganoderma spores, the cAMP and mtDNA levels in hippocampus of the new-born rats and 30-day post-natal rats recovered almost to the levels of normal control rats, and the ATPase activity and pgc1 alpha expression level were also increased significantly. CONCLUSION: Ganoderma spores may regulate the levels of mitochondria-related molecular substances in hippocampus of young rats birthed by rats with gestational hypertension.
5.Primary study on proteomics about Ganoderma lucidium spores promoting survival and axon regeneration of injured spinal motor neurons in rats

OBJECTIVE: To detect some proteins associated with the effect of ganoderma lucidium spores (GASP) on promoting the survival and axon regeneration of injured spinal motor neurons in rats. METHODS: The rats were divided into normal control group, untreated group and GASP-treated group, and the rats in the last two groups received ventral root avulsion. GASP preparation was fed to the rats in the GASP-treated group for 14 days. The gray matter tissues of the lumbar spinal were sampled from rats in each group after 14 days following ventral root avulsion, and the extracted proteins from these tissues were detected by using 2-dimensional electrophoresis. Matrix assisted laser desorption/ionization time-of-flight mass spectroscopy (MALDI-TOF MS) was utilized to identify the differentially expressed proteins among these three groups. RESULTS: There were six kinds of proteins differentially expressed among the three groups, which were collapsin response mediator protein 2 (CRMP-2), F-actin capping protein beta subunit (FCP-beta), isocitrate dehydrogenase [NAD] subunit beta (IDH-beta), ATPase, glutamate oxaloacetate transaminase-1 (GOT1) and M2 pyruvate kinase (M2-PK). The expression levels of CRMP-2, IDH-beta, ATPase and GOT1 were higher in the GASP-treated group than those in the untreated group, while the expression levels of FCP-beta and M2-PK were lower than those in the untreated group. CONCLUSION: GASP maybe promotes the survival and axon regeneration of injured spinal motor neurons in rats by virtue of up- or down-regulating the expression levels of the proteins mentioned above.
6.Research progress in spore-based in-situ restoration technology of concrete with microorganisms.

Concrete is the most widely used modern building material. It is easy to crack under the action of stress, which makes the concrete structure permeable, affecting the durability and integrity of the structure, and thus shortening its service life. Microbial in-situ remediation technology is a low cost, effective and green way for concrete crack repairing. Due to its excellent biocompatibility, service life elongation, economic losses and environmental pollution reduction, microbial in-situ remediation technology has been intensively investigated. Bacillus has attracted much attention because of its excellent biomineralization ability, extremely strong environmental tolerance and long-term survival ability of its spores. In order to promote the research, development and large-scale application of microbial in-situ healing of concrete, the paper reviews the mechanism of spore-based in-situ healing of concrete, the survival of spores exposed in concrete, the influence of spores and external additives on the mechanical properties of concrete, progress in research and development of healing agent as well as healing effects. Moreover, future research focuses such as improving the survival ability of spores in the harsh environment of concrete, reducing the influence of external additives on the mechanical properties of concrete, and strengthening the healing effect of actual field applications are also summarized.

As a strategic emerging industry of China, the biotechnology industry develops rapidly in recent years, which significantly increased the demand for creative and capable talents. As a core curriculum of bioengineering specialty, biotechnology equipment plays an important role in fostering such talents. To address the problems in biotechnology equipment course teaching such as limited equipment availability, limited engineering practice, and lack of learning motivations, curriculum reform and optimization were performed based on curriculum resource development, virtual reality-physical combined engineering training, and boosting learning motivations. The optimized teaching contents focus on fostering morality, intelligence, and creative practice abilities by connecting new requirements of social development, introducing new progress in biotechnology research, as well as new practices in research and development (R & D). Measures such as teaching methods innovation, assessment and evaluation methods optimization, cutting-edge R & D progress, diverse resources integration, and online-offline combined teaching, were developed to boost the learning motivation and foster the innovation competence of students. By above exploration and practice, the practice and innovation competence of students were significantly enhanced.
