1.In vitro Proliferation and Cytotoxicity of PBMC Induced by Autologous Tumor Specific Antigen Costimulated with IL-2

To observe its proliferation and cytolytic activity in vitro induced by autologous soluble tumor antigen and IL-2, PBMC costimulated with TSA extracted from gastric tumor by biochemical procedure and IL-2 was cultured. The proliferation and cytotoxicity were evaluated by MTT method and the phenotype was analysed by APAAP method. TSA alone did not induce PBMC proliferation while TSA (10?g/ml) with IL-2 presented syngenic stimulating effect which was time and concentration dependent. The cytolytic unit of PBMC induced with IL-2/TSA was 86.3% vs 48.6% with IL-2 alone. The phenotypic measurement demonstrated that TSA enhanced CD25 and CD8 expression significantly. The observation offered experimental data for further research work about tumor activated killer(TAK) cells. The results indicated that TAK cells would be a new type of immunoactive cells in adoptive immunotherapy against tumor.

3.Study on assessment indicator system of function for standardized patients in the objective structured clinical examination for nursing specialty

Objective To develop an assessment indicator system of function for standardized patients in the objective structured clinical examination for nursing specialty,which provides the basis for the standardized training and assessment of standardized patients.Methods The Delphi method was used to determine this indicator system.Results After two rounds of the Delphi procedure,the assessment indicator system was drawn-up,which included 3 first-class indicators and 12 second-class indicators and 35 third-class indicators with weights.Conclusions The achievements of this study may provide reference for nursing educators to screen standardized patients strictly and improve the quality of standardized patients continuously,and also provide the basis for development of scale or instrument in the further.
4.The endothelial function under various blood glucose levels and the protective effect of PGC-1α on macrovasculature in patients with diabetes mellitus

Objective To explore the vascular endothelial function in individuals with normal glucose tolerance (NGT),impaired glucose tolerance (IGT),and type 2 diabetes mellitus,and to study the role played by PPARγcoactivator 1 α (PGC-1 α) in diabetic macrovascular function.Methods A total of 94 subjects with NGT,87with IGT,and 306 with type 2 diabetes mellitus were recruited in the study.Carotid flow-mediated vasodilatation (FMD),glyceryl trinitrate dilatation (GTN),and carotid intima-media thickness (CIMT) were assessed by highresolution B-mode color vascular Doppler instrument.Finally,correlation analysis was conducted.NO,Akt-Ser473phosphorylation levels,IL-β,and PGC-1 α mRNA expression levels were measured by means of nitrate reductase,western blot,and reverse transcriptase real-time fluorescence quantitative PCR assay methods in peripheral blood mononuclear cells respectively.Results FMD in newly-diagnosed type 2 diabetic group was significantly lower than that in NGT and IGT groups(P<0.01).GTN and CIMT showed no difference.PGC-1 α mRNA transcription level in diabetic group was significantly lower than that in non-diabetic groups (P<0.01).Correlation analysis showed that PGC-1 α was positively correlated with FMD and GTN (P<0.05 or P<0.01).Logistic regression analysis revealed that PGC-1 α was independently associated with FMD (P < 0.01) after adjusting other influencing factors.Conclusion FMD is a more sensitive early warning indicator of endothelial function than CIMT and GTN.As indices of atherosclerosis,CIMT and GTN are more or less focused on the progression of macrovascular complications.PGC-1 α is a protective factor of macro-vasculature in diabetes mellitus,and its effect may be achieved through the protection of the vascular endothelium.
5.Correlation analysis of serum exsomal miRNA-155-5p expression with prognosis in patients with esophageal squamous cell carcinoma

Objective:To investigate the relationship between the expression of serum exsomal miRNA-155-5p (miR-155-5p) and prognosis in patients with esophageal squamous cell carcinoma (ESCC).Methods:A total of 336 samples from ESCC patients in Fujian Provincial Cancer Hospital from October 2014 to December 2015 were collected. The relative expression levels of serum exsomal miR-155-5p were detected by using real-time fluorescent quantitative polymerase chain reaction (qRT-PCR). Cut-off value of the expression levels of serum exsomal miR-155-5p was determined by using X-tile software. Based on the optimal cut-off value, patients were divided into miR-155-5p low expression group and miR-155-5p high expression group. The survival curve was drawn by using Kaplan-Meier method and Cox proportional hazards regression model was used to make survival analysis.Results:The cut-off value of serum exsomal miR-155-5p expression level was 2.340. According to the cut-off value, patients were divided into miR-155-5p low expression group (<2.340) of 51 cases and miR-155-5p high expression group (≥2.340) of 285 cases. There were no statistical differences in age ( χ2 = 0.020, P = 0.887), gender ( χ2 = 0.283, P = 0.595), tumor location ( χ2 = 0.063, P = 0.977), differentiation grade ( P = 0.474), clinical staging ( χ2 = 3.996, P = 0.136) and surgery treatment ( χ2 = 0.941, P = 0.332) of patients in both groups. ESCC patients in serum exsomal miR-155-5p high expression had a higher risk of death compared with patients in miR-155-5p low expression group ( HR = 1.763, 95% CI 1.049-2.961, P = 0.032). Conclusion:The high expression level of serum exsomal miR-155-5p is associated with poor prognosis in ESCC patients and it could be used as a prognostic new marker in ESCC patients.
6. Relationship among loneliness at workplace,job burnout and turnover intention in prison policemen

OBJECTIVE: To explore the relationship among loneliness at workplace, job burnout, and turnover intention in prison policemen. METHODS: A convenience sampling method was used to select a total of 290 prison policemen officers as the study subjects in Guangxi Zhuang Autonomous Region. The Workplace Loneliness Scale, Job Burnout Scale and Turnover Intention Scale were used to investigate the loneliness at workplace, job burnout and turnover intention respectively.RESULTS: The scores of turnover intention of 31-40 age group were higher than that of 41 or over age group(P<0.01). The scores of turnover intention in the length of service 1-5 years group and 6-10 year length of service group were higher than that of length of service over 16 years or more(P<0.05). The scores of turnover intention of section group policemen were higher than that of division group(P<0.05). The loneliness in the workplace was positively correlated with job burnout[Spearman rank correlation coefficient(r_S)=0.343, P<0.01], and turnover intention(r_S=0.392, P<0.01), and job burnout was positively correlated with turnover intention(r_S=0.408, P<0.01). Job burnout was the intermediate variable between loneliness in the workplace and turnover intention with a mediating effect rate of 60.2%. CONCLUSION: The job burnout of prison policemen plays a partial role in mediating the influence of loneliness at workplace on turnover intention.
