1.Effect of captopril on prolongation of action potential duration and reduction Ik in ischemia pig ventricular myocytes

AIM: To evaluate the effect of captopril on action potential duration and outward delayed rectification potassium current (Ik). METHODS: Action potentials were recorded using a conventional glass microelectrode filled with 3 mol/L KCl solution. Membrane patch clamp whole cell recording technique was used to investigate the Ik current maximum in the holding potential - 50 mV, lasting time 100 ms, command potential + 40 mV. RESULTS: The action potential duration of 30%, 50% repolarization (APD30, APD50) and ERP were significantly prolonged, but APD90 wasn' t prolonged significantly when captopril group compared with ischemic group. The amplitude of Ik increased significantly in ischemic group, but significantly decreased in captopril group and in captopril + ischemic group. The shapes of current - voltage relationship were unchanged among groups, but significantly upward in ischemic group and downward in captopril and captopril + ischemic group. CONCLUSION:Captopril exerts electrophysiologic action due to decreasing delay outward rectification potassium current and prolonging action potential duration of APD30, APD50 and ERP.
2.Comparing study of echocardiography and X-ray angiography on measurement of patent ductus arteriosus

Objective To measure the sizes of patent ductus arteriosus (PDA) by echocardiography,compare the results with those by X-ray angiography(XA),to analysis the correlation of measurements between echocardiography and XA,to explore the value of echocardiography on screening the size of PDA suitable for transcatheter occlusion.Methods One hundred and forty-one participants was included.The parameters of PDA including the length of PDA(PDAL),the aortic side diameter (PDADao) and the pulmonary side diameter of PDA(PDADpa) were measured from the parasternal great artery short-axis view (PSSA) and the suprasternal descending aorta long-axis view(SSLA) of echocardiography and left lateral projection of XA,and the other parameters such as the left ventriclular end-diastolic diameter index (LVEDDI),the main pulmonary artery diameter index(MPADI),the pressure drop of PDA shunt (△P) and the pulmonary artery systolic pressure index(PASPI) were measured by echocardiography.The sizes of PDA from PSSA,SSLA and XA were compared,and the correlations of the sizes of PDA between echocardiography and XA,the correlations between the other parameters by echocardiography and the PDADpa index(PDADIpa) by XA(PDADIpa-XA) and PSSA(PDADIpa-PSSA) were analyzed.Results ①The difference of the sizes of PDA:Between PSSA and SSLA had difference on mersurement of PDA(all P < 0.05),except SSLA had no difference on measurement of PDADao with XA (P >0.05),others echocardiography parameters all had difference on measurement of PDA with XA (all P < 0.05).②Correlation:Both PSSA and SSLA had positive correlations with XA (all P <0.01).And among these parameters,PDAL by SSLA,PDADao by SSLA and PDADpa by PSSA had better correlations with XA (r =0.92,0.87 and 0.91,respectively,all P <0.01).LVEDDI,MPADI and PASPI had positive correlations with PDADIpa (all P <0.01),the LVEDDI and MPADI had better correlations with PDADIpa-PSSA(r =0.76,0.72,all P <0.01),but the PASPI had a better correlation with PDADIpa-XA(r =0.70,P <0.01).PASP had a negative correlation with △P (r =-0.79,P <0.01).Conclusions There was difference between the sizes of PDA at different echocardiographic views,PDADpa by PSSA and the PDAL and the PDADao by the SSLA were more close to those parameters by XA.Multiplane views of echocardiography can evaluate all the more accurately the size of PDA and play an important role in guiding transcatheter occlusion of PDA.
3.Esketamine reduces ischemia-reperfusion injury of skeletal muscle of rats

Objective To investigate the role of low dose esketamine in pretreated limb ischemia-reperfusion inju-ry.Methods The rats were divided into sham-operated group(Sham group),ischemia-reperfusion group(I/R group,3 h of ischemia and 2 h of reperfusion),esketamine group(ESK group,ip,5 mg/kg).The plasma con-centrations of creatine kinase(CK)and lactate dehydrogenase(LDH)were measured.The wet/dry weight ratio of skeletal muscle was immediately detected.The gastrocnemius muscle was harvested and the level of malondial-dehyde(MDA)and superoxide dismutase(SOD)was detected by colorimetric assay.The expression of nuclear factor erythroid 2-related factor 2(Nrf2)and heme oxygenase-1(HO-1)were detected by Western blot and im-munohistochemical staining.Results Compared with Sham group,the index of wet/dry weight ratio,MDA,CK,LDH,Nrf2 and HO-1 were all increased,but SOD was decreased in I/R group(P<0.05).Compared with I/R group,the index of wet/dry weight ratio,MDA,CK and LDH were significantly lower,but SOD and Nrf2 and HO-1 were significantly higher in ESK group(P<0.05).Conclusions ESK may increase Nrf2 in the nucleus,thereby increase the HO-1 protein as an antioxidant agent.
4.Preparation of lactoferrin modified nanogel dropping pills and study on drug transport into brain

Objective To improve the water solubility of natural drugs and the efficiency of their transport into the brain,to prepare nanogel dropping pills modified with lactoferrin for loading poorly soluble drugs and investigate the preparation process,and to provide a reference for drug development in the field of neurodegenerative diseases.Methods Using the insoluble drug icariin as the payload,the lactoferrin-modified zinc alginate nanogels were prepared by the reverse-phase microemulsion method.The drug loading and encapsulation rates of icariin in the nanogel were determined by HPLC.Taking the utilizing parameters such as roundness,weight difference and disintegration time as evaluation indicators,the best preparation process of dropping pills was optimized by single factor experiments and Box-Behnken response surface methodology.It was characterized by Fourier transform infrared spectroscopy,the release behavior of nanogel in dropping pills was explored by basket rotating method,and the brain entry efficiency between sublingual and oral administration of drip pills were compared by fluorescence tracing method.Results The drug loading rate of the nanogel was(2.67±0.05)%,and the encapsulation rate was(84.74±0.03)%by HPLC.The optimal preparation process for lactoferrin modified nanogel dropping pills was as follows:the ratio of matrix to nano gel was 5:1,the drop distance was 5.5 cm,and the condensation temperature was 8.5℃.Using the modified lactoferrin as a marker,its cumulative release rate in vitro reached 92.25%.Fluorescence tracing studies showed that compared to the oral group,sublingual administration of dripping pills facilitated the faster and more effective passage of drugs through the blood-brain barrier,significantly enhanced the efficiency of drug transport into the brain.Conclusion The prepared lactoferrin-modified nanogels can be used as a loading platform for poorly soluble drugs and enhance the drug solubility.When further formulated into dripping pills,it significantly enhances the efficiency of drug transport into the brain after sublingual administration.This also enhances medication compliance among neurodegenerative patients and fulfills the clinical demand for the drug.
5.Impact of surgical approach on the efficacy and safety of patients with primary central nervous system lymphoma

Objective:To investigate the effects of different surgical approaches on the efficacy and safety of patients with primary central nervous system lymphoma(PCNSL).Methods:The clinical data of 130 PCNSL patients admitted to The First Affiliated Hospital of University of Science and Tecnology of China between July 2012 and May 2023 were retrospectively analyzed.Patients were assigned into resection and biopsy groups according to the surgical approach.The safety and efficacy of the two groups were compared,including the objective re-sponse rate(ORR),overall survival(OS),and progression-free survival(PFS)after chemotherapy.Results:ORR and 2-year OS and PFS rates of the resection group were not significantly different from those of the biopsy group(ORR:63.2%vs.62.8%;2-year OS rate:68.6%vs.73.7%;2-year PFS rate:35.2%vs.40.7%,all P>0.05).The incidence of postoperative complications was significantly higher in the resection group than in the biopsy group(29.3%vs.11.4%,P=0.04).The difference in the length of hospitalization between the two groups was not statistic-ally significant.Treatment of the biopsy group was less expensive(P<0.01).Conclusions:In patients with PCNSL,biopsy and resection had comparable efficacy;however,the safety profile was better,and hospitalization was less costly in biopsy than in resection.
