Sixty cases of acute lumbar sprain were treated by puncturing Yintang(Ex-HN 3) toward Shenting(GV 24), among which, 50 cases were cured, 9 were improved and only one failed, and the total effective rate was 98.3%.

BACKGROUND: Changes of leukocytes are related with some postoperative complications after renal transplantation. OBJECTIVE: To observe dynamically the level of white blood cell (WBC) and its morphological changes using automatic blood cell analyzer to evaluate its relationship with infection, immunosuppressive intoxation and acute rejection. DESIGN, TIME AND SETTING: Observation and controlled trial was performed at Second Xiangya Hospital, Central South University from June 2004 to October 2005. PARTICIPANTS: Sixty-eight patients who received renal transplantation were selected. According to clinical symptoms and laboratory biochemical indexes, X-ray, B ultrasound, and renal puncture biopsy, the patients were divided into stable group (n=36), infection group (n=9), intoxation group (n=6) and acute rejection group (n=13). METHODS: The blood routine was tested with Sysmex XE-2100 automatic blood analyzer following renal transplantation and WBC manual differentiation was done to observe peripheral blood morphology, their liver function. In addition, renal and liver functions were detected by automatic biochemical system 7600A. MAIN OUTCOME MEASURES: WBC count and its morphological changes in peripheral blood and liver and renal function after renal transplantation. 3.Clinical significance of thrombospondin 4 expression in gastric carcinoma

Objective The aim of this study was to measure quantitatively the TSP-4 mRNA expression and its significance in gastric carcinoma was evaluated by correlating its expression with clinicopathological features, microvessel density(MVD) and MMP-9.Methods Eighty-two patients with gastric carcinoma were recruited in this study. TSP-4 mRNA expression in gastric carcinoma and adjacent tissue was detected by real-time PCR. CD34 and MMP-9 protein expression were examined by immunohistochemistry. MVD was determined according to CD34-positive tubular structures. Results The expression level of TSP-4mRNA in gastric carcinoma was obviously higher than those of adjacent tissue(P=0.03) and it was associated with tumor size, histological type, lymph node metastasis, MVD and MMP-9(P=0.002, P=0.031, P=0.014,r=0.67 P<0.01, P=0.008),but not sex, age and depth of invasion. (P=0.53,P=0.57,P=0.15).Conclusion TSP-4 may play an important role in occurrence and progression of gastric carcinoma and that the effect of TSP-4 on tumor growth and metastasis may be exerted through regulation of angiogenesis and MMP-9 expression in gastric carcinoma.

5. Quantitative Analysis of Relationship Between Gut Flora Dysbiosis and Severity of Liver Cirrhosis

Background: The incidence of liver cirrhosis is increasing year by year in China. Decompensated liver cirrhosis and its related complications seriously threaten people's health. Aims: To investigate the changes of gut flora in cirrhotic patients and its correlation with the severity and prognosis of the disease. Methods: One hundred patients with liver cirrhosis admitted from Dec. 2010 to Jul. 2011 at the Nanfang Hospital, Southern Medical University were enrolled. Fifty healthy subjects were served as controls. Using genus-specific primers and genomic DNA extracted from the fecal samples, a quantitative analysis of the gut flora was conducted by real-time PCR. Relationships between the changes of gut flora and grade of liver function, prognostic score, inflammatory indicator and complications of cirrhosis were analyzed. Results: Compared with healthy subjects, fecal Enterobacterium was significantly enriched, while fecal Lactobacillus, Bacteroides and Clostridium were significantly reduced in cirrhotic patients (all P<0.05). Enterobacterium was positively correlated with the grade of liver function, Child-Turcotte-Pugh (CTP) score, model for end-stage liver disease (MELD) score, C-reactive protein (CRP), and the occurrence of spontaneous peritonitis, respectively (all P<0.05), while Clostridium was negatively correlated with the above mentioned indices (all P<0.05). Conclusions: Gut flora dysbiosis, including increase in Enterobacterium and decrease in Clostridium, is correlated with the severity and prognosis of patients with liver cirrhosis. It may aggravate liver cirrhosis by promoting an inflammatory response.
6. Quantitative Analysis of Major Shifts in Gut Microbiota Before and After Helicobacter pylori Eradication

Background: Helicobacter pylori (Hp) infection occurs in about half of the world's population, and Hp eradication therapy is an important method for treating Hp infection-related diseases. Aims: To explore the major shifts in gut microbiota before and after Hp eradication by using bismuth-containing quadruple therapy. Methods: Fifty Hp-positive patients in whom the Hp was successfully eradicated by bismuth-containing quadruple therapy from January 2016 to January 2017 at the Third People's Hospital of Shenzhen were enrolled. Using genus-specific primers and genomic DNA extracted from the fecal samples collected before and within 3 days after eradication therapy, a quantitative analysis of the major shifts in gut microbiota was accomplished by real-time PCR. Results: Compared with those before Hp eradication, fecal Enterobacterium and Enterococcus were significantly increased and Bifidobacterium, Bacteroides, Clostridium and total bacteria were significantly decreased after Hp eradication (P<0.05). No significant change was observed in Lactobacillus (P>0.05). Conclusions: Eradication of Hp infection by bismuth-containing quadruple therapy may cause changes in gut microbiota composition and resulting in dysbiosis.
7.The application of antiskid steel plate on supination-eversion ankle fracture

Objective To study the application effect of antiskid steel plate internal fixation via exposing lateral and posterior ankle fracture through fibula posterolateral approach in the treatment of Lauge-Hansen supination-eversion ankle fracture.Methods From January 2009 to December 2012,exposed the lateral and posterior ankle fracture through fibula posterolateral approach of 36 patients suffering from the Lauge-Hansen supination-eversion ankle fracture,and fixed the lateral and posterior ankle by the antiskid steel plate.Adopt Olerud-Molander ankle function scoring system to evaluate the postoperative effect of the ankle.Results All the patients were followed up for 7-27 months,with an average follow-up of 19 months.There was no complications such as incision infection and cutaneous necrosis.Only 1 patient occurred with peroneus myotenositis,and the symptom was relieved after taking out the steel plate.The fracture was healed within 16 weeks.Among the 36 cases,22 cases were excellent,9 cases were good,4 cases were medium and 1 case was poor.The fine rate was 86.1%(31/36).Conclusion Using the antiskid steel plate to treat the lateral and posterior ankle fracture through fibula posterolateral approach,can fully expose a single incision,fix the lateral and posterior ankle and reduce the tension in suture,improve the incision healing rate and fracture stability,provide a new method for treating ankle fracture.
8.Expression of human FLT3 ligand gene in human bone marrow stromal cells.

OBJECTIVETo construct a retroviral-mediated vector of FLT3 ligand (FL) and express it in human bone marrow stromal cells.
METHODSFL cDNA was inserted into the retroviral vector pLXIN by gene recombination technology. The recombinant plasmid pLFIN was transferred into retrovirus packaging cell line PA317 by lipofectamine, and the positive clones were selected by G418. The mRNA expression in human stromal cells and integration of genome DNA were assayed by reverse transcriptase-polymerase chain reaction (RT-PCR) and genomic DNA-PCR. The expression of FL protein and its biological activities in the culture were investigated by ELISA and mouse bone marrow CFU-GM assay.
RESULTSThe recombinant plasmid pLFIN was successfully constructed. In genome of these transfected target cells, Neo gene and FL gene were integrated, FL mRNA was transcripted and FL protein was expressed at 4.35 ng/ml/24 h. The specific activities of FL in the culture indicated that human bone marrow stromal cells transfected with FL could significantly express FL in vitro.
CONCLUSIONThe retroviral-mediated FL gene was expressed in bone marrow stromal cells and the biological activities of FL were detectable in the supernatant of the transfected cells. These results provide a basis for studies on hematopoietic regulation by gene transfected bone marrow stromal cells.

Trauma is the leading cause of death for people under 40 years old in the world. At present, the rescue and treatment system of trauma patients in China is not yet well established, and the mortality of trauma patients is higher than those in the developed countries. Improving the treatment system is the key to reducing the trauma mortality. In order to innovate the service mode of trauma first aid, further promote the establishment of regional trauma first aid system, improve the ability of trauma treatment, reduce the mortality and disability rate of trauma patients in Jiangxi Province, recently Health Commission of Jiangxi Province and the First Affiliated Hospital of Nanchang University have reached a consensus on the establishment of Jiangxi trauma first aid center. In order to provide reference for the construction of trauma treatment system, the author analyzes the following aspects including functional positioning, basic requirements, organization management, and evaluation of core indicators.
