The histological and biomechanical aspects of the osteogenesis in metaphysioto-my lengthening of the tibia were observed in 45 immature rabbits, which were divided into 3 groups as follows:(1)Group A:After metaphysiotomy, the periosteum and endosteum of the transected boneends were well-preserved.(2)Group B:After metaphysiotomy, the periosteum about 0.7 cm in width on the 2 bone ends was circumcised. Then the 2 bone ends were aligned without disturbing the endosteum.(3)Group C:After metaphysiotomy,a part of the bone marrow and endosteum in the 2 bone ends wwere removed. Then the 2 bone edns were well aligned and the periosteum was sutured up.In all the experimental animals, the tibia was lengthened by 1mm/day.It was found that in Group A,the formation and remodelling of the new bone in the lengthened area occurred quite early after metaphysiotomy and the new bone was good in its biomechanical quality;in Group B,the formation of new bone was retarded and the biomechanical quality of the new bone was not satisfactory;and in Group C,new bone formation was sluggish in the early stage after metaphysiotomy but it wwas active in the late stage, and the neww bone formed was similar to that of Group A in its histological morphology and biomechanical quality. The periosteum could grow synchronously with the slow traction without any tearing. In addition, osteotomy and mechanical traction could stimulate and accelerate periosteal osteogenesis. It is believed that an intact periosteal cuff is of significance in the osteogenesis of the lengthened area since it serves as a restriction framework for the adherent growing of the connection between the new bones of the 2 bone ends and periosteal osteogenesis plays a more important role than endosteal osteogenesis in the bony healing of the lengthened area.

OBJECTIVE:To evaluate therapeutic efficacy of tolvaptan combined with levosimendan in the treatment of severe congestive heart failure(CHF). METHODS:100 severe CHF patients were randomly divided into basic treatment group, tolvaptan group, levosimendan group and drug combination group, with 25 cases in each group. All groups were given Digitalis,ACEI,spi-rolactone,furosemidum and other comprehensive treatment;tolvaptan group was additionally given tolvaptan(15 mg/time,qd);le-vosimendan group given levosimendan [0.1 μg/(kg·min)] for consecutive 24 h;drug combination given tolvaptan (15 mg/time, qd)and levosimendan [0.1 μg/(kg·min)] for consecutive 24 h. Blood pressure, heart rate, dyspnea, the improvement of systemic clinical symptoms and 24 h urine volume were observed in 4 groups before and 7 d after treatment. The venous blood samples were collected to determine the change of Na+,K+,renal function,BNP and other related indicators;the changes of LVEDd,LVESd, LVEF,CI and other indicators were determined by UCG. RESULTS:7 days after administration, compared with basic treatment group, the blood pressure, heart rate,BNP,LVEDd and LVESd of tolvaptan group, levosimendan group and drug combination group decreased significantly, while dyspnea, systemic clinical symptoms and LVEF were improved significantly, and 24 h urine volume and CI increased significantly, with statistical significance(P<0.05). The change of drug combination group was more sig-nificant,there was statistical significance, compared with tolvaptan group and levosimendan group(P<0.05). There was no signifi-cant difference in serum electrolytes and renal function. CONCLUSIONS:Tolvaptan combined with levosimendan is effective and better than drug alone in the treatment of severe CHF.

Objective Interleukin-21(IL-21) is likely to contribute to the development of liver fibrosis, but up to now, no study has been reported on the relationship between IL-21 and intrauterine adhesions ( IUA) .This study aimed to establish a rat model of IUA induced by mechanical injury and lipopolysaccharide (LPS) infection (dual injury), determine the expression level of serum IL-21, and confirm the association of serum IL-21 with the formation of IUA. Methods Forty healthy female SD rats were randomly divided into four groups of equal number, control, mechanical injury, LPSinfection, and dual injury. At 7 days after IUA modeling, uterine tissue-swere collected from all the animals for observation of the endometrial glands, detection of the degree of IUA by Masson staining, measure-ment of the serum IL-21 level by radioimmunoassay, and analysis of the correlation between the number of endometrial glands and the de-gree of fibrosis. Results The number of endometrial glandswas significantly smallerin the dual injury group (3.59±1.20) than in the mechanical injury (11.66±2.34) and LPSinfection group(11.59±1.47)(P<0.05), while the proportion of fibrosis area wassignificantly higher in the former group(0.65±0.03) than in the lattertwo(0.30±0.07 and 0.32±0.08)(P<0.05).The level of serum IL-21 was signifi-cantly increased in the dual injury group ([286.21±27.80]pg/mL) as compared with those in the control ( [ 118.65 ±22.55 ] pg/mL ) , mechanical injury([176.20±19.05]pg/mL), and LPS infection group ([187.98±16.51]pg/mL) (P<0.05), with a positive correlation be-tween the IL-21 level and theproportion of fibrosis area ( r=0.271, P<0.05) . Conclusion A rat model ofIUAwas successfully established by mechanical injury and lipopolysaccharide (LPS) infection. The evi-dent increase of serum IL-21 in the IUA model was positively correlated with the percentage of fibrosis area, suggesting that IL-21 may be involved indirectly in the formation of IUA.

Objective:To optimize and establish the experimental methods for the determination of 2,4-dihydroxybenzophenone (BP-1) in mouse brain. Methods:BP-1 was determined by high performance liquid chromatography (HPLC) and separated by Waters Symmetry? C18 (4. 6 mm × 250 mm, 5 μm) using isocratic elution, and the sample preparation conditions were optimized by orthogonal experiment design. The mobile phase was methanol-water (volume ratio 3∶1) containing 3% (volume fraction) ace-tic acid (pH 3. 40) at a flow rate of 1. 0 mL/min, and ultraviolet (UV) detection wavelength was set at 290 nm. Retention time was used for qualitative analysis and internal standard method for quantitative analysis. Results: Under the optimized experimental conditions, the calibration curve was linear with a correlation coefficient of 0. 999 8 over the concentration range of 0. 2-10. 0 mg/L. The recoveries of BP-1 were between 96. 8% and 104. 5%. The intra-day and inter-day precision of BP-1 were 3. 5% -5. 7% and 4. 5% -6. 4%, respectively. The extraction recoveries of BP-1 at three concentrations (0. 5, 2. 0, 8. 0 mg/L) in the mouse brain were 90. 5%, 89. 5%, and 97. 7%, and the matrix effect of BP-1 at these three concentrations were 102. 9%, 102. 7%, and 90. 9%, respectively. Conclusion:The method is simple, ac-curate, and suitable for determination of the contents of BP-1 in mouse brain.

Objective To amplify the 16S RNA fragments of 7 clinically isolated strains of Brucella spp. by PCR-RFLP technique, so as to provide experimental basis for the studies on diagnostics, genetics and epidemiology of Brucella spp. Methods According to the gene sequence of ATCC 25840 standard strain in GenBank, special primers for the 16S RNA conservative area in the Brucella spp. were designed. DNA extraction and PCR amplification of the 16S RNA fragments were performed with the 7 isolated strains. PCR products were then sequenced and RFLP analysis was conducted with appropriate restricted enzymes to study the homology and the mutation sites in those strains. Meanwhile, the clinical data of infected patients were retrospectively analyzed to evaluate the relationship between the clinical features and genotypes of Brucella infection. Results The amplified target fragments were about 1500bp in length and consistent with what was expected. The sequencing and homology analysis showed a 98.88% homology and 11 mutation sites among the 7 isolated strains. Four genotypes were identified by RFLP. Retrospective analysis of the clinical data indicated that no obvious relationship existed between the genotypes and the clinical features. Conclusions Amplifying 16S RNA fragments by PCR technique is a feasible method to make an early diagnosis of Brucella infection. The 7 clinically isolated strains are different in genotypes and 16S RNA fragment is a highly conservative fragment in bacterial genome with some mutations. The research provides evidence for the genetics and epidemiology of brucellosis.
