1.Clinical Efficacy of Xiaoer Chiqiao Qingre Granule for Child Wind-heat Common Cold Fever(Stagnant Type)

OBJECTIVE:To observe the curative effect of Xiaoer chiqiao qingre granule in treating child common cold fever (stagnant type).METHODS:A total of 118 cases with child common cold fever(stagnant type) were randomly assigned to receive Xiaoer chiqiao qingre granule (treatment group,n=60) or ribovirin tablet,Ertangganmao granule(control group,n=58) tid for 3 days.The antipyretic time, clinical effective rate, and improvement in clinical symptoms were compared between the two groups. 2.Social support and subject well-being of only-child-lost people:the mediating role of self-efficacy

Objective To investigate the relationship between social support and subjective well-be-ing in only-child-lost ( OCL ) people, and the mediating effect of self-efficacy in this relationship. Methods By stratified cluster sampling,206 only-child loss people were investigated with Social Support Rating Scale ( SSRS) ,General Self-Efficacy Scale ( GSES) and Index of Well-Being ( IWB) ,and data were analyzed with SPSS19.0 software and Amos17.0 software. Results ①Scores of SSRS,GSES and IWB in only-child-lost people were(31.78±8.18),(24.12±0.67) and (8.38±3.05). ②Correlation analysis showed that scores of GSES,SSRS and 3 dimensions of SSRS were positively correlated with scores of IWB ( r=0. 542,0.332,0.214,0.332,0.216;P<0.05) . ③Self-efficacy partially mediates the relationship between social support and subjective well-being through mediating effect test(χ2/ df=1.986,GFI=0.985,AGFI=0.942, RMSEA=0.069),mediating effect accounted for 16.08%.Conclusion Social support is an important factor which plays a direct or indirect role in subject well-being. Self-efficacy is the intermediate link between social support and subject well-being.
3.Sensitivity of human gastric cancer cells to TRAIL is not associated with caspase-8 promoter methylation status

Objective To study the effect of methylation state of C5 of the cytosine in the CpG di nucleotide of caspase 8 promoter on tumor necrosis factor related apoptosis inducing ligand (TRAIL) antitumor activity in gastric carcinomas. Methods The methylation states of the caspase 8 promoter region of 5 kinds of gastric carcinoma strains were measured by methylation specific PCR method. The antitumor activity of TRAIL protein was measured by MTT method. Results No methylation of caspase 8 promoter was found in gastric carcinoma cells. Treatment with demethylation agent 5 Aza 2′ deoxycytidine (5 Aza CdR) increased sensitivity of gastric cancer cells to TRAIL, but did not change methylation status of caspase 8 promoter in gastric carcinoma cells. Conclusion Caspase 8 promoter methylation status is not associated with TRAIL antitumor activity.
4.TLC Identification of Pomegranate Seed and Terminalia Chebula, HPLC Determination of Cinnamal and Piperine in Tibetan Medicine Shi-Wei Xiao-Shi-San

This study was aimed to establish a method for quality control of Tibetan medicine Shi-Wei Xiao-Shi-San (SWXSS). Thin layer chromatography (TLC) was established for the qualitative determination. Cinnamal and piperine were determined by HPLC. The separation was performed on Phenomenex C18 column (4.6 mm × 250 mm, 5μm). The results showed that TLC can be used in the identification of pomegranate seed and Terminalia chebula. The linear ranges of cinnamal and piperine were within the range of 0.06-0.37μg and 0.05-0.33μg, respectively. And the standard line was Y=9.273 2×106X-2.348 2×105, r=0.999 8;Y=7.315×103X-3.857, r=1.000 0. It was concluded that the identification method was specific, accurate and practical, which can be applied in the quality control of SWXSS.
6.Enhanced TRAIL antitumor activity on human gastric cancer cells induced by 5-Aza-CdR is involved in up-regulation of caspase-8

Objective To study the effects of demethylation agent 5-Aza-2′-deoxycytidine (5-Aza-CdR) on the antitumor activity of tumor necrosis factor (TNF)-related apoptosis-inducing ligand (TRAIL) against gastric carcinoma. Methods Expression of caspase-8 mRNA was examined by RT-PCR. Antitumor activity of TRAIL protein was measured by MTT method. Results The inhibition rates of treatment with 200 ng/ml TRAIL for 72 h on gastric cell lines SGc 7901, Kato 3, and AGS were 9.83%, 11.94%, and 4.04%. After treatment with 5-Aza-CdR, the inhibition rates of 200 ng/ml TRAIL on gastric cell lines increased to 38.98%, 52.42%, and 30.72%. Before exposure to 5-Aza-CdR, expression of caspase-8 mRNA was low and an increased expression of the caspase-8 was found in the three gastric cancer cells after treatment with 5-Aza-CdR. Conclusion Treatment with 5-Aza-CdR can increase TRAIL antitumor activity on human gastric cancer and its mechanisms might be involved in the up-regulation of caspase-8 gene.
7.Role of mitochondrial pathway in apoptosis of gastric cancer cell line SGC-7901 induced by Helicobacter pylori

Objective To evaluate the role of mitochondrial pathway in apoptosis of gastric cancer cell line SGC-7901 induced by Helicobacter pylori(H.pylori).MethodsApoptosis was evaluated in SGC-7901 cells by flow cytometry.RT-PCR and Western blotting were used to measure the expressions of genes and proteins associated with mitochondrial pathway.Effect of caspase inhibitors on apoptosis induced by H.pylori strain NCTC11637 was investigated.ResultsNCTC11637 directly induced apoptosis in SGC-7901 cells.Apoptotic rate was 6.30%,11.57%,8.63% and 7.22% respectively at 6,12,24 and 48 h after coculture with H.pylori.H.pylori upregulated the expression of Bax,and induced a time-dependent activation of caspase-9 and-3.Apoptosis was inhibited significantly by pre-incubation with the inhibitors of caspase-9 and-3.Caspase-8 inhibitor reduced H.pylori-induced apoptosis by 20%.ConclusionH.pylori infection upregulates the expressions of Bax mRNA and protein in gastric cancer cell line SGC-7901,and induces activation of caspase-9 and-3.Mitochondrial pathway may be the major one in H.pylori-induced apoptosis in SGC-7901.
8.Mediation role of self-efficacy between social support and depression of only-child-lost people

Objective:To investigate the relationship between social support and depression of only-child-lost (OCL) people,and the mediation role of self-efficacy in this relationship.Methods:By stratified cluster sampling,214 OCL people were enrolled,with 80 males and 134 females,ages from 49 to 83 years old.They were assessed by General Self-Efficacy Scale,Social Support Rating Scale,and Self-rating Depression Scale.Results:Univariate analysis showed that there were significant differences in age groups (t=2.85,P<0.05),with or without spouse (t=5.62,P<0.05),family location (t=3.95,P<0.05),per capita monthly income (F=3.48,P<0.05) among the social support scores.There was significant difference between the per capita monthly income and self-efficacy scores in QCL people (F=5.46,P<0.05).Correlation analysis showed self-efficacy and social support were positively correlated (r=0.26,P<0.01).Self-efficacy (r=-0.59,P<0.01) and social support (r=-0.59,P<0.01) negatively correlated with depression in OCL people.Self-efficacy partially mediated the relationship between social support and depression.Conclusion:The person who is <60 years old,with spouse and the high per capita monthly income,and lives the rural area,would have high social support levels among QCL people.The person who has high per capita monthly income would have high self-efficacy.Self-efficacy is one of the direct prediction for depression,and plays an indirect role between social support and depression.Intervention of depression among OCL people could be applied to change their cognition,and to enhance their self-efficacy.

Abstract: In order to investigate immune protection against swine-origin influenza virus (S-OIV) A H1N1, the helper-dependent adenovirus vector (HDAd) system was exploited to construct recombinant HDAd encoding hemagglutinin (HA). The HA gene was synthesized and cloned to the HDAd backbone. Then, the HDAd/HA DNA molecules were transfected into 293Cre4 cells with calcium phosphate. The cells were infected by helper virus 16 hours after the transfection. The 293Cre4 cells were coinfected with HDAd/HA and the helper virus for large-scale preparation of HDAd/HA. The HDAd/HA was obtained and purified twice with CsCI density ultracentrifugation and observed morphologically under transmission electron microscope, and the expression of HA protein was analyzed with RTPCR. Recombinant HDAd/HA expressing HA protein was successfully constructed which could pave the way for in vivo investigation on immunogenicity and efficacy against S-OIV A H1N1 infection.

10.Reasons for revision in 33 patients after total hip replacement

BACKGROUND:The number of complications after hip replacement, such as infection, implant loosening, fracture prosthesis wear, osteolysis, and recurrent dislocation, had drastical y increased. These complications would induce the increased occurrence of total hip revision.
OBJECTIVE:To analyze the causes and treatment measures of revision after total hip replacement. METHODS:The reasons for revision, the prosthesis selection, the treatment of bone defect and the postoperative rehabilitation were discussed in 33 cases after total hip replacement. The prosthesis for revision included general metal ring and lining (21 cases), large head and cup (8 cases), polyethylene cup (4 cases), general femoral components (15 cases, including 11 cases fixed by bone cement), and lengthening femoral components (18 cases, including 9 cases fixed by bone cement and 6 cases of combined components).
RESULTS AND CONCLUSION:Al 33 patients were fol owed up for 24-60 months, averagely 36.5 months. After revision, wound healed perfect, and the prosthesis was reliable. No case suffered from infection or dislocation. Hip joint function was greatly improved. Harris score was increased from 37.1 preoperatively to 91.3 postoperatively. Medium-or short-period clinical fol ow-up results demonstrated that if the indication of revision was right, bone defects were handled perfectly, prosthesis was chosen correctly, one-stage total hip revision can get a good clinical efficacy.
