1.Surveillance of suspected adverse events following immunization in Wuyi County from 2018 to 2021

Objective:
To investigate the occurrence of suspected adverse events following immunization (AEFI) in Wuyi County, Zheijang Province from 2018 to 2021, so as to provide insights into safety assessment of vaccination. Methods The AEFI data in Wuyi County from 2018 to 2021 were collected from the AEFI Information Management System and Immunization Information Management System of National Immunization Information Management System in China. The reported incidence of AEFI and epidemiological characteristics of cases with AEFI were analyzed in Wuyi County from 2018 to 2021 using a descriptive epidemiological method.
Methods:
The AEFI data in Wuyi County from 2018 to 2021 were collected from the AEFI Information Management System and Immunization Information Management System of National Immunization Information Management System in China. The reported incidence of AEFI and epidemiological characteristics of cases with AEFI were analyzed in Wuyi County from 2018 to 2021 using a descriptive epidemiological method.
Results:
A total of 870 007 vaccine doses had been administered in Wuyi County from 2018 to 2021, and a total of 482 AEFI were reported, with annual mean reported incidence of 55.40/105. The incidence of AEFI associated with vaccines included in the national immunization program was significantly higher than those not included in the national immunization program (67.75/105 vs. 44.00/105; χ2=20.334, P<0.001). There were 435 individuals with general reactions (50.00/105 incidence), with high fever, local redness and swelling and solid nodes; 39 cases with abnormal reactions (4.48/105 incidence), including 87.18% allergic reactions; 7 cases with coincident syndrome and one case with psychogenic reaction; however, no vaccine quality or vaccination events were reported. The five vaccines with the greatest incidence of AEFI included 13-valent pneumococcal polysaccharide conjugate vaccine (269.11/105), diphtheria and tetanus toxoids and acellular pertussis, inactivated poliovirus and Haemophilus b conjugate vaccine (204.23/105), 23-valent pneumococcal polysaccharide vaccine (175.85/105), tetanus, diphtheria, acellular pertussis vaccine (128.39/105) and combined live vaccine for measles, mumps and rubella (124.36/105). The reported incidence rates of AEFI were 10.89/105, 49.88/105, 56.07/105 and 57.78/105 following oral, intradermal, subcutaneous and intramuscular administrations, respectively. In addition, AEFI were predominantly found to occur in patients at ages of 0 to <2 years (333 cases, 69.09%), and AEFI predominantly occurred <1 day following immunization (336 cases, 69.71%).
Conclusions
General reactions were predominant types of AEFI following vaccination in Wuyi County from 2018 to 2021, and the reported incidence of AEFI associated with vaccines included in the national immunization program was significantly higher than those not included in the national immunization program. AEFI mainly occurred among children under 2 years of age.
2.The effect of Fe3O4 nanometer magnetic fluid induced hyperthermia on implanted liver cancer in nude mice

Objective To study the therapeutic effect of Fe3O4 nanometer magnetic fluid-induced hyperthermia on implanted liver cancer in nude mice under alternating magnetic field. Methods Nude mice model bearing implanted HepG2 was established. Mice were then randomly divided into 3 groups: the blank control group; the magnetic field group; nanometer magnetic fluid group. The magnetic field group were just put under the magnetic field; Nanometer magnetic fluid group received injection of PEG-PEI/Fe3O4 nanometer magnetic fluid under the alternating magnetic field. At the frequency of 40 kHz, and magnetic field of 5 kA/m, 15 minutes one day in the next 14 days. On the 7th day and the 15th day, the changes of tumor volume and weight were recorded, cell apoptosis were observed and recorded and pathological examination was done. Results On the 7th and the 15th day, in the nanometer magnetic fluid group, tumors' volume was smaller and the weight was lighter than other groups, and the tumor inhibitory rate of 54. 20% (t = 14. 506,P <0. 01 ) was significantly higher than the control group and the magnetic field group 22. 66% ( t = 7.497, P < 0. 05 ). In the control group, tumor cells grew well, high density, the nucleus engrained, the shape irregular, the nuclear fission clear; compared with the control group, in the magnetic field group, tumor cells scatter thinly, intercellular substance increases, and necrosis area formed;in the nanometer magnetic fluid group, many of tumor cells died, their cell nucleus broke up and vanished,the blood vessel reduced obviously, and the tumor cell spread thinly. Conclusions Under the alternating magnetic field, PEG-PEI/Fe3O4 nanometer magnetic fluid inhibits liver cancer growth in nude mice model of HepG2.
3.Application of the Cerebral Palsy Rehabilitation System in the Digital Community on the Thumb Adduction Deformity

Objective:To explore the effect of digital rehabilitation system on the recovery of infants with cerebral palsy.Methods:Twenty-one children with cerebral palsy were treated with residual cerebral palsy in Putuo District,and 21 children with cerebral palsy were followed up.The patients were divided into two groups (n =21).The control group was treated by routine OT training by the parents,and the treatment group was treated with the digital rehabilitation system.Three months later,the efficacy was evaluated and compared.Results:After 3 months of treatment,the total effective rate (effective rate and effective rate) of the two groups was 90.5% and 81%,respectively,and the treatment group was significantly higher than the control group (P <0.05).The PROM of the two groups was improved (P <0.01),and the PROM in the treatment group was significantly higher than that in the control group (P <0.05).FMFM was significantly higher than that before treatment (P <0.01),and FMFM was significantly higher in the treatment group than in the control group (P <0.01).Conclusion:Family (community) digital rehabilitation system can effectively improve the rehabilitation of children with cerebral palsy.
4. Effect of nano-SiO_2 on the survival and PARP-1 expression in 16HBE cells

OBJECTIVE: To explore the effects of nano-silicon dioxide( SiO_2) on the survival and poly( ADP-ribose)polymerase-1( PARP-1) expression in human bronchial epithelial cells( 16 HBE cells). METHODS: i) The 16 HBE cells were treated with nano-SiO_2 at concentrations ranging from 0 to 100 mg/L for 24. 0 hours,and CCK-8 assay was used to examine cell viability. ii) The 16 HBE cells were divided into 6 groups: solvent control group( equal volume solvent treatment),micro-SiO_2 control group( treated with 20 mg/L micro-SiO_2),5,10,and 20 mg/L nano-SiO_2 groups( treated with the corresponding final dose of nano-SiO_2),and curcumin group. The curcumin group was given pretreatment with curcumin at a final concentration of 10 μmol/L for 2. 0 hours followed by treatment with a final concentration of 20 mg/L of nano-SiO_2. Cells in each group were harvested at time points of 4. 0,12. 0 and 24. 0 hours after treatment. The relative expression of PARP-1 mRNA and protein in 16 HBE cells was detected by quantitative real-time polymerase chain reaction and Western blotting respectively. RESULTS: i) The survival of 16 HBE cells decreased with increasing nano-SiO_2 treatment dose,showing a dose-effect relationship( P < 0. 01). ii) The expression of PARP-1 mRNA and protein in 16 HBE cells were dose-dependently decreased after nano-SiO_2 stimulation at the 12. 0 and 24. 0 hours time points( P < 0. 01). The expression of PARP-1 mRNA and protein in 5,10,and 20 mg/L nano-SiO_2 groups decreased at the above mentioned time points( P < 0. 05),compared with the solvent control group at the same time points. The expression of PARP-1 mRNA and protein in 20 mg/L nano-SiO_2 group was lower than that in the micro-SiO_2 control group at the same 12. 0 and 24. 0 hours time point( P < 0. 05). The above two indexes of cells were higher in curcumin group than that of 20 mg/L nano-SiO_2 group at the 12. 0 hours time point( P < 0. 05). CONCLUSION: Nano-SiO_2 stimulation can lead to decrease survival of 16 HBE cells in a dose-dependent manner and down-regulation of PARP-1 expression may be one of the mechanisms of proliferation and inhibition of 16 HBE cells induced by nano-SiO_2. Curcumin has certain protective effect on nano-SiO_2-induced 16 HBE cell injury.
