1.An Intracerebral Hemorrhage Model Established by Autologous Blood Injection Three Times in Rat

ObjectiveTo establish an animal model of intracerebral hemorrhage (ICH) in rat and to observe the changes in behavior, brain edema and tissue structure during absorption of the hematoma.MethodsAn animal model of ICH was established by stereotactically infusing 100 μl (the first time 20 μl, the second time 40 μl, the third time 40 μl) of autologous caudal artery blood into caudate nucleus in the rats. The behavior of rats with ICH was assessed by Bederson score, beam-walking test, bilateral forepaws grasp and measurement of forelimb placing. The brain edema and the change in histology were observed.ResultsThe successful ratio of this model was 75%. There were significant behavioral changes and brain edema at 6 h after the ICH in rats of the experimental group compared to those of the control group. The worse behavioral changes and brain edema was at 48~72 h after hemorrhage. In the earlier period, the nerve cells were swelling. A lot of glial cells appeared around the hematoma in the late period.ConclusionThis ICH model is reproducible, and the obvious behavioral change and histological changes are similar to clinical patient with ICH.
2.The effects of tetrandrine on activity of collagenase derived from human hypertrophic scar.

OBJECTIVETo observe the effect of tetrandrine on activity of collagenase derived from human hypertrophic scar for the sake of clarifying the mechanism as tetrandrine acting on scar.
METHODSThe experimental concentration was controlled below that of cell proliferation inhibited, SDS-PAGE electrophoresis was adopted to separate collagenase from extracellular matrix, and then activated by trypsin analyzed the activity of collagenase with density scanning apparatus. At the same time quantity of extracellular collagen was measured using improved chloraseptine T oxidizing assay, moreover analyzed correlation between activity of collagenase and quantity of extracellular collagen.
RESULTSIn the concentration below the lever of inhibiting fibroblast proliferation, the total activity of collagenase could be significantly increased by tetrandrine with dosage-dependence associated with quantity of extracellular collagen reduced, which was much greater than that of triamcinolone.
CONCLUSIONIncreasing activity of collagenase on degradation of collagen even in a lower concentration was one of the mechanisms of tetrandrine treating hypertrophic scar.

3. Effect of Lycium barbarum soup on histological structure, inducible nitric oxide synthase activity, Caspase-3 and nuclear factor-κB protein expression in retina of mice injured by heroin

Objective To explore the effects of Lycium barbarum soup on retina histological structure and related active substances in mice injured by heroin. Method Totally 120 Kunming mice were randomly divided into control group, heroin group, Lycium barbarum 1 group and Lycium barbarum 2 group. The mice were intraperitoneal^ injected escalation with doses (2, 4, 6 g/L) heroin 0.2 ml continuous (1-5 N 6-10 and 11-15 days) for 15 days (twice a day). One hour after injected heroin, Lycium barbarum 1 and 2 group was intragastric administration with (1.0 g/kg, 2.0 g/kg) Lycium barbarum soup for 0.2 ml. The changes of retina tissue structure were observed by biomicroscopy, the changes of plasma lactate dehydrogenase (LDH) activity and retina inducible nitric oxide synthase (iNOS) activity were detected by colorimetry, the changes of Caspase-3 and nuclear factor (NF)-kappa;B protein expression in retina was detected by immunohistochemistry method. Result Compared with the control group, the retina tissue of the heroin group was damaged in varying degrees, ganglion cells decreased, vacuolated degeneration occurred, the structure of the inner and outer nuclear layers was disordered, loose and reticular. The LDH activity of plasma increased significantly(P<0.05 or P<0.01). The iNOS activity of retina increased significantly (P<0.01). The Caspase-3 and NF-kappa;B protein expression of retina increased, and the average optical density increased significantly (P<0.05 or P<0.01). Compared with the heroin group, the ganglion cell layer vacuolation of the retina in Lycium barbarum 1 and 2 groups was reduced, the inner and outer nuclear layers were arranged orderly. The LDH activity of plasma decreased significantly (P<0.05 or P<0.01). The iNOS activity of retina decreased significantly (P<0.05 or P<0.01). The Caspase-3 and NF-kappa;B protein expression of retina decreased significantly (P<0.01). Conclusion Lycium barbarum soup can improve the anti-oxidation and anti-inflammatory ability of cells, inhibit Caspase-3 expression and NF-kappa;B activation of retina. It can effectively reduce the damage of heroin on the retina and has a certain protective effect on the retina.
