1.Urocortin-postconditioning protects heart from ischemia-reperfusion injury through interfering mitochondria-dependent apoptotic pathway

Objective:To investigate the effects of Urocortin-postconditioning on mitochondria-dependentapoptotic pathway.Methods: 24 Wistar rats were randomly divided into 3 groups(n=8):Control group(CON),I/R group(I/R),Urocortin group(UCN).The left ventricular samples in the Control group were collected as pre-ischemia controls through thoracotomy.After isolated heart Langendoff and Neely models were established,the rat hearts in the I/R group and the UCN group were continuously perfused with Krebs-Henseleit (K-H)buffersolution for 30 min,then the rat hearts were arrested by cardioplegia,St.Thomas-2 solution(STH-2)for 30min.After that,the rat hearts in the I/R group were reperfused with K-H buffer solution for 90 min.The rat hearts in the UCN group were reperfused with K-H buffer solution containing 10-8 mol/L Urocortin for 90 min.The releasing of Cyto C protein into cytosol was detected by Western blot.The expression of Caspase-3 protein in cardiomyocyte was detected by immunohistochemical assay.The apoptosis index (AI)of cardiomyocyte was detected by TUNEL.Results:Compared with Control group,the expression ofCaspase-3 and Cyto Cprotein in the I/R group and the UCN group were significantly increased(P
2.Mechanisms of childhood hemophagocytic lymphohistiocytosis:A bioinformatic analysis

Objective Hemophagocytic lymphohistiocytosis (HLH) is a life-threatening condition characterized by excessive inflammation, with a high incidence in children and a death rate of 40%.This study was to analyze the gene expression profile in child-hood HLH and explore the important pathways of childhood HLH using bioinformatic methods . Methods The childhood HLH gene ex-pression profile data GSE26050 were obtained from the Gene Expression Omnibus (GEO) database of the National Center for Biotechnolo-gy Information.Differentially expressed genes were identified with the GEO 2R online analysis tools released recently .The key pathways of the differentially expressed genes were investigated using the Kyoto Encyclopedia of Genes and Genomes ( KEGG) pathway enrichment a-nalysis. Results A total of 184 differentially expressed genes were identified , 126 upregulated and the other 58 downregulated .They were enriched in 3 pathways, including cytokine-cytokine receptor interaction , hematopoietic cell lineage and NOD-like receptor signaling pathways. Conclusion Bioinformatic tools allow the identification of the key genes and pathways associated with the development and progression of childhood HLH and point out the potential directions for researches on the mechanisms of childhood HLH .
3.Heat shock protein 27 and its role in retinal ganglion cell apoptosis in rat high intraocular pressure model

Background Glaucoma is common blinding eye diseases characterized by chronic loss of retinal ganglion cells(RGCs).Currently glaucoma pathogenesis is not completely understood,heat shock protein 27 ( HSP27 )may be associated with the pathogenesis of glaucomatous optic neuropathy. Objective Through the establishment of a rat model of high intraocular pressure,detection of the expression of HSP27 antibody in serum and RGCs apoptosis to investigate the role of HSP27 in RGCs apoptosis. Methods Fifty-one clean Wistar rats were divided into high intraocular pressure group (34 rats)and sham operation group( 17 rats)using a random number table.An animal model of high intraocular pressure was established in the right eye by the application of bipolar underwater electrocoagulation on vein of sclera surface in the experimental group,and rats with conjunctiva incision only without electric coagulation were served as sham operation (control).The intraocular pressure of rats of the both groups including experimental and control rats was measured 1,2,4,6,8 weeks after operation and then the rats were sacrificed.1 ml serum was collected from these rats to determine the concentration of HSP27 antibody.The retinas of the rats were isolated and homogenated for the extraction and analysis of the retinal protein by Western blot.Apoptosis of RGCs were assayed by TUNEL.The use of the experimental animals followed the Regulations for the Administration of Affair Concerning Experimental Animals by Kunming Medical Collegc. Results Intraocular pressure was elevated significantly after modeling and remained a high value during the expcrimental duration,showing a significant difference among the different groups ( F =318.502,P<0.01 ).However,no significant change in intraocular pressure before and after operation in the sham-operation group ( F =2.076,P > 0.05 ).The serum concentration of HSP27 antibody was increased significantly overtime from 1,2,4,6 to 8 weeks after the surgery applied,with a significant difference among various time points( F =154.221,P<0.01 ),but no obvious difference was seen in the shamoperation group( F =0.422,P>0.05 ).Importantly,Apoptosis rate of RGCs in the experimental eyes was significant higher as the time prolong after intraocular elevated(x2=856.12,P<0.05 ).The positive rate of RGCs apoptosis in the rats with high intraocular pressure group was elevated significantly compared to the rats in the sham operation group(P<0.05). Conclusions High intraocular pressure induces the expressions of HSP27 in retina and the accumulation of HSP27 antibody in rats serum;the elevated HSP27 is associated with the increased cell death in RGCs.
4.THE INFLUENCE OF POLYTETRAFLUOROETHYLENE MEMBRANE ON CELL GROWTH OF PERIODONTAL MEMBRANE AND EXPERIMENTAL REGENERATION OF PERIODONTAL TISSUE

The effects of polytetrafluoroethylene (e PTFE) membrane on periodontal ligament cell (PDLC) growth and periodontal tissues and new cement regeneration were studied, and the prospects of clinical application of e PTFE membrane were discussed. The results showed that PDLC grew well on the e PTFE membrane and no toxicity or inhibition was found. Close adhesion of the cells to the membrane was observed by scanning electron microscope. Histological studies suggested that the peculiar porous structure of e PTFE membrane protects and promotes healing of alveolar bone defect when the membrane edge was sealed perfectly on the crown side,exhibiting inhibition of growth of epithelium towards tooth root. The membrane was beneficial to regeneration of new alveolar bone and cement and establishment of new adhesion,and was regarded as a good substitute of old materials.
5.Determination of Eugenol in Xiao’er Fuxie Waifu Gel by RP-HPLC

Objective To establish a method for the determination of eugenol in Xiao’er Fuxie Waifu Gel.Methods RP-HPLC method was developed with Lichrospher-C18 analysis column,using methyl alcohol-water(65:35) as mobile phase.The detection wavelength was 280 nm and flow rate was 1.0 mL/min.Results The linear range of eugenol was from 0.108 to 1.724 ?g(r= 0.999 9).The average recovery was 99.1 %and RSD was 1.4 %.Conclusion The method is convenient and accurate.It can be used for the quality control of Xiao’er Fuxie Waifu Gel.
6.Achillis Tendon-Sliding Procedure and Subtalar Joint Capsula Release by the Same Cut and Individualization in Treatment of Congenital Clubfoot

Objective To explore achillis tendon-sliding procedure and subtalar joint capsula release by the same cut and individualization treatment used in the correction of congenital clubfoot.Methods Forty eight cases (76 feet) of congenital clubfoot who were first visit were treated by achillis tendon-sliding procedure and subtalar joint capsula release by the same cut,meantime,footplate fascia release and transfer of the anterior tibial tenden were made by individualization.Results All cases were followed-up,the average time follow-up was 1 year and 6 months,excellent were 71% and good were 15.7%.Five cases were recurrence and its rate was 6.94%.Conclusions Achillis tendon-sliding procedure and subtalar joint capsula release by the same cut can solve rotation of displacement between calcaneus and talus and individualization treatment can also solve high arch deformity and adduction of foot.This technique is a simple,safe and effective treatment for congenital clubfoot.
7.Finite element analysis of kyphosis in ankylosing spondylitis treated by different osteotomy methods

BACKGROUND: Kyphosis in ankylosing spondylitis is sagittal or coronal imbalance, but there is a lack of study on its orthopedic biomechanics, and biomechanics is of great significance for the reconstruction of spinal stability after orthopedic surgery.OBJECTIVE: To establish a three-dimensional (3D) finite element model of kyphosis in ankylosing spondylitis treated by osteotomy on software, and to analyze its biomechanical properties, thus providing theoretical basis for clinical practice.METHODS: A 3D finite element model of kyphosis in ankylosing spondylitis was established based on CT data, and the predetermined angle of the osteotomy at L2 was measured. Afterwards, vertebral column decancellation and vertebral column resection were stimulated, and then the biomechanical parameters were analyzed. RESULTS AND CONCLUSION: (1) The 3D finite element models of kyphosis in ankylosing spondylitis treated by vertebral column decancellation or vertebral column resection at L3 were established successfully. (2) The stress on the screw and contact at each segment in the vertebral column decancellation group was significantly higher than that in the vertebral column resection group except for S1. (3) To conclude, both two methods can reconstruct the sagittal balance,but vertebral column decancellation exhibits significantly higher stress on the screw. Indeed, the incidence of internal fixation failure and complications in vertebral column decancellation is higher than that in vertebral column resection at the same segment and angle.

The study was o investigate the effectiveness of problem-based learning and scenario simulated teaching in Pathological experiment course.Clinical common,multiple cardiovascular diseases related experiments were selected for reform.The case was in advance handed out to the second year students of clinical medicine,stomatology,imaging medicine and otherspecialty,requiring them to complete the data access,group discussion,PPT production,diagnoses and treatment scenarios simulation video shooting tasks.Every group showed their PPT and video in class.They were asked between groups.Summative evaluation was given before class.Curriculum effectiveness questionnaire was issued and recycled,and according to students' performance teacher gave their scores and reward.The study showed that the comprehensive application of the above teaching methods and means could effectively improve students' learning enthusiasm and initiative,cultivate their medical humanistic spirit,and help to improve the teaching quality of pathology experiment.
9.CT characteristics of solitary fibrous tumor of the mediastinum and analysis of 5 misdiagnosed cases

Objective To investigate CT features of solitary fibrous tumor of the mediastinum(SFTM),and to analyze the causes of misdiagnosis of this disease.Methods CT features of 5 patients of SFTM confirmed operatively and pathologically were retrospec-tively analyzed.All patients underwent before operation but were misdiagnosed.Results 3 cases were located in right mediastinum with oval-shap,well-defined margin,and “pleural tail sign”,2 cases were located in right thoracis cavity with unwell-defined margin, and in the adjacent lung were oppressed.In all 5 patients,the mean median of tumors was 1 1.5 cm(8.6-1 5 cm),non-homogeneous density of tumors were seen on plain CT scans,and obvious non-homogeneous enhancement with“geographic pattern”on contrast-en-hanced CT.Conclusion CT can clearly reveal the size and the appearance of SFTM,adjacent architectures,the final diagnosis de-pends on histopathological and immunohistochemical findings of the tumors.To un-know is the main caused of misdiagnosis of medi-astinal SFT.
10.Study on the time of amounted to peak of human adipose-derived stromal cells differentiation into neural precursor cells in vitro

Objective To reseach the time point of the highest percentage of neural precursor cells derived from adipose stromal cells (ADSCs) in vitro, and to observe the ultrastructure features of neural precursor cells. Methods Used the β-mercaptoethanol to induce ADSCs to differentiate into neural precursor cells and neuron-like cells. The morphology of the uninductedcells and inducted cells were observed with inverted phase contrast microscope. The expression of nestin which was the marker of neural precursor cell in each group was detected using immunofluorescence staining method. The ultrastructural feature of cells which was induced for 3 hours were observed. Results The highest ratio of positive expression of nestin was 3 hours following induction,with the ratio ( 86.25 ± 4.82) %. There were many protuberance on the cell membrane under transmission electron microscopy.There were plenty of organelles in the neural precursor cells. The neural precursor cells had a large size nucleus,large nucleoplasmic index, much extended chromatin,and less condensed chromatin. The nucleus had double-layer nuclear envelope, more nuclear pore on the nuclear envelope. Conclusion The time point of the highest percentage of neural precursor cells derived from ADSCs is 3 hours,and the ultrastructral feature of induced neural precursor cells confirm that cells at this time point are in a state of split active period.
