1.Comparison and selective preference of preparation methods of platelet-rich gel

BACKGROUND:Platelet-rich plasma gel contains some kinds of growth factors, which play important roles in repair of local trauma and bone defects. Platelet-rich plasma gel is now more and more studied as tissue engineering scaffolds, but there are no unified preparation methods.
OBJECTIVE:To summarize the studies about the preparation methods of platelet-rich plasma gel and the growth factors in platelet-rich plasma gel in recent years, and to compare different methods in order to find which one is better.
METHODS:A online search of PubMed database from January 1980 to January 2013 and China National Knowledge Infrastructure from January 1980 to January 2013 was performed with the key words of“platelet-rich plasma, platelet-rich plasma gel, tissue engineering”in English and Chinese, respectively. The studies about detection and preparation methods of platelet-rich plasma were included and those unrelated to platelet-rich plasma were excluded. Final y, 322 articles were gained and 67 articles were included in result analysis.
RESULTS AND CONCLUSION:Al preparation methods have their advantages. We should select the suitable method according to the goal of study. With the further study of platelet-rich plasma, it wil become more and more important for bone defect repair and tissue engineering research.
2.High resolution melting and its application in identity study of traditional Chinese medicine.

High resolution melting (HRM), based on melting curve analysis, requires not only saturating dyes that fluoresce in the presence of double-stranded DNA, but also higher resolution detection equipment. The melting curve is a novel method for sequence matching, genotyping and mutation scanning. The technology is simple, accurate, rapid, closed-tube, low-cost, and high-throughput, which make it gain more and more applications. This review article presents the basic principles, key factors and both the advantage and limitations of HRM. The potential application is discussed in the study of molecular identity of traditional Chinese medicine.

4.Study on chemical constituents in stems of Nelumbo nucifera by UPLC-ESI/Q-TOF-MS/MS.

This paper employed UPLC-Electrospray Ionization /Quadrupole-Time of Flight-Mass /Mass Spectrometry( UPLC-ESI/Q-TOF-MS/MS) to analyze the chemical constituents in the stems of Nelumbo nucifera. The stems of N. nucifera were extracted with 75% methanol, and we applied an Agilent Zorbax SB-Aq column (2.1 mm x 100 mm, 1.8 μm) to UPLC analysis with water methanol-water( containing 0.05% formic acid) in gradient as mobile phase. The eluates were then detected by ESI-Q-TOF-MS/MS. Results indicated that 22 benzylisoquinoline alkaloids were indendified. Among them, one alkaloid may be a new compound and a component was found in the Lotus for the first time. We fully identify the composition of the Lotus stems for the first time, Which could provides theoretical foundation for further study and utilization of the medicinal resources.

5.Clinical analysis of nine cases of paroxysmal exercise-induced dystonia.

This study was aimed to analyze the clinical features of paroxysmal kinesigenic dyskinesia (PKD) and extend the understanding of this disease. From August, 2008 to October, 2010, 9 patients were diagnosed with PKD in the Department of Neurology of the First Affiliated Hospital of Zhejiang University, China. The data involving clinical demographic characteristics, somatosensory evoked potentials, results of electromyography, video electroencephalography (EEG), brain magnetic resonance imaging (MRI) and computerized tomography (CT) were collected. All PKD patients exhibited unilateral or bilateral recurrent episodic dyskinetic attacks triggered by sudden voluntary movements. The duration of the attacks ranged from several seconds to one minute. The attack frequency ranged from approximately once in several months to more than 10 times in a day. Patients suffered from no conscious disorders during the attack, and no neurological signs were found during the period between attacks. No abnormal somatosensory evoked potentials were found. Routine EEG, video EEG monitoring or brain imaging showed normal findings. Classical treatment for anti-epilepsy, including carbamazepine and topiramate, was administered to the patients and proved to be effective. It was concluded that PKD is characteristically triggered by sudden voluntary movement; no abnormal electroneurophysiological findings are observed in PKD, and antiepileptic drugs are effective in treating the disorder.
6.Quantitative evaluation of myocardial microvascular endothe lial dilatation by time-intensity curve with myocardial contrast echocardiography in dogs

Objective To investigate the changes of mi crovascular endothelial dilatation in stunned myocardium. Methods Thirteen healthy and adult mongrel dogs were randomly organized into two groups, group A (n=6) and group B (n=7). Each dog in group A and B underwent 15 min and 30 min acute ligation at left anterior descending coronary artery (LAD), respectively. Myocardial contrast echocardiography was implemented via intra-femoral vein bolus injection of C 3F 8-exposed sonicated dextrose albumin at different examining time.Peak-intensity (PI) and area under curve (AUC) were derived from time-intensity curve (TIC) after aortic root injection of acetylcholine (ACH) or nitroglycerin (NG),and then PI ratio (PIR) and AUC ratio (AUCR) of pre- to post-ACH or pre- to post- NG were calculated at each of the corresponding examining times. Myocardial endothelium-dependent relaxation (EDR) and non-EDR were analyzed at different examining times. Results ①At baseline (before LAD ligation), PI and AUC in group A and B increased considerably via aortic root injection of ACH and NG. ② After ACH injection,PIR in group A decreased sharply at 5,30 and 60 min, whose P values were less than 0.05 compared with baseline. And all these recovered completely at 120 min reperfusion. However,PIR in group B recovered slowly and reached finally to baseline at 150 min reperfusion. Meanwhile, the variation trends of AUCR in both group A and B were the same as those of PIR. Overall, the dynamic tendencies of PIR and AUCR in these two groups for post-NG at different examining times were much similar to those of post-ACH. ③ The decreased magnitudes of PIR and AUCR in group A and B after ACH injection were bigger than those of NG injection at 5,30 and 60 min reperfusion(P

8.Color Doppler Flow Imaging in Assessment of the Resectability for Pancreato-ampullar Diseases

The reseclability of 45 cases with pancreato-ampullar diseases was evaluated preoperatively with color Doppler flow imaging(CDFI).In 43,the situation was confirmed by surgery and pathology with obvious involvement of the peripancreatic vessels and/or liver metastases as the criteria for nonresectability. 22 patients considered to be not resectable on the basis of CDFl were proved at surgerY.While in 21 patients without sign of obvious involvement of peripancreatic vessels on CDFI,the resectability rate was 85.7%.The overall correlation rate of CDFl was 93%.It is concluded that CDFI plays a definite role in evaluating the resectability of panereato-ampullar disease.The positive signs of vessel involvement are discussed.
9.Laparoscopic Total Mesorectal Excision Combined with Pull-through Anastomosis for Rectal Cancer:Report of 8 Cases

Objective To evaluate the advantage of laparoscopic total mesorectal excision (TME) and the clinical value of pull-through anastomosis in the treatment of rectal carcinoma. Methods From November 2005 to December 2006, laparoscopic TME combined with pull-through anastomosis was carried out on 8 patients with rectal cancer. Results In all of the 8 patients, TME was completed under a laparoscope without additional incision, and the pull-through anastomosis was performed by hand.The mean operation time was 220 minutes (range, 180 to 300). The patients were followed up for 8-18 months (mean, 13 months), during which none of them developed implantation at puncture sites or local recurrence. Conclusions Laparoscopy provides a direct, magnified and reliable view for TME. Pull-though anastomosis is a convenient and low-cost method.
10.Influence of IgG in bullous pemphigoid blister fluid on the secretion of chemokines by human keratinocytes

Objective To investigate the effect of immunoglobulin G (IgG) in bullous pempbigoid (BP) blister fluid on the secretion of chemokines by human keratinocytes. Methods IgG was obtained from the blister fluid of patients with bullous pemphigoid and sera of normal human controls, then purified by sequential precipitation with caprylic acid and ammonium sulfate. The immunological activity of blister fluid was tested before and after the purification by BP180 ELISA kit. Keratinocytes were isolated from the foreskin tissue of yong adults, and subjected to primary culture. After 3 passages, the primary keratinocytes were harvested and subcultured in the presence of purified IgG of 0.5, 1, 2, 4 and 8 g/L, respectively, for 24 hours, or IgG of 4 g/L for 3, 6, 12, 24 hours, respectively, followed by the detection of levels of eotaxin, monocyte chemoattractant protein (MCP)-1 and interleukin (IL)-8 in the supemate of keratinocytes by ELISA. Results The valence of IgG remained unchanged after the purification with caprylic acid and ammonium sulfate. Compared with IgG from the sera of normal controls, that from bullous pemphigoid blister fluid sig- nificantly enhanced the secretion of IL-8 by keratinocytes in a time- and dose-dependent manner (both P < 0.01 ). Neither eotaxin nor MCP-1 was detected in the supemate of control IgG-treated, BP IgG-treated or untreated keratinocytes. Condusions The IgG in BP blister fluid has been proved to stimulate the secretion of IL-8 by cultured human keratinocytes, which may be involved in the pathogenesis of BP.
