2.The clinical study of the effect of the treatment of the knee joint replacement with Rivaroxaban

Objective To explore the effect of the application of Rivaroxaban in the treatment of the patients with total hip replacement.Methods 114 patients with total knee arthroplasty were randomly divided into control group and observation group,57 cases in each group.All patients were operated by the standard operation in the department of orthopedics.The control group was given low molecular weight heparin before operation.The patients in the observation group were treated with 8h after the operation,and the color Doppler ultrasound was performed on the patients with double lower limbs and lungs after 2 courses of treatment.Observation of drainage,two groups of patients after total blood transfusion,blood index,adverse reactions and deep vein thrombosis cases.Results In the observation group,the deep vein embolism rate (1.75%)was significantly lower than that in the control group of 12.28% (χ2 =4.25,P <0.05 );The postoperative drainage volume,blood transfusion and blood index of the two groups had no significant differences (t =0.57,0.82,1.03,all P >0.05).During the treatment,the patients in the two groups were adverse reactions.Conclusion The effect of oral administration of Rivaroxaban after hip replacement surgery is remarkable,which can effectively reduce the incidence of deep venous thrombosis,and it is safe,and it is worthy of clinical application.
3.One case report of pharyngeal bursa invasive fungal disease with lower cranial nerve involvement as the first manifestation.

To increase the identification of pharyngeal bursa invasive fungal disease with lower cranial nerve involvement, reduce the misdiagnosis and improve the awareness of invasive fungal disease. We report the clinical data of a case with lower cranial nerve involvement as the first manifestation and reviewed the related literature.

4.PRELIMINARY STUDY ON MUTUAL ANTIGENS BETWEEN LUMBRICUS TERREST RIS AND SCHISTOSOMA JAPONICUM

Objective To study preliminar il y the mutual antigens between Lumbricus terrestris[WT5”BZ ] and Schistosoma japonicum. Methods The antigen s of Lumbricus terrestris (L.terrestris) and [ WT5”BX]Schistosoma japonicu m (S.japonicum) were analysed by SDS-PAGE technique, and reacte d with the sera of patients with schistosomiasis and the sera of rabbits immunized with LtAg by Dot-ELISA and Western-blot. Results [WT5” B Z]There were many similar molecular weight antigens between LtAg and SjAg. Spe cially some antigens between 30-90 kDa form LtAg had intensive cross reactio n with the sera of patients with schistosomiasis. SjAg was also reacted with the sera of r abbits immunized with LtAg. Conclusion There are at least three immunocompetent antigens with similar molecular weight between L.terrestris and S.japonicum. [
5.Preparation of constant negative pressure drainage tube and the evaluation of its effect

Objective To explore the drainage effect of drainage tube with different materials on the basis of a constant nega tive pressure drainage,and its impact on flap survival.Methods The most common clinical used Polyvinyl chloride resin (PVC)pipe,latex tubes,silicone tube drainage tubes,and flaps were compared at different phases of the drainage,to determine the best clinical flap with drainage material.After three groups of flaps were designed,including a constant negative pressure drainage device,without a constant negative pressure drainage system,and drainage alone (no negative pressure) drainage flap,the flap survival was observed at different phase space.Three different drainage drainage ways of the flap and the flap survival area of effect were statistically analyzed.Results The silicone tube drainage was more fully and effectively.Constant negative pressure drainage volume of liquid was up to the flap with drainage to the fullest,the highest rate of skin flap healing,and better healing.Conclusions The constant negative pressure in the drainage of the flap makes drainage more fully,and promotes rapid healing of wounds,while the silicone tube is suitable for flap drainage.

7.Clinical and evidence-based study of coagulopathy after traumatic brain injury

Coagulopathy after traumatic brain injury is a coagulation dysfunction caused by brain tissue injury, which is characterized by over activations of coagulation, fibrinolysis and anticoagulation. It is the result of the interaction of multiple factors and multiple links with complicated pathophysiology. This study reivewed the clinical features and evidence-based evidences of coagulopathy after traumatic brain injury in order to summarize and discuss the latest progress in the diagnosis and treatment of coagulopathy after traumatic brain injury.
8.Diagnostic value of quantitative detection of SLIT2 methylation for cervical high grade precancerous lesion

Objective To explore the clinical diagnostic value of quantitative detection of the slit homologue 2 (SLIT2) methylation for cervical high grade precancerous lesions. Methods According to histopathologic diagnostic results, 178 patients infected with high-risk HPV were divided into normal cervix group (n=45), low-grade lesion group (n=50) and high-grade lesion group (n=83). The cervical exfoliated cells were collected in three groups. The methylation levels of SLIT2 were measured by pyrosequencing in three groups. The diagnostic threshold of SLIT2 in high grade precancerous lesions was estimated by receiver operating characteristic (ROC) curve. Results The percentages of SLIT2 methylation were (4.53 ± 1.37)%, (5.81 ± 2.26)% and (11.80 ± 8.47)% in normal cervix group, low-grade lesion group and high-grade lesion group, respectively. And the differences between three groups were statistically significant (F=27.61, P<0.001). The percentage of SLIT2 methylation was significantly higher in high-grade lesion group than that of normal cervix group and low-grade lesion group (P<0.001). There was no significant difference in the percentage of SLIT2 methylation between normal cervix group and low-grade lesion group (P=0.297). The area under the ROC curve was 0.895 and optimal cut-off value was 6.41%. The sensitivity and specificity were 80.7% and 83.2%, respectively for the detection by SLIT2 methylation. Conclusion The quantitative detection of SLIT2 gene methylation level in cervical exfoliated cells by pyrosequencing can effectively diagnose cervical high grade precancerous lesions.
9.Inhibition of expression of bcl-2 gene by siRNA in glioma cell line U251

AIM: To evaluate the effectiveness of small interference RNA on inhibiting bcl-2 gene expression. METHODS: With Ambion's software and kit, we designed and synthesized siRNA targeted bcl-2, which were transfected into astrocytoma cell line U251 with lipofectamine. The non-transfected cells and treatment with antisense drug G-3139 were taken as controls. MTT was used to detect the inhibitory rate of cell growth. Flow cytometric method was used to detect the change in cycle of the cells. The inhibitory effect of siRNA on mRNA level was detected by RT-PCR and on protein level was by immunohistochemical method. RESULTS: For the living rate of cell, siRNA 2, 3, 5, 6 groups were significantly lower than siRNA 1, 4 groups, lipofectamine and control group at 24, 48, and 96 h. siRNA 1-6 groups only had statistic difference with antisense group at 24, 48 h. As for PCR and immunohistochemical method, the expression of bcl-2 on siRNA 2, 3, 5, 6 groups were significant lower than other groups. The results of flow cytomytric method showed the cells transfected with siRNA 1-6 and antisense were blocked at S stage. CONCLUSION: siRNA inhibited bcl-2 gene expression more than 50%.
