2.Analysis of postoperative complications after transrectal ultrasound-guided 24-core transperineal prostate biopsy

Objective To analyze the complication rates in 210 patients undergoing transrectal ultrasound-guided 24-core transperineal prostate biopsy.Methods Clinical data of 210 patients who underwent transrectal ultrasound-guided 24-core transperineal prostate biopsy from 2010 to 2012 were collected.A retrospective analysis of complications after prostate biopsy was conducted.Results The main postoperative complications were hematuria,bloody stool,dysuria,urinary retention,fever (>38℃),urinary infection,vasovagal syncope,and their complication rates were 43.3 % (91/210),10.9% (23/210),23.3% (49/210),25.2% (53/210),14.8% (31/210),21.9% (46/210),0.48% (1/210) respectively.In addition,5 patients (2.5%) were hospitalized due to biopsy related complications.Conclusions Transrectal ultrasound guided 24-core transperineal prostate biopsy is an relatively safe and effective puncture method with common minor complications and less severe complications.Adequate attentions should be paid to its complications.
3.Interleukin-17 in apical exudates of periapical periodontitis treated with minocycline controlled-release formulation

BACKGROUND: Experimental proof for the efficacy, safety, and immunological assessment is needed when minocycline is used for root canal disinfection.OBJECTIVE: To investigate the effect of minocycline for root canal disinfection on levels of interleukin-17 in apical exudates of periapical periodontitis and periapical exudate volume.METHODS: Sixteen patients with acute periapical periodontitis (16 teeth) scheduled for root canal therapy were enrolled and randomly divided into calcium hydroxide and minocycline groups, respectively, followed by root canal disinfection.One week after disinfection, periapical index, periapical exudate volume and interleukin-17 level were detected prior to the root canal filling. Another 16 patients with normal pulp vitality (16 teeth) scheduled for single root canal filling were enrolled as control group, in which periapical index, periapical exudate volume and interleukin-17 level were detected.RESULTS AND CONCLUSION: The periapical exudate volume and interleukin-17 level in the calcium hydroxide and minocycline groups were significantly higher than those in the control group (P < 0.05). The periapical index and interleukin-17 level in the calcium hydroxide and minocycline groups were decreased significantly at 1 week after root canal disinfection (P < 0.05), while there was no difference between these two experimental groups in the periapical index, periapical exudate volume and interleukin-17 level. To conclude, the use of minocycline significantly reduces interleukin-17 level and periapical exudate volume, and thus achieves effective outcomes in periapical disease.
4.Molecular Characterization of Two Rice cDNAs Encoding GDP-Mannose-3', 5'-Epimerase and Their Expression Patterns

GDP-mannose-3', 5' -epimerase (GME), which converts GDP-mannose into GDP-L-galactose, is essential for the biosynthesis of L-ascorbic acid in higher plants. The molecular characterization of two GME genes from rice has been reported. Firstly,both cDNAs were isolated from the rice mature leaves using RT-PCR technique. By comparing their sequences with homologues from other plants, it was found that GME genes were highly conserved among plant species, though phylogenetic study showed that all known GMEs could be divided into two distinct groups corresponding to monocots and dicots. Secondly, the genomic organization of rice OsGME genes was investigated, and a similarity of splice patterns was revealed. Finally, the expression patterns of the two cDNAs have been studied in various tissues and under different stress conditions by semiquantitative RT-PCR assay. The results showed that the OsGME1 transcript was up-regulated in response to cold stress, and gibberellin might regulate L-ascorbic acid levels by affecting transcription of both OsGME genes.
5.Expression of livin and caspase-3 and their clinical significance in pancreatic carcinoma

Purpose To study the expression of livin and caspase-3 protein in pancreatic carcinoma and their correlation with clinicopathologic features and survivial time.Methods High-throughput tissue microarray technique and immunohistochemistry were used to detect the expression of livin and caspase-3 protein in 88 pancreatic carcinoma specimens without a history of chemo-radiation therapy and 11 non tumorous pancreatic tissues,and the relationship of these subjects was analyzed with Spearman rank correlation. Among these 88 cases, 54 cases were followed-up for 2 months to 6 years, whom were used for the prognosis analysis.Results In the 88 cases, the positive rate of livin was 76.1% (67/88); however, the positive expression of caspase-3 was 90.9%(10/11) in non-tumorous pancreatic tissue, which was much higher than that in the cancer (40.9%,36/88), with statistically significant differences(P<0.01). There were significant relationships of livin protein expression with sex, TNM stage and lymph node metastasis (P<0.05); the expression of caspase-3 protein expression was significantly related to sex, histological grade, TNM stage and lymph node metastasis (P<0.01). Spearman rank correlation showed a significant correlation between livin and caspase-3 protein (P<0.01). In 54 cases with followed-up data, univariant analysis revealed that both of livin protein expression and lymph node metastasis were related to the survival rate (P<0.05), but caspase-3 protein expression, sex, age, tumor location, size, histological grade and TNM stage were unrelated to prognosis (P>0.05). Furthermore,COX stepwise proportional hazards analysis showed that only livin status retained independent significance in prospecting prognosis of pancreatic carcinoma.Conclusions The expression of livin is associated with clinicopathologic features and prognosis in pancreatic carcinoma. High expression of livin protein indicates poor prognosis, which could become an independent index for the evaluation of prognosis.caspase-3 protein may be involved in the regulation of apoptosis and the progression of pancreatic carcinoma.
6.Study on Characteristics of Pointing Movement Time for Targets with Different Position in Front of Human Body

Objective To study the characteristics of pointing movement time for targets with different position in front of human body,to detect the effects of targets location on pointing movement time.Methods A pointing movement experiment of human arm was designed and carried out.By the statistic analysis of the experimental data,the relationship between pointing movement time and target position was discussed.Results It was shown that the starting point position and target position extremely affected the pointing movement time.Conclusion When a right-handed person touches the different targets,both the direction to target and the starting point position(target-circle location) would affect the pointing movement time extremely.When the position of the starting point changed by horizontal or vertical,the pointing movement time is more changeful by horizontal than that by vertical.To compare the pointing movement time for each target on a target-circle,the pointing movement time for right-upper target is the shortest.When the starting point is located on the right side of human body,all pointing movement time of that target-circle is the shortest.
7.Effects of breviscapine on Na~+,K~+-ATPase activity of proximal tubule in early diabetic rats

Aim To investigate the effects of breviscapine(Bre)on Na+,K+-ATPase activity of proximal tubule(PT) and its renal protection in early diabetic rats. Methods The rats were divided into three groups: diabetic model (DM group), Bre treatment (Bre group) and normal control (NC group). Rats were administrated with Bre(20 mg?kg-1?d-1, ip) in Bre group, with normal saline(ip) in DM and NC groups for four weeks after diabetes induction with streptozotocin (STZ 65 mg?kg-1, ip) in Bre and DM groups. The urine and blood samples were collected from two intra-ureteral cannulas and the heart, respectively, under anesthesia four weeks after diabetes induction. After one of the renal arteries being perfused, the renal cortex was incubated and PT segments were microdissected freehand under microscope before the Na+,K+-ATPase activity in the segments were assessed by liquid scintillation counter. The blood glucose, levels of creatinine in serum and in urine were assayed. The microalbumin , ?2-microglobulin(?2-MG) of urine and endogenous digitalis-like substance (EDLS) of serum were measured respectively by radioimmunoassay. Results The PT Na+,K+-ATPase activity in NC group was(959.11?117.35) pmolPi?mm-1?h-1, and that in DM group was significant higher for (1893.53?383.90) pmolPi?mm-1?h-1 than it(P
8.A Study of Pointing Movement Characteristics and Ergonomics on Front Board

Objective To study pointing movement characteristics and ergonomics when human upper limb touches different targets on the front board. Methods An experiment was designed, in which the data of motion duration, peak velocity, trajectory length and peak power at shoulder and elbow joint were collected. Results By analysis of the experimental results, the pointing movement characteristics among the different regions on the front board were disclosed preliminarily. Conclusion When a right-handed person touches a target on the front board, the pointing movement ergonomics on left-upper region is the best, while that on right-lower region is the worst.
9.Lidocaine effects on increasing levels of malondiadehyde and lactate in rats cortex brain cells cultured under anoxic condition

Objective: To determine the mechanism of protective effects of lidocaine on brain during ischemia. Method: Wistar rats within 24h after birth were enrolled. The brain cortex cells were separated and then cutured under normal or anoxic condition. The OD value of these cells were detected with rapid colorimetric assay after they were cultured 24h and 48h. According to the above values, other cells were cultured in different condition and divided intc three groups: control group(N) cultured under normal condition (37℃, 5%CO_2+95%air); anoxic group(A) and anoxic+ lidoeaine group(L) cultured under anoxic condition(37℃, 5%CO_2+95%N_2). 6.9?10~(-5)mol/L lidocaine was added to the medium in group L. Cells of these three groups were cultured under different condition for 48h, then malondiadehyde and lactate levels of each group were measured. Result: The survival cells of anoxic group were significantly lower than that of control group after cultured for 48h, the malondiadehyde levels were as follows (nmol/ml): group N(4.93 ?0.43, n=12), group A(5.56?0.34, n=12), group L(4. 80?0.25, n=11); the lactate levels (mmol/L): group N(8.05?1.64, n=21), group A (11.86?2.58, n=22), group L(9.96?1.88, n=21), Conclusion: Lidocaine may protect brain from ischemia through inhibiting malondiadehyde and lactate production induced by anoxia.
10.Clinical analysis of severe pneumocystis pneumonia in 7 children with non-human immunodeficiency virus infection

Objective:To better understand the clinical characteristics of children with pneumocystis pneumonia(PCP).Methods:Seven cases of non-human immunodeficiency virus infected children with PCP admitted to pediatric intensive care unit(PICU) at Xi′an Children′s Hospital from January 1, 2019 to December 31, 2019 were enrolled.Clinical findings, laboratory examination results, diagnosis, treatment, and outcome data were retrospectively reviewed.Results:There were seven children enrolled in this study including six boys and one girl.The average age was(6.09±3.80)years.Six patients received a long-term steroid or immunosuppressive therapy, and the average duration of medication was(80±30)days.One patient was eventually diagnosed with immunodeficiency disease despite no history of steroid and immunosuppressive therapy.Fever, cough and progressive dyspnea were the main manifestations.The pulmonary signs were not obvious, however, the chest radiographs showed serious ground glass changes.Laboratory examination showed that white blood cell counts were normal, lymphocyte counts were decreased to varying degrees, while C-reactive protein, calcitonin and lipopolysaccharide increased slightly or normal.Lactate dehydrogenase significantly increased and PaO 2/FiO 2 decreased, respectively.All children combined with different degrees of heart, liver and digestive tract damage.Pneumocystis jirovecii infection was diagnosed by metagenomics next-generation sequencing(mNGS), of which one case was detected by bronchoalveolar lavage fluid, and the remaining six cases were detected by peripheral blood.After entering PICU, mechanical ventilation, intravenous injection of human immunoglobulin, and continuous renal replacement therapy were performed.None of the children used TMP-SMZ for prophylaxis outside the hospital.Among them, one case was diagnosed with PCP before entering PICU and was given TMP-SMZ for 20 days, while the remaining six patients started to receive TMP-SMZ after entering PICU.Finally, one patient was cured, three died and another three cases died after giving up treatment.The average hospitalization days were(11.28±8.78)days, and the average hospitalization costs were (78, 000±24, 000) RMB. Conclusion:Children with non-human immunodeficiency virus infected PCP usually have underlying diseases or immunodeficiency diseases.Due to non-specific manifestations, rapid progression and high mortality, clinicians should pay more attention to it.It is necessary to dynamically observe the respiratory symptoms, perform mNGS as soon as possible, and give TMP-SMZ treatment in time.TMP-SMZ prophylaxis can be given to patients who are highly suspected of PCP but unable to perform pathogenic examinations.
