1.Evaluation on the acceptability of method performance validation for quantitative tests

We compared the total errors observed from both precision and accuracy validation experiments with defined medically allowable errors for each quantitative test,and used Westgard's method evaluation decision chart for initial judgement of the acceptability of method validation performance. Moreover,we need to judge the acceptability of other method validation parameters,so that we could integratedly assess and judge whether the testing system is acceptable to use in the laboratory.
2.Resveratrol protects against peroxide toxicity in rats induced by arsenic trioxide

Objective To study the effects of resveratrol against toxicity induced by arsenic trioxide in rats.Methods Arsenic trioxide was given to SD rats intraperitoneally for 4 weeks.Resveratrol was injected intraperitoneally to the animals 4 h before arsenic trioxide challenge at dose of 30,60,or 120 mg?kg-1?d-1.Four weeks later,the contents of malondialdehyde(MDA),superoxide dismutase(SOD),nitric oxide(NO),glutathione peroxidase(GSH-Px) in serum,lung,spleen,liver and brain tissues were measured.Results Compared with the normal control rats,arsenic trioxide challenge resulted in increases of NO and MDA contents in serum,lung,spleen,liver and brain tissues,and decreases of GSH-Px and SOD levels in the rats received arsenic trioxide only.Resveratrol significantly inhibited these changes induced by arsenic trioxide in rat serum,lung,spleen,liver and brain tissues in a dose-dependent manner.Conclusion Resveratrol can protect antiperoxidase activity and inhibit peroxide productions.
3.Intra-articular injection of synovial mesenchymal stem cells to treat articular cartilage injury

BACKGROUND:Among various seed cel s, synovial mesenchymal stem cel s have unique advantages in the repair of articular cartilage injury. OBJECTIVE:To review the progress of synovial mesenchymal stem cel s and its intra-articular injection in the treatment of articular cartilage injury. METHODS:The first author searched PubMed and CNKI by computer to retrieve articles published from January 2004 to December 2004 using the keywords of“synovial mesenchymal stem cel s;intra-articular injection;cartilage repair”in English and Chinese, respectively. Final y, 57 articles were included in result analysis. RESULTS AND CONCLUSION:It is easy to isolate and culture synovial mesenchymal stem cel s, which has great advantages in cartilage repair. What’s more, intra-articular injection therapy for articular cartilage injury is feasible and safe. Intra-articular injection of synovial mesenchymal stem cel s is a very promising treatment for cartilage damage, but there are stil many problems to be solved in the future.
4.The significance of lipoprotein-associated phospholipase A2 measurement in type 2 diabetes mellitus patients combined with atherosclerosis

Objective To investigate the clinical significance of lipoprotein-associated phospholipase A2(LP-PLA2) measure-ment for type 2 diabetes mellitus(T2DM) patients combined with atherosclerosis .Methods The concentrations of LP-PLA2 ,cho-lesterol(CHO) ,triglyceride (TG) ,low density lipoprotein(LDL) ,high density lipoprotein(HDL) were measured for 47 T2DM pa-tients combined with atherosclerosis(patient group) and 94 healthy people who just underwent healthy examination and were proved to be healthy(control group) ,meanwhile the age and gender information of those people were collected .Then the tests results were compared between the two groups .Results There was no significant difference in age ,gender ,CHO ,HDL ,TG ,LDL between the two patient and control group(P>0 .05);Plasma LP-PLA2 level in patient group was higher than that in control group ,and the difference was statistically significant(P<0 .05) .Non-conditional Logistic regression analysis showed that the LP-PLA2 level was the risk factor with statistical significance(P<0 .05) .Conclusion LP-PLA2 concentration is an independent risk factor for athero-sclerosis in T2DM patients ,and its measurement has certain value for the diagnosis of atherosclerosis in T2DM patients .
5.Enrichment process of scoparone from Yinchenhao Decoction with macroporous resin

Object To study the optimal conditions and parameters for the enrichment process of scoparone in Yinchenhao Decoction (YCHD) with macroporous resin. Methods With the enrichment degree of scoparone in YCHD as an index, the optimal conditions of the enrichment process were investigated. Results The optimal conditions were that the aqueous extract of YCHD was taken into the macroporous resin column, and kept for 30 min, then the resin was washed by five times of distilled water to get rid of impurity and three — four times of 70% ethanol to elute scoparone. Conclusion The process is feasible to enrich effective ingredient scoparone from YCHD.
6.Effects of puerperium pelvic floor muscle training on pelvic floor muscle strength

Objective To analyze the effects of puerperium pelvic floor muscle training on pelvic floor muscle strength and its clinical significance. Methods One hundred postpartum women were included, with full-term singleton pregnancies and with complete follow-up records from Obstetrics Department of Peking University First Hospital between March 1, 2013 and October 31, 2013. Women with vaginal birth and cesarean birth commenced pelvic floor muscle training twice a day from 24 and 72 h after delivery, respectively. According to the different training frequencies, the subjects were divided into three groups: never-training group, occasional-training group (<6 times per week) and regular-training group (≥6 times per week). All patients received pelvic floor muscle strength measurement 6-8 weeks after parturition. And the strength of type Ⅰ and type Ⅱ pelvic floor muscle fiber was divided into 0, Ⅰ, Ⅱ, Ⅲ, Ⅳ and Ⅴgrade. The abnormal strength of typeⅠand typeⅡpelvic floor muscle fiber standed for the grades lower thanⅢ. We compared the general conditions, delivery modes and abnormal ratio of type Ⅰ and type Ⅱ pelvic floor muscle fiber among the three groups, analyzed the relativity between the level of pelvic floor muscle fiber strength and pelvic floor muscle training frequency, and analyzed the influential factors of pelvic floor muscle fiber strength. Statistical analyses were performed by one-way ANOVA, rank-sum test, Pearson χ2 test, Kruskal-Wallis nonparametric test, Nemenyi test, Spearman rank correlation analysis and multivariate Logistic regression analysis. Results There was no statistical difference in age, gestation at delivery, parity, body mass index before delivery, neonatal birth weight and delivery mode among the never-training group (21 cases), occasional-training group (30 cases) and regular-training group (49 cases) (all P > 0.05). There was no statistical difference in labor time of first, second and total stage and episiotomy rate among the vaginal birth cases of the three groups. The abnormal ratio of type Ⅰ muscle fiber strength among the three groups was 100% (21/21), 77% (23/30) and 6% (3/49), respectively, while that of type Ⅱ muscle fiber strength was 100% (21/21), 53% (16/30) and 20% (10/49), respectively. And there were significant statistical differences among the three groups (F=119.16 and 77.84, both P<0.01). Spearman rank correlation analysis indicated that the level of typeⅠand typeⅡmuscle fiber strength had a significant positive correlation with pelvic floor muscle training frequency (r=0.88 and 0.79, both P<0.01). Multivariate Logistic regression analysis showed that pelvic floor muscle training was an influential factor for pelvic floor muscle strength level (95%CI:0.000-0.193, P<0.01). Conclusions Puerperium pelvic floor muscle training can help enhance the tension force of pelvic floor muscle, might benefiting the postpartum recovery of pelvic floor muscle function.
7.Influence of tamoxifen on the level of cellular proliferation on cultured human pituitary adenoma cells and its mechanism

Aim To investigate the effects of tamoxifen on the proliferation and DNA synthesis of cultured human pituitary adenoma cells and make a further investigation of the mechanism of the inhibitory effect of tamoxifen on the proliferation of pituitary adenoma cells. Methods The techniques of MTT colorimetry, 3H-TdR, flow cytometry, PKC activity detection and cAMP/ cGMP levels detection were used to detect or observe the effects of tamoxifen on proliferation, DNA synthesis, cell cycle, PKC activity and cAMP/ cGMP levels of cultured human pituitary adenoma cells, respectively.Results ①Tamoxifen (0.1,1 and 10 ?mol?L -1) inhibited the proliferation and DNA synthesis of cultured human pituitary adenoma cells in a dose-dependent manner.② tamoxifen (1,10 and 20 ?mol?L -1) increased the ratio of G_1 phase of pituitary adenoma cells, and decreased the ratio of S and G_2 phase markedly;②compared with control, PMA, a PKC activator, increased the activity of membrane and total PKC in human pituitary adenoma cells. However, after a 15-min treatment with tamoxifen (10 ?mol?L -1),a significant reduction of the activity of cytoplasm, membrane and total PKC in human pituitary adenoma cells was observed;③tamoxifen (1 and 10 ?mol?L -1) increased the amount of cAMP in the cytoplasm of human pituitary adenoma cells, but had no effect on that of cGMP. Conclusion These data provide an important clue to explore the molecular mechanisms of the inhibitory effect of tamoxifen on the proliferation of pituitary adenoma cells, and suggest that the modulating effect of tamoxifen on the proliferation of pituitary adenoma cells results from interactions of several cellular signaling pathways.
8.Neurolymphomatosis, a case report

Neurolymphomatosis(NL) is characterized by lymphomatous infiltration of the peripheral nervous system. We report a case of neurolymphomatosis(NL) which was confirmed by sural nerve biopsy. Sural nerve specimen from a 49-year-old female patient with weakness of limbs was examined with routine histochemical and immunohistochemistry staining, in which the first antibodies against CD3, CD20, CD45, CD45RO and CD68 were used. Numerous T-lymphoma cells invaded in the adipose tissue of epineurium of sural nerve. The nerve biopsy showed marked axonal degeneration of myelinated fibers. The clinical and histopathologic findings confirmed the diagnosis of neurolymphomatosis.
9.Analysis methods of chromatographic fingerprints of Panax gingseng C.A.Mey.

AIM: To establish chromatographic fingerprint of Panax gingseng C.A.Mey.by HPLC. METHODS: Separation was performed on an Agilent ZORBAX SB-C_(18)(4.6 mm?250 mm,5 ?m) analytical column with mobile phase consisting of acetonitrile and water with gradient elution with the flow rate 1.0 mL/min and the column temperature at 40 ℃.The UV wavelength used for detection was set at 203 nm and the analysis time was 90 min. RESULTS: 18 co-peaks on the HPLC fingerprints of Panax ginerseng C.A.Mey.were indicated.The similarities were determined by cosine.The results of similarity analysis were 0.90~1.00. CONCLUSION: Perfect fingerprints were obtained which can be used for the quality control of Panax gingseng C.A.Mey.
10.Morphological changes of apoptotic brain neuroglioma cells induced with methyl-mercuric chloride and significances

Objective To study the anti-neuroglioma effect of methyl-mercuric chloride(MMC) by observing the morphological changes of apoptotic neuroglioma cells induced with MMC in rats with brain neuroglioma.Methods The rat models of neuroglioma were established,and divided into two groups.The rats in experimental group were lavaged with MMC 1 week after injected with C6 glioma cells,10 mg?kg-1every day,the rats in control group were treated with sodium chloride at the same dose.24 d after inoculation all rats were sacrificed except natural death,the brain tissues were obtained,and the pathohistological changes were observed under light microscope and transmission electron microscope.Results The macropathological result showed that the tumor volume in experimental group was smaller than that in control group.Under light microscope,in experimental group the growth density of C6 ghioma cells was lower than that in control group,and the apoptotic cells with smaller volume and karyopyknosis were found.The result of transmission electron microscope showed that in experimental group,the glioma cells had some changes such as karyopyknosis,chromoplasm margination,nuclear fragmentation and vacuolar degeneration and so on.Conclusion MMC has inhibitory effect on the proliferation of C6 glioma in rats in vivo,its mechanism may be related to inducing the apoptosis of neuroglioma cells.
