2.Research advances in relationship between diabetes and pancreatic cancer
Journal of Clinical Hepatology 2014;30(8):737-739
Pancreatic cancer (PC)is one of common malignant digestive diseases. It is mostly diagnosed at advanced stage, with an ex-tremely poor progression. The relationship between diabetes and PC was shown by numerous epidemiological studies for decades.Retrospec-tive clinical studies and research on molecular mechanisms in recent years have led to a new understanding of the relationship between diabe-tes,especially new-onset diabetes, and PC,the effect of antidiabetic medication on PC,and the molecular mechanisms underlying the con-nection between diabetes and PC.It is suggested by recent data that long-standing diabetes is one of the risk factors for PC development, new-onset diabetes may facilitate early diagnosis of PC,diabetes may have an impact on the prognosis of PC,the option of antidiabetic medication may influence the incidence of PC,and exploring the molecular mechanisms underlying the association between diabetes and PC may help to identify the new therapeutic target for PC.

Acta Medicinae Universitatis Scientiae et Technologiae Huazhong 2017;46(4):404-409
Objective To explore the effect of carnosic acid combined with tamoxifen on breast cancer cells and the mechanism. Methods Breast cancer cell lines MCF-7,T47D were divided into 4 groups:PBS was given to control group, carnosic acid(CA)was added to CA group, tamoxifen(TAM)was added to TAM group, and CA+TAM group was treated with carnosic acid and tamoxifen. The proliferation ability was measured by MTT assay, the colony formation capacity was measured by colony formation assay, the invasion ability was measured by Transwell chamber assay, the migration ability was measured by wound-healing assay, the expression of Caspase-8,Caspase-3 and PARP protein were measured by Western blotting. Results CA,TAM and CA+TAM suppressed breast cancer cell proliferation with a dose-dependent manner.Compared with CA group and TAM group, the proliferation inhibitory rate of CA+TAM group was significantly increased(P<0.05).In MCF-7 and T47D cell lines, compared to control group, the rate of colony fromation, number of invasive cells and cell migration rate of CA,TAM,CA+TAM groups were significantly decreased(all P<0.05).These indexes mentioned above in CA+TAM group were significantly lower than those in CA group and TAM group(all P<0.05),but there were no significant differences between CA group and TAM group (all P>0.05).In two breast cancer cell strains, the expression of cleaved Caspase-8 protein between control,CA,TAM and CA+TAM groups was not significantly different(P>0.05).Compared to control group, the expression levels of cleaved caspase-3,cleaved PARP protein in CA,TAM and CA+TAM groups were significantly increased(P<0.05),and these protein levels were significantly higher in CA+TAM group than in CA and TAM group(all P<0.05).But there were no significant differences between CA group and TAM group(all P>0.05).Conclusion Carnosic acid combined with tamoxifen can suppress the proliferation, invasion and migration of breast cancer cells, and its mechanism may be related to the activation of Caspase-3/PARP signaling pathway.

7.Advance in Metabolism of n-Alkane and Metabolic Engineering of Candida tropicalis
Microbiology 1992;0(02):-
Metabolism of n-alkane mainly includes ?-oxidation pathway and ?-oxidation pathway in Candida tropicalis, which are both catalyzed by a series of enzymes. The transformation from n-alkane to fatty acid is accomplished via ?-oxidation pathway, and the degradation of fatty acid is involved in ?-oxidation pathway. The mechanism on metabolism of n-alkane and studies on related enzymes are summarized in the review. Furthermore, some molecular genetics and metabolic engineering techniques related to Candida tropicalis developed in recent years are introduced.

Objective To investigate and analyze the behavior and therapeutic status of rheumatoid arthritis (RA) patients. Methods Out patients diagnosed with rheumatoid arthritis in our hospital from May to August 2007 were enrolled. The data including sex, age of onset, site of first hospitalization and medication status were collected and analyzed. Results In this 181 RA patients, the mean age of onset was (53±11) year-old, mean history duration was (10±8) years, the ratio of male to female was 1:4.2. The orthopedics department was the most common site of first hospital visit (32.0%, 58/181) and rheumatology department was the most common site to clarify the diagnosis (62.4% ,113/181). The diagnosis delay caused by patients themselves was (5.9±17.2) months and the delay caused by doctors was (9.0±22.0) months. More than half of the patients were not treated appropriately before they came to our hospital. Methotrexate was the most commonly used DMARDs (67.3%), followed by leflunomide (46.4%), sulfasalazine (37.5%) and hydroxyehloroquine (19.6%). Conclusion In this cohort, the proportion of patients who come to rheumatology department immediately after disease onset is low. There is delay between symptoms and final diagnosis. More than half of the patients are not treated appropriately.
