1.Tissue-engineered neural complex seeded in modified collagen scaffold for treatment of acute spinal cord injury in adult rats

Objective To repair the injured spinal cord in adult rats using tissue-engineered neural complex constructed in vitro by tissue engineering techniques and discuss the treatment effect.Methods Neural stem cells (NSCs) from rats were cultured separately and modified collagen scaffold was also prepared to construct complex of NSCs /modified collagen scaffold in vitro.Embryo spine cord extract was administered to induce differentiation of NSCs,the structure of which was revealed by histochemical technique and scanning electron microscope.The rat models of hemisected spinal cord injury were built and grafted with tissue-engineered neural complex.Functional recovery of the models was evaluated every two weeks postoperatively.Effect in repair of spinal cord injury was assessed by histological method at postoperative three and eight weeks.Results After tissue-engineered neural complex planted in the modified collagen scaffold was grafted into the injured spinal cord in adult rats,the seed cells survived and further differentiated into functionally active neurons.The differentiated glia cells presented quite regular reticular structure.Motor function below the level of the injured spinal cord in adult rats obtained obvious recovery.Conclusion Tissue-engineered neural complex seeded in the modified collagen scaffold takes effect in structural reconstruction and functional recovery of the injured spinal cord in adult rats.
2.Significance of transcatheter arterial chemoembolization combined with sorafenib for the treatment of primary hepatocellular carcinoma

Transcatheter arterial chemoembolization (TACE) was the preferred method of non-operation treatment for hepatocellular carcinoma (HCC).Radical resection of HCC remains difficult,extrahepatic metastasis was not easy to deal with,and repeated treatment aggravated the liver injury,so the long-term efficacy was poor.Sorafenib could control tumor angiogenesis and block the proliferation of tumor cells.A male patient with primary HCC and in the stage Ⅱb according to the Chinese clinical liver cancer staging system was treated by TACE combined with sorafenib and antiviral treatment in Henan Cancer Hospital.After the treatment,the intrahepatic lesions were inactive,and the pulmonary metastasis was partially relieved.The patient was followed up till May 2012,and the survival time was 39 months.The hepatic function was normal,and the hepatitis B virus (HBV) replication was negative.No intervention treatmentrelated complications were detected,and the KPS score was 100.
3.Change of energy metabolism and body composition before and after chemotherapy in patients with colon cancer

Objective:To evaluate the nutritional status before and after chemotherapy in patients with colon cancer by measuring energy metabolism and body composition.Methods:Fifty-one patients with colon cancer were included and were investigated using self-control method (before and after chemotherapy).Resting energy expenditure (REE) was determined by bed metabolic instrument,and body composition was measured by body composition analyzer.Results:For all patients,the REE on the 2nd day after chemotherapy was decreased significantly compared with that before chemotherapy (P < 0.05).After chemotherapy,body fat percentage and body moisture percentage were increased significantly (P < 0.05),and lean body mass percentage was decreased significantly (P < 0.05).Conclusion:After chemotherapy,the REE and lean body mass percentage of the patients with colon cancer were decreased,and body fat percentage and body moisture percentage were increased.
4.Characteristics and feasibility of bone marrow-derived mesenchymal stem cells labeled with 5-bromodexyuridine

BACKGROUND: The indicant of successful transplanted bone marrow stem cells is that the labeled transplanted ceils can survive in the target organs and can their biological functions. Up to date, at cell level, there are several labeled methods such as enzyme-linked, 3H-TdR, fluorescent and 5-bromodexyuridine method, etc.OBJECTIVE: To observe the biological characteristics of bone marrowderived mesenchymal stem cells labeled with 5-bromodexyuridine. DESIGN: Observations in single kind of sample SETTING: Department of Thoracic Cardiac Surgery, Affiliated Hospital of North China Coal Medical College MATERIALS: The experiment was carried out in the Experimental Center of North China Coal Medical College from July, 2003 to November,2004 on six Japanese long-eared rabbits ,with the age of 3 months , of either gender and the body mass of (3.00±0.25)kg.METHODS:①Monocytes were isolated from the bone marrow with Percoll reagent of the concentration of 1 073 g/L. The mesenchymal stem cells were cultured and proliferated with Eagle's medium, which was improved by adding 10% low sugar Dulbecco's to fetal bovine serum, so that their purity could reach about 95%. Secondly, in the experimental groups, the percentage of bone marrow-derived mesenchymal stem cells labeled with 5-bromodexyuridine was detected after 24 hours, 48 hours, 72 hours and 96hours, respectively. The negative-controlled group was composed of bone marrow-derived mesenchymal stem cells non-labeled with 5-bromodexyuridine while the blank-controlled group was made by substituting phosphate buffer or normal mice serum for primary antibody.MAIN OUTCOME MEASURES: In three different groups, 200 cells of each group were detected and observed at different time. Outcomes of the immunochemical stauning and counting of bone marrow-derived mesenchymal stem cells labeled with 5-bromodexyuridine were also determined.RESULTS: The positive bone marrow-derived mesenchymal stem cells labeled with 5-bromodexyuridine were observed in all the experimental groups. The positive materials were brown, granular and dfiffusedly scattered in the nucleus while no positive cell was observed in the controlled groups. After 24 hours, 48 hours, 72 hours and 96 hours, the numbers of bone marrow-derived mesenchymal stem cells labeled with 5-bromodexyuridine were 48±2, 100±4, 173:t:2, 178±3 respectively. The labeling percentage was raised gradually with the elongation of time. 72 hours later,the labeling percentage is above 85%. The numbers in negative-controlled and blank-controlled groups were all zero.CONCLUSION: The appropriate time of bone marrow-derived mesenchymal stem cells labeled with 5-bromodexyuriding is 72 hours. The sensibility of post-labeling detection is high due to the results being observed in the low power microscope, which makes this method suitable to quantitative analysis in massive tissue. These results show that the method of bone marrow-derived mesenchymal stem cells labeled with 5-bromodexyuridine is feasible.
5.In vivo magnetic resonance imaging tracking of bone marrow-derived mesenchymal stem cells via intracoronary administration: Consistency to pathohistological results

BACKGROUND: Recent trials and clinical studies have shown that intracoronary transplantation of bone marrow-derived mesenchymal stem cells (MSCs) improves cardiac function following acute myocardial infarction (AMI). However, whether homing of MSCs into the infarcted myocardium or not is still unknown.OBJECTIVE: To study the homing of MSCs intracoronary administration in porcine myocardial infarction model using in vivo magnetic resonance imaging tracking.METHODS: Porcine MSCs were isolated and cultured by the whole bone marrow method. Following labeling by superparamagnetic iron oxide (SPIO), MSCs were treated with trypsinization to adjust the concentration at 10~(10)/L. Myocardial infarction was induced in all 10 pigs. At one week after modeling, the labeled MSCs were delivered via intracoronary infusion with standard over-the-wire (OTW) balloon angioplasty catheters. Prussian blue staining was used to evaluate labeling efficiency, and double echo steady state was used to scan four-chamber and cor biloculare at long axis view, which was considered as locating phase to obtain image of left ventricle at short axis view. RESULTS AND CONCLUSION: MSCs could be efficiently and safely labeled with SPIO. Intracoronary transplantation of MSCs is able to home the sites of myocardial injury and the border between infarcted and normal tissue. MRI can track SPIO-labeled MSCs delivered through intracoronary and were confirmed on pathology. After 5 weeks the injected labeled cells could still be detected with MRI.

Phaffia rhodozyma is a good strain for astaxanthin production. An over-producing mutant YB-20-29 was obtained by means of Cs137-?ray and N-methy1-N'-nitro-N-nitrosoguanidin (NTG) treatment. The biomass for this strain by shake culture was 36.32 g/L, the pigment content was 1216.0 ?g/g, an increase of 308% compare to o-riginal strain. The astaxanthin content in broth was 30.9?g /mL. It was a potential strain for astaxanthin over-production.

Objective To investigate blood gas analysis and lactic acid evaluation in aerobic forearm exercise and the significance of aerobic forearm exercise for the auxiliary diagnosis of mitochondrial myopathy and encephalopathy patients.Methods Forty-two patients with mitochondrial myopathy and encephalopathy patients, 40 healthy control, and 40 patients control were studied.They performed a protocol under aerobic exercise conditions, consisting of intermittent forearm exercise for 4 minutes at 40% of intented maximal voluntary contraction force.Blood samples were collected to monitor blood gas and plasma lactate before, during arid after exercise.Results During exercise venous PO_2(mm Hg, 1 mm Hg=0.133 kPa)decreased in mitochondrial myopathy and encephalopathy patients from 41.2?12.6 to 39.5?16.2, whereas PO_2 fell from 50.5?14.4 to 30.8?13.1 in healthy control and from 50.1?7.9 to 44.3?35.5 in patient control.Venous PO_2 decreased much more in healthy control group than the other 2 groups(F= 6.34,P
8.Idiopathic generalized myokymia:Diagnosis and treatment

ObjectiveTo probe the clinical features,diagnosis and treatment of idiopathic generalized myokymia.MethodsSeven patients with idiopathic generalized myokymia were analysed retrospectively.ResultsAll 7 patients showed prominent myokymia characterized by undulating and vermicular movements spreading across the muscle surface. The myokymia in gastrocnemius muscles in all cases. The myokymia also appeared in both upper extremities in 5 patients,and in faces,waist,back,abdomen and all extremities in 2 patients. Muscle rippling movement was induced and increased by exercise,and persistent during sleep. The vermicular myokymia could be observed easily in the relaxation of the muscles. Electromyography tests showed myokymic discharges in 5 patients,but normal in 2 patients. 5 patients of them were cured with carbamazepine and phenytoin sodium.ConclusionThere are typical clinical features and effective treatment in the patients with idiopathic generalized myokymia.

The common clinical manifestations of the primary agiitis of the central nervous system include burst of headache, dementia, change of aptitude, paralysis of cranial nerves, and recurrent focal depletion of the neural function. Lptomeningeal and brain biopsy are still the gold criteria for diagnosis. The prognosis may be improved after cortin and immunosuppressant therapy.

10.Analysis of BRAF gene mutations in 80 patients with malignant melanoma in Xinjiang Uygur Autonomous Region

Objective To assess the relationship between BRAF gene mutations and clinical phenotype of malignant melanoma.Methods Tissue specimens were collected from the lesions of 80 patients with malignant melanoma,and from the normal skin of 30 patients with trauma in the Department of Plastic Surgery or General Surgery,and subjected to paraffin embedding and DNA extraction.PCR was performed to amplify the exon 11 and 15 of BRAF gene followed by DNA sequencing.Chi-square test and Fisher's exact test were carried out to assess the relationship between BRAF gene mutations and clinical phenotypes of malignant melanoma.Results BRAF gene mutations were found in 19 (23.8%) of the 80 malignant melanoma specimens.Among the 19 mutationpositive specimens,17 (88.2%) carried mutations in exon 15 of BRAF gene with V600E as the most frequent (88.2%,15/17) mutation type,and 2 (10.5%) carried mutations in exon 11.No mutation was found in any of the normal skin tissue specimens.The average age at onset was 57.5 years in these patients.The frequency of BRAF gene mutation was significantly higher in patients younger than 60 years than in those older than 60 years (37.1% vs.13.3%,x2=6.613,P < 0.05).A significant difference was observed in the frequency of BRAF gene mutation among tissue specimens of mueosal,acral and non-aeral malignant melanoma (18.2% (4/21) vs.14.7%(5/34) vs.41.7% (10/24),x2=6.167,P < 0.05).There was no significant association between BRAF gene mutation and gender,race or lymph node metastasis (all P > 0.05).Conclusions BRAF gene is a hot spot for mutations in patients with malignant melanoma in Xinjiang Uygur Autonomous Region,with V600E point mutation in exon 15 as the most frequent mutation type.BRAF gene mutations appear to be closely correlated with the age at onset of and lesional sites in,but uncorrelated with gender and race of or lymph node metastasis in,patients with malignant melanoma.
