Objective To evaluate the toxicological safety of Shanze weight-reducing food in order to provide some toxicological data for its application. Methods According to the national standard of safety evaluation, the following experiments of acute toxicity test through mouth in rats, the genetic toxicity test, thirty days feeding test and teratogenicity test in mice were performed. Results LD 50 was more than 20.0 g?kg -1, equivalent to 467 times of the recommended dosage, in acute toxicity test through mouth in both male and female rats, which fell into the scale of non-toxicity. Under the dosage range of 1.05 g?kg -1 to 4.20 g?kg -1, equivalent to 100 times of the recommended, the negative results were observed in the genetic toxicity test of micronucleus test of bone marrow cells, sperm shape abnormality test in mice and the Ames test. The negative impacts on the growth, development, hematology, biochemistry, histology and the ratio of organ to whole body were not found on the experimental rats of thirty days feeding. Under the same dosage range, no teratogenic effects were found in the experiments of teratogenicity test. Conclusions Shanze can be used as a kind of health weight-reducing food with toxicological safety under the experimental dosage range of 1.05 g?kg -1 to 4.20 g?kg -1.

Background Cardiomyocytes apoptosis was related to oxidative stress which has been shown to play an important role in ventricular remodeling after myocardial infarction(MI).Probucol is a hypolipidemic and antioxidant agents.Several reports demonstrated its cardioprotective effect after MI.However, the exact effect of probucol on the modification of the apoptosis related gene Bcl-2 and Bax is not clear. Objective To investigate the effects of probucol on mRNA and protein expression of Bcl-2,Bax and oxidative stress in myocardial infarcted rats. Methods Forty-one SD rats that survived 24 h after ligating left anterior descending coronary artery were randomly to receive placebo-saline(5 mL/d,n=20)or probucol(probucol 60 mg/kg?d,n=21).Twelve rats underwent sham operation were served as control(n=12).Six weeks after treatment, hemodynamic pararmeters and left ventricular function were measured with catheterization.Cardiomyocytes apoptosis were determined by TUNEL method.Myocardium mRNA of Bcl-2 and Bax expression levels in the non-infarcted myocardium were as- sessed by RT-PCR.The myocardium protein expression of Bcl-2 and Bax in the non-infarcted myocardium were de- termined by Western blot.Colorimetry was used to determine oxidative metabolism index in myocardium. Results 1)Compared with the sham rats, all MI rats showed marked decrease in Bcl-2 mRNA and protein expression in myo- cardium with increase of Bax mRNA and protein expression and apoptosis index(P

Objective To evaluate the effect of penehyclidine hydrochloride (PHC) on the level of angiopoietin-1 (Ang-1) and tyrosine kinase receptor-2 (Tie-2) during endotoxin-induced acute lung injury (ALI) in rats. Methods Forty adult male Sprague-Dawley rats, weighing 180-220 g,were randomly divided into 4 groups using a random number table (n =10 each):control group (group C),ALI group,low-dose PHC group (group L-PHC) and high-dose PHC group (group H-PHC).ALI was induced with iv injection of lipopolysaccharide 5.0 mg/kg via the tail vein.In L-PHC and H-PHC groups,PHC 0.6 and 2 mg/kg were injected, respectively, via the tail vein at 1 and 24 h after lipopolysaccharide injection. The rats were sacrificed at 48 h after the initial injection of PHC to measure the lung water content, protein concentration in bronchoalveolar lavage fluid (BALF),and the expression of Ang-1,Tie-2 and phosphorylated Tie-2 in lung tissues. The morphological changes of lung tissues were observed under light microscope and the ultrastructural changes of alveolar epithelial barrier under transmission electron microscope. Results Compared with group C,the lung water content and protein concentrations in BALF were significantly increased, and the expression of Ang-1 and phosphorylated Tie-2 was down-regulated in the other three groups (P < 0.05).Compared with group ALI,the lung water content and protein concentrations in BALF were significantly decreased, and the expression of Ang-1 and phosphorylated Tie-2 was up-regulated in H-PHC group (P < 0.05),and no significant changes were found in the parameters mentioned above in group L-PHC (P >0.05).The damage to lung tissues was significantly reduced in group H-PHC as compared with group ALI.Conclusion PHC can improve the permeability of pulmonary microvascular and reduce injury to alveolar epithelial barrier, thus ameliorating endotoxin-induced ALI in rats, and the effect is dose-related and up-regulation of Ang-1 expression and inhancement of Tie-2 activity are involved in the mechanism.

Chinese Journal of Obstetrics and Gynecology 2008;43(5):336-340
Objective To explore the secretion of chemokine regulated upon activation normal T cell expressed and secreted(RANTES)influenced by the complex microenvironment in the peritoneal cavity of women with endometriosis and investigate chemotaxis of RANTES on the peritoneal monocytes. Methods The contact and non-contact co-culture systems including three target cells of ectopic tissue were established. The three target cells were endometrial stromal cells(ESC),human peritoneal mesothelial cells(HPMC)and monocytes. After collection of the supernatant of co-culture systems, the levels of RANTES were detected by enzyme linked immunosorbent assay(EUSA).Migration of U937 cell,a monocyte line, was detected by chemotaxis assay. Results ESC,HPMC,and U937 cultured alone secreted slight RANTES,(5.0±0.5),(4.0±0.3),and (254±40)ng/L. Compared with the culture of the target cell alone, the levels of RANTES in each co-culture system increased significantly, with the highest level in the contact culture system of E-H-U(2250±96)ng/L. RANTES secretion of non-contact co-culture of three cells were higher than contact co-culture of two cells(P<0.01):U/E-H(912±93) vs E-H(50±40)ng/L,H/E-U(1201±93) vs E-U(243±192)ng/L,and E/H-U(1519±96) vs H-U(1251±73)ng/L. ESC,HPMC,and ESC-HPMC co-culture improved significantly migration of U937 cells [ number of cell migration respectively(6.0±0.3),(6.2±0.3),(10.0±0.3)×103,P<0.01],which could be inhibited efficiently by anti-RANTES neutralizing antibody.Condusion The target cells in the peritoneal cavity of patients with endometriosis promote the secretion of RANTES in autocrine and paracrine manners and migration of monocytes.

Objective To investigate the expression of HIF-1α protein in the placenta bed and the concentration of von Willrand factor (vWF) in maternal peripheral blood from pre-eclampsia and normal pregnancy, and to determine the effect of HIF-1α and vWF on the pathogenesis of pre-eclampsia. Methods Forty pre-eclampsia patients (20 mild and 20 severe) were recruited as 2 study groups, and another 20 normal pregnant women were served as a normal group.Western blot was used to detect the expression of HIF-1α protein in the placenta bed. ELISA was adopted to detect the concentration of vWF in the maternal peripheral blood.Correlation between HIF-1α protein expression and vWF level was analyzed by Spearman method. Results The expression of HIF-1α protein in the placenta bed was the highest in the severe pre-eclampsia patients among 3 groups, followed by the mild pre-eclampsia patients and the normal controls. There was significant difference (among) 3 groups (P<0.001).The concentration of vWF in the maternal peripheral blood was the highest in the severe pre-eclampsia patients, followed by the mild pre-eclampsia patients and the normal controls. There was significant difference among the 3 groups (P<0.05). Postive correlation was found between the expression of HIF-1α protein in the placenta bed and the concentration of vWF in the maternal peripheral blood in patients with pre-eclampsia (r_1=(0.65,) P<(0.001)),and between the concentration of vWF in the maternal peripheral blood and the pathogenetic condition degree of pre-eclampsia (r_2=(0.61,)P<0.001).Conclusion HIF-1α in coordination with vWF may play an important role in the pathogenesis of pre-eclampsia.
