Lysophosphatidic acid (LPA) is a newly discovered multi-function "phospholipid messenger" which is primarily from platelet activation,and is involved in a variety of biological effects,and closely correlated with carcinoma growth,invasion and metastasis.LPA plays an important role in carcinoma development.There is a certain degree of clinical significance of LPA in some carcinoma diagnosis and prognosis.
2.The effect of type 2 diabetes on collagen content in different organs in OLETF rats

Objective To research the effect of type 2 diabetes on collagen content in different organsinOLETFrats.Methods 24maleOLETFratsweregivenhigh‐caloriedietfor30weekstoset up the T2DM. At the 30th week ,16 male T2DM OLETF rats were selected as case group ,and 12 LETO rats were used as control group. The skin ,liver ,and femur tissue were collected at the same time. HE and Masson staining and immunohistochemical test were performed to measure the content of collagen and HPLC was used to detect hydroxyproline. Results Compared with the NC group ,DM group showed the collagen fibers were decreased in femoral and skin tissue ,but increased in liver tissue. The collagen content of the skin tissue was significantly lower in DM group than in NC group [(149.9 ± 18.4) vs (266.0 ± 54.8)mg/g ,P<0.01] ,and the collagen content of the femur tissue was also lower in DM group than in NC group [(185.60 ± 19.38) vs (243.27 ± 22.19)mg/g ,P<0.05]. In contrary ,the collagen content of liver tissue was higher in DM group than in NC group [(134.54 ± 18.78) vs (64.52 ± 21.51) mg/g ,P<0.01]. Conclusion The effect of type 2 diabetes on the collagen in different organs is different. The collagen content decreases in the femoral and skin tissue but increases in the liver.
3.The antibacterial activity of imipenem in combination with cefoperazone-sulbactam against carbapenem-resistant Acinetobacter baumannii

Objective To investigate the interaction of in vitro antibacterial activity between cefoperazone-sulbactam and imipenem against carbapenem-resistant Acinetobacterbaumannii.Methods The minimum inhibitory concentrations (MIC) of imipenem and cefoperazone-sulbactam against Acinetobacter baumannii were detected and the fractional inhibitory concentration (FIC) index of imipenem/cefoperazone-sulbactam combination was calculated by broth micro-dilution method.The interactions of imipenem and cefoperazone-sulbactam were assayed by K-B agar plate method.Results The MIC50 of cefoperazone-sulbactam and imipenem were both 16 mg/L,and MIC90 of the two drugs were both 64 mg/L.Among the 26 strains of Acinetobacter baumannii,16 strains had FIC indexes ≤0.5,and 10 had FIC indexes≥2.0.K-B agar plate assay showed that the cefoperazone-sulbactam/imipenem combination was synergistic for strains with FIC indexes≤0.5,and antagonistic for strains with FIC indexes ≥ 2.0.Conclusions Imipenem in combination with cefoperazone-sulbactam can be either antagonistic or synergistic against carbapenem-resistant Acinetobacter baumannii.The clinical use of cefoperazone-sulbactam/imipenem combination for the treatment of carbapenem resistance Acinetobacter baumannii infections should be cautious in case of antagonism.
4. Chemical constituents in root of Petasites tricholobus Franch. and their anti-inflammatory activity

Objective: To study the chemical constituents in the root of Petasites tricholobus Franch, and evaluate their anti-inflammatory activity. Methods: The primary extraction was done with 95% ethanol and subsequently with other agents including ethyl acetate. The compounds extracted with ethyl acetate were isolated by silica gel column chromatography and purified by means of recrystallization. The structures of the compounds were elucidated by employing chemical and spectral methods. The anti-inflammatory activities of the compounds were evaluated through their inhibitory effect on the trachea contraction of guinea pig induced by histamine in vitro. Results: Six compounds were isolated from the root of Petasites tricholobus Franch., namely, homofukinolide(1), bakkenolide-B(2), stigmasterol(3), and β-sitosterol(4), bakkenolide-D(5), and β-sitosterol-3-O-β-D-glucopyranoside(6). Bakkenolide-B and bakkenolide-D demonstrated inhibitory effect on the trachea contraction induced by histamine in vitro. Conclusion: Homofukinolide, β-sitosterol, and β-sitosterol-3-O-β-D-glucopyranoside are isolated for the first time from Petasites tricholobus Franch. Bakkenolide-B and -D have obvious anti-histamine activities.
5.Construction of DC-SIGN eukaryotic expression vector and establishment of BHK21 cell line stably transfected by the recombinant vector

Objective To construct a eukaryotic expressing vector harboring human DC-SIGN, and establish a BHK21 cell line stably and highly expressing DC-SIGN. Methods The DC-SIGN gene fragment which contained Not I and BamH I sites was amplified by PCR from pUNO-hDCSIGN1Aa plasmid, digested with Not I and BamH I, and then cloned into an eukaryotic expression vector pIRES-neo to construct eukaryotic expression vector pIRES-neo-DC-SIGN. The recombined plasmid was identified with Not I and BamH I enzyme digestion and sequencing, the latter was then transfected to BHK21 cells by LipofectamineTM 2000. After screening culture by G418, BHK21 cell line stably expressing DC-SIGN was established. The expression of DC-SIGN was identified by flow cytometry, Western blotting and immunofluorescence method. Results The gene sequence of DC-SIGN was consistent with that of design. PCR and double enzyme digestion analysis showed that the recombinant plasmid pIRES-neo-DC-SIGN was constructed successfully. After transfection, positive clones were selected with G418. After limiting dilution assay, BKH21 cell lines stably expressing DC-SIGN were established. The detection result of flow cytometry showed that the expression ratio of DC-SIGN positive clones was close to 90%. The result of immunofluorescence displayed that the expression of DC-SIGN was mostly located on the surface of cell membrane. Western blotting displayed the specific band of DC-SIGN protein. It showed that the BHK21 cells stably expressing DC-SIGN were successfully established. Conclusion DC-SIGN eukaryotic expression vector has been successfully constructed. The successful establishment of BHK21 cell lines which can stably express DC-SIGN provides a substantial foundation for further study on the DC targeting vaccines.
7.Lateral decubitus versus supine position in treatment of intertrochanteric fractures with proximal femoral nail antirotation

Objective To compare the lateral decubitus and supine position in the treatment of femoral intertrochanteric fractures with femoral proximal nail antirotation (PFNA).Methods A retrospective analysis was performed of the 85 patients with intertrochanteric fracture who had been treated at our hospital between September 2013 and September 2014.They were 24 men and 61 women,28 to 99 years of age (average,78.5 years).By Evans-Jensen classification,one case was type Ⅰ,15 cases were type Ⅱ,40 type Ⅲ,23 type Ⅳ,and 6 type Ⅴ.All the patients were treated with internal fixation with PFNA.Lateral decubitus position was adopted in 45 cases and supine position in 40.The 2 groups were compared in terms of operation time,intraoperative bleeding volume,difference between preoperative and postoperative hemoglobin values,tip-apex distance (TAD),calcar tip-apex distance (Cal-TAD),fracture healing time,and Harris score of the hip joint.Results The 85 patients were followed up for 9 to 21 months (average,13.6 months).Both 2 groups obtained fracture healing,with no screw cut-out.The lateral decubitus group had significantly less operation time (60.3 ± 17.5 min) and intraoperative bleeding volume (70.8 ± 37.8 mL) than the supine position group (72.7 ± 19.7 min and 90.3 ± 20.9 mL,respectively) (P < 0.05).The former also had a significantly smaller difference between preoperative and postoperative hemoglobin values (24.1 ± 5.7 g/L) than the latter (28.2 ± 8.5 g/L) (P < 0.05).There were no significant differences between the 2 groups in TAD,Cal-TAD,fracture healing time (3.7 ± 0.9 months versus 3.7 ± 1.0 months),or Harris hip score (73.4 ± 10.7 versus 75.5 ± 9.5) (P > 0.05).Conclusion In the treatment of femoral intertrochanteric fractures with PFNA,lateral decubitus position may be associated with shorter operation time and less intraoperative blood loss compared with supine position.
8.Clinical Study on Treatment of Elderly Herpes Zoster with Moxibustion on Heat-sensitive Acupoints Combined with Erwei Badu Powder

Objective To study the curative effect and mechanism of moxibustion on heat-sensitive acupoints combined with Erwei Badu Powder in the treatment of elderly herpes zoster (HZ) with type of stagnant heat of liver channel. Methods Sixty-five patients with HZ were randomly divided into treatment group (35 cases) and control group (30 cases). Two groups were orally given Longdan Xiegan Decoction and aciclovir conventionally, the treatment group was given moxibustion additionally on heat-sensitive acupoints combined with Erwei Badu Powder, and the control group was only given external application of Erwei Badu Powder for 10 days. The symptom scores, levels of T lymphocyte subsets and serum interleukin-2 (IL-2) were observed before and after treatment, the curative effect was evaluated, and PHN was followed up. Results The effective rate was 85.71%(30/35) in the treatment group and 66.67%(20/30) in the control group. The curative effect of treatment group surpassed the control group (P<0.05). Incidence of PHN was lower (P<0.01). The levels of CD4, CD4/CD8, and IL-2 increased and the level of CD8 decreased significantly in two groups compared with those before treatment (P<0.05). All indexes were better in the treatment group than those in the control group (P<0.05). Conclusion The curative effect of moxibustion on heat-sensitive acupoints combined with Erwei Badu Powder in treatment of elderly HZ is better than that of Erwei Badu Powder only, particularly in reducing the occurrence of PHN, improving neuralgia and insomnia. The mechanism may be related to increasing levels of CD4, CD4/CD8 and IL-2, and improvement of body’s cell-mediated immunity.
9.Application of video-assisted thoracoscopic surgery for thoracic traumas

Objective To research the feasibility and superiority of video-assisted thoracoscopic surgery(VATS) in the diagnosis and treatment of thoracic traumas.Methods Video-assisted thoracoscopic surgery was used in 40 patients with thoracic traumas,including thoracic exploration,repair of pulmonary laceration,and evacuation of clotted hemothorax.[WTHZ]Results All the 40 patients were cured,including simple VATS in 31 patients,thoracoscopy-assisted mini-incision surgery in 8 patients,and conversion to open surgery in 1 patient.The time of operation was 79.9?33.1 min.The amount of blood clots and noncondensing blood cleared was 567.5?177.8 ml.The closed thoracic drainage tube was removed at 24~48 h postoperatively,with a drainage volume of 220?45.6 ml.The length of hospital stay was 4~13 d(mean,8.7 d).The sutured wound healed by first intention in all the patients.No postoperative complications were observed.Follow-up reviews in 35 patients for 6~12 months(mean,8.6?2.6 months) revealed uneventful recovery and no trauma-related complications.Conclusions As compared with conventional open surgery,VATS has shown advantages of exact diagnosis,timely management,little invasion,and quick recovery for patients with thoracic traumas.
10.Effects of Four-week Intermittent Fasting on Skeletal Muscle Mass and Autophagy in Rats

Objective To observe the changes of body weight,fat mass and skeletal muscle mass of rats after 4 weeks of intermittent fasting,and explore relationship with autophagy in skeletal muscle,so as to provide theoretical basis for intermittent fasting.Method Twenty male Sprague Dawley rats were randomly divided into a control group (Con) and an intermittent fasting group (IF),each of 10.The rats of IF group were forbidden to eat food every Wednesday and Friday,and the body weight of both groups was recorded weekly.After 4 weeks,Dual-Energy X-Ray Absorption (DEXA) was used to analyze the body fat mass,then the bilateral soleus was separated to record the wet weight and measure the cross-sectional area of the soleus fibers by testing laminin with immunofluorescence confocal laser scanning microscope.The form of autophagic vacuole of soleus was observed using a transmission electron microscopy.The expression of autophagy-related protein LC3,p62 and regulating protein AMPK,pAMPK and ULK1 were measured using Western blotting.Result After 4 weeks of intermittent fasting,the weight and fat mass of IF were significantly lower than those of Con (P<0.01),but there were no significant differences between them in wet weight and cross-sectional area of soleus (P>0.05).The expressions of AMPK,p-AMPK,ULK1 in IF were significantly higher than those in Con (P<0.01).Compared with Con,the expression of LC3-Ⅱ/LC3-Ⅰ of IF increased significantly,while the expression of p62 decreased significantly (P<0.01).Conclusion Four weeks of intermittent fasting decreases the fat mass significantly,and control the weight efficiently.Intermittent fasting can maintain the skeletal muscle mass by promoting moderate autophagy through the AMPK-ULK1 pathway.It should be a potential lose weight method for further research.
