1.Effect of tilting table exercise on orthostatic hypotension after spinal cord injury in rabbits

ObjectiveTo establish an experimental model of orthostatic hypotension (OH) after spinal cord injury (SCI) in rabbits and investigate the effect and relevant mechanism of tilting table exercise on OH.MethodsRabbit model of OH was established by completely transecting animal's spinal cord at the T5 segment and putting animal on a 60° tilting table. Tilting table exercise was applied to training group for 28 days starting from 4th day after operation. Systolic blood pressure (SBP) and pulse rate (PR) were observed by non invasive sphygmobolometer of rabbit at 3rd, 10th, 17th, 24th and 31st day after the operation. The concentrations of plasma renin activity (PRA) and angiotensinⅡ (AngⅡ) of different position were detected with radio immunoassay at 31st day after operation.ResultsSBP at 60° head up tilt increased and difference of SBP between two positions decreased significantly in training group at 31st day. The concentrations of PRA and AngⅡ of SCI group and training group were higher than that of sham group. There were no significant differences in the concentrations of PRA and AngⅡ between training group and SCI group.Conclusion T5 complete SCI can induce OH in rabbits. Tilting table exercise can partially improve OH after SCI in rabbit, which may be not related to activation of the circulating renin angiotensin system.
3.Effect of Tilting Table Exercise and Shengmaiyin on Orthostatic Hypotension after Spinal Cord Injury in Rabbits

ObjectiveTo develop an experimental model of orthostatic hypotension (OH) after spinal cord injury (SCI) in rabbits and investigate the effect of tilting table exercise and Shengmaiyin on it. MethodsRabbit model of OH after SCI were induced by complete transection of spinal cord at the T5 spinal segment and raised to 60° head up tilt (HUT) on a tilting table. Tilting table exercise and Shengmaiyin were applied to different groups for 28 d from 4 days after operation. Systolic blood pressure (SBP) and pulse rate (PR) were observed with non-invasive sphygmobolometer of rabbit 3, 10, 17, 24, 31 d after the operation. Liquid chromatography with electrochemical detection(HPLC-ECD) was used to detect the concentrations of plasma norepinephrine (NE) and epinephrine(E) with horizontal position and HUT 60° on the 31st day. ResultsSBP with HUT 60° in rabbits of combined treatment group was significantly higher than that in rabbits of SCI group on the 24th day after operation,and the drop in SBP from horizontal position to HUT60° was significantly lower than that in rabbits of SCI group. SBP with HUT 60° in rabbits of exercise group and combined treatment group was significantly higher than that in rabbits of SCI group on the 31st day after operation. The drop in SBP from horizontal position to HUT 60° was significantly lower in rabbits of Shengmaiyin group, exercise group and combined treatment group than that in rabbits of SCI group on the 31st day after operation. The concentration of E and NE of SCI group, Shengmaiyin group, exercise group and combined treatment group was lower than that of sham group, but there was no significant difference among these groups. ConclusionThe treatment with tilting table exercise and Shengmaiyin can improve OH after SCI partially in rabbits without involving norepinephrine and epinephrine significantly. The period of treatment can be shorten when both therapies used together.
4.Effect of Tilting Table Exercise and Shengmaiyin on Alpha 1 Adrenogenic Receptor after Spinal Cord Injury in Rabbits

Objective To observe the effect of tilting table exercise and shengmaiyin on alpha 1 adrenogenic receptor (α1AR) in the superior mesenteric artery after spinal cord injury (SCI) in rabbits.Methods 46 rabbits were randomly divided into the sham group (n=6), SCI group (n=10), shenmaiyin group (n=10), exercise group (n=10) and combined treatment (shenmaiyin plus exercise) group (n=10). The rabbit model of SCI was established by complete transection of spinal cord at the T5 spinal segment. Tilting table exercise and shengmaiyin were applied to different groups for 28 days from 4 days after operation. The expression of α1aAR,α1bAR,α1dAR mRNA in the superior mesenteric artery was detected by RT-PCR at 31d after SCI.Results The mRNA expressions of α1aAR, α1bAR, α1dAR subtypes increased to different extent in rabbits of the shengmaiyin group, exercise group and combined treatment group compared with those of SCI group.Conclusion The role of tilting table exercise and shengmaiyin improving orthostatic hypotension after SCI in rabbits may be related to up-regulation of α1aAR, α1bAR, α1dAR mRNA in the superior mesenteric artery.
6.Golgi dispersal in irradiated cells and the protective effect of vanillin derivatives

Objective To determine the Golgi dispersal in radiation damaged cells and the protective effect of vanillin derivatives.Methods Immunofluorescence, cell cycle analysis of flow-cytometry,Western blot,and clone formation were used.Results Immunofluorescence observation showed that the Golgi dispersal caused by 2 Gy 60 Coγ-ray was significantly increased in a dose-dependent manner in the range of 4-10 Gy as was demonstrated by the fact that the Golgi area was significantly increased. When the irradiated cells were treated with the radioprotective agent VND3207, a vanillin derivative,the Golgi dispersal induced by radiation was significantly reduced.The radiation-induced Golgi dispersal was also displayed in a pattern of time-course after irradiation in the HeLa cells, and persisted at least to 36 h post-irradiation. Cell cycle test results indicated that the Golgi dispersal was not associated with the G2/M arrest triggered by radiation-induced DNA damage response.VND3207 could promote cell survival by plate colony formation assay.Conclusion The Golgi dispersal can be caused byγ-ray irradiation in a dose-and time-dependent manner, and VND3207 can provide a good protection against radiation injury associated with inhibited Golgi dispersal.
7.A case of clivus chordoma with first appearance of sudden deafness.

The patient was hospitalized for sudden hearing impairment for one day.
PHYSICAL EXAMINATION:
the blood pressure was 150/90 mm Hg, the tympanic membranes in both ears were complete and otopiesis. Audiogram showed total deafness in the right ear and slight sensorineural deafness at speech frequency and 80 db for high tone air conduction and 70 db for bone conduction at high frequency in left ear. Tympanogram showed "A" type in both ears and the ipsilateral and contralateral acoustic reflex in both ears were not induced. BAEP showed that the V wave threshold on the right was not induced and it was 50 dbnHL on the left. CT showed a limited low density area in the clivus. MRI showed a space-occupying lesion behind the basilar clivus and ahead of brain stem. Pathological examination showed CK(+), EMA(+), S-100(+) according to immunohistochemistry, which was in accordance with chondroid chordoma.

8.The role of SIRT1 in the treatment of diabetic nephropathy.

Diabetic nephropathy presents an increasing trend worldwide. It has been an attractive area to find novel targets for the treatment of diabetic nephropathy. SIRT1 (Sirtuin 1), a member of deacetylation enzymes, regulates cell senescence, metabolism, and apoptosis. In last ten years, lots of studies showed that SIRT1 exerts a protective effect in the progression of the diabetic nephropathy by promoting reconstruction of energy homeostasis, modulating cell redox state, resisting cell apoptosis, inhibiting inflammation and ameliorating renal fibrosis. SIRT1 has become a potential new target for the treatment of diabetic nephropathy.

9.Mitochondrial retrograde signaling: a new option for the treatment of mitochondrially related diseases

Mitochondria play a key role in cell metabolism. In addition to synthesizing ATP, they also participate in many physiological and pathological processes, including apoptosis, inflammation, oxidative stress, neuronal disease, tumor development, and aging. Most gene transcription of mitochondrial proteins occurs in the nucleus, so the biogenesis of mitochondria and the maintenance of mitochondrial homeostasis mainly depend on the expression of nuclear genes (nDNA) and mitochondria-nucleus interactions. Conversely, mitochondria can affect the expression of nuclear genes through nuclear transcription factors, a process called mitochondrial retrograde signaling. This review summarizes the research progress on mitochondria-nucleus retrograde signaling and its regulation, including the ways by which mitochondria regulate nuclear genes and affect biological processes, and discusses new strategies for the treatment of diseases that involve mitochondrial retrograde signaling in disease pathology.
10.Rat intestinal absorption trait of peimine and peiminine in Thunberg fritillary bulb extract.

To study the in situ intestinal absorption kinetics and compatibility influence of peimine and peiminine in rats, the absorption of peimine and peiminine in small intestine (duodenum, jejunum and ileum) and colon of rats was investigated using in situ single-pass perfusion method and the drug content was measured by HPLC-ELSD. Perfusion rate, pH, concentration of drug, gender and bile duct ligation can significantly affect the absorption of peimine and peiminine, the Ka, and Papp values in the condition of pH 6.8 and pH 7.4 had significant difference (P<0.01), as drug concentration irlcreased, the absorption parameters of peimine and peiminine decreased, Ka and Papp between low concentrations and middle concentrations was significant difference (P<0.01). Verapamil can not affect Ka and Papp of peimine and peiminine which are in the extract (P> 0.05). Bitter almonds and licorice can significantly reduce the absorption of peimine and peiminine with the usual dose (P<0.01), extracted separately and together had no significant difference on Ka and Papp (P> 0.05). Experimental results show that the absorption features of peimine and peiminine are basically the same, both of them could be absorbed at all segments of the intestine in rats and had no special absorption window, and with significant differences between male and female individuals. The absorption of peimine and peiminine complies with the active transport and facilitated diffusion in the general intestinal segments. Bitter almond and licorice can reduce the intestinal absorption rate ofpeimine and peiminine.
