1.The mechanism of protective effects of melatonin on global ischemia/reperfusion induced brain injury in rats

Objective: To study the effects of melatonin(MT) on inducible nitric oxide synthase (iNOS) expression in hippocampal CA1 at 24 hours and TDT-mediated dUPT nick end labling (TUNEL) positive cells in hippocampal CA1 at 48 hours after global ischemia(20 min)/ reperfusion in rats. Methods: MT was injected intraperitoneally at 0 h, 1 h, 2 h and 6 h after ischemia(20 min) induced by the“occlusion of four arteries”, 2.5 mg*kg-1 or 10 mg*kg-1 each time, respectively. Researching the expression of iNOS in hippocampal CA1 with the immunocytochemistry method 24 hours after reperfusion and the number of TUNEL positive cells in hippocampal CA1 48 hours after reperfusion utilizing TUNEL. Results: At these doses, MT could decrease iNOS expression in hippocampal CA1 at 24 h after global ischemia (20 min)/reperfusion and the number of TUNEL cells in hippocampal CA1 at 48h after reperfusion in rats. Conclusion: Decreasing the expression of deleterious iNOS maybe one of the mechanisms involved in protective action of MT on ischemia-vulnerable brain region.
2.Preparation of medical polypropylene anti-bacterial mesh coated with triclosan and detection of its nature

Objective To explore the production of medical polypropylene anti-bacterial mesh coated with triclosan,and study the properties of its controlled-release drugs,as well as the efficiency of the mesh coated with triclosan.Methods A medical polypropylene anti-bacterial mesh coated with triclosan was produced by the method of solvent evaporation,with triclosan as antibacterial agent and ethylene-vinyl alcohol copolymer as carrier.The meshes were divided into four groups: A,B,C and D.The meshes in A,B and C(experimental) groups were coated with triclosan,and D group was control group.The ultrastructure of meshes of the 4 groups was observed under electron microscope.The slip diffusion method was used to observe the diameter of bacteriostasis-rings in different time periods.At the same time,we observed the maximum tensile strength of the mesh in transverse and longitudinal axis when they were pulled off before and after the experiment.Results The surfaces of B group were uniform and smooth,and the coating could continuously release drugs during 7 days in sodium chloride.The diameter of bacteriostasis-rings in B group was significantly larger than that in control group(P
4.Expression, purification and bioactivity of a recombinant fusion protein consisting of anti-HBs single chain Fv and interleukin-2

Objective To study the expression, purification and bioactivity of a recombinant fusion protein consisting of anti-HBs single chain Fv and interleukin-2. Methods The engineering bacterium M15[pQE-ScFv-IL-2] which can express the fusion protein consisting of anti-HBsAg single chain Fv and interleukin-2, was induced by IPTG, then a 43kD recombinant protein was identified by SDS-PAGE and Western-blot analysis. Results The ratio of target protein to total protein of host reached 18%. Further analysis confirmed that the recombinant protein formed inclusion body in the cytoplasm of bacteria. 95% purity could be achieved after two-step purification of ScFv-IL-2, including Ni metal chelating chromatography (the first) and ion-exchange chromatogram (the second). The bioactivity assay of the purified product showed that the antibody-cytokine fusion protein could bind to HBsAg specifically and stimulate the proliferation of CTLL-2. Conclusion These results suggest that the fusion protein retains the bioactivity of its parental molecules, and may be a potential gene-engineering targeting drug for the treatment of chronic hepatitis B and other relevant diseases.
5.Experimental Study of Red Blood Cells after Cold Storage at -80℃

The changes in shape, structure and function of red blood cells (RBC) after cold storage were observed by using slow cold storage technique. Before and after cold storage, RBC mean corpuscular volume (MCV), RBC fragility (RF), whole blood viscosity (WBV), RBC deformity (RBCD), LFU, SatO2, hematocrit (HCT), RBC recovery rate (RBCRR) and RBC immune function were measured. The result showed that after resuscitation, RBC had vital resistance to hypotonic solution. There was no significant difference in MCV, RBCD and LFU (P>0.05). HCT and WBV were decreased and RBCRR reached 84.1 %(all P<0.01). SatO2 was increased from 30.5 % to 98.2 %. RBC immune function showed no significant difference (P>0.05). It was suggested that quality of RBC didn't change after cold storage and RBC could be cryopreserved for a long time.
6.Pharmaceutical Care in Disaster Assistance in Large Earthquake

OBJECTIVE:To provide information for carrying out pharmaceutical care for future emergency relief work. METHODS: Based on our own experiences,the pharmaceutical care-related work involved in the medical relief in "May?12" Wenchuan earthquake was investigated and analyzed summarily. RESULTS: Scientific emergency plan could create order for the busy relief work,and creative work could make up the deficiency of the plan. It is advisable to attach great importance to the role of the clinical pharmacists in the earthquake relief work. Good command and powerful measures contribute to the smooth proceeding of emergency medical rescue. CONCLUSION: The pharmaceutical care plays an important role in the emergency medical rescue.
7.Application of Bayesian Mixed Treatment Comparison Method in Pharmacoeconomics Evaluation

OBJECTIVE:To explore the basic principles,advantages and application prospects of Bayesian mixed treatment comparison (MTC),and provide theoretic support for making reasonable decisions of pharmacoeconomics. METHODS:The prob-lems existing in model and method that had been used in the study of pharmacoeconomics as starting point,literature research was used to sort out the domestic development and applications of MTC and study its application and development prospect. RESULTS &CONCLUSIONS:The evaluation methods of pharmacoeconomics play an important role in selection of essential medicines,medi-care insurance directory medicines,basic health and basic public health programs,the results directly affect the total effective rate of medical resources’configuration. The current research models cannot effectively solve the problem,for example,decision tree model is difficult to make evaluation of long-term treatment;Markov model and the use of multiple attribute utility theory are restricted by the model parameters;discrete event simulation is limited because it depends on the epidemiological study and clinical trial. MTC, an extension approach of the traditional Meta-analysis,is an effective new method developed recently to analyze and compare the di-rect or indirect evidence of several different clinical treatment factors in order to make a comprehensive evaluation of the various inter-ventions in the absence of evidence support. So the MTC can make up for the limitation of current evaluation methods.

9.The phenotype changes of SGC7901 gastric cancer cell-dendritic cell fusion vaccine

Objective To obtain SGC7901 gastric cancer cell-dendritic cell fusion vaccines by cell fusion technology. The phenotype changes of the fusion cells were detected to provide experimental data for the following research work. Methods The SGC7901 gastric cancer cells and dendritic cells were induced to be fused by PEG and the pure fusion cells were obtained by selective culture with the HAT/HT culture system. The phenotype changes of the fusion cells were detected by flow cytometer. Results The fusion cells had special morphologic characteristic and the phenotypes of the fusion cells were remarkably higher than the parental dendritic cells. Conclusion The SGC7901 gastric cancer cell-dendritic cell fusion cells kept special morphologic characteristic and their phenotypes were significantly higher than the parental dendritic cells, which may be the cellular base for their stronger biological effects.
10.Cytotoxic effects of gastric cancer cell-DC fusion vaccines

Objective To study on the cytotoxic effects in vivo and in vitro of the T lymphocytes which were activated by the DC-gastric cancer cell fusion vaccines. Methods The mononuclear cells were separated from the peripheral blood of gastric cancer patients and co-cultured with granulocyte-macrophage colony stimulating factors, interleukin-4 and tumor necrosis factor-? to generate mature dendritic cells. The human gastric cancer SGC7901 cells and the dendritic cells were fusioned by using polyethylene glycol, and the pure fusion cells were screened out by culturing with HAT and HT selective culture systems. The mixture of SGC7901 and DC without fusion and the T lymphocytes served as controls. The ability of fusion cells activated by T lymphocytes to kill SGC7901 gastric cancer cells was investigated by MTT in vitro. The anti-tumor effects were evalutated by detecting the growth of the new planted tumors in the nude mice, the apoptosis and proliferation of the planted cancer cells, the pathological changes of the tumor tissues and the prohibitive rate for planted tumors before and after the application of the fusion cell vaccine. Results The mature dendritic cells were obtained from peripheral blood mononuclear cells of gastric cancer patients successfully. Dendritic cells and SGC7901 cells were fusioned and the pure fusion cells were got.
