1.Research progress on tumor-associated macrophage in gastric cancer

It has been confirmed that tumor invasion and metastasis is not only dependent on the charac -teristics of the tumor ,but also dependent on the tumor microenvironment .Tumor-associated macrophage ( TAM) , which constitutes a large number of the tumor -infiltrating immune cells,is stimulated into M1 macrophage and M2 macrophage by different signals in the tumor microenvironment .Whereas the vast majority of studies with nu-merous tumor types show that the presence of M 2 phenotypes in the tumor microenvironment is associated with a worse prognosis ,most studies in gastric cancer also claim the same results .Hypoxia and coagulation factors may both contribute to it .It has identified that the function of TAM could be changed with their location of tumor ,but it has nothing to do with cell differentiation and cell type of gastric cancer .In this review,we summarize the differ-ent and controversial effects of TAM in gastric cancer .
2.Intravenous transplantation of GDNF gene-modified CD34~+ cells from human umbilical cord blood for cerebral ischemie-reperfusion inury in rats

Objective To observe the neurologicaI functional recovery after intravenous transplantation of human umbilical cord blood(HUCB)CD34~+ cells transfected with glial cell-derived neurotrophic factor (GDNF) and green fluorescent protein (GFP) in SD rats with middle cerebral artery occlusion (MCAO).and tO investigate the survival,migration and neural differentiation of the graft cells.Methods (1)CD34~+ cells were isolated from HUCB using centrifuge combined with immune beads and then identified by flow cytometry,and transfected by the recombinant plasmid of GDNF.GFP or GFP plasmid by liposome method.(2) Sixty aduh male SD rats with MCAO were randomly divided into three groups (n=20 in each group):GDNF-GFP-CD34~+ cells group,in which the GDNF-GFP-CD34~+ cells were transplanted intravenously at the 24th h after the establishment ofmodels of MCAO;GFP-CD34~+ cells group:in which the GFP-CD34~+ cells were transplanted intravenously at the Same time;Normal saline group,in which normal saline was injected at the same time.Fifteen SD rats served as sham-operated group.(3) Neurological functional measurements were performed using the modified neurolc'gical severity score.Quantitative histological determinations of infarct volume were performed using standard TTC staining and quantitative image analysis. The GDNF level in the cell culture or the cerebral tissue was measured by ELISA. Meanwhile, the survival and migration of GFP-labeled CD34~+ cells and the expression of astrocytie marker-GFAP and the neuron marker-neuronal nuclei (NeuN) were detected by immunohistochemical and fluorescent staining. Results (1) The GDNF level in the cell culture was significantly higher in GDNF-GFP-CD34~+ cells than in GFP-CD34~+ cells (P<0. 05). (2) No significant difference was found in the modified neurological severity score at the day 7 after transplantation among the three groups with MACO, but at day the 28 after transplantation, the neurological function in GFP-GDNF-CD34~+ group was improved significantly (5.0±1. O) as compared with the GFP-CD34~+ cells (5. 9 + 1.4) or saline groups (7. 0±1.7) (P<0. 05). The cerebral infarct volume in GFP-GDNF-CD34~+ group (142±44mm~3) was significantly decreased as compared with the GFP-CD34~+ group (196±58 mm~3) (P<0. 05) or saline group (233<50 mm~3 ) (P<0. 01 ). The GDNF level in cerebral tissue in GFP-GDNF-CD34~+ group was significantly increased as compared with the GFP-CD34~+ group (P<0. 05) at the 28th day after treatment. At the 28th day after treatment, the NeuN positive cells (6.7±2.0), and GFAP positive ceils (14.1±3.3) in GFP-GDNF-CD34~+ group were significantly increased as compared with the GFP-CD34~+ group (P<0. 05), but there were no positive cells in sham-operationgroup. Conclusion Intravenous transplantation of GDNF gene-modified CD34~+ cells from human umbilical cord blood could improve the neurological function in rats with MCAO. The increased GDNF level in cerebral tissue was one of possible mechanisms responsible for the different improvements.

4.Analysis of Ifected Instances By Hp of Civil Servant in Beijing

Objective To observe the infected instances of civil servant by Hp in Beijing and study the relationship between Hp and age,sex.Methods 3275 people were divided into four groups based on age:yong group,midlife group,pre-agedness group and agedness group;each group based on sex were divided into two groups.Every one was detected by 13C-urea breath test.If the resalt is over four,it is positive,otherwise it is negative.Measure dates were analised by statistical software.The infecting rate of Hp is denoted by persent,Chi square test was used in the every group.Results The mean infecting rate of Hp was 41.47%.The women in midlife group was 46.02%,it is the highest.The men over 75 in agedness group was 38.48%,it is the lowest.The infecting persent of yong group was higher than that of agedness group.The persent of midlife group and pre-agedness group was meaning result.The difference in age and sex had no statistical meaning.Conclusion The infecting rate of civil servant by Hp in Beijing is obviously lower than of people in Beijing,which is no relationship with sex and age.
5.Correlation between the alleles polymorphism of CYP2A6 and CYP2B6 and blood concentration of Valproate sodium

Objective To investigate the correlation between the alleles polymorphism of CYP2A6 and CYP2B6 and blood concentration of Valproate sodium(VPA).Methods The 165 epilepsy patients received simplex VPA and without dysfunction of liver and kidney were chose in this study.The alleles polymorphism of CYP2A6 in 95 cases and CYP2B6 in 70 cases were detected by polymerase chain reaction(PCR).Fluorescence polarization immunoassay(FPIA) was used to measure the blood concentration of VPA.Results The frequence of allele CYP2A6*4 in 95 cases was 13.2%.The blood concentration of VPA in patients with allele CYP2A6*4[(4.23?0.27)mg/ml]was significantly higher than that in patients without allele CYP2A6*4[(3.35?0.38)mg/ml](P

7.Helicobacter pylori infection induces cyclooxygenase-2 expression in gastric mucosa

Objective Adenocarcinoma of the stomach has been established as a consequence of gastric infection with Helicobacter pylori(H.pylori).The specific role of H.pylori in the pathogenesis is unknown yet.Recent studies indicate that expression of mitogen inducible cyclooxygenase-2(COX-2) occurs in gastrointestinal tumors.The purpose of the present study was to investigate expression of COX-2 protein in the human stomach with or without H.pylori infection.Methods Twenty-seven subjects who were asymptomatic referred to the hospital for healthy examination including endoscopic screening.Biopsy specimens were obtained from the subjects without any macroscopic lesions,such as peptic ulcer or gastric malignancies.H.pylori infection was determined by rapid urease test(CLO test),bacterial culture and histology(Giemsa staining).Expression of COX-2 was evaluated by immunohistochemistry using the avidin-biotin-peroxide complex(ABC) method The association between COX-2 expression and H.Pylori infection was assessed by Fisher's exact test.A P value less than 0.05 was considered to be statistically significant.Results specific immunostaining for COX-2 was observed in antral mucosa of 18 subjects infected with H.pylori COX-2 was expressed in gastric mucosal epithelia,mainly in the foveolar epithelial cells.Furthermore,COX-2 was also observed in the neck cells of the gastric glands and inflammatory mononuclear cells beneath the mucosal epithelia.Expression of COX-2 was never found in the gastric mucosa of H.pylori-negative subjects.A positive association of H.pylori infection with COX-2 expression was statistically significant(P
8.Analysis of results of sample surveys of blood for clinical use in Hunan Province from 1997 to 2001

Objective To strengthen the control of blood quality, ensure the safety of blood for clinical use and effectively contain the occurrence of transfusion transmitted disease. Methods Using the method of random sample survey, appraisal was made once again of the blood for clinical use in the blood stations of 14 cities(regions) of Hunan Province in 1997~2001, blood that had passed reexamination by relevant blood collecting and supplying institutions. The appraisal, conducted in the proportion of 10% of the yearly use of blood, focused on 5 indexes, including HBsAg, Anti HCV, Anti HIV, RPR, and ALT. Results 6 211 blood samples were checked, with the total positive rate being 7.89‰. Among theses, the positive rate of samples sent for testing was 7.78‰, the positive rate of samples collected and tested on the spot was 9.35‰, the positive rate of HBsAg was 2.59‰, the positive rate of Anti HCV was 3.86‰, the positive rate of Anti HIV after primary screening was 0.64‰(ascertained to be negative through Western Blot testing), the rate of ALT≥25 IU was 0.81‰, and the testing of syphilis was all negative. Conclusion Serious problems remain to be solved with regard to the safety of blood for clinical use.
9.Analysis of indocyanine green angiography and fundus fluorescein angiography for high myopia

-6.00D), selected randomly and consecutively, were examined by ICGA and FFA synchronically.ResultsThe result of early phase of FFA showed hypofluorescence of the background in 25 eyes,while of late phase showed subretinal neovascularization (SRNV) in 10 eyes and streak formation in 40 eyes. The result of ICGA showed choroidal retrobulbar arteries in 8 eyes, hypofluorescence of the background in 35 eyes, SRNV in 8 eyes, and streak formation in 52 eyes.ConclusionThe iconographic characteristics of ICGA and FFA of high myopia include hypofluorescence of the background, SRNV and streak formation. ICGA can give more exact information on the lesions of choroid in high myopia. The synchronic examination of ICGA and FFA may act as a guide to the therapy for high myopia.
10.Recent advances in DNA vaccines against allergic airway disease: a review.

DNA vaccine is used in infectious diseases initially, and later is applied in neoplastic diseases, allergic diseases and other fields with the further understanding of DNA vaccine and the development of genetic engineering. DNA vaccine transfers the genes encoding exogenous antigens to plasmid vector and then is introduced into organism. It controls the antigen proteins synthesis, thus induces specific humoral and cellular immune responses. So it has a broad application prospect in allergic diseases. Compared with the traditional protein vaccines used in specific immunotherapy, DNA vaccine has many advantages, including high purity and specificity, and improvement of patients' compliance etc. However, there are still two unsolved problems. First, the transfection rate of unmodified naked DNA plasmid is not high, Second, it's difficult to induce ideal immune response. In this study, we will review the progress of DNA vaccine applications in respiratory allergic diseases and its various optimization strategies.
