1.Preventive effects of ginseng fiber on hepatic fibrosis induced by bone loss in mice

AIM: To study the relationship between hepatic fibrosis and osteoporosis, and observe the preventive effects of ginseng fiber on bone loss. METHODS: Forty PCR mice were randomly divided into 4 groups. The mice in control group were treated by daily oral gavage with vehicle. The mice in other three groups were given SC injection of 40% CCl_4 10 ml?kg -1 and treated by either daily oral gavage with vehicle, colchicine, or ginsen hair for 42 d. The liver injury indexes were measured and the mineral elements and hydroxyproline of femur were determined. RESULTS: Compared with the control group, the serum enzyme activity of alanine aminotransferase (ALT), aspartic acid aminotransferase (AST) markedly increased and serum albumin (Alb) and A/G distinctly decreased in CCl_4 group whose liver slides also showed typical liver cirrhosis. The dried weight of femur markedly reduced and the bone calcium content and bone hydroxyproline content significantly decreased in CCl_4 group. Bone copper and bone magnesium increased in CCl_4 group. Ginseng fiber markedly decreased the serum enzyme activity of ALT and increased the bone calcium content and bone hydroxyproline content. The preventive effects of ginseng fiber was similar to that of colchicine. CONCLUSION: The bone mass is lost in mice with chronic hepatic injury induced by CCl_4. Ginseng fiber can prevent bone loss and hepatic fibrosis induced by CCl_4 in mice.
2.Grey correlative analysis of bone volume and factors in ovariectomized rats

AIM: To study the factors that affect bone volume in ovariectomized rats. METHODS: The bone volume and factors were analyzed by the grey system theory method in ovariectomized rats. RESULTS: Serum estradiol was the most important factor for the bone volume, followed by the bone contents of calcium, phosphorus and magnesium. Serum calcium, phosphorus and the bone contents of hydroxyproline were the less important factors for the bone volume. CONCLUSION: Serum estradiol and bone contents of calcium are the most important factors that affect bone volume in ovariectomized rats.KEY WOLDS grey correlative analysis; bone volume; factors; ovariectomized rats; osteoporosis

METHODS: Forty PCR Mice were randomly divided into 4 the control group, the NS group, the colchicine group and the Ginseng fiber groups. Rats in control group were treated by daily oral gavage with vehicle. Rats in other three groups were given SC injection of 40% CCl 4 10 ml?kg -1 and treated by either daily oral gavage with vehicle, or colchicine at 0.1 ml?kg -1, or Ginseng fiber at 10 g?kg -1 for 42 d. The liver injury indexes were measured. RESULTS: Compared with control group, the serum enzymes of alanine aminotransferase (ALT), aspartic acid aminotransferase(AST) were markedly increased but serum albumin (Alb) and A/G were decreased distinctly in CCl 4 group whose liver slides also showed typical liver cirrhosis. Ginseng fiber markedly prevented CCl 4-induced increases in liver weight, serum ALT and TP. Ginseng fiber lightened the hepatic pathological necrosis resulting from CCl 4. The preventive effect of Ginseng fiber was identical to that of colchicine. CONCLUSION: Ginseng fiber can prevent hepatic fibrosis induced by CCl 4 in mice.
4.Application of ROC curve for evaluating risk evaluation value of serum LDH, BXTM, β2-MG,CRP and DD in bone marrow infiltration of non-Hodgkin's lymphoma

Objective To investigate the relationship between the indexes change of lactate dehydrogenase(LDH),serum tumor associated material(BXTM),β2-microglobulin(β2-MG),C-reactive protein(CRP) and D-dimer(DD) with lymphoma cell bone marrow infiltration(BMI) degree in the patients with non-Hodgkin's lymphoma(NHL).Methods Seventy-four cases of initially treated NHL were retrospectively analyzed and divided into the bone marrow invasion group(BMI,26 cases),lymphoma cell leukemia group(LMCL,31 cases) and non-BMI group(N-BMI,17 cases).The related indexes levels in each group were detected.ROC curve was drawn and analyzed.Results The levels of LDH,β2-MG,CRP and DD levels in the BMI group and LMCL group were higher than those in the N-BMI group(P<0.05).Based on the ROC curve analysis,the optimal diagnostic cut-off points of each index in BMI were determined.The CRP and LDH had good sensitivity(100.00 %),specificity(80.00 %-100.00 %) and diagnostic accuracy(80.00 % 100.00%).In the LMCL group,the diagnostic accuracy of DD,BXTM and β2-MG was 63.30 %-100.00%.Conclusion The changes of LDH,CRP,β2-MG and DD in the patients with NHL are consistent with the BMI severity of lymphoma cells,which has a good clinical value in staging,curative effect judgment and prognosis.
5.Changes of bone in female Sprague-Dawley rats aged 4.5 and 7.5 months

AIM: To observe the changes of bone in female Sprague Dawley rats aged 4.5 and 7.5 months. METHODS: Forty 4 month old virgin female Sprague Dawley rats were randomly divided into 4.5 month group and 7.5 month group. Bone histomorphometric analysis of the proximal tibial metaphysis (PTM), tibial shaft (Tx) and the fifth lumbar vertebral body (LV5) was performed in undecalcified sections. RESULTS: There was no significant change in bone volume of PTM, LV5 and Tx between 4.5 and 7.5 months of age. However, the bone formation parameters (%L.Pm, MAR, BFR/TV, BFR/BV, BFR/BS) of LV5 and Tx fall rapidly between 4.5 and 7.5 months of age. CONCLUSION: There is no significant change in cancellous and cortical bone mass, but the bone formation of LV5 and Tx decreases in female Sprague Dawley rats aged from 4.5 to 7.5 months.

Objective To approach on the expression pattern of MMP-2 and E-Cadherin in the effects of gastrin on gastric canc-er cells ,so as to investigate their role in the metastasis and invasion of gastric cancer .Methods Genomic DNA modified by sodium bisulfite was used as a template .RQ-MSP detect the methylation status of the gene promoter .The relative amount of Methylation status was performed with comparative threshold cycle (2- △ t ) method .Results MMP-2 and E-Cadherin genes in gastric cancer cells were at hemimethylated status in gastrin before and after the intervention .Gastrin can reduce the methylation levels of MMP-2 gene(P<0 .05) ,but the could be effected by proglumide receptor antagonist to some certain extent (P>0 .05) .Gastrin can pro-mote the methylation levels of MMP-2 gene ,but proglumide decrease that effect (P<0 .05) .Conclusion Gastrin plays a certain role in the biological behavior of gastric cancer impact by changeing on the expression pattern of MMP-2 and E-Cadherin in the effects of gastrin on gastric cancer cells .

Objective To study the anti-HCV-IgG antibody and serum HCV-RNA value in the diagnosis of hepatitis c.Methods From December 2015 to December 2006,76 patients with hepatitis C were enrolled in this study.All patients were tested for serum anti-HCV-IgG antibody and serum HCV-RNA,and the positive detection rate of hepatitis C was detected by two methods alone.Results 76 specimens of anti-HCV IgG antibody testing positive rate was 27.63%,the detection of serum HCV-RNA positive rate was 25.00%,the two kinds of joint detection positive rate was 22.37%,the three methods positive rate were relatively close,but there was no significant difference(P>0.05).According to different loads interval differentiate HCV-RNA virus detection rate of rising,anti-HCV-IgG five interval were 60.00 %,83.33 %,100.00 %,100.00 %,100.00 %.Conclusion The detection of single anti-HCV IgG antibody or serum HCV RNA in the diagnosis of hepatitis c have limitations,the joint detection can effectively reduce the missed diagnosis and improve positive rate and provide a reliable basis for clinical diagnosis of hepatitis C.
9.Levels of serum FOXO1 and ATG7 in elderly patients with severe pneumonia and their relationship with short-term prognosis

Objective To explore the levels of serum forkhead transcription factor 1(FOXO1)and autoph-agy-related gene 7(ATG7)in elderly patients with severe pneumonia and their relationship with short-term prognosis.Methods A total of 122 elderly patients with severe pneumonia admitted to the hospital from Au-gust 2020 to August 2022 were selected as the case group,and 105 elderly people who came to the hospital for physical examination during the same period were selected as the control group.The serum levels of FOXO1 and ATG7 were detected by enzyme-linked immunosorbent assay(ELISA).The patients were divided into the survival group(n=91)and the death group(n=31)according to whether they died within 28 days after the diagnosis of severe pneumonia.Receiver operating characteristic(ROC)curve was used to evaluate the predic-tive value of serum FOXO1 and ATG7 levels for short-term mortality in elderly patients with severe pneumo-nia.The influencing factors for short-term mortality in elderly patients with severe pneumonia were explored by multivariate Logistic regression analysis.Results The serum FOXO1 and ATG7 levels in the case group were higher than those in the control group,and the differences were statistically significant(t=7.615,29.591,P<0.05).The serum expression levels of FOXO1 and ATG7 in the death group were higher than those in the survival group,and the differences were statistically significant(t=10.029,9.399,P<0.05).The area under the curve(AUC)of serum FOXO1 and ATG7 for predicting short-term mortality in elderly pa-tients with severe pneumonia was 0.733(95%CI:0.673-0.799)and 0.826(95%CI:0.756-0.893),respec-tively,and the AUC of FOXO1 combined with ATG7 for predicting short-term mortality in elderly patients with severe pneumonia was 0.908(95%CI:0.862-0.949).FOXO1 and ATG7 were positively correlated with acute physiology chronic health evaluation(APACHE Ⅱ)score(r=0.693,0.627,P<0.05).The results of multivariate Logistic regression showed that APACHE Ⅱ score ≥22.56(OR=3.589,95%CI:1.714-7.515),FOXO1 ≥10.67 ng/mL(OR=2.529,95%CI:1.232-5.193),ATG7 ≥16.35 pg/mL(OR=2.875,95%CI:1.414-5.845)were risk factors for death in elderly patients with severe pneumonia(P<0.05).Con-clusion Serum levels of FOXO1 and ATG7 are significantly increased in elderly patients with severe pneumo-nia,and both could be used to evaluate the short-term prognosis of elderly patients with severe pneumonia.
