1.Use of zebrafish models for the research of traditional Chinese medicine.

Traditional Chinese medicine (TCM) exhibits a broad range of effects on biological activity that is probably due to interactions of complex chemical constituents with multiple targets in the body. Understanding the active chemical constituents in TCM is very important in providing rationales for the clinical usage of TCM. The zebrafish (Danio rerio) has recently become a popular model in the field of drug screening, specifically emerging as an important vertebrate model for in vivo high-content drug screening of multiple efficacy parameters and whole-organism toxicity. The authors also discussed the advantages of the zebrafish model for evaluating drug metabolism. Zebrafish usage in TCM screening should be a viable approach that helps identify active chemical markers, biological pathways and mechanistic actions of TCM.
2.Glycogen synthase kinase 3β and leukemia

Glycogen synthase kinase 3β (GSK3 β) is a conserved cytoplasmic serine/threonine protein kinase.It can not only negatively regulate the classical Wnt and Akt signal pathways which are associated with leukemia,but also positively regulate the NF-κB,non-classical Wnt and lysosomal apoptosis signal pathways,thereby regulating the proliferation,differentiation and apoptosis of leukemia cells.At present,a variety of GSK.3 β inhibitors and activators have been reported in the researches of leukemia,which play important roles in reducing the adverse reactions of chemotherapy,reversing multidrug resistance and enhancing killing effect to leukemia cells.GSK3β is expected to become a target of leukemia molecular targeted therapy.
3.ESTABLISHMENT OF XENOGRAFT MODEL OF HUMAN PROMYELOCYTIC LEUKEMIA MUTANT CELLS (HL-60-AR) IN NUDE MICE

The present study is designed to establish a xenograft model of human promyelocytic leukemia cell mutant (HL-60-AR) deficient in hypoxanthine guanine phosphoribosyltransferase (HGPRT) in nude mice. A solid leukemia sarcoma developed after subcutaneous inoculation with HL-60-AR cells. Comparative studies of HL-60-AR/Nu tumor cells in nude mice and cultured HL-60-AR cells in vitro revealed virtual identity as shown by light microscopic morphology, ultrastructure of cell, cytochemistry, chromosome analysis, LDH isoenzyme pattern, genetic markers and differentiated characters assay. Up to now, twelve generations haw been transmitted in rive by inoculating with the solid tumor Cells developed in nude mice. This nude mice model in which human leukemia cells grew could be considered as a useful model for in rive studies of human leukemic cells proliferation, differentiation and the screening for anti-leukemia drugs.
4.Differential gene expression pattern between autosomal dominant polycystic kidney tissue and normal kidney tissue: a DNA microarray study

Objective:To study the differential gene expression pattern between autosomal dominant polycystic and normal kidney tissue,and to deduce the etiological factor and treatment for autosomal dominant polycystic kidney disease(ADPKD).Methods: The cDNA probes were prepared by labelling normal kidney tissue mRNA and ADPKD tissue mRNA with Cy3-dUTP and Cy5-dUTP separately through reverse transcription.The probes were then hybridized with the DNA microarrays(PCR products of(4 096) human cDNAs onto specially treated glass slides) and the fluorescent signals were scanned with ScanArray 4000 scanner.Semi-quantitive RT-PCR was performed to test the expression level of 4 related genes.Results: Of the 4 096 genes screened,463 genes showed obvious changes.Expression of 206 genes was upregulated in the polycystic kidney tissue,especially cyclin D_(2),MMPs,TIMP1 and fibroblast activation protein;expression of 257 genes was downregulated,especially phosphatase 1A and acid phosphatase 1.The expression of genes tested by RT-PCR was in accordance with those detected by cDNA microarray.Conclusion: ADPKD may be related to the upregulation of cyclin,MMPs,and various kinds of growth factors,and drugs like inhibitors of CaM and MMPs might have therapeutical effects on ADPKD.
5.Effects of fructose-1,6-diphosphate on preservation of isolated rat heart

Objective To evaluate the effects of fructose-1,6-diphosphate (FDP) on the preservation and high-energy phosphate metabolism of hypothermic SD rat hearts. Methods Sixteen SD rats were randomly divided into 2 groups. Their hearts were harvested and preserved in UW solution at 4℃ for 12 hours. In the study group (n=8), FDP (5mmol/L) was added to the UW solution, whereas in the control group, FDP was not added. Langendorff perfusion was established with the isolated hearts, and cardiac functions were examined, as well as the activities of lactate dehydrogenase (LDH) and creatine kinase (CK) in the perfusion fluid collected from coronary. Then the high-energy phosphate concentration, ATP, ADP, and AMP contents in the myocardium were determined with HPLC. 0.05).The acute rejection rates in non-dialysis group was significantly lower than that of dialysis group(19.53% vs 34.27%,P0.05).The 1 year survival rates of patient/graft were 98.44% in non-dialysis and(97.72%/)95.96% in dialysis group(no significance);the 3 year survival rates of patient/graft were 96.23% in nondialysis and(94.4%/88%) in dialysis group(no significance).Conclusion:Renal transplantation without dialysis can avoid the dialysis complications and transfusion-induced sensitization,and reduce the risk of hepatitis infection as well as the acute rejection rate;while its patient/graft survival rate is similar to that of dialysis renal transplantation,making it feasible for clinical application.
7.The Implementation and Generalization of Matching between Computerized Physician Order Entry and Charging Items

Methods: Standard data of computerized physician order has been established, in which physician order items and charging items have been matched. It has effectively solved the problems in the process of computerized physician order entry. Results and Conclusion: The implementation of matching computerized physician and costs has effectively controlled the errors and missing of costs, which released the complaint of patients and received the good reputation and welcome, the compliance of doctors using computerized physician order entry has been obviously improved, meanwhile, it has provided new application experiences for implementing the following charging of national medical services prices in hospitals.
8.Experimental study of elbow joint stability following ulna coronoid process fractures

Objective To evalvate the influence of elbow joint stability following ulna coronoid process fractures. Methods Three cadaveric upper extremities were used. The height, width, thickness, diameter and opening angle of ulna coronoid process were measured in six arms, while the coronoid process fragments at the level of 2 mm, quarter, half, three quarter and whole height were resected consecutively, the changes of the range of the elbow joint movement were recorded. Another sixteen specimens were divided into two groups: the anterior bundle of the ulnar collateral ligaments (UCL) in one group was unrepaired; While the other repaired, then the angle of cubitus valgus between two groups was compared. Results If the osteotomy bone was within quarter of ulna coronoid process, the elbow joint function was good. However, in cases of osteotomy on half of coronoid process, the elbow extension function was limited and the elbow became unstable. After reduction with internal fixation, the angle of cubitus valgus was greater in the second group, and the difference of elbow joint in flexion 0?, 30?, 60?, 90? and 120? had statistical significance(P
9.Clinical study on renal retransplantation

Objective To summarize the experiences of kidney retransplantation from 1978 to Dec. 2002.Methods The clinical data of 86 cases subject to first renal transplantation and 86 cases to second renal transplantation were retrospectively analyzed. The main causes of failure of the first renal transplantation were CAN, while those of the second renal transplantation were HAR, and other complications such as AR, graft rupture and serious graft T.B. PRA or CDC in 31 cases was positive before retransplantation. The clinical data such as antibody-inducing therapy, the protocols of immunosuppression and the survival rates of patients and grafts in these patients were analyzed.Results Survival rates at 1, 3, 5 year of the patient/graft of retransplantation group were 84.8 %/61.6 %, 79.1 %/45.3 % and 58.1 %/41.9 % respectively, while those at 1, 3, 5 years of the patient/graft with the first renal transplantation were 89.5 %/79.1 %, 81.4 %/74.4 % and 67.4 %/58.1 % respectively. 10.Analysis on the risk factors for delayed graft function in cadaveic kidney transplantation

Objective To investigate the risk factors for delayed graft function (DGF) under-(going) cadaver renal transplantation.Methods Twenty-four relative variables were analyzed by SPSS11.0 statistical software. Using chi-square analysis method, the influence of these variables on DGF was retrospectively studied. Logistic regression analysis was done to assess and rank the relative risk of potential variables.Results The overall incidence of DGF was 5.01 %. Chi-square analysis (revealed) that the risk factors for DGF were identified as: cold ischaemia time, warm ischaemia time, recipient hypovolaemia, number of previous transplants, allosensitisation of the recipient, ATN, (accelerated) rejection, cyclosporine nephrotoxicity, ureteral leakage obstruction. Logistic multivariate analysis showed that there were 8 risk factors contributing to DGF.Conclusion DGF is a common (allograft) complication in the immediate posttransplant. There are many risk factors contributing to this condition. The basic change is ischemia-reperfusion injury. It is very important to comprehend the risk (factors) thoroughly in order to prevent them effectively.
