1.Research on improving the surgery skills of the interns of clinical medicine

Objective To research the value of using constant circling individual training program to improve surgery clinical skills of clinical medical specialty interns.Method 42 interns were divided into 3 groups:A(excellent),B(medium) and C(poor) according to their scores in entrance clinical skill test and each group would perform respective training program including conventional training,unified intensive training and intensive training in person.Their performance would be scored six times during the whole process of training.Results Compared with the final scores with beginning scores,the number of students scored A increased significantly meanwhile the number of students scored C decreased obviously.Conclusion The constant circling individual training program combined with effective unified test can benefit improving the surgery clinical skills of the interns of clinical medicine.
2.The influence of deep sea water on wound healing of mice

Objective This experiment aimed to study the influence of deep sea water (DSW) on wound healing of mice .Methods 24 Mice were randomly divided into two groups :group DSW(n=12) and group sterilize tap water(STW)(n=12) ,freely feeding for 14 days respectively ,and calculated the amount of food and water .On the 15th day ,1 cm × 1 cm size of wound was established on the back area of mice ,and continued to feed with DSW and STW respectively .Tracking the wound healing rate .Specimen was taken in the edge of wound tissue on postoperative 3 ,5 ,7 days ,then observed histopathological changes .Results Compared group DSW with group STW ,there was no significant difference in the total amount of food and water .5 days after the formation of wounds ,the wound healing rate of group DSW was significantly higher than group STW .Histological observation :compared with group STW ,vascular endothelial cells and new capillaries of the group DSW was increased ,and group DSW had less inflammatory cell and more fibroblast cells proliferation .Conclusion deep sea water can promote wound healing .
3.Psychological rehabilitation of paraplegic patients with spinal fracture

Objective To study the effects of psychological rehabilitation in the treatment of paraplegic patients with spinal cord injury (SCI).Methods A hundred paraplegic spinal fracture survivors were divided into a research group and a control group with 50 cases in each.Systematic rehabilitation was given to the patients in the control group,while this was combined with individualized psychological rehabilitation protocols for the patients in the research group.The daily treatment lasted 10 weeks,6 days a week.All of the patients were assessed with the Barthel index (BI),a functional independence measure (FIM),the Hamilton depression scale (HAMD) and the Hamilton anxiety scale (HAMA) pre-treatment and post-treatment.Results Before the intervention there was no significant difference between the 2 groups in any of the assessments.After ten weeks the average BI,FIM,HAMD and HAMA results in the research group were significantly better than those pre-treatment and also significantly better than those in the control group.Conclusions Psychological rehabilitation can distinctly improve the functioning and psychological state of paraplegic patients after SCI.
4.The anti-cancer effects of physiological deep-sea water combined with hyperthermia for hepatocellular carcinoma in vitro

Objective To explore the anti‐cancer effects of physiological deep‐sea water(PDSW) combined with hyperther‐mia for hepatocellular carcinoma in vitro .Methods Deep‐sea water (DSW) from the south Chinese sea was processed ,and made in‐to PDSW ,detection of some elements .In vitro ,the cultured normal liver cells and human hepatoma QGY‐7703 cells were randomly divided into PDSW group and normal saline(NS) group ,the NS group received saline ,the PDSW group received different concentra‐tions of PDSW .Two groups were heated respectively to 6 h of 40 ℃ or 1 h of 43 ℃ ,24 ,48 ,72 h after the administration of PDSW or saline ,the normal liver cells and QGY‐7703 cells proliferation capacity and toxicity were investigated by MTT assay .At the same time testing PDSW and NS in 40 ℃ 6 h for 10 d state of human liver QGY‐7703 cell clone formation rate .Results The results of MTT assay showed that tumor inhibitory rate were time and concentration dependent in tow groups .Tumor inhibitory rate of PD‐SW group in different time was significantly higher than NS group (P<0 .05) .On the other hand ,the inhibitory of hepatocyte for PDSW group in different time were significantly lower than NS group .In addition ,the clone formation rate of PDSW group was lower than those of NS group(P<0 .05) .Conclusion PDSW can improve the heat tolerance of normal liver cells .When combine with heat ,it can obviously inhibit the growth of human liver cancer QGY‐7703 cells .
5.Effect of Proprioceptive Training on Lumbar Disc Herniation

Objective To investigate the effects of proprioceptive training on lumbar disc herniation (LDH). Methods 50 patients with LDH were divided into experimental group (n=25) and control group (n=25). Both groups received physiotherapy and core stability exercise, and the experimental group received proprioceptive training with BIODEX Balanced System in addition. They were assessed with the Visual Analogous Scale (VAS) of pain, Oswestry Disability Index (ODI), and posture stability and limits of stability of BIODEX Balanced System. The incidence of relapse was followed up in a year. Results The scores of VAS, ODI, posture stability and limits of stability improved in both groups after treatment (P<0.05), and the ODI, posture stability and limits of stability improved more in the experimental group than in the control group (P<0.05). There were 5 cases relapsed in the control group, and 2 cases in the experimental group. Conclusion The proprioceptive training may further improve the function of lower back, and motor control in patients with LDH, and prevent the relapse.
6.Effect of Different Psychological Rehabilitation Time on Patients with Paraplegic Spinal Cord Injury

Objective To study the effect of different psychological rehabilitation time on patients with spinal cord injury (SCI). Methods 80 patients with SCI caused by spinal fracture were divided into early psychological rehabilitation group (n=40) and late psychological rehabilitation group (n=40). The routine treatment was administered to both groups, and psychological rehabilitative therapy was also administered immediately and 3 weeks after admission respectively. Their function, psychological state and quality of life were evaluated. Results The scores of modified Barthel index (MBI), Functional Independence Measure (FIM), and psychology, independence, social relations and the total score in World Health Organizations Quality of Life (WHOQOL-100) were higher, and the scores of Hamilton Self-rating Scale for Depression (HAMD) and Hamilton Self-rating Scale for Anxiety (HAMA) were lower in the early psychological rehabilitation group than in the late psychological rehabilitation group (P<0.05). Conclusion Early psychological rehabilitation is beneficial for functional and psychological state recovery in patients with SCI.
7.The neuroprotective effects of cyclin-dependent kinase-5 inhibition in mice with Niemann-Pick disease type C.

In order to investigate the neuroprotective effects of cyclin-dependent kinase-5 (cdk-5) inhibition in mice with Niemann-Pick disease type C (NPC) (npc(-/-)), recombinant adeno-associated virus (rAAV) carrying the small interfering RNA (siRNA) specific for cdk-5 gene was injected into 3-day-old npc(-/-) mice intracerebroventricularly. The rAAV-GFP-injected age-matched npc(-/-) mice and non-surgery age-matched npc(-/-) mice were employed as controls (n=6-10/group). From the 4th to 8th week after the treatment, mice were weighed, and evaluated for limb motor activity by using the coat hanger test once a week. Eight-week-old npc(-/-) mice were sacrificed by decapitation, and brains were quickly dissected and halved sagittally. Immunohistochemistry, Western blotting, and HE staining were used to evaluate the neuropathology in npc(-/-) mice. The results showed that rAAV-cdk-5-siRNA-GFP significantly reduced the number of axonal spheroids, delayed the death of Purkinje neurons, ameliorated motor defects in npc(-/-) mice, and significantly attenuated the hyperphosphorylation of tau proteins. These data suggested that inhibition of cdk-5 activity has neuroprotective effect on neurons in NPC mice.
8.Observation of curative effect of gastric bypass on type 2 diabetes: a study with eight cases and literature review

Objective To investigate the effects of gastric bypass on glycometabolism and improvement of islet β cell function and insulin resistance in patients with type 2 diabetes. Methods Eight patients with type 2 diabetes combined with gastric carcinoma who treated with gastric bypass were studied prospectively. Fasting and postprandial plasma glucose levels, fasting and postprandial insulin C-peptide levels, and body mass index (BMI) were measured right before the surgery and at intervals of 1 week, 2 weeks, 1 month and 3 months after the surgery. Glycosylated hemoglobin (HbA1c) levels were measured before and 3 months after the surgery. The outcome of the diabetes after 3 months of the surgery was also monitored. Results Fasting and postprandial plasma glucose levels decreased (P < 0.05) and fasting and postprandial insulin C-peptide levels increased (P < 0.05) after the surgery. HbA1c levels also decreased (P < 0.05) after 3 months of the surgery. There was no significant change of BMI at all intervals after the surgery(P> 0.05). All of the 8 patients reached the total effective standard and 6 patients reached the clinical remission standard after 3 months of the surgery. Conclusions It suggests that gastric bypass can significantly lower plasma glucose levels in type 2 diabetes, which does not depend on the loss of weight. The control of plasma glucose by gastric bypass may be due to the improvement of islet β cell function and increasing secretion of endogenous insulin.
9.Effect of physiological deep-sea water on the hyperthermal tolerance of mice

Objective To explore the effects of physiological deep-sea water(PDSW) on hyperthermal tolerance of Kunming (KM ) mice in the 45 .0 ℃ environment .Methods Deep-sea water from the south Chinese sea was processed ,and the metallic ele-ments dissolved in the DSW were analysed .The mice were randomly divided into 2 groups :the control group received tap water ;the experimental group treated with PDSW for 15 d .And then the mice were fed in the 45 .0 ℃ conditions .The survival time and histo-morphometric analyses of the brain ,lung ,heart ,liver and kidney were investigated .Results The survival time in PDSW-fed group was significantly longer than that of the control group (P< 0 .05) .Moreover ,histomorphometric analyses showed that PDSW could protect the brain ,lung ,heart ,liver and kidney of KM mice from the 45 .0 ℃ conditions .The results of western blot revealed that ex-pression of HSP72 of liver tissues for PDSW-fed group substantially increased ,when compared with the control mice(P< 0 .05) . Conclusion PDSW could improve hyperthermal tolerance of KM mice ,which maybe in the relation with expression of HSP72 pro-moted by PDSW .
10.SP1-induced lncRNA MCF2L-AS1 promotes cisplatin resistance in ovarian cancer by regulating IGF2BP1/IGF2/MEK/ERK axis

Objective:
Cisplatin resistance is a huge problem encountered in ovarian cancer treatment. Our study probed the roles and the underlying mechanisms of lncRNA MCF2L-AS1 in ovarian cancer cisplatin-resistance.
Methods:
SKOV3 and IGROV-1 cells were subjected to gradually increasing concentrations of cisplatin to construct ovarian cancer cisplatin-resistance cells. Cell proliferation was evaluated by cell counting kit-8 and colony formation assays. Cell apoptosis was assessed using Annexin V and PI staining. The relationships between SP1, MCF2L-AS1 and insulin-like growth factor-2 mRNA binding protein 1 (IGF2BP1) were verified by RNA pull-down, RIP, ChIP and dual-luciferase reporter gene assay, respectively. Tumor xenograft experiment was employed to evaluate the effects of MCF2L-AS1 silencing on ovarian cancer cisplatin-resistance in vivo. TUNEL staining and immunohistochemistry were performed in tumor tissue.
Results:
MCF2L-AS1 and IGF2BP1 were upregulated in cisplatin-resistant cells. MCF2L-AS1 silencing suppressed cell proliferation of cisplatin-resistant cells, while promoted the apoptosis, suggesting that MCF2L-AS1 knockdown suppressed ovarian cancer cells cisplatin-resistance. Meanwhile, MCF2L-AS1 silencing enhanced cisplatin sensitivity in ovarian cancer parental cells and IGF2BP1 overexpression impaired cisplatin sensitivity of parental cells. MCF2L-AS1 activated IGF2/MEK/ERK pathway through interacting with IGF2BP1. Transcription factor SP1 activated MCF2L-AS1 expression. MCF2L-AS1 knockdown inhibited ovarian cancer cisplatin-resistance in vivo.
Conclusion
SP1-induced MCF2L-AS1 promoted ovarian cancer cisplatin-resistance through activation of IGF2/MEK/ERK pathway via interacting with IGF2BP1.
