1.Study on the application of EPC in management of hospital energy saving

Objective: Through the analysis of the current situation of energy consumption and the energy management problems that hospital faced, elaborat the high demands of energy management in hospital effective logistics management, the saving mechanism of Energy Performance Contracting (EPC) is proposed. Methods: The process of introducing EPC to China and related concepts are described, and the main mode and implementation processes as well. Results: To explore the feasibility of the integration in energy-saving reformation and the mode of energy runing management what is the core to EPC, tell people what need to pay attention to in the implementation process, and provide scientific methods for energy management of efficiency operation and sustainable development in hospital. Conclusion:At last, pointing out that saving mechanism of EPC is suitable for hospitals and will bring significant saving benefit.
2.RAPD Analysis of genetic relationship among Dioscorea zingiberensis with different diosgenin

Objective To study the genetic relationship among Dioscorea zingiberensis with different diosgenin content at DNA level.Methods DNA of all types was amplified by 12 random primers from 40 random primers,the polymorphic bands of RAPD were counted.And the results of types were analysed by POPGENE 1.31(Tool for Population Genetic Analysis 1.31) software.Results A total of 73 bands was obtained,64 of total bands(87.67%) was polymorphic.The results indicated that there were apparent and abundant genetic variations in D.zingiberensis. Conclusion The formation and accumulation of diosgenin could be affected significantly by the genetic basis of D.zingiberensis.
3.Temperature and time change of moxa cone moxibustion with different purities of moxa.

OBJECTIVETo explore the relationship between the temperature and time in moxa cone moxibustion with different purities of moxa.
METHODSAccording to the purities, the moxa were divided into a 10 : 1 group, a 20 : 1 group, a 30 : 1 group and a 40 : 1 group, 30 moxa cones in each group. With the VICTOR DM6902 electronic thermometer, the temperature of the undersurface center at different time points during the moxa cone moxibustion with different purities of moxa was measured. Once the moxa cone was ignited, the results were recorded at the end of each second. Also the temperature peak of the undersurface center and the time when the peak occurred were recorded.
RESULTThe undersurface temperature was increased in all the groups; the time of moxa cone reaching the lowest peak temperature was significantly different in all the groups (all P<0. 05), which was the 10 : 1 group, 20 : 1 group, 30 : 1 group and 40 : 1 group from slow to fast. 50 s, 60 s, 70 s, 80 s and 90 s after moxa cone was ignited, the temperature of moxa cone at the same time point was significantly different in all the groups (all P<0. 05), which was the 10 : 1 group, 20 : 1 group, 30 : 1 group and 40 : 1 group from slow to fast. Conclusion Among the moxa with purity of 10 : 1, 20 : 1, 30 : 1, and 40 : 1, the temperature change rate of the low-purity moxa cone is smaller than that of higher purity, and the stimulating duration of the former is longer than the latter. It is believed that the moxa with purity of 40 : 1 is suitable for scarring moxibustion; the moxa with purity of 30 : 1 and 20 : 1 is suitable for the non-scarring moxibustion; the moxa with purity of 10 : 1 is suitable for gentle moxibustion therapy. The high-purity moxa can also be applied to the field of the low-purity moxa.

4.Expressions of survivin and caspase-9 in human primary hepatocellular carcinoma tissues and their significances

Objective To study the expressions of survivin and caspase-9 in human primary hepatocellular carcinoma(HCC) tissues and their relationship with apoptosis.Methods Immunohistochemical staining was uesed to detect the expressions of survivin and caspase-9 in paraffin sections of 96 cases including 42 HCC tissues,42 paired adjacent noncancerous tissues and 12 normal liver tissues. Apoptosis was detected by TUNEL.Results No immunoreactivity of survivin was seen in normal liver tissues.The positive rates of survivin in HCC and paired adjacent noncancerous tissues were 73.81% and 4.76%,respectively,there was significant difference between two groups(P0.05).The expressions of survivin and caspase-9 in HCC tissues were not associated with age,sex and the size of tumor,but they were directly associated with tumor histological grade and metastasis.Conclusion High expression of survivin in HCC may inhibit apoptosis through depressing the expression of caspase-9,and it is an indicator of independent poor prognosis.
5.Inducement of tetraploid Dioscorea zingiberensis

Objective To create new tetraploid resource of Dioscorea zingiberensis in the hope of potential breeding materials with high yield and high level of diosgenin. Methods Callus with tiny green buds were directly soaked in colchicine solution or cultured in medium plus with colchicine to prohibit the isolation of chromosomes and induce tetraploid mutants. Results Tetraploids were achieved successfully. The most efficient treatment for inducing tetraploid was soaking the tiny buds in 1?103 mg/L colchicine solution for 24 h, the inducing rate could be up to 35. 2%. The induced tetraploids exhibited notable difference with common wild type (diploids) in morphology, physiology, and microscopic structure. ConclusionThe tetraploid plants show the advantages of gigantic size and vigorous growth. Thus, the established technique system to induce tetraploid from tissue cultured callus would provide an efficient alternative pathway for medicinal plants of Dioscorea L. breeding.
6.Effect of alum on immune response in mice induced by HBoV1 VP2 VLPs

Objective:To study the effect of alum on immune response in mice induced by HBoV1 VP2 VLPs.Methods:BABL/c mice were randomly divided into VLPs experimental group, alum adjuvant experimental group, PBS control group and alum adjuvant control group,the experimental group mice were intramuscular immunization with HBoV1 VP2 VLPs and HBoV1 VP2 VLPs added alum,control group mice were immunization with alum or PBS buffer,then to study the effect of alum on immune response in mice induced by HBoV1 VP2 VLPs by cellular and humoral immune strength.Results: Alum adjuvant decreased cellular immune response induced by HBoV1 VP2 VLPs(P<0.001),enhance the HBoV1 VP2 VLPs immuned serum IgG titer(P<0.05)and IgG activity(P<0.01).Conclusion:Alum adjuvant can enhance humoral immune response induced by HBoV1 VP2 VLPs,but weaken cellular immune response induced by HBoV1 VP2 VLPs,when HBoV1 VP2 VLPs used as a prophylactic vaccine it should add alum adjuvant,when used as a therapeutic vaccine,it should not add alum adjuvant.
7.Effects of Yiqi Jiedu Compound on the Expression of Aire and ROR-γt/Foxp3-Related Inflammatory Factors in the Thymus of Experimental Autoimmune Myasthenia Gravis Rats

Objective To study the effect of Yiqi Jiedu compound on autoimmune regulator(Aire)and transcription factor retinoic acid-related orphan receptor(ROR-γt)/Effects of forkhead wing helix transcription factor 3(Foxp3)-related inflammatory factor expression.Methods The EAMG model was replicated by immunization-induced injection of mouse-derived AchR-α subunit 97-116 peptide,including normal group,model group,positive control group(prednisone),and Yiqi Jiedu compound high-dose,medium-dose and low-dose groups.Group of 10,the behaviors and Lennon scores of the rats in each group were observed.ELISA method was used to detect serum transforming growth factor-β1(TGF-β1)and interleukin-6(IL-6)levels.Western blot was used to detect the thymus tissue of EAMG rats.The expression of Aire and Foxp3,and the expression of RORγt in the thymus tissue of rats in each group was detected by RT-PCR.Results Compared with before treatment,the Lennon grading score of each drug group was significantly decreased;compared with the prednisone group,there was no significant difference in the Lennon score of each dose group of Yiqi Jiedu compound(P>0.05).RT-PCR detection results showed that compared with the normal group,the relative expression of RORγt mRNA in the thymus of the model group was significantly increased(P<0.01).Western blot detection results showed that the expressions of Aire and Foxp3 in the model group were significantly lower than those in the normal group(P<0.05);The levels of Aire and Foxp3 proteins in thymus of the rats in each drug group were significantly higher than those of model group(P<0.01).ELISA results showed that serum IL-6 content in model group was significantly higher than that in normal group(P<0.01),while TGF-β1 content was significantly lower than that in normal group(P<0.01).After intragastric administration,the content of IL-6 in all drug groups was significantly decreased,and the content of TGF-β1 was significantly increased(P<0.01).Conclusion Yiqi Jiedu compound can improve the symptoms of muscle weakness in EAMG rats,and its mechanism may be through up-regulating the expression of Aire protein,regulating immune inflammatory response,affecting the differentiation of Th17/Treg,thereby regulating the immune balance of ROR-γt/Foxp3.The role of autoimmune response may be one of the mechanisms of its treatment of MG.
8.Analysis of epidermal growth factor receptor mutation characteristics in patients with non-small cell lung cancer in Fuyuan of Yunnan Province

Objective:To investigate the epidermal growth factor receptor (EGFR) mutation rate, mutation characteristics and distribution characteristics of different mutation types in patients with non-small cell lung cancer (NSCLC) in Fuyang of Yunnan province, to provide the clinical individualized targeted therapy of NSCLC in this region.Methods:A total of 328 NSCLC patients whose native place were Fuyuan and who underwent EGFR test in Fuyuan County People's Hospital in Yunnan Province from January 2018 to August 2020 were selected, and their clinical data such as gender, age, ethnicity, pathological type and the results of EGFR test were collected for statistical analysis.Results:The EGFR mutation rate of NSCLC patients was 40.55% (133/328). The EGFR mutation rate of female patients was higher than that of males ( P < 0.01). The EGFR mutation rate showed a downward trend with age, the EGFR mutation rate of patients ≤ 60 years old was higher than that of patients > 60 years old ( P = 0.014). The EGFR mutation rate in ethnic minority was not statistically different from Han nationality ( P = 0.789). The EGFR mutation rate of patients without smoking history was higher than that of patients with smoking history ( P<0.01). Patients with adenocarcinoma had a higher EGFR mutation rate than squamous cell carcinoma ( P = 0.002). The EGFR mutation rate in patients with stage Ⅰ-Ⅱwere higher than that in patents with stage Ⅲ-Ⅳ ( P = 0.013). The EGFR mutation rate in tissue samples were higher than that in peripheral blood samples ( P = 0.009). In 328 patients the EGFR single-point mutation rate was 24.70% (81/328), and the compound mutation rate was 15.85% (52/328); the common mutation rate was 17.07% (56/328), and the rare mutation rate was 23.48% (77/328). The top 5 mutation types were L858R (10.06%), G719X+S768I (7.32%), 19-Del (7.01%), G719X+L861Q (6.40%), and G719X (4.21%). In 133 patients with EGFR mutation, the proportion of patients with rare mutation [57.89% (77/133)] was higher than the proportion of patients with common mutation [42.11% (56/133)]. Conclusion:The EGFR mutation rates of female, adenocarcinoma, non-smoking and young NSCLC patients in Fuyuan area are high, and the rare mutation rate is high.
9.The levels of serum interferon gamma-induced protein 10 in patients with acute pathogen infection

Objective To observe the levels of interferon gamma-induced protein 10(IP-10)in children with acute viral and bacterial infection.Methods There were three groups:acute viral infection group(51 ca-ses),acute bacterial infection group(52 cases),and healthy control group(51 cases).Serum IP-10 levels were detected by enzyme linked immunosorbent assay(ELISA),and serum C reactive protein(CRP)were deter-mined with BNⅡ automatic protein analyzer.Results The level of serum IP-10 and CRP were significantly different in different groups(P<0.05).Compared with control group,the level of IP-10 and CRP were higher in acute viral infection group or in acute bacterial infection group(P<0.05).The level of IP-10 was higher in acute viral infection group than that in acute bacterial infection group(P>0.05).The level of CRP was higher in bacterial infection group than that in acute viral infection group(P<0.05).The IP-10 levels in viral Infec-tion patients were not correlated with the CRP levels.The area under curve(AUC),sensitivity and specificity of IP-10 and CRP were 0.688 and 0.873,35.0% and 79.6%,96.1% and 98.0%.When cut-off value of predic-tive probability was 0.713,sensitivity and specificity were increased to 82.5% and 100.0%.Conclusion Ser-um IP-10,CRP and predictive probability are valuable in the diagnosis of acute pathogen infection in children.
10.Effects of Chrysophanol on the Activation of Microglia and the Expression of Inflammatory Factors in Cerebral Ischemia Model Rats

OBJECTIVE:To study the effects of chrysophanol on the activa tion of microglia and the expression of inflammatory factors in cerebral ischemia model rats. METHODS :SD rats were randomly divided into sham operation group , model group and chrysophanol high ,medium,low dose groups [7.88,3.94,1.97 mg/(kg·d)],with 20 rats in each group (the number was complemented in cases of death or unsuccessful modeling during modeling process ). Except for sham operation group , middle cerebral artery occlusion model was established in other groups by improved thread method. After 2 hours of ischemia , sham operation group and model group were intraperitoneally injected with 1 mL normal saline ,and each administration group was intraperitoneally injected with 1 mL corresponding drug ,once a day ,for 7 consecutive days. After last medication ,the score of neurological impairment was recorded ;cerebral infarction of rats was observed by TTC staining ,and the percentage of cerebral infarction area was calculated. The expression of Iba- 1 positive cells in ischemic penumbra of rats was observed by immunofluorescence staining. The expression of Notch- 1,TNF-α and ICAM-1 in the ischemic penumbra of rats were detected by Western blotting assay. RESULTS :In sham operation group ,there was no infarction area in the brain tissue ,and the Iba- 1 positive cells in the ischemic penumbra were few and branched. Compared with sham operation group ,the infarction area of cerebral tissue in rats was obvious in model group ; the 052)number of Iba- 1 positive cells in ischemic penumbra were 〔ZQ2017003〕) increased significantly ,and they were in amoeba or round shape;the neurological impairment score ,the percentage of cerebral infarction area , relative expression of Notch- 1, TNF-α and ICAM-1 protein in ischemic penumbra were increased significantly (P<0.05). Compared with m odel rats ,the infarction area of cerebral tissue in each dose group of chrysophanol was reduced to different extent ;the number of Iba- 1 positive cells in ischemic penumbra was decreased ;neurological impairment score ,the percentage of cerebral infarction area ,relative expression of Notch- 1,TNF-α and ICAM-1 protein were significantly decreased (P<0.05 or P<0.01). CONCLUSIONS :Chrysophanol has a certain protective effect on the brain tissue of cerebral ischemia model rats ,and can relieve the nerve injury. Its mechanism may be associated with inhibiting the activation of microglia and expression of inflammatory factors mediated by Notch pathway.
