3.Analysis on RAPD Genetic Polymorphism of HIV Related Oral Candida Species

60 Candida strains were analyzed with randomly amplified polymorphic DNA(RAPD) from HIV-infected patients' oral cavity.The results showed that RAPD-PCR fingerprinting appeared from zero to five bands whose size rang from 300 bp to 2 kb with P2 primer,Candida albicans have three characteristic bands of 300 bp,400 bp and 600 bp in size,non-Candida albicans have the similar bands.They can be clas-sified five genetic clusters and 14 genotypes by Cluster Analysis,two Candida albicans(P385&P403) with fluorocytosine-resistance were classified into C1 genotype for "Squared Euclidean Distance" being 0.115,one Candida albicans(P321) and one Candida glabrata(P522) who were resistant against Amphotericin B were classified into D1 genotype for "Squared Euclidean Distance" being 0.221.So,an abundant genetic polymorphism appeared in Candida species related HIV,RAPD could be as a reference for genotype of Candida albicans;different Candida speices had some special bands,different Candida strains had similar bands in the same species;it is perhaps that some genotypes were relevant to drug resistance.
4.Clinical observation of gefitinib in treatment of female patients with adenocarcinoma of lung WU

Objective To evaluate the efficacy and adverse effects of gefitinib in the treatment of fe- male patients with advanced adenocarcinoma of lung who had failed to previous chemotherapy.Methods These patients received 250mg of gefitinib orally,once daily until disease progression or development of intol- erable toxic reaction.They were evaluated one month after treatment and every other month thereafter.Results Among the 27 evaluable patients,there were 1 CR(3.7%),11 PR(40.8%),10 SD(37.0%)and 5 PD(18.5%). The overall response rate was 44.5%(95% CI 29%~68%);and 22 patients(81.5%)gained profit(CR+PR+ SD)from the clinical therapy(95% CI 62%~94%);the mean TTP was 7.2 months.Symptomatic improvement rate was 80.0%.The main adverse effects were mild rash and diarrhea.Conclusion gefitinib has significant efficacy in the treatment of female patients with advanced tung cancer who had failed to previous chemother- apy.Adverse effects are mild.gefitinib is a suitable therapy for these patients.
5.Interactions between mycoplasma lipid-associated membrane proteins and the host cells.

Mycoplamas are a group of wall-less prokaryotes widely distributed in nature, some of which are pathogenic for humans and animals. There are many lipoproteins anchored on the outer face of the plasma membrane, called lipid-associated membrane proteins (LAMPs). LAMPs are highly antigenic and could undergo phase and size variation, and are recognized by the innate immune system through Toll-like receptors (TLR) 2 and 6. LAMPs can modulate the immune system, and could induce immune cells apoptosis or death. In addition, they may associate with malignant transformation of host cells and are also considered to be cofactors in the progression of AIDS.

7.Associated factors for HIV stigma among the patients on antiretroviral therapy in Wenshan Prefecture

Objective:
To learn HIV related stigma and its associated factors among the patients on antiretroviral therapy ( ART ) in Wenshan Prefecture, Yunnan Province, so as to provide evidence for eliminating HIV discrimination.
Methods:
A total of 419 subjects were recruited by convenience sampling from three ART clinics in Wenshan City and Maguan County between October 2017 and January 2018. HIV/AIDS Related Stigma and Discrimination Scale developed by Li Xianhong et al was employed. The multivariate linear regression model were used to explore the influencing factors for HIV stigma.
Results:
The median scores of disclosure concern, public rejection, family stigma, internalized stigma, health service providers' stigma were 24.00, 6.00, 10.00, 20.00, 2.00, respectively, and the overall was 68.00. The multivariate linear regression analysis showed that female patients ( standardized β=0.135 ) , patients with opportunistic infection ( standardized β=0.120 ), patients had no HIV infected family member ( standardized β=-0.128 ) , patients without family support ( standardized β=-0.175 ) , patients received gift from ART clinics ( standardized β=0.124 ) , patients scored lower in ART knowledge ( standardized β=-0.117 ) were likely to scored higher in HIV stigma.
Conclusions
The stigma on disclosure concern and internalized stigma dimensions are grievous among ART patients in Wenshan Prefecture. Gender, opportunistic infection, HIV infection in family, family support, receiving incentive gifts from clinics and awareness of ART are associated with HIV stigma.
8.Parents willingness to vaccinate their children aged 3-11 years with the COVID-19 booster vaccine in Nanjing

Objective:
To investigate parents acceptance to the COVID-19 vaccine booster shots for their children aged 3-11 years in Nanjing based on the theory of planned behavior, and to provide a scientific theoretical basis for carrying out COVID-19 booster vaccination among children of this age group.
Methods:
A total of 1 286 parents of children aged 3-11 years in Nanjing were selected by multistage stratified cluster sampling. A questionnaire survey based on the theory of planned behavior was used to investigate their willingness to vaccinate their children with the COVID-19 booster dose, and structural equation model was used to conduct data analysis.
Results:
About 90.2% of parents were willing to give their children a booster dose of the COVID-19 vaccine. The model constructed based on the theory of planned behavior could explain 42.2% of the variance of vaccination acceptance. Attitudes and perceived behavioral control had a direct positive effect on parents acceptance to booster dose of the COVID-19 vaccine (path coefficients were 0.47 and 0.18, P <0.01); The direct effect of subjective norms on vaccination acceptance was not statistically significant; Perceived behavioral control and subjective norms could have indirect positive effects on vaccination acceptance through attitudes (path coefficients were 0.27 and 0.13, P <0.01).
Conclusion
Parents in Nanjing have a higher acceptance to vaccinate their children aged 3-11 years with the COVID-19 vaccine booster dose. The theory of planned behavior shows a good explanatory ability on parents acceptance to vaccinate their children, and attitude plays an important role in the formation of vaccination acceptance.
9.Comparison of effect on blood glucose variability of intravenous infusion of low dosage fructose and glucose in type 2 diabetic patients in stable phase

Objective To compare the effect of intravenous infusion of low dosage fructose with glucose infusion on blood glucose variability in type 2 diabetic patients in stable phase.Methods Data of 22 patients (male:female,18:4)with type 2 diabetes mellitus in stable phase were collected for the study.From 9am to 11 am,all the subjects were administrated one of the following solutions at constant speed on consecutive days randomly:5% glucose injection 200 mL with insulin 3 U,0.9% normal saline 200 mL or 5%fructose injection 200 mL. The blood glucose concentrations were detected every 5 minutes after infusion by confinuous glucose monitoring system(CGMS).The variability of blood glucose was compared using these 3 kinds of solutions.A two-tailed P value of ≤0.05 was considered statistically significant. Statistical analysis was carried out with SPSS version 13.0.Results No significant difference was found among these 3 kinds of solutions.In addition,no adverse effects were founded.Conclusion Low dosage infusion of neither fructose nor glucose had apparent effect on blood variability of patients with type 2 diabetes mellitus in stable phase.
10.Use of allogenic acellular dermal matrix combined with autologous epidermal cells for the repair of tissue defect.

OBJECTIVETo investigate a method for the repair of tissue defect.
METHODSAllogenic acellular dermal matrixes (ADM) were implanted to full-thickness skin defects made on the dorsa of rats. Two weeks later, autologous suspended epidermal cells were transplanted on to the surface of vascularized ADM. Respectively, neoepidermis was macroscopically observed 2, 3, 5 weeks after grafting, and samples were taken to make routine paraffin sections for microscopical examination, and immunohistochemical staining for type IV collagen was also performed.
RESULTSThe vascularized ADM could support proliferation and differentiation of epidermal cells, and also could promote the formation of dermal-epidermal junction. Suspended epidermal cells in an artificial culture system in vivo could develop into mature epidermis. The reconstructed skin not only looked like the normal one in appearance in which hair was removed, but also revealed a better function.
CONCLUSIONSFull-thickness skin defect can be repaired by transplanting autologous epidermal cell suspension on to vascularized ADM.
