1.Detection of HPV16 and p53 Protein in Tumors of Nasal Cavity and Sinus

Objective To explore the role of HPV16 and p53 protein in the pathogenesis of tumors of nasal cavity and sinus of Chinese people. Methods The expression of HPV16 and p53 in 58 tumor samples of nasal cavity and sinus was detected by immunohistochemistry. Results There was no p53 expression in inverted pipillomas(IP). HPV16 infection rate in IP was 62.5%. The positive rate of p53 protein in the malignant tumors and IP with atypical hyperplasia was 56.0% and 44.4%, respectively, and HPV16 infection rate in the both tissues was 24.0% and 66.7%, respectively. The positive rate of p53 and HPV16 simultaneous expression in IP with atypical hyperplasia were 33.3%, which was higher than that in the malignant tumors and IP without atypical hyperplasia. Conclusion The high-risk HPV16 infection may be primary cause of IP. p53 gene mutation may be close relation with the pathogenesis of nasal cavity and sinus malignant tumors. HPV16 and p53 may have synergic effect in the malignant transformation of nasal cavity and sinus benign tumors.

3.Arthrolysis and a hinged external fixator for posttraumatic elbow stiffness

Objective To explore the clinical effect of arthrolysis and a hinged external fixator for treatment of posttraumatic elbow stiffness.Methods From April 2007 to April 2012,26 cases of posttraumatic elbow stiffness patients were treated with arthr o lysis and a hinged external fixator after operation,and the effect was observed.Results After operation,26 patients were followed up for 15(8-24) months,the ranges of elbow joint motion increased from (42 ± 17) ° to (119 ± 17) ° after operation.There was significant difference (t =152.85,P< 0.05).The MEPS scores increased from (36 ± 13) points to (82 ± 14) points after operation,15 cases were excellent,5 cases were good,3 cases were fair and 3 cases were poor,the rate of excellent and good cases was 76.9% (20/26).Twenty-three cases (88.5%,23/26)elbow function improved.Postoperative infective complication occurred in 1 case,postponed incision healing occurred in 1 case and 1 patient had cubital nerve paralysis,the symptom disappeared after 3 months and the patient showed no instability in elbow joint.Conclusion Arthrolysis and a hinged external fixator can deep stabe the elbow and its specific position,allowing early rehabilitation to promote functional recover of the elbow.
4.Efficacy of Using a Semiconductor Laser Combined with Short-wave Ultraviolet Rays to Treat Herpes Zoster

Objective To observe the efficacy of using a semiconductor laser combined with short-wave ultraviolet rays(UV)to treat patients with herpes zoster. Methods Forty-two patients with herpes zoster in the Department of Dermatology of our hospital were randomly divided into two groups,with 21 patients per group. Patients in the control group were treated with conventional dermatology drugs and short-wave UV,whereas patients in the observation group were treated with conventional dermatology drugs and a semiconductor laser combined with short-wave UV. Re-sults After treatment for 7 days,the efficacy rate in the control group was 80.95%,whereas that in the observation group was 95.24%. The analge-sia time,scab time,and scab removal time were significantly shorter in the observation group than in the control group(P<0.05). The results of the pain assessment(visual analog scale )showed that the score of the observation group after treatment was significantly lower than that of the con-trol group(P<0.05). Conclusion Therapy involving a semiconductor laser combined with short-wave UV for patients with herpes zoster can be more effective than therapy with only a short-wave UV laser in promoting herpes healing,relieving neuralgia,and improving quality of life.
5.The A-204C Polymorphism in CYP7A1 Gene Affects Its Promoter Activity

cholesterol 7α-hydroxylase gene ( CYP7A 1 ) plays a key role in the catabolism of cholesterol into bile acids. To investigate whether the A-204C polymorphism in CYP7A1 gene affects the gene expression,using luciferase as the reporter gene, four recombinants were constructed by inserting forward or reverse sequence with A or C allele at the polymorphism site into the promoter-less vector pGL3-basic. The constructs were then transfected into four cell lines and the luciferase activity of each expression vector was examined by dual luciferase reporter gene assay system. The results showed that activities of the forward sequence of both genotypes were higher than that of reverse sequence. Promoter activity of the recombinants with A allele was about one third lower than that with C allele. According to the analysis with TRANSFAC database, there may exist a Zic3 binding site when there is the C allele at -204. Our study indicates that the A-204 C polymorphism in CYP7A1 promoter region decreases its promoter activity and thus represses the gene expression, possibly due to the lack of a potential Zic3 binding site.
6.Efficacy of modified 270°spinal canal decompression combined with one-stage pedicle screw fixation and vertebral reconstruction for severe thoracolumbar fractures

Objective To investigate the efficacy of modified 270°spinal canal decompression combined with one-stage pedicle screw fixation and vertebral reconstruction for severe thoracolumbar fractures.Methods A retrospective case-control study was made on 44 cases of severe thoracolumbar fractures treated from January 2009 to June 2014.There were 34 males and 10 females, with age range of 20-68 years (mean, 37.9 years).The injured vertebrae included T11/12 in one case, T12-L1 in 10, L1/2 in 21, L2/3 in nine and L3/4 in three.Injury type was all AO type B2.According to the American Spinal Injury Association (ASIA), neurological deficit was Grade A in three cases, Grade B in five, Grade C in 22, and Grade D in 14.Load sharing score (LSC) was 7-9 points (mean, 8.1 points), while the thoracolumbar injury classification and severity score (TLICS) was 7-9 points (mean, 8.2 points).According to the difference of surgical procedures, all cases were divided into traditional group (21 cases) and improved group (23 cases).The patients in traditional group were treated by 270° spinal canal decompression combined with one-stage pedicle screw fixation and vertebral reconstruction, and those in improved group were treated by modified 270°spinal canal decompression combined with one-stage pedicle screw fixation and vertebral reconstruction.Between-group differences were compared with regard to operation time, intraoperative blood loss, postoperative drainage, allogeneic blood transfusion, pain visual analogue scale (VAS) at postoperative 72 hours, Cobb angle, anterior vertebral height, spinal canal compromise, Denis score, work state, neurological function, bone graft fusion and complications.Results Duration of follow-up was (32.4±15.8)months (range, 12-60 months).No wound infection or deterioration of neurological function was found after operation.Operation time was (2.4±0.5)hours in traditional group, less than (2.8±0.6)hours in improved group (P<0.05), while there were no significant differences in intraoperative bleeding, postoperative draining and blood transfusion between the two groups (P>0.05).VAS was (3.2±0.9)points in traditional group and (3.3±0.9)points in improved group at postoperative 72 hours (P>0.05).Cobb angle, anterior vertebral height and spinal canal compromise in both groups obtained well recovery and maintained after operation (P<0.05), but the recovery of spinal canal compromise was better in improved group than traditional group [(2.1±1.8)% vs.(11.8±6.1)%] (P<0.05).Denis score and work state were similar between the two groups (P>0.05).At the last follow-up, ASIA Grade A was noted in three cases, Grade C in two, Grade D in 23, and Grade E in 16.Bone union was achieved in all cases, with no implant loosening or breakage observed.Thirteen cases in traditional group were found with residual bone fragments in the spinal canal after surgery, and two of them received revision surgery.Conclusions Either modified or traditional 270° spinal canal decompression combined with one-stage pedicle screw fixation and vertebral reconstruction can attain satisfactory clinical results in treatment of severe thoracolumbar fractures.However, the modified 270°spinal canal decompression can achieve more thorough decompression without reducing stability of the spine.
7.Comparative analysis of albumin and bilirubin levels in patients with nephrotic syndrome and postoperative gastroparesis syndrome

Objective Hypoalbuminemia is a typical symptom of nephrotic syndrome ( NS) , which may result from the loss of much protein with urine.Hyperbilirubinemia is also a common symptom in patients with NS.This study is intended to reveal the relationship between hypoalbuminemia and hyperbilirubinemia in patients with NS by investigating urine bilirubin, albumin( ALB ) and 24-hour urine protein in the NS group, chronic glomerulonephritis ( CGN ) group and postoperative gastroparesis syndrome ( PGS) group ( ALB<35 g/L) .Methods Totally 187 patients with NS, 70 patients with CGN and 64 patients with PGS ( ALB <35 g/L ) were recruited before ALB, urinary protein ( UPR ) , urinary microalbuminuria/creatinine(Umalb/cr) and total bilirubin(TBIL) were detected.SPSS 17.0 Software was used to analyze the difference between the three groups and to reveal the correlations between TBIL and UPR, ALB.Results TBIL, ALB, UPR and Umalb/Cr levels were significantly different between NS, CGN and PGS groups ( one-way ANOVA test, P <0.05), and TBIL was positively correlated with ALB but negatively correlated UPR and Umalb/Cr in both NS and CGN groups (Spearman′s rho test,P<0.05);but no correlation was found between these items in PGS group (Spearman′s rho test,P>0.05) .Conclusion Serum bilirubin of patients with NS is at a low level and shows significant correlations with serum albumin and urinary protein levels.No similar association is found with the other two groups.The results in this study show that the causes of low serum bilirubin in patients with NS may related to the large amount of protein lost in urine.
8.Advances in modulation of mammalian sperm function by fertilization promoting peptide.

Both fertilization promoting peptide and adenosine stimulate capacitation but inhibit spontaneous acrosome loss by modulation of the adenylyl cyclase (AC)/cAMP signal transduction pathway. This is a review aimed at analyzing the function of fertilization promoting peptide during this process. The possible molecular basis is also discussed.

9.Analysis of C2ORF71 gene variant in a Chinese patient with retinitis pigmentosa.

OBJECTIVE:
To explore the genetic basis for a Chinese patient with retinitis pigmentosa (RP).
METHODS:
Whole exome sequencing (WES) was carried out to screen potential variant in the proband. Candidate variants were determined by taking consideration of clinical phenotype. Sanger sequencing was used to verify the variant in the proband and his parents.
RESULTS:
The proband was found to harbor compound heterozygous variants of c.8G>A (p.Cys3Tyr) and c.958_959insA (p.Arg320Glnfs*29) in the C2ORF71 gene, which has derived from his father and mother, respectively. Both variants were unreported previously. Based on the ACMG guidelines, they were predicted to be likely pathogenic and pathogenic, respectively.
CONCLUSION
The novel compound heterozygous variants of the C2ORF71 gene probably underlay the pathogenesis of RP in the proband. Above finding has enriched the spectrum of C2ORF71 gene mutations and facilitated genetic counseling for the family.

OBJECTIVETo develop a simple and reliable method for intensifying the hybridization signals of gene chips.
METHODSThe authors added EDTA and another FAM-labeled probe to the normal PCR products, denatured the mixture by heat, and then let the mixture hybridize with the fastened probes on the chip.
RESULTSWith the use of EDTA and another FAM-labeled probe, the hybridization signals increased by 6 times or greater.
CONCLUSIONAdding EDTA and another probe to the normal PCR products is a simple and efficient method to intensify the hybridization signal of chips.
