1.Biological characteristics of porous tantalum:short-term application is clinically safe but the long-term effect needs to be further studied

BACKGROUND:Porous tantalum, characterized as high porosity, low elastic modulus, high friction coefficient, stable biological properties, good compatibility and typical structural properties, has been the focus in medicine, especial y in orthopedics. <br> OBJECTIVE:To review the biological characteristics of porous tantalum, including its mechanical properties, compatibility, and biological activity. <br> METHODS:A computer-based search of PubMed, CSTJ, Wanfang and VIP databases was done for articles relevant to porous tantalum published from 1990 to 2014 using the keywords of“porous tantalum, biological character, orthopedic applications”both in English and Chinese. <br> RESULTS AND CONCLUSION:Porous tantalum, with high porosity, low elastic modulus, high friction coefficient, and good compatibility, has become increasingly popular in medicine, especial y in orthopedics. At present, porous tantalum is used in the preparation of the integrated acetabular cup, total hip arthroplasty acetabular cup, acetabular reinforcing pad, porous tantalum metal rod, tibial plateau prosthesis and patel ar prosthesis and tibial plateau prosthesis. The short-term fol ow-up of porous tantalum implants in orthopedic surgery has been reported. And clinical data, radiology, histological analysis of the removed material have proved the practicability and safety of the porous tantalum. But the long-term effects of porous tantalum remain to be confirmed.
2.Therapeutic effect of aerobic exercise on blood pressure and vasoactive mediator in patients with isola-ted diastolic hypertension

Objective:To observe therapeutic effect of aerobic exercise on blood pressure and vasoactive mediators in patients with isolated diastolic hypertension (IDH).Methods:A total of 93 recently diagnosed IDH patients were se- lected and randomly divided into exercise intervention group (n=47,received aerobic exercise,twice/d,50~80 min/time for six months)and control group (n=46,didn't receive aerobic exercise).Office blood pressure (OBP), ambulatory blood pressure monitoring (ABPM)and levels of vasoactive mediators were measured and compared be- tween two groups before and six months after intervention.Results:There were no significant difference in levels of OBP,ABPM indexes and related vasoactive mediators between two groups before intervention (P>0.05 all).Com- pared with before intervention,after six-month intervention,there were significant reductions in levels of office di-astolic blood pressure (ODBP),mean DBP (mDBP),daytime DBP (dDBP)and nighttime DBP (nDBP)in both groups (P<0.05 or <0.01);compared with control group,there were significant reductions in levels of ODBP [(89.23±14.70)mmHg vs.(79.26±13.18)mmHg],mDBP [(86.51±14.30)mmHg vs.(77.20±12.45)mm- Hg],dDBP [(87.93±12.16)mmHg vs.(78.02±11.27)mmHg]and nDBP [(83.18±12.57)mmHg vs.(74.22 ±11.36)mmHg]in exercise intervention group,P<0.05 all.Compared with control group,after six-month inter- vention,there were significant rise in levels of adrenomedulin (AM)[(36.75±5.13)ng/L vs.(47.59±6.82)ng/L]and prostaglandinI2 (PGI2)[(89.55±16.31)ng/L vs.(99.24±18.20)ng/L],and significant reductions in levels of angiotensin Ⅱ (AngⅡ)[(98.30± 15.65)ng/L vs.(91.28±17.40)ng/L]and thromboxane A2 (TXA2 )[(93.61±15.27)ng/L vs.(80.24±12.31)ng/L]in exercise intervention group,P<0.05 all.Conclusion:Aer- obic exercise can significantly lower blood pressure and correct vasoactive mediator level disorder in IDH patients.
3.Porous tantalum in orthopedic applications:joint replacement and cartilage reconstruction scaffolds

BACKGROUND:Porous tantalum with high porosity, low elastic modulus, high friction coefficient has stable biological characters and good biocompatibility, which is considered as an ideal implant material in orthopedic surgery. OBJECTIVE:To review the application situation of porous tantalum used in artificial joints, spine, bone necrosis, defects of bone and so on. METHODS:A computer-based search of PubMed, Wanfang, and VIP databases was performed for relevant articles published from 1990 to 2014 using the keywords of “porous tantalum, biological character, orthopedic applications” in English and Chinese. RESULTS AND CONCLUSION:Porous tantalum is a kind of good orthopedic implant material, with high porosity, low elastic modulus, high friction coefficient, and good compatibility. Porous tantalum is mostly used in the preparation of integrated acetabular cup, total hip arthroplasty acetabular cup, acetabular reinforcing pad, porous tantalum metal rod, tibial plateau prosthesis, patelar prosthesis and tibial plateau prosthesis, which has achieved good clinical effects.
4.Bone marrow mesenchymal stem cells combined with VEGF gene for the treatment of limb ischemia in rabbits

Objective To evaluate bone marrow mesenchymal stem cells combined with VEGF gene in the treatment of limb ischemia in rabbits.Methods The right hind limb ischemia model of New Zealand rabbit was established by superficial femoral artery excision and deep femoral artery ligation.Rabbits then were divided randomly into 4 groups: empty plasmid control group(EP group),bone marrow mesenchymal stem cells group(BMSC group),VEGF gene therapy group(VEGF group),combination bone mesenchymal stem cells and VEGF gene therapy group(BV group).There were 8 rabbits in each group.Angiogenesis was detected by arteriography on day 28 after treatment and expression of VEGF was detected by immunohistochemical staining on day 30 after treatment.Results There were no differences of collateral vessel count between the EP group,BMSC group and VEGF group.The collateral vessel count in BV group was higher than that of the other three groups.Immunohistochemistry of VEGF showed that the integrated optical density(IOD)in BMSC and VEGF groups increased significantly compared with the EP group; the IOD in BV group was the highest compared with the other three groups.Conclusions Combination bone marrow mesenchymal stem cells and VEGF gene in the treatment of limb ischemia in rabbits can obtain stable and effective expression of VEGF along with significant improvement of limb ischemia.
5.Functional outcomes of cochlear implantation in patients with malformed inner ear

OBJECTIVE The aim was to explore the clinical experiences and access the auditory performance and speech intelligibility of cochlear implantation in patients with inner ear malformations. METHODS Among 700 patients who received multi-channel cochlear implantation from 1997 to 2007 in Beijing Tongren Hospital, 108 patients were diagnosed with malformed inner ear. A retrospective study was performed about the surgical characteristics and the mapping characteristics after implantation. The long term follow-up of the rehabilitative efficacy will also be presented. RESULTS ①All patients restituted auditory after operation. ②After the regular rehabilitation, the long term follow-up of the auditory performance and speech intelligibility were very positive. Everyone has shown improved listening and communication skills evaluated by Categories of Auditory Performance and Speech Intelligibility Rating. ③Most of the children are studying or working and are actively involved in their local communities. CONCLUSION The cochlear implantation can be performed safely in patients with malformed inner ear. The results suggest that cochlear implantation could also provides long-term benefits to those malformed inner ear cases.
6.Determination of methyl nonyl ketone in volatile oil from herbs of Houttuynia cordata by GC-MS.

OBJECTIVETo develop a GC-MS method for the determination of methyl nonyl ketone in the volatile oil from the herbs of Houttuynia cordata.
METHODThe sample was split in the 240 degrees C injection port, with 20:1 split ratio, and separated on a DB-5 (30 m x 0.25 mm, 0.25 microm film thickness) fused silica column with helium as the carrier gas. The temperature program was as follows: 100 degrees C for 2 min, the 5 degrees C x min(-1) to 150 degrees C, then 15 degrees C x min(-1) to 200 degrees C, and kept for 10 min. The MS transfer line temperature was set to 250 degrees C, the MS source temperature was set to 200 degrees C. The ionization mode was electron ionization (EI) and the selective ion monitor mode was used.
RESULTA good linear relationship was constructed over the injection amount range of 5.5-110 ng of methyl nonyl ketone. The average recovery was 98.9%, and RSD was 2. 2%.
CONCLUSIONThe developed method was sensitive, accurate, and can be used for the determination of methyl nonyl ketone in the volatile oil and for the quality control of H. cordata.

7.Study on serum S100A12 level in patients with obstructive sleep apnea hypopnea syndrome

Objective To investigate the correlation and significance of S100A12 with obstructive sleep apnea hypopnea syndrome (OSAHS).Methods Fifty-three patients with OSAHS were chosen as OSAHS group and 46 healthy volunteers were chosen as control group.The levels of S100A12 and hs-CRP in the two groups were compared,and its relationship with those of epworth (ESS),apnea hypopnea index (AHI),and minimum blood oxygen saturation (L-SpO2) were analysised.Results The scores of ESS,BMI,A HI,L-SpO2,hs-CRP and S100A12 in two groups were statistically significant (P<0.05).The severity of hs-CRP in severe OSAHS group was significantly higher than that of mild OSAHS group (P<0.05).There was no significant difference in hs-CRP between moderate OSAHS group and mild OSAHS group and severe group (P>0.05).The level of S100A12 in severe OSAHS group was significantly higher than that of moderate OSAHS group and OSAHS mild group(P<0.05).The level of S100A12 in moderate moderate group was significantly higher than that of mild group(P<0.01).hs-CRP was negatively correlated with ESS and AHI (r=0.822,0.787,P<0.01),was positively correlated with L-SpO2 (r=-0.740,P<0.01),S100A12 was positively correlated with ESS and AHI (r =0.707,P < 0.01),and negatively correlated with ESS and AHI (r =0.707,0.807,P<0.01),and negatively correlated with with L-SpO2 (r=-0.670,P<0.01).Conclusion S100A12 is associated with OSAHS.The higher the severity of OSAHS,the higher the S100A12 value,which can be used as a new predictor of cardiovascular disease risk in OSAHS patients.

Magnetotactic bacteria (MTB), a group of phylogenetically diverse organisms that use their unique intracellular magnetosome organelles to swim along the Earth's magnetic field, play important roles in the biogeochemical cycles of iron and sulfur. Previous studies have revealed that the bacterial actin protein MamK plays essential roles in the linear arrangement of magnetosomes in MTB cells belonging to the Proteobacteria phylum. However, the molecular mechanisms of multiple-magnetosome-chain arrangements in MTB remain largely unknown. Here, we report that the MamK filaments from the uncultivated 'Candidatus Magnetobacterium casensis' (Mcas) within the phylum Nitrospirae polymerized in the presence of ATP alone and were stable without obvious ATP hydrolysis-mediated disassembly. MamK in Mcas can convert NTP to NDP and NDP to NMP, showing the highest preference to ATP. Unlike its Magnetospirillum counterparts, which form a single magnetosome chain, or other bacterial actins such as MreB and ParM, the polymerized MamK from Mcas is independent of metal ions and nucleotides except for ATP, and is assembled into well-ordered filamentous bundles consisted of multiple filaments. Our results suggest a dynamically stable assembly of MamK from the uncultivated Nitrospirae MTB that synthesizes multiple magnetosome chains per cell. These findings further improve the current knowledge of biomineralization and organelle biogenesis in prokaryotic systems.
