1.Effects of infrasound on learning and memory and the expression of ryanodine receptor in hippocampi and temporal cortex of rats

Objective To investigate the effects of 8Hz infrasound on spatial learning ability of rats and its the underlying mechanism. Methods Forty male SD rats were randomly divided into a control group and an experiment group (consisting of the 8Hz, 90dB, 100dB and 130dB subgroups). The experimental subgroups were exposed to 8Hz, 90dB, 100dB or 130dB infrasound for 2 hours daily, respectively, for a total of 6 days. All of the rats were taken out of the infrasonic depot once a day and given Morris maze training. And at last the brain of the rats were harvested and treated with immunohistochemical staining for detection of ryanodine receptor (RyR). The content of RyR in hippocampi and temporal cortex were detected under an optical microscope. Results The rats in the 8Hz, 90dB, 100dB and 130dB subgroups showed prolonged latency in Morris training ( P =0.010, P =0.001 and P = 0.000 ). The hippocampi and temporal cortex of rats in 8Hz, 90dB, 100dB and 130dB subgroups showed less positive neurons of RyR ( P =0.000). There was a negative correlation between the latency period of rats finding the hidden platform and the content of RyR in hippocampus and temporal cortex. Conclusion Infrasound of 8Hz, 90dB, 100dB and 130dB could decrease the spatial learning ability of rats, and this was possibly related to the reduce of RyR in hippocampus and temporal cortex.

Objective To study the preventive effect of paeonol on restenosis after angioplasty in rabbit balloon injury model and its mechanism.Methods Male New Zealand white rabbits with clean grade were selected.Balloon injury rabbit abdominal aorta combined with high cholesterol diet were used to establish rabbit atherosclerosis model.We used balloon to dilate the narrow abdominal aorta.The animals were randomly divided into four groups (control group,low concentration paeonol group,high concentration paeonol group and rapamycin group).Animals in different group were orally given normal saline 2 ml · kg-1 · d-1,low concentration of paeonol 75 mg · kg-1 · d-1,high concentration of paeonol 150 mg · kg-1 · d-1,rapamycin administrated with 1.25 mg · kg-1 · d-1 for six weeks respectively.After 6 weeks,the animals were killed by air embolism,and immediately dissected to remove the abdominal aorta.We used hematoxylin and eosin(HE) staining and Masson staining to detect the thickness of the arterial intimal hyperplasia and the gross morphological changes of the narrow artery.Immunohistochemical staining was used to detect changes of proliferating cell nuclear antigen (PCNA) expression level.Western blotting was used to discover P27kip1 protein expression in the narrow artery.Results Paeonol and rapamycin reduced the degree of intimal hyperplasia.The neointima areas in the control group,low concentration paeonol group,high concentration paeonol group and rapamycin group were (0.16±0.03),(0.09±0.01),(0.05±0.01) and (0.04±0.01) respectively; the ratio of neointimal area to media area was (90.62 ±3.43),(42.30 ±2.94) (33.72±3.19) and (28.78±6.67) respectively; the intimal area proliferation rate was (33.3 ± 3.1) %,(23.2 ± 2.2) %,(17.5 ± 1.5) % and (16.2 ± 1.7) % respectively.There were significant differences in the neointima area,ratio of neointimal area to media area,and the intimal area proliferation rate among the four groups (F=47.98,257.25,74.60,all P<0.05).The expression level of PCNA and P27kip1 protein were (0.20±0.04),(0.06±0.02),(0.05±0.02) and (0.08± 0.02),and (0.47±0.09),(0.78±0.20),(1.29±0.12) and (1.83±0.13) in the control group,low concentration paeonol group,high concentration paeonol group and rapamycin group respectively,and the differences were statistically significant between the four groups (F=37.22 and 107.69,both P<0.01).Conclusions Paeonol plays a role in anti-vascular smooth muscle cell proliferation.This effect may be through inhibiting the expression of PCNA,and increasing the expression of P27kip1 in the cell cycle.
5.Application of self-made negative pressure drainage device for extremity soft tissue trauma

Objective To investigate the clinical value of self-made dressing negative pressure drainage technique in the treatment of the four limbs soft tissue trauma.Methods A total of 60 patients with the four limbs soft tissue trauma in our hospital from December 2013 to December 2014 were randomly divided into the observation group and control group.The observation group with 30 cases were treated with the self-made negative pressure drainage technique,and the control group with 30 cases received the conventional treatment.The pain score,wound healing rate,complication rate between two groups were compared.Results The pain score of the observation group was(1.17±0.22),which was lower than(3.17±0.49) of control group,the difference was statistically significant(P<0.05).The wound healing rate of observation group(73.33%) was higher than that of the control group(40%),the difference was statistically significant(P<0.05).The complication rate of observation group(3.33%) was lower than that of control group(23.33%),the difference was statistically significant(P<0.05).Conclusion The self-made dressing negative pressure drainage technique applied to the patients of four limbs soft tissue trauma can reduce the pain,shorten the healing time,and reduce the incidence of complications.
6.Effects of infrasound on hippocampus ultrastructure of rats

ObjectiveTo investigate effects of 8Hz 90dB and 8Hz 130dB infrasound on hippocampi ultrastructure of SD rats.Methods60 male SD rats were randomly divided into the control group and experimental group. Rats of experimental group were exposed to 8Hz 90dB or 8Hz 130dB infrasound 2 hours every day for 1,7,14, 21,28,35 and 42 days. As soon as the exposure finished,the right hippocampus of rat was taken and examined by electron microscopy.ResultsHippocampus ultrastructure of rats was changed after only one day exposure to infrasound and the longer exposure to infrasound, the worse ultrastructure changed. But during the latter period,damages of hippocampus ultrastructure became lighter. Different rules were observed between 90dB and 130dB groups.ConclusionInfrasound of 8Hz 90dB and 8Hz 130dB can make hippocampus ultrastructure of rats damaged mainly as denaturalizations. These changes are nonliner with time, and the hippocampus of rats has adaptability to infrasound.
7.Significance of tyrosine kinase receptor gene and its variant expressions in bladder tumor tissues

ObjectivesTo explore the clinical significance of tyrosine kinase receptor RON mRNA expression and it's splicing variant in bladder tumors. Methods Sixty-three cases of transitional cell carcinoma of the bladder (TCCB), including 30 cases of pathological grade I , 15 cases of pathological grade Ⅱ and 18 cases of pathological grade Ⅲ (44 cases of clinical stage Tis + T1, 15 cases of T2 + T3 +T4), 7 inverted papilloma of the bladder ( IPB), 9 cases of papillary urothelial neoplasm of low malignant potential (PUNLMP) and 12 cases of normalbladder mucosa RT-PCR was employed with the internal standard (GAPDHmRNA) to detect the expression of RON mRNA. PCR and direct sequencing was then utilized to identify the potential RON mRNA splicing variants. Finally, the variants' positive rates of expression were analyzed among the different tissues, diverse TCCB pathological grades and clinical stages. ResultsThe expression levels of RON mRNA/GAPDH mRNA among TCCB, IPB, PUNLMP and normal control were 4. 9 × 10-3 ( 1. 8 × 10-3-1.0 × 10-2 ), 3. 8 × 10-3 (2. 4 × 10-3-1.7 × 10-2 ), 4. 9 ×10 -3 ( 1.7 × 10 -3-1.1 × 10 -2 ) and 1.0 × 10-3 (4. 5 × 10-4-2. 8 × 10-3 ) respectively. The difference had a statistical significance (x2K-W = 17. 278 ,P <0. 05 ). The expression levels among pathological grade I, Ⅱ andⅢ were 3.7 × 10-3( 1.3 × 10-3-7.5 × 10-3) , 4. 9 × 10-3(1.9 × 10-3-1.1 × 10-2) and 8.9 × 10-3(2. 7 ×10 -3-8.0 × 10 -2 ) respectively. The erpression levels among the clinical stage Tis + T1 and T2 + T3 + T4were 3.5 × 10-3 ( 1.2 × 10 -3-7. 7 × 10-3 ) and 9. 7 × 10 -3 ( 2. 9 × 10-3-5. 3 × 10-2 ). The differences between expression levels were of statistical significance among the different pathological grades ( x2k-W =7. 341, P <0. 05 ) and clinical stages ( Z = - 2. 306, P < 0. 05 ). The positive rates of exon 11deletion(E11△) in TCCB, IPB and PUNLMP were 71% (45/63), 57% (4/7) and 67% (6/9) respectively, andthe total positive rate in bladder tumor tissues was 70%. Meanwhile, expression of the novel RON variant wesnot detected in the normalbladder mucosa. The positive expression rate of E1 1△ has no significant correlationamong the different clinical pathological tissues (x2 = 0. 620, P > 0. 05 ). There was no statistical significancein expression positive rate between different pathological grades ( Z =0. 221, P >0. 05 ) and clinical stages( Z = 0. 538, P > 0. 05) as well. A novel RON splice variant, deletion of RON exon 11 3 476 - 3 539 ( E3476 -3539△) was fond in the pathological tissue. The positive expression rates of E3 476 -3 539 in TCCB,IPB and PUNLMP were 57% (36/63), 43% (3/7) and 56% (5/9) respectively, and the total positive expression rate was 56% (44/79). The positive rates of E3 476 -3 539△ in pathological grade I , Ⅱ and Ⅲ were 40% ( 12/30), 67% (10/15) and 78% (14/18), and it's positive rates in clinical stage Tis +T1and T2 +T3 + T4 were 48% (21/44) and 80% (12/15). The differences in each group had significantly statistical significance ( Z = 7. 285, 5. 041, P < 0. 05 ) . However, the positive rates amongdifferent pathological tissues had no significance (x2 = 0. 517, P > 0. 05 ). Conclusions The expression level of RON mRNA is significantly associated with histological grading and clinical stage. RON may play an important role in the progression ofTCCB. Compared with the normal control, the increased RON variant expression may contribute to the carcinogenesis of the bladder tumor.
8.CT and MRI manifestations of the intrathoracic ganglioneuroma

ObjectiveTo evaluate CT and MRI findings of the intrathoracic ganglioneuroma and to improve its diagnosis and differential diagnosis ability.MethodsClinical,CT( n = 14),MRI (n = 6) and pathology manifestations of 20 patients with the intrathoracic ganglioneuroma were retrospectively analyzed.All 20 cases had chest CT and MRI plain scanning and multiphase enhance scanning before operation.ResultsSeventeen of 20 lesions were located in posterior mediastinum,2 in pleura side and 1 in right thorax cavity.The CT value of the plain scans ranged from 20 to 40 HU ( mean 30.5 HU),Tubercle calcification were detected in four masses,one case with fat density was showed on CT scanning.After injecting contrast media,CT value ranged from 0 to 12 HU (mean 6.2 HU) in artery phase,ranging from 10 to 20 HU ( mean 14.3 HU) in delay phase.Five of 6 cases of MRI signals were homogeneously low intensity on T1 WI,1 case with fat signal was imhomogeneously low intensity on T1WI.Six cases were imhomogeneously high intensity on T2WI.A whorled appearance was visualized in one tumor on T2WI.The post-contrast enhancement MR images was slight enhancement imhomogeneously in artery phase and gradual increasing enhancement in delay phase.ConclusionOn CT and MR imaging,no enhancement or slight enhancement in artery phase and gradual increasing enhancement in delay phase are characteristic manifestations of ganglioneuroma in the thorax.
9.The relationship between positive experience and social support of spouses' of community stroke patients

Objective To investigate the relationship between positive experience and social support of spouses' of community stroke patients.Methods 100 stroke patients and their spouses in two communities were conveniently selected and investigated about their positive experience and social support with General Information Questionnaire (GIQ),Modified Barthel Index(MBI),Positive Aspects of Caregiver (PAC),Perceived Social Support Scale (PSSS).Results The mean score of the spouses' positive experience was (22.74±5.85); the dimension of serf-affirmation was (15.82±3.84),higher than the dimension of life expectation(6.92±2.95).The scores of the spouses' positive experience were significantly different among age,gender,education of spouses and age and ability of daily life of stroke patients; the score of the spouses' positive experience was positively correlated with social support,subjective support,utility of support;multiple linear regression showed that age of spouses,ability of daily life of stroke patients and utility of support were influential important factors of spouses' positive experience.Conclusions The spouses' positive experience was below average level,we must pay more attention to improve it,utility of support can improve spouses' positive experience.
10.A bone filling mesh container for treating senile osteoporotic vertebral compression fractures combined with intravertebral cleft sign

BACKGROUND: Percutaneous vertebroplasty is an effective method for osteoporotic thoracic vertebral compression fracture, but bone cement leakage is easy to occur in patients with intravertebral cleft. OBJECTIVE: To explore the therapeutic efficacy of bone-filling mesh containers in elderly patients with osteoporotic vertebral compression fractures combined with intravertebral cleft sign. METHODS: From October 2017 to July 2018, 62 patients aged from 60 to 80 years with osteoporotic vertebral compression fractures combined with intravertebral cleft sign were admitted at the Affiliated Hospital of Qinghai University. Among them, 31 cases were treated with unilateral percutaneous vertebroplasty (control group), and the other 31 cases were treated with bone-filling mesh containers (study group). The leakage of bone cement, the time of operation and the times of fluoroscopy were recorded in the two groups. The visual analogue scale score and Oswestry disability index score were compared between the two groups before and 7, 30, 60 days after treatment. The height changes of injured vertebrae were evaluated by X-ray before treatment and 7 days after treatment. The quality of life was evaluated by the MOS item short from health survey (SF-36) before and 60 days after treatment. RESULTS AND CONCLUSION: (1) The time of operation and the times of fluoroscopy in the study group were less than those in the control group (P < 0.05), and the leakage rate of bone cement in the study group was lower than that in the control group (6% vs. 39%, P < 0.05). (2) The visual analogue scale score and Oswestry disability index score of the two groups were significantly improved at 7, 30 and 60 days after treatment. The visual analogue scale scores of the study group were lower than those of the control group at different time points after treatment (P < 0.05). There was no difference in Oswestry disability index scores between the two groups at different time points after treatment (P> 0.05). (3) The height of injured vertebrae 7 days after treatment in both groups was significantly higher than that before treatment (P < 0.05), and there was a significant difference between the two groups (P < 0.05). (4) The quality of life of the two groups was significantly improved at 60 days after treatment (P < 0.05), but there was no difference between the two groups (P> 0.05). These findings indicate that compared with unilateral vertebroplasty with bone cement injection, bone-filling mesh containers with bone cement injection could reduce the incidence of cement leakage, relieve pain and increase the height of injured vertebrae in elderly patients with osteoporotic vertebral compression fracture combined with intravertebral cleft sign.
