1.Effect of right stellate ganglion block on heart rate variability in patients with carbon dioxide pneumoperitoneum

Objective To invetstigate the effects of right stellate ganglion block (SGB) on heart rate variability (HRV) in patients with carbon dioxide pneumoperitoneum.Methods Forty ASA Ⅰ or Ⅱ patients of both sexes,aged 39-75 yr,weighing 47-75 kg,undergoing elective laparoscopic cholecystectomy,were randomly divided into 2 groups ( n =20 each):control group (group C) and right SGB group (group SGB).Right SGB was performed with 1% lidocaine 7 ml at 10 min before induction of anesthesia.The low frequency power ( LF),high frequency power (HF),total power (TP) and LF/HF ratio were recorded before SGB (baseline,To ),at 5 and 30 min of pneumoperitoneum (T1,2),at 5 min after deflation (T3) and at the end of operation (T4 ).The LF in normalized units (LFnu) and HF in normalized units (HFnu) were calculated.Results Compared with the baseline values at T0,LF,LFnu and LF/HF ratio were significantly increased at T1-4,HF was significantly decreased at T3,4,and HFnu was significantly decreased at T2-4 in group C,and HF was significantly increased at T1-4,HFnu was significantly increased at T2-4 and LF/HF ratio was significantly decreased at T1-4 in group SGB ( P < 0.05 or 0.01 ).LF,LFnu and LF/HF ratio were significantly decreased at T1-4,and HF at T1-4 and HFnu at T2-4 were significantly increased in group SGB as compared with group C ( P < 0.05 or 0.01 ).Conclusion Right SGB can not only inhibit the sympathetic activity induced by carbon dioxide pneumoperitoneum,but also increase the vagal tone.
2.Improvement of memory function of mice by Funaoling Oral Liquid

ObjectiveTo explore the effect of Funaoling Oral Liquid on improving memory function of mice.Methods30 mice were injected with scopolamine 1mg/kg to make model of memory obstacle after treated by Funaoling Oral Liquid,piracetam or saline for 10 days.The escape test was used to measure the memory function.The blood circulation obstacle of the mice in mesentery was made by norepinephrine,and then, Funaoling Oral Liquid, Happy-hundred-year Oral Liquid and saline were given to observe the recovery time. The serum superoxide dismutase(SOD) activity in mice was measured after using different dosage of Funaoling Oral Liquid(75g/kg,37.5 g/kg and 18.75g/kg),piracetam or saline for 10 days.ResultsFunaoling Oral Liquid improved the memory function and blood circulation of mesentery significantly compared with that of the model(P<0.05 or P<0.01). The serum SOD activity in mice which was treated by Funaoling Oral Liquid 18.75g/kg were higher than that of model(P<0.05).ConclusionsThe Funaoling Oral Liquid can improve the memory and blood circulation of mice after injured,as well as to enhance the activity of SOD.
3.Clinical study of early intervention treatment of aneurysmal subarachnoid hemorrhage in the elderly

Objective To investigate the safety and feasibility of early intervention treatment for aneurysmal subarachnoid hemorrhage in the elderly. Methods Within 6 to 72 hours after hemorrhage,89 aneurysms were detected by overall brain angiogram in 84 elderly patients(aged above 60 years)Who were diagnosed as subarachnoid hemorrhage on CT.HUNT-HESS grade was as following:26 cases of grade Ⅱ,31 cases of grade Ⅲ,25 cases of grade Ⅳ,2 cases of grade Ⅴ.Interventional embolization treatment was performed immediately.Aceording to the structure of the aneurysm,pure coil embolization,balloon assisted technique and combined with stent technique were performed.Anhydration,fluid expansion,vessel dilatation,lumbar puncture and/or lumbar drainage were performed after procedure.At the same time,the complications were treated. Results Eighty-three aneurysms were embolized successfully in 89 patients.The Success rate of the technique was 93.3%.The Glasgow Outcome Seale(GOS)score at 3 months post procedure showed a good result in 68 patients.medium disability in 7 patients,severe disability in 4 patients,death in 5 patients (1 case was related to the technique). Conclusions Interventional embolization iS a safe and effective therapeutic approach in elderly patients with subarachnoid hemorrhage.
4.The role of miR-34a in AngⅡinduced myocardial hypertrophy via alteration of AMPK/mTOR signal pathway

Objective To explore the effects of miR-34a and the alteration of AMPK/mTOR signal pathway on angiotensin (Ang)Ⅱ-stimulated cardiomyocytes hypertrophy. Methods Neonatal rat cardiomyocytes were cultrued in vitro and were divided into 3 groups:negative control for lentivirus carrying miR-34a group (NC), AngⅡplus lentivirus carrying negative control group (AngⅡ+NC) and AngⅡplus lentivirus carrying miR-34a group (AngⅡ+miR-34a). The relative cell area was detected by confocal microscopy. The expression of miR-34a and hypertrophy-related genes (ANP and β-MHC) were analyzed by real time PCR. The AMPK/mTOR signal pathway was measured by Western blot assay. Results Compared to NC group, the relative cell area was increased in AngⅡ+NC group (P<0.05). But compared with AngⅡ+NC group, the relative cell area was decreased in AngII+miR-34a group (P<0.05). Moreover, compared with NC group, the expression of miR-34a was decreased, and the expression of hyperthophy-related genes(ANP and β-MHC) was up-regulated in AngⅡ+NC group. However, the expression of miR-34a was decreased, and the expression of hyperthophy-related genes (ANP and β-MHC) was down-regulated (P<0.05). Finally, compared to NC group, the ratio of phosphop-AMPK/AMPK was significantly induced in AngII + NC group, but the ratio of phosphop-mTOR/mTOR was significantly decreased (P<0.05). However, compared to AngⅡ+NC group, the ratio of phosphop-AMPK/AMPK was significantly decreased in AngII + miR-34a group, but the ratio of phosphop-mTOR/mTOR was significantly increased (P<0.05). Conclusion miR-34a is able to inhibit myocardial hypertrophy of neonatal rat cardiomyocytes, and its mechanism is partly carried out by the alteration of the signal pathway of AMPK/mTOR.
5.Investigation on in-stent recurrent stenosis after stenting of the extra-and intracranial artery in the elderly

Objective To investigate the distribution of in-stent recurrent stenosis (1RS) after balloon-angioplasty and stenting in elderly cerebral ischemic patient and its correlative factors.Methods Totally 158 elderly cerebral ischemic patients with extra-and/or intracranial artery stenosis were treated with 172 stents in our hospital.The inhibition of platelet function was assessed by using the thrombus elasticity chart system in perioperative period,and the anti-platelet agents were adjusted correspondingly.150 patients were followed up by digital subtraction angiography (DSA) and the other 8 patients were followed up by CT angiography,during an average follow-up duration of 8.3 months.Results There were 11 (6.4%) IRS lesions,among which 10 lesions were at the vertebral artery origin and 1 lesion was at common carotid artery bifurcation.23.8% of IRS happened after vertebral artery origin stenting,and all in-stent recurrent stenosis were in the origin of non-dominant vertebral artery,whose IRS rate was 66.7%.Conclusions IRS are more common found in vertebral artery origin than in other parts of vertebral artery and are more common found in nondominant side than in dominant side.
6.The study of executive function in patients with mild traumatic brain injury

Objective To explore the executive function in patients with mild traumatic brain injury (mTBI). Methods The evaluation of executive function was conducted in 63 mTBI patients in 1 week, 1, 3 and 6 month after injury and 60 health controls. The executive function was evaluated using Trail Making Test (TMTA), Hopkins Verbal Learning Test-revised (HVLT-R), Brief Visuospatial Memory Test-revised (BVMT-R), Stroop Color Word Tes and Con-tinuous Performance Test (CPT). Results There were significant differences between mTBI patients and controls in TM-TA, HVLT-R, BVMT-R, Stroop Color Word Test and CPT a week following TBI (P<0.05). There were a significant in-crease in scores of TMTA and a significant decrease in HVLT-R, BVMT-R, stroop and CPT-IP at 1, 3 and 6 month compared with 1 week following TBI (P<0.05). There were a significant decrease in TMTA and a significant increase in BVMT-R and stroop 1 month compared with 1 week following TBI (P<0.05). There were a significant increase in stroop, BVMT-R and CPT-IP at 3 month compared with 1 month following TBI (P<0.05). There was a significant increase in stroop and CPT-IP at 6 month compared with 3 month following TBI (P<0.05). Conclusions Patients with mTBI have executive function impairment. The recovery times of different executive function are different in patients with mTBI.
7.Role of miRNA-204 targeted LC3B in Ang Ⅱ induced myocardial hypertrophy

Objective To explore the effects of miRNA-204 targeted LC3B expression on Ang Ⅱ induced cardiomyocytes hypertrophy.Methods The primary neonatal rat cardiomyocytes served as the research objects and divided into the control group,AngⅡ group,combination-treated group 1 (cardiomyocytes were given Ang Ⅱ stimulation,meanwhile infected by negative control lentivirus vector),combination-treated group 2 (cardiomyocytes were given Ang Ⅱ stimulation,meanwhile infected by lentivirus carrying miRNA-204 overexpression vector) according to different treatments.About 48 h to 72 h after intervention treatment,the cardiomyocyte hypertrophy change was detected by confocal microscopy,the expression of miRNA-204 was analyzed by real time PCR,the protein expression of LC3B was measured by Western blot and targeted gene of miRNA-204 was demonstrated by dual-luciferase reporter assay system.Results Compared with the control group,the cardiomyocyte relative surface area in the Ang Ⅱ group was significantly enlarged,the protein expression of LC3B was significantly increased,the expression of miRNA-204 was upregulated,the differences were statistically significant (P<0.05).Whereas comparing the combination-treated group 1 with combination-treated group 2,the protein expression of LC3B in the latter was down-regulated and the cell area was reduced (P<0.05).The further luciferase activity report gene experiment results suggested that miRNA-204 was able to bind to LC3B 3'-UTR and decreased the luciferase activities (P<0.05),but not to bind its mutated fragment for inactivating luciferase activity(P>0.05).Conclusion miRNA-204 is able to inhibit Ang Ⅱ induced cardiomyocytes hypertrophy,its action is realized by targeting the expression of LC3B.

This study was designed to prove simultaneously blocking both EGFR and COX-2-medi-ated pathways may be an efficient means of inhibiting cancer cell growth in NPC. Method: A combination of tarceva (EGFR-selectivetyrosine kinase inhibitors) with celecoxib(Cox-2 inhibitor) was studied on its effects on cell growth, cell cycle progression, apoptosis and protein expression in CNE-2 cell lines by cell growth assay, flow cy-tometric analysis assay and Western blotting. Result: ①The inhibition rate of cell growth was higher in the group treated with a combination of two agents than that the sum of rates of the two groups treated with only one agent (P<0.05). ②The combination of tarceva with celecoxib significantly induced G_1 arrest(P<0.05) ,but did not in-crease apoptosis rate(P>0.05). ③The group of combination showed less expressions of p-EGFR and COX-2 than any other group. Conclusion Simultaneously blocking EGFR and COX-2 mediated pathways would significantly in-hibit the growth of CNE-2 cell line, increase G_1 arrest and reduce the expression levels of p-EGFR and COX-2.
9.Percutaneous induced thrombosis closure on small patent ductus arteriosus solely under echocardiography guidance

Objective To investigate the efficacy and safety of percutaneous induced thrombosis closure on small patent ductus arteriosus (PDA) solely under echocardiography guidance.Methods From June 2013 to September 2016,276 PDA patients were treated by femoral artery retrograde approach under echocardiography guidance.Twenty-five patients (14 boys,11 girls) among them with small PDA received percutaneous induced thrombosis closure because the delivery sheath could not pass PDA,with mean age (3.7 ± 1.6) years,mean body weight (16.6 ±5.4) kg and the mean diameter of the most narrow PDA was (1.3 ± 0.4) mm.All patients were assessed by transthoracic echocardiography,chest radiography and electrocardiography preoperatively.The intraoperative and postoperative effects were evaluated by echocardiography.Outpatient assessment was performed at 1,3,6 and 12 months after operation with electrocardiography and echocardiography,and yearly thereafter.Results Twenty-five patients were successfully treated with percutaneous induced thrombosis closure solely under echocardiography guidance.The mean operation time was (35.2 ± 6.1) minutes.The immediate residual shunt rates and 24 hours after operation were 28.0% (7/25 cases) and 16.0% (4/25 cases),respectively.All patients recovered and were discharged from hospital.The average hospitalization stay was (3.0 ± 0.6) days.The mean follow-up was (16.8 ± 12.3) months,residual shunt was observed in 2 patients (8.0%),and it decreased from 1.5 mm and 2.0 mm preoperatively to 0.8 mm and 1.0 mm postoperatively,respectively.All patients survived without peripheral vascular injury,cardiac perforation,PDA rupture or thrombembolia.Conclusions Percutaneous induced thrombosis closure of small PDA solely under echocardiography guidance is safe and effective.It can avoid occluder implantation and radiation injury.However,the residual shunt after operation needs following up closely.
10.In vitro experimental study of killing Hep-2 cells of laryngeal cancer with suicide fusion gene CDglyTK.

OBJECTIVE:
To study the killing effect of suicide gene CDglyTK combined with GCV or 5-FC on the human laryngeal carcinoma Hep-2 cell line in vitro.
METHOD:
Constructed plasmid pcDNA3.1 (-) CMV. CDglyTK was verified by enzyme digestion of Xho I /Hind III and automatic sequence analysis, then it was introduced into Hep-2 cells by electroporation to yield cells expressing CDglyTK stably after selecting with G418(400 ng/L) for 14 da. The expression of CDglyTK mRNA in transfected Hep-2 cells was tested by RT-PCR. Compared with Hep-2 cells transferred with pcDNA3.1(-), in vitro chemosensitivity of CDglyTK-expressing Hep-2 cells to 5-FC, GCV or 5-FC + GCV was detected by MTT assay.
RESULT:
The recombinant plasmid contained full-length coding region sequence of CD and TK gene. A anticipated 707 bp fragment was amplified from total RNA of CDglyTK-expressing Hep-2 cells by RT-PCR and a fusion protein of 59 000 was detected in cell extract from transfected Hep-2 cells. In vitro study growth of CDglyTK-positive Hep-2 cells were inhibited by 5-FC, GCV or 5-FC + GCV respectively, and the antitumour effect of 5-FC + GCV is superior to 5-FC or GCV.
CONCLUSION
CDglyTK may be a candidate for treating human laryngeal cancer.
