1.Caveolae and multidrug resistance of tumor cells

Caveolae is a specified micro-domain of plasma membrane, which consists of caveolin and many lipid molecules and membrane proteins. Caveolae plays important roles in internalization of extra-membrane molecules, transmembrane signal transdution and transport of cholesterin. Recent studies indicated that caveolae and its components were up-regulated in multidrug resistant tumor cells and might participate the development of multidrug resistance of tumor cells. This paper concentrated on the role of caveolae in tumor multidrug resistance.
2.Intervention effects of atorvastatin calcium tablets on soluble CD40 ligand and C-reactive protein in patients with cardiac angina

ObjectiveTo investigate the intervention of level of atorvastatin calcium tablets (lipitor) on soluble CD40 (CD40L) and high sensitivity C-reactive protein (hs-CRP) in patients with angina.Methods 65 patients with coronary heart disease were divided into stable angina (SA group,n= 30) and unstable angina (UA group,n= 35).The vein blood was collected in the hospitalized morning,2,4,6 weeks after treatment and serum levels of hs-CRP and CD40L were measured and compared between the two groups.ResultsThe levels of soluable CD40L [(20.52± 2.91)μg/L] and CRP [(7.96±1.69) mg/L] in UA group were higher than SA group [(7.96±-1.35) tg/L and ( 1.58 ± 0.91 ) mg/L] (t = 21.705、18.493,both P< 0.05) before treatment,and after treatment,their levels in the two groups were significantly lower than pre-treatment (P<0.01).ConclusionsCD40L and hs-CRP may involved in the pathophysiology of unstable angina process and can be used as an indicator reflecting vulnerable plaque.Lipitor might enhance stability of atherosclerotic plaque and prevent acute coronary events by reducing levels of CD40L and hs-CRP.

4.The efficacy and safety evaluation of tolterodine tartrate in treating patients with early overactive bladder symptoms after transurethral resection prostate

ObjectiveTo evaluate the efficacy and safety of tolterodine tartrate in treating patients with early overactive bladder(OAB) symptoms after transurethral resection prostate (TURP).Methods Thirty-one patients who received TURP and were found OAB at recent follow-up were enrolled and divided by random digits table method into study group ( 16 cases) and control group ( 15 cases).The patients in study group were treated with 2 mg tolterodine tartrate twice a day for four weeks,while the changes of OAB in control group were observed.The OAB symptoms score (OABSS),maximum flow rate(Qmax),24 h urination and adverse reactions during the period of medication of two groups before and after treatment were recorded and observed.Results The second evaluation,all the indexes except Qmax in study group improved significantly compared with those of the first evaluation in study group and control group the second evaluation.The indexes included urination times[(6.8 ± 1.0) times vs.( 12.5 ± 1.5) times,(11.8 ± 1.2)times],urgency times[(1.4 ± 1.1) times vs.(4.1 ±2.2) times,(4.1 ±2.3) times],urine volume [(214 ±36) ml vs.( 177 ± 46) ml,( 178 ± 44) ml ],nocturia times [ ( 1.9 ± 0.7) times vs.(2.9 ± 1.3 ) times,(2.8 ±1.4) times ],urge incontinence times [ (0.6 ± 0.5 ) times vs.( 1.6 ± 1.0) times,( 1.5 ± 1.0) times ].OABSS in study group the second evaluation was significantly lower than that in the first evaluation in study group and the second evaluation in control group [ (3.6 ± 1.8 ) scores vs.( 7.6 ± 3.3 ) scores,(7.4 ± 3.2) scores,P < 0.01 or < 0.05 ].There was no statistical significance in all the indexes in control group between the first evaluation and the second evaluation (P > 0.05).There were 2 cases with adverse reactions which performed as dry mouth which could be tolerant.ConclusionTolterodine tartrate is efficient and safe in treating patients with early OAB symptoms after TURP.
5.Effects of stretch load on the ultramicrostructure and c-kit gene expression of the bladder ICCs in rats

Objective To investigate the effects of stretch load on the ultramicrostructure and c-kit expression of the bladder interstitial cells of cajal (ICCs) in rats. Methods Rat model with detrusor overactivity due to partial outflow obstruction with bladder neck ligation were established. The effects of stretch load on the changes of ultramicrostructure of the bladder ICCs were observed by scanning electron microscopy. The changes in c-kit gene expression of ICCs in stable, overactive, and normal detrusors were analyzed with RT-PCR. Results The ultramicrostructure of the ICCs changed significantly. 6.Expression of Bcl-2 in Gastric Cancer Cells Transducted with Antisense Bcl-2

By molecular cloning technique, the expressing plasmids pDOR-SV40-Bcl-2 cDNA were successfully constructed. The reconstructed plasmids with lipofectamine were transduced into gastric cancer cell line SGC7901 and then the positive clones which contained the reconstructed plasmids were choosed by G418. Circa 150 positive clones were choosed from 2 x 10_5 gene transduced cells, which suggested that the transducing efficiency was more than 1%o; 2 positive clones were expanded and passage, then 1 drug-resistance cell strain (SGC7901 anBcl-2 cells) was obtained. The bloting results suggested, Bcl-2 cDNA were expressed in both gene transduced and not transduced cell strains, but the expressing level of mRNA and protein in gene transduced cell strain was very low than that in gene not transduced cell strain were positive by the means of Southern blot, Northern blot and Western blot. This results showed that normally in gastric cancer cells the expressing level of Bcl-2 gene was very high, and the cDNA fragment of antisense Bcl-2 were successfully transduced into gastric cancer cells, and in gene transduced cell strains the expressing of Bcl-2 gene was effi-cientlv blocked.
7.Comparison of two absorbable sutures in abdominal wall incision

BACKGROUND:In resent year,as the biological materials improving,all kinds of sutures are used widely in clinic. It is an important problem how to select the exact one for surgeon. OBJECTIVE:To compare the effect of antibacterial and poly (p-dioxanone) absorbable sutures (PDS) in abdominal wall incision. DESIGN,TIME AND SETTING:A contrast observation was performed at the Department of General Surgery,the Second Affiliated Hospital of Shantou University Medical College between January 2005 and August 2007. PARTICIPANTS:A total of 450 laparotomy patients,including 230 males and 220 females aging 18-78 years with the mean age of (40?9) years were collected in this study. METHODS:The 450 patients were randomly divided into three groups,including the antibacterial absorbable suture (AAS; abdominal wall incision except the skin was successively sutured with triclosan-coated antibacterial absorbable suture),PDS group (abdominal wall incision except the skin was successively sutured with PDS suture),and control group (abdominal wall incision was treated with common suture),with 150 cases in each group. All patients were followed up for 12-24 months. MAIN OUTCOME MEASURES:The visual analogue scale (VAS; point 0:indolence; point 10:severe pain),the incision heal classification and complications including incisional infection,rejection reaction,scar formation,incisional disruption and incisional hernia. This paper reviewed and analyzed the management and performance of 43 key clinical pmjeets in recent ten years in Fudan University(the clinical part of original Shanghai Medical University).It was found that hardware equipments of clinic,the ability of disease prevention,diagnosis and treatment obviously improved,the talent training and personnel structure received outstanding outcome,the academic level of basic researches got obvious promotion,the preponderant disciplines established gradually and consolidated,and the center radiation effect showed preliminary success under the aid of key project fund.Constructing the key clinical projects should boost actively amalgamation of medicine and sciences and pay a great attention to develop the medicine auxiliary discipline to further enlarge the clinical research achievements and fully display the guidance function for the development of our country~clinical medicine discipline under the new situation.
9.Effect of nicotinic acid on hyperphosphatemia in maintenance hemodialysis patients

Objective To assess the effect of nicotinic acid on hyperphosphatemia in maintenance hemodialysis (MHD) patients.Methods Twenty-six MHD patients with stable disease condition and hyperphosphatemia that could not be successfully controlled by conventional methods were selected.All patients were treated with oral nicotinic acid for 20 weeks.The changes of serum calcium,high sensitivity Creactive protein (hs-CRP),albumin (ALB),aspartate aminotransferase (AST),alanine aminotransferase (ALT),gamma-glutmyltransferase (GGT),total bilirubin (TBIL),derect bilirubin (DBIL),high density lipoprotein cholesferol (HDL-C),low density lipoprotein cholesferol (LDL-C),triacylglycerol (TG),total cholesterol (TC),intact parathyroid hormone,hemoglobin,platelet count,etc were observed before and after treatment.Results Two patients withdrew from this study because.of pruritus and diarrhea after two weeks of oral nicotinic acid.In 24 cases,compared with that before treatment,the serum phosphate,calcium-phosphorus product was decreased [(1.49 ± 0.19) mmol/L vs.(2.13 ± 0.16) mmol/L,(4.40 ± 0.49)mmol2/L2 vs.(5.45 ± 0.36) mmol2/L2] and HDL-C was increased [(1.45 ± 0.14) mmol/L vs.(1.18 ± 0.15)mmol/L] after treatment,and there was significant difference (P < 0.05).There was no significant difference in the serum calcium,iPTH,LDL-C,TG,TC,ALB,AST,ALT,GGT,TBIL,DBIL,hs-CRP,platelet count,hemoglobin between before and after treatment (P > 0.05).Conclusions Nicotinic acid is effective as all adjuvant measure in control of hyperphosphatemia in patients on MHD and can increase HDL-C.It provides a new approach for the treatment of hyperphosphatemia.
