1.Trends in incidence of malignant tumors in Yuyao City from 2011 to 2018

Objective:
To investigate the trends in incidence of malignant tumors in Yuyao City, Zhejiang Province from 2011 to 2018, so as to provide insights into formulation of the malignant tumor control strategy.
Methods:
Data pertaining to the incidence of malignant tumor in Yuyao City from 2011 to 2018 were captured from the Chronic Disease Surveillance Module of Ningbo Municipal Digital Disease Control and Prevention Platform. The crude incidence, Chinese population-standardized incidence, global population-standardized incidence, cumulative incidence (0 to 74 years of age), and truncated age-standardized incidence (35 to 64 years of age) of malignant tumors were estimated, and the trends in incidence of malignant tumors were analyzed in Yuyao City from 2011 to 2018 using average annual percentage change (AAPC).
Results:
The crude incidence, Chinese population-standardized incidence, global population-standardized incidence, cumulative incidence (0 to 74 years of age), and truncated age-standardized incidence (35 to 64 years of age) of malignant tumors were 433.78/105, 289.37/105, 224.22/105, 25.83% and 83.34/105 in Yuyao City from 2011 to 2018, with AAPC of 5.90%, 3.31%, 3.31%, 2.84% and 4.24%, respectively (all P<0.05), and the incidence of malignant tumors appeared an overall tendency towards a rise. The incidence of malignant tumors increased with age in Yuyao City from 2011 to 2018, and the incidence of malignant tumors was significantly higher in women than in men at ages of 20 to 54 years, while higher incidence was seen in men than in women at ages of 55 years and older. The ten most common malignant tumors included lung cancer, gastric cancer, colorectal cancer, liver cancer, breast cancer, esophageal cancer, thyroid cancer, pancreatic cancer, prostate cancer and cervical cancer, accounting for 78.84% of all malignancies, and the the incidence of lung cancer, colorectal cancer, breast cancer, thyroid cancer, pancreatic cancer and prostate cancer appeared a tendency towards a rise year by year, with AAPC of 8.37%, 5.36%, 6.42%, 24.81%, 5.49% and 18.78%, respectively (all P<0.05).
Conclusions
The of malignant tumors appeared a tendency towards a rise in Yuyao City from 2011 to 2018, and lung cancer, thyroid cancer, specific female cancers, male gastrointestinal and prostate cancers should be given a high priority for cancer control in Yuyao City in the future. Early screening and early diagnosis of cancers should be facilitated among high-risk populations.
2.Therapeutic evaluation of 89SrCl2 combined with 99Tc-MDP in treat-ing bone pain of patients with breast cancer and osseous metastasis

Objective:To evaluate the clinical value of radioactive nuclide strontium chloride (89SrCl2) combined with 99Tc-MDP in treating patients with breast cancer and osseous metastasis. Methods:A total of 80 patients with breast cancer and experiencing bone pain from osseous metastasis were randomly categorized into three groups. 22 patients were treated with 99Tc-MDP (99Tc-MDP group), 30 were treated with 89SrCl2 (89SrCl2 group), and 28 were treated with the combination therapy of 89SrCl2 and 99Tc-MDP (combination group). The analgesic effect, remission of bone metastases, and quality of life of patients in the three groups were observed before and after treatment. Side effect was also monitored. Results:In the combination group, the overall pain relief rate and the increase rate of life quality score were 92.9%(26/28) and 78.6%(22/28), respectively. The combination group was statistically significantly different from the two single-treatment groups (P<0.05). The effective rate of metastatic lesions was 46.4%(13/28) in the combination group, which was significantly higher than that in the 99Tc-MDP group (18.2%, 4/22;P<0.05). The effective rate of metastatic lesions was not significantly different between the combination and 89SrCl2 groups (33.3%, 10/30) (P>0.05). Conclusion:The treatment of 89SrCl2 com-bined with 99Tc-MDP can increase the analgesic effect and significantly improve the curative effect without overt side effects in patients with breast cancer and bone metastasis.
3.Relationship between pulse wave velocity and the NYHA classification of coronary insufficiency

Objective To investigate the relationship between brachial-ankle pulse wave velocity (baPWV)and different stage of cardiac dysfunction.Methods 253 consecutive patients with coronary atherosclerotic heart disease were enrolled from August 2006 to February 2007.Patients were grouped according to the functional classification of New York Heart Association(NYHA).The baPWV was measured non-invasively with a VP1000 automated PWV/ABI analyzer(PWV/ABI,Colin CO.Ltd.,Komaki.Japan).At the sanle time,BNP,EF,LAEDV and LVESV were measured in all the patients.Results Brachial-ankle PWV was significantly correlated with cardiac function classification of NYHA(r=0.444.P<O.001),BNP(r=0.394,P<0.001)and left ventriculat end diastolic volume(r=-0.130,P<0.05),The under area of receiver operating charachateistic(ROC)curve was 73.9%.The ROC curve demonstrated that when the value of brachial-ankle PWV was equal to or larger than 1717 cm/s(≥1717 cm/s),the sensitivity of diagnosing mild cardiac dysfunction was 72.9%and specifity 61.8%.When its value was equal to or larger than 1900 cm/s(≥1900 cm/s),the sensitivity and specificity were 61.5%and 81.9% respectively.Conclusion Brachial-ankle PWV is significantly correlated to the NYHA classification of heart failure and it may be a prospective index to diagnose early stage of cardiac dysfunction.
4.Effect of stellate ganglion block on heart rate variability during thyroidectomy performed under cervical plexus block

Objective To investigate the effect of stellate ganglion block(SGB)on heart mte variability (HRV)during thyroidectomy performed under cervical plexus block.Methods Forty-five ASA Ⅰ or Ⅱ pafients of both sexy8(8 males,37 females) aged 20-60 yr,weighing 40-70 kg undergoing elective thyroidectomy under cervical plexus block were randomly divided into 3 groups(n=15 each):group Ⅰ control,group Ⅱ left SGB and group Ⅲ right SGB.The parameters of HRV included low frequency(LF),high frequency(HF),LFnu,HFnu and LF/HF ratio.SBP,DBP,HB and HRV parameters were recorded before anesthesia(To,baseline),immediately(T1) and at 5 and 10 rain after cervical plexus block(T2,T3),at the beginning and end of operation (T4,T5).Results SBP and DBP were significantly increased at different time points after cervical plexus block in all 3 groups.Compared with the baseline at T0,LFnu and LF/HF ratio were significantly increased while HR,HF and HFnu decreased during operation in group Ⅰ,LF,HF and HFnu were significantly decreased while HR increased during operation in group Ⅲ(P<0.05),but no significant change in HRV parameters was found during operation in group Ⅱ(P>0.05).Compared with group Ⅰ,HR and LFnu were significantly decreased while HF and HFnu increased in group Ⅱ(P<0.05),but no significant change was found in the indexes mentioned abovein group Ⅲ (P>0.05).Conclusion SGB can adjust cervical plexus block-induced sympathetic excitability and maintain the balance between sympathetic and parasympathetic tension.Left SGB outweighs right SGB.
5.An association between glucose metabolism status and brachial-ankle pulse wave velocity in Chinese patients with coronary artery disease

Objective To investigate the association between glucose metabolism status and brachial-ankle pulse wave velocity(baPWV)in Chinese patients with coronary artery disease(CAD).Methods baPWV values were measured in 198 CAD patients.Plasma glncose,serum lipids,hsCRP and other baseline data were obtained in all the patients.According to the glucose level,the participants were divided into 3 groups:normal glucose tolerance(NGT)group,impaired glucose regulation(IGR)group and diabetes mellitus(DM)group.ResultsbaPWV values in the DM group(1807±381)cm/s were significantly higher than those in the NGT group[(1615±248)cm/s,P=0.000]and IGR group[(1674±277)cm/s.P=0.035].Multiple stepwise linear regression analyses indicated that higher baPWv values were associated with aging and hsCRP levels in 198 patients.In DM group,higher baPWV values were independently associated with aging and HbA1 C levels.Conclusions In patients with coronary artery disease.baPWV values increase with different glucose metabolism status from NGT to IGR then DM.baPWV values in the patients with DM were significantly higher than those in the other two groups.Glycemic control may ameliorate arterial stiffness.
6.Change and Clinical Significance of Peripheral Blood DN Tregs in Patients with Systemic Lupus Erythematosus

Objective To study the expression and function of TCRαβ+ CD3+ CD4-CD8-T (Double negative regulatory T cells,DN Tregs) in peripheral blood of Systemic lupus erythematosus (SLE) patients,and investigate their function in pathogenesis of SLE.Methods TCRαβ+ CD4-CD8 T cells were quantified as percentage of total CD3+ T cells in peripheral blood from 20 SLE patients and 20 healthy controls by flow cytometry.Used ELISA to test the cytokine levels (IFN-γ,TNF-α,IL-6,IL-17A) in the plasma of SLE patients.And analyzed the relationship between the percentage of DN Tregs and cytokines levels.Results The ratio of DN Tregs in TCRαβ+ T cells was significantly increased in SLE patients compared to healthy donors (t=3.54,P<0.01).The levels of IFN-γ,TNF-α and IL-6 in plasma of SLE patients were higher than healthy donors (t=2.824,2.085,2.304,P<0.05).The frequency of DN Tregs was found to correlate with IFN-γ (r=0.52,P=0.02) but not TNF-α (r=0.17,P=0.16) and IL-6 (r=0.47,P=0.49).Conclusion This study reveals frequency of DN Tregs in peripheral blood of SLE patients was higher than healthy controls,the frequency of DN Tregs was also found to correlate with IFN-γ levels,which means that DN Tregs may play an important role in pathogenesis of SLE.
7.In vitro establishment of a three-dimensional hepatocyte culture system using collagen hydrogel as scaffolds

BACKGROUND:Rapid loss of liver-specific functions of the cultured hepatocytes limits the development of hepatocyte-based therapies. OBJECTIVE:To establish a three-dimensional culture system based on col agen hydrogel that enables to enhance liver-specific functions for a long period during culture of hepatocytes. METHODS:Hepatocytes were isolated from Sprague-Dawley rats and then encapsulated into liquid type Ⅰcol agen solution that was premixed with hepatocyte growth factor and Dulbecco's modified Eagle’s medium to create hepatocyte/col agen hydrogel compounds. The compounds were inoculated into a 96-wel plate. After gelation, culture medium was added. Light microscope, hematoxylin-eosin staining and transmission electron microscopy were used to examine the morphological characteristics and ultrastructure of the cultured hepatocytes. The cellsupernatant was col ected and tested for albumin secretion and urea synthesis. Periodic acid-schiff staining, immunofluorescence staining and quantitative real-time PCR were also used to further clarify liver-specific phenotype or function of the hepatocytes.RESULTS AND CONCLUSION:(1) Light microscope revealed that hepatocytes were round shape and distributed uniformly in col agen hydrogel. The three-dimensional hepatocyte culture system exhibited similarities to liver-like structure and tight junction were formed between hepatocytes after 14 days of culture. (2) Within the three-dimensional culture system, hepatocytes remained positive for periodic acid-schiff staining, albumin and hepatocyte nuclear factor-4αpositive after 14-day culture, which provided the convincing evidence of highly differentiated primary hepatocytes with functions of glycogen and albumin synthesis. (3) The albumin and urea productions in the three-dimensional culture system had a significantly higher level than in the two-dimensional culture, and could remain at a high level at least for 15 days. (4) The expression levels of hepatocyte-specific genes including Albumin, HNF-4α, Claudin-3, CYP1A1, CYP3A1 and G6P were significantly improved in the three-dimensional culture as compared with the two-dimensional culture. The col agen hydrogel based three-dimensional culture system provides a valuable model to enhance the hepatocyte functional maintenance and lay the foundation for the development of hepatocyte-based therapy for liver disease.
8.Genetics analysis of two childhood acute myeloid leukemia patients with variant t(8;21)

Objective To report two childhood acute myeloid leukemia (AML) patients with t(8;20)(q22;q13) and t(1;8;21)(q32;q22;q22) respectively,as variant t(8;21).Methods Chromosome preparation of bone marrow cells were made using short-term culture and karyotypic analysis was carried out using R and G-banding techniques.Interphase-fluorescence in situ hybridization (I-FISH) and metaphase-FISH (M-FISH) were performed using dual color,dual fusion AML1-ETO probe to detect the AML1-ETO fusion gene.Multiplex RT-PCR was used to demonstrate the expression of AML1-ETO fusion transcript.Results The karyotype of bone marrow cells for these two childhood AML patients were 45,X,-Y,t(8;20)(q22;q13)[12]/46,XY[3](case 1) and 46,XX,t(1;8;21)(q32;q22;q22)[18]/46,XX[2](case 2),respectively.I-FISH and M-FISH confirmed that they all had the AML1-ETO fusion gene and variant t(8;21).The AML1-ETO fusion transcript in both patients was detected by RT-PCR.Conclusion t (8;20)(q22;q13) and t (1;8;21)(q32;q22;q22) are variant t (8;21) in nature.It is important to combine the conventional karyotypic analysis with D-FISH and multiplex RT-PCR to determine the nature and prognosis of AML patients with variant t(8;21).
9.Elevated serum uric acid level as a predictor for cardiovascular and all-cause mortality in Chinese patients with high cardiovascular risk

Objective To assess the predictive value of serum uric acid levels for cardiovascular and all-cause mortality in a large prospective population based study.Methods The study was based on 3648 participants in Shanghai and Beijing,who were inpatients with high cardiovascular(CV) risk at baseLine (2004.7 to 2005.1),and blood was taken.Follow-up for death from cardiovascular disease and any cause was complete until January 1,2006.Results The mean follow-up was 1 years.There were 303 deaths during follow-up,of which 121 were cardiovascular.Crude mortality rates were 8.3 % for all patients,6.8% for female patients (116/1715),and 9.7% (187/1933) for male patients.Among men,patients in the lower and higher uric acid groups had increased cardiac and overall mortality risks compared with patients in the normal uric acid groups.Similar relation was found in women but not statistically significant.After adjusting for other conventional risk factors (age,diabetes,hypertension,diuretic use and smoking),baseline uric acid level was still associated with increased risk for death from cardiovascular disease (P=0.005),or death from all causes (P=0.014) Conclusion Our data suggest that abnormal serum uric acid levels are independently and significantly associated with risk of cardiovascular and all-cause mortality.(J Geriatr Cardiol 2008;5:15-20)
10.Expression of homosapiens chromosome segregation gene in homogenous nasopharyngeal carcinoma tissue cells

OBJECTIVE To study the expression of CSE1L gene sifted from the whole genome expression profiling in the aRNA from homogenous human nasopharyngeal carcinoma (NPC) and non-NPC biopsy tissue samples using semi-quantitative RT-PCR (sqRT-PCR). METHODS CSE1L gene was sifted from the whole genome expression profiling from homogenous NPC and non-NPC biopsy tissue samples. RNA later RNA Stabilization Reagent was used to preserve the tissue samples harvested from the nasopharynx of NPC and non-NPC patients. The samples were microdissected to get the homogenous tissue cells and then the total RNA was isolated from them. The antisense RNA (aRNA) was amplified from the total RNA by “in vitro transcription” (IVT). CSE1L gene expression in the homogenous tumor cells of NPC and the pure epithelial pillar cells of normal nasopharyngeal tissue was investigated by sqRT-PCR. RESULTS The high quality total RNA could be harvested from the microdissected homogenous tissue cells, and then the sufficient aRNA was amplified from it. The expression of CSE1L gene was identified using these aRNA by sqRT-PCR. There were significantdifference between the average expression value of CSE1L in NPC tissue (1.740?1.105) and in non-NPC tissue (0.618?0.183; df=30, t=3.159, P=0.004). The expression of CSE1L gene in the whole expression profiling were 1.056?0.296 in NPC tissue and 0.465?0.835 in non-NPC tissue respectively (df=16, t=4.317, P=0.001) . CONCLUSION The whole genome expression profiling with sqRT-PCR could be used to sift the marker genes from biopsy tissue samples. CSE1L may be as a candidate oncogene in NPC.
