1.The effect of sodium selenite on heat induced aggregation of lens crystallin

Objective To study the effect of sodium selenite on heat aggregation of lens crystallin. Methods 12 fresh and intact bovine lenses were incubated with different concentrations of sodium selenite for 72 hours. The lenses of the control group were incubated without sodium selenite for the same length of time. After removing the capsule homogenate, the ?-, ?- and ?-crystallin from the lenses were separated by gel filtration chromatography. After the separation and purification, the crystallin were heated at 60℃ for 30 minutes. Then the absorption value at 360nm was measured again. The difference in the percentage of the absorption value after aggregation was analyzed. Results Comparing with the control group, the lenses incubated with sodium selenite showed earlier opacification in a concentration-dependent manner. After the separation by gel filtration chromatography, the ?-crystallin peak showed an antelocation on the chromatogram, suggesting that there was some high molecular aggregation formation. 2.Effects of dexmedetomidine on rabbit sinus node and atrioventricular node

Objective To investigate the effects of different doses of dexmedetomidine(Dex) on rabbit sinus node and atrioventricular node.Methods A total of 24 healthy male rabbits weighing 1.5-2.8 kg,were divided into 3 groups randomly according to random number table (n =8).Group C (control),critical dosage of Dex causing sinus bradycardia D1 (loading dose of Dex was 10 μg/kg, continual pumping dose was 5 μg · kg-1 · h-1 ),six times of critical dosage of Dex causing sinus bradycardia D2 (loading dose of Dex was 60 μg/kg,continual pumping dose was 30 μg·kg-1 ·h-1 ). Rabbits were anesthetized,the right femoral artery was separated and catheterized followed by real-time monitoring of arterial blood pressure.Right external jugular vein was searched and separated,bi-polar stimulating electrode were inserted to the junction of superior vena cava and right atrium,the index of sinus node and atrioventricular node were observed by means of programmed stimulation.Si-nus node recovery time (SNRT),corrected sinus node recovery time (CSNRT),total recovery time (TRT),and atrioventricular node 2∶1 point were recorded before Dex infusion (T0 ),1 5-20 min after infusion of Dex (T1 )and 50-60 min after perfusion of Dex (T2 ).Results SNRT,CSNRT,TRT and 2∶1 point had no statistical significance.Compared with T0 ,SNRT,CSNRT and CSNRT were signifi-cantly prolonged at T1 and T2 .2∶1 point in group D1 and D2 was shortened obviously at T1 than that at T2 (P <0.05).SNRT,CSNRT and TRT of group D1 at T2 were significantly prolonged,2∶1 piont was shortened compared with T1 (P <0.05).SNRT,CSNRT and TRT of group D1 and D2 were pro-longed both at T1 and T2 than those of group C.2∶1 point was shortened in group D1 and D2 at T1 than that in group C (P <0.05).Compared with group D1,SNRT,CSNRT and TRT of group D2 at T1 and T2 were prolonged,2∶1 point was shortened obviously (P <0.05).Conclusion Load capacity of 10 μg/kg Dex apparently inhibits the function of rabbit sinus node and atrioventricular node,which is partially recovered within a short time (≤ 1 h).The inhibiting effect is more continously and re-markably in load capacity of 60 μg/kg Dex.
3.Effects of different doses of dexmedetomidine on atrioventricular node conduction function in healthy volunteers

Objective To evaluate the effects of different doses of dexmedetomidine on atrioventricular node (AVN) conduction function in the healthy volunteers.Methods Sixteen healthy volunteers of both sexes,aged 18-30 yr,with body mass index of 19-26 kg/m2,were included in the study.Dexmedetomidine was infused in a loading dose of 1.0 μg/kg over 10 min,followed by an infusion of 0.5 μg · kg-1 · h 1 for 50 min (Dose Ⅰ);1-2 weeks later,dexmedetomidine was infused in a loading dose of 1.5 μg/kg over 10 min,followed by an infusion of 0.75 μg · kg-1 · h-1 for 50 min (Dose Ⅱ).Before infusion of dexmedetomidine (T0) and at 15 and 35 min of infusion (T1.2),AVN Wenckebach point,AVN 2 ∶ 1 block point,AVN relative refractory period (AVNRRP),and AVN effective refractory period (AVNERP) were measured.Results AVN Wenckebach point and AVN 2 ∶ 1 block point were significantly decreased,and AVNRRP and AVNERP were significantly prolonged at T1,2 compared with those at T0 (P<0.05).Compared with Dose Ⅰ,AVN Wenckebach point at T2 and AVN 2 ∶ 1 block point at T1,2 were significantly decreased,and AVNRRP and AVNERP were significantly prolonged at T1,2 in the subjects receiving Dose Ⅱ] (P<0.05).Conclusion Dexmedetomidine can inhibit AVN conduction function in the healthy volunteers,and the inhibitory effect is enhanced with the increasing doses.
4.Effect of dexmedetomidine on rabbit heart rate: in vitro and in vivo experiments

Objective To evaluate the effect of dexmedetomidine on heart rate (HR) of rabbits through in vitro and in vivo experiments, and investigate the mechanism by which dexmedetomidine lowered HR.Methods In vitro experiment Healthy adult rabbits of both sexes, weighing 2.0-2.5 kg, aged 8-10 weeks, were studied.The 24 isolated hearts passively perfused in a Langendorff apparatus were randomly divided into 3 groups (n =8 each) using a random number table: control group (group C) , and dexmedetomidine 3 and 30 ng/ml groups (D1 and D2 groups).The isolated hearts were continuously perfused with K-H solution for 45 min in group C.After 15 min of equilibration, the isolated hearts were perfused for 30 min with K-H solution containing dexmedetomidine 3 and 30 ng/ml in D1 and D2 groups, respectively.At 15 min of equilibration, and at 15 and 30 min of perfusion with K-H solution containing dexmedetomidine, HR and left ventricular systolic pressure (LVSP) were recorded.In vivo experiment Twenty-five healthy adult rabbits of both sexes, weighing 2.0-2.5 kg, aged 8-10 weeks, were randomly divided into 5 groups (n=5 each) using a random number table: dexmedetomidine 3, 6, 9, 12, and 15 μg/kg groups (D3, D6, D9, D12, D15groups), to receive the corresponding doses of dexmedetomidine which was intravenously infused over 10 min.HR and mean arterial pressure were monitored and recorded before administration (T0) , and at 15 and 40 min after administration (T1,2).The correlation between doses of dexmedetomidine and change rate of HR was tested by Spearman correlation analysis.Results In vitro experiment Compared with group C, no significant changes were found in HR and LVSP at each time point in D1 and D2 groups (P>0.05).In vivo experiment Compared with those at T0 , HR at T1 in D6 and D9 groups, HR at T1,2 in D12 and D15 groups, and mean arterial pressure at T1,2in D6, D9, and D12 groups were significantly decreased (P<0.05) , and no significant change was found in HR at each time point in group D3 (P>0.05).The correlation coefficient between doses of dexmedetomidine and change rate of HR was 0.944 (P<0.05).Conclusion The mechanism by which dexmedetomidine lowers HR of rabbits is not related to direct inhibition of sinoatrial nodes, but associated with the balance of autonomic nervous system.
5.Effects of dexmedetomidine on lung ischemia-reperfusion injury in rats

Objective To evaluate the effects of dexmedetomidine on lung ischemia-reperfusion (I/R) injury in rats.Methods Ninety-six healthy SPF male Wistar rats,weighing 250-350 g,aged 8-12 weeks,were randomly divided into 4 groups (n =24 each):sham operation group (S group),I/R group,low dose dexmedetomidine group (DL group) and high dose dexmedetomidine group (DH group).In DL and DH groups,dexmedetomidine 100 and 500 μg· kg-1 · d-1 were injected intraperitoneally once a day for 2 consecutive days,while the equal volume of normal saline was given in S and I/R groups.Lung·I/R was induced by clamping the left hilum of lung for 45 min followed by reperfusion at 30 min after administration on 2nd day.At 45 min of ischemia,and 60 and 120 min of reperfusion,6 rats were sacrificed,and lungswere removed for determination of TNF-α (tumor necrosis factor-a) content and myeloperoxidase (MPO) activity in lung tissues.The percentage of damaged alveolar in lung tissues was detected at 120 min of reperfusion.Another 6 rats were lavaged and the broncho-alveolar lavage fluid (BALF) was colleted for determination of the total protein concentrations.Results Compared with S group,the TNF-α content,MPO activity,percentage of damaged alveolar,and total protein concentrations in BALF were significantly increased in I/R,DL and DH groups.Compared with I/R group,the TNF-α content,MPO activity,percentage of damaged alveolar,and total protein concentrations in BALF were significantly decreased in DL and DH groups.Conclusion Dexmedetomidine can alleviate the lung I/R injury in rats,and the mechanism is related to inhibiton of the inflammatory responses.
6.The study on expression of survivin protein in salivary gland tumor

Objective To discuss the Survivin protein expression in different salivary gland tumor.(Methods)This experiment was based on tissue chip technology.Expression of Survivin protein in the salivary gland tumor was analyzed with immunohistochemical methods.Results ① Survivin expression showed significant difference in the salivary gland benign and malignant tumors(P0.05).The gland around the malignant tumor showed diversity compared with malignant tumor(P0.05).Conclusion Survivin expression has correlation with the relay tumor canceration,and the gland around malignant tumor has correlation with occurrence,progress,prognosis in the salivary gland malignant tumor,and it is a reliable marker of earlier period diagnosis and prognosis of the malignant tumor of salivary gland.
7.Clinical Application of 64-slice Spiral CT Colonography

Objective To evaluate the techniques and application of 64-slice helical CT colonography in colonic lesions. Methods Thirty-eight patients (including 12 colonic carcinomas,10 polypi, 9 colon multiple scrobiculus, 2 multiple diverticula, 2 negative, 1 congenital bowel malrotaion,1 ulcerative colitis and tunica mucosa glandular organ hyperplasy,1 colocolic anastomosis of terminal ileum and sigmoid colon) underwent volume scanning using 64-slice helical CT after cleaning colon. Six types of reconstruction including CT virtual colonoscopy(CTVC), volume rendering(VR), multiple planar reconstruction(MPR), 360?sectional view, RaySum,and navigation were gained.CTVC appearances were compared with that of conventional colonoscopy(CC).Results In 12 colonic carcinomas and 10 polypi,the lesions’ morphology,number,size, were satisfactorily shown by CTVC. The lesions’ location,range,and 1 intestinal canal obviously stenosis were also exactly shown by RaySum.The relationship between tumor and environment, 3 liver or retroperitoneal lymph node metastasis were satisfactorily shown by MPR and 2D transection image. On 360?sectional view,the distance between tumor and anus was exactly measured and the result was in conformity to that of CC. Of the CTVC appearances,1 sigmoid carcinoma and 1 transverse colon polyp was in unconformity to that of CC;1 ulcerative colitis and descending colon carcinoma was diagnosed as ulcerative colitis and tunica mucosa glandular organ hyperplasy by pathology.Conclusion 64-slice helical CT is a valuable imaging technique for detecting colon diseases. CTVC can obtain more clinical information than CC combining MPR,RaySum and VR.
8.Significance of exposing and recognizing Zuckerkandl's tubercle during thyroidectomy

Objective To Explore the significance of successful exposing and recognizing Zuckerkandl's tuhercle(ZT)during thyroidectomy.Methods Three hundred and seventy patients(501 sides) underwent lobectomy or total thyroidectomy from January 2009 to June 2011 were included in this study.The ZT was assessed in terms of its presence or absence,size and anatomical association with the recurrent laryngeal nerve(RLN)and superior parathyroid(SP).Results ZTs were found in 412 of 501 sides ( 82.2% ),among which 368(89.3% ) ZTs were located in the middle third of the lateral lobe of the thyroid gland.ZTs passed over the RLN in 379 of 412 sides(92.0% ).When the ZTs were located in the middle or lower third of the lateral lobe of the thyroid gland,the SPs were all located in the cranial portion of ZT.The SP was adhered to the ZT in 80.1% of the cases.RLN damage rate was 0.40%,and no SP damage occurred.Conclusion Exposing and recognizing Zuckerkandl's tubercle during thyroidectomy is of important clinical significance,which helps to identify and protect RLN and SP,so as to reduce surgical complications.
9.Effects of taurine-magnesium coordination compound on sodium channel in rat cardiomyocytes of arrhythmia induced by aconitine

Aim To investigate the antiarrhythmic mechanism of taurine-magnesium coordination compound on sodium current in single rat ventricular myocytes of arrhythmia induced by aconitine.Methods Whole-cell patch clamp was used to record INa in normal cardiomyocytes and single rat ventricular cardiomyocytes of arrhythmia induced by aconitine.Results In ventricular cardiomyocytes of rat,INa was blocked by 100~400 ?mol?L-1 TMCC in a concentration-dependent manner.INa was increasd from(45.56?1.96)pA/pF to(59.19?11.49)pA/pF by 1 ?mol?L-1 aconitine,while decreased to(34.23?1.33)pA/pF by 24.24 ?mol?L-1 amiodarone.TMCC(100,200,400 ?mol?L-1)could restore INa to(51.61?5.96)pA/pF,(40.91?6.73)pA/pF,(41.50?5.50)pA/pF respectively.Amiodarone could restore INa to(40.22?1.47)pA/pF.Conclusions TMCC can restore INa,which is increased by aconitine,and the effect is equal to that of amiodarone.TMCC blocks INa of ventricular cardiomyocytes,which may be one of its antiarrhythmic mechanisms.
10.Development of monoclonal antibodies against the gp90 protein of reticuloendotheliosis virus and mapping of their recognition regions.

In order to develop monoclonal antibodies (McAbs) against the gp90 protein of reticuloendotheliosis virus (REV), the His-tagged gp90 protein of REV was used to immunize BALB/c mice. Hybridomas were generated by fusing mouse myeloma cells SP2/0 with the splenocytes from the immunized mice. After screening and 3 rounds of cloning process, 3 hybridomas (3G5-B8, 3G5-A10 and 1G12) that stably secreted McAbs against the REV-gp90 were obtained. The isotypes of the McAbs were determined to be IgG1, IgG1 and IgG2b. The McAbs specifically bound to gp90 in REV-infected DF-1 cells, as demonstrated by Western blotting and indirect immunofluorescence assay. The recognition regions on gp90 that were recognized by 3G5-B8/3G5-A10 and 1G12 were located between amino acids 200 to 245 and 230 to 235, respectively, as demonstrated by Western blotting analysis. These McAbs will be useful in the diagnosis and pathogenesis study of REV.
