1.Application of molecular immunologic technique in study of Chinese integrative medicine

Molecular immunologic technique gets quick development in recent years, especially the fast development of the immuno-labelling technique, which found its indispensable status in the study of traditional Chinese medicine (TCM). With the development of the study of TCM, medical immunology connecting TCM with the Western medicine has become one of the most indispensable conjunctive subjects. Scientists can study the theories and prevention or treatment mechanisms of TCM not only at holistic and cell levels, but also at molecular and gene levels. Appropriate application of the molecular immunologic techniques according to their characteristics will play an important role in the study of Chinese integrative medicine.
2.Reproduction and evaluation of 6-OHDA-induced rat model of Parkinson's disease

In the site of lesion in SN, the number and volume of dobaminergic neurons were decreased, and the tissue structure of SN became indistinct, the levels of dopamine (DA), dihydroxyphenylaceticacid (DOPAC), homovanillicacid (HVA) 5-hydroxytryptamine (5-HT) and 5-hydroxyindoleaceticacid (5-HIAA) were more conspicuously reduced than that in uninjured side. TH-immunoreactive cells of substantia nigra compact part were lessened obviously on injured side. The abnormal rotatory behavior of PD rats did not spontaneously disappear during 10 months of continuous observation. Conclusion The PD rat model, which is reproduced by injecting 6-OHDA to selectively destroy dopaminergic neurons in the substantia nigra of rat, is stable and reliable, and demonstrated similar primary pathological changes of PD.
3.ACTH-independent Cushing syndrome(report of 89 cases)

Objective To study the diagnosis and treatment of adrenocorticotrophic hormone (ACTH) independent Cushing syndrome. Methods A retrospective review was done on 89 patients with ACTH independent Cushing syndrome, including 85 cases of adrenal tumors with hypercorticism, 2 cases of adrenocortical adenocarcinoma and 2 cases with ACTH independent macronodular adrenal hyperplasia. Results Favorable results were observed from the surgery for 85 cases of adrenal tumors combined with hypercorticism.Two cases with ACTH independent macronodular adrenal hyperplasia were cured by bilateral adrenolectomy. Adrenal cortical adenocarcinoma was unresectable for both the patients who survived 7 and 9 months respectively. Plasma ACTH,dexamethasone suppression test,B ultrasonography and CT scan were useful for the diagnosis and differentiated diagnosis of ACTH independent Cushing syndrome. Conclusions Prognosis of adrenal tumors with hypercorticism and ACTH independent macronodular adrenal hyperplasia is favorable after surgery,whereas the prognosis of adrenal cortical adenocarcinoma is poor.
4.The effects of different doses of propofol infusion on real-time monitoring of recurrent laryngeal nerve

Objective To evaluate the effects of different doses of propofol infusion on real-time monitoring of recurrent laryngeal nerve. Method 30 ASA Ⅰ-Ⅱ patients who underwent thyroid gland surgery were given propofol intravenously in different concentrations. BP, HR and real-time monitoring of recurrent laryngeal nerve were recorded with the propofol dosage varying from 3 to 6, 9, 12, 15, 18?g/(kg?min). Results The amplitude of recurrent laryngeal nerve reflex had no change with different doses of propofol infusion. The HR and BP were elevated with low doses of propofol infusion, but there were no changes when propofol concentration was 15 or 18?g/(kg?min). Conclusion Propofol infusion i.v. does not interfere with real-time intraoperative monitoring of recurrent laryngeal nerve, and it does not inhibit the action potential of recurrent laryngeal nerve, but can effectively decrease the cardiovescular reaction.
5.Effect of Pingchuanlingchongji on the Auricular Microcirculation in Mice

Objective To observe the effect of Pingchuanlingchongji (PCL) on the auricular microcirculation of mice and to investigate the effect mechanism of the treatment for asthma. Methods 50 mice were randomly divided into five groups, group 1 and group 2 were given 0.9% NS (25 mL/kg) as control group; group 3 was given aspirin (0.25 g/kg), 4 group was given low-dose (1.1 g/kg) PCL, group 5 was given high-dose (2.2 g/kg) PCL. Last for seven days. We injected Posterior Pituitary Injection (0.5 U/mL) on the rear vein of 2-5 group mices after the last time of given druy. The diameter of micro-artery in mice auricular, the diameter of micro-vein and blood flow speed were observed. 6.Study on the mechanism of therapeutic effect of "Yichantang" in the treatment of Parkinson disease

Objective To explore the mechanism of therapeutic effect of Yichantang in the treatment of Parkinson′s disease (PD). Methods A rat model of PD was replicated. The experimental animals were divided into a normal control group,a normal saline control group and a Yichantang group. Changes in the rats' behavior pattern was obrerved. Using the technique of intracerebral dialysis,the content of catecholamine like substances in the nigral extracellular fluid(NECF)was measured by means of high performance liquid chromatography(HPLC). Changes in the affinity and quantity of M and DA receptors were determined too. Results Yichantang markedly improved the rats' rotation behavior,and enhanced the Bmax and KD of DA and M receptors in the brain tissues on the side of lesion. 7.Effects of Astragalus Polysaccharides on Damage of Keratinocytes in UVB Radiation Skin

Objective To investigate the effects of Astragalus polysaccharide (APS) on damage of keratinocytes in ultraviolet B (UVB) radiation skin in HaCaT cells; To preliminarily explore its mechanism of action.Methods HaCaT cells were treated by UVB irradiated for modeling. Blank control group, model group, UVB irradiation group and low-, medium- and high-dose APS interventional groups were set up. Each medication group was given relevant medicine. MTT assay was used to determine the cell activity; GSH-Px, CAT, SOD activity and MDA content were detected by biochemical colorimetric method; contents of TNF-α and IL-6 were detected by ELISA.Results Compared with the control group, the contents of SOD, CAT and GSH-Px in the model group decreased significantly, while the contents of MDA, TNF-α and IL-6 increased significantly (P<0.05); compared with the mode group, the contents of SOD, CAT and GSH-Px in the medication groups increased significantly, while the contents of MDA, TNF-α and IL-6 were reduced significantly (P<0.01).Conclusion APS can reduce the oxidative stress injury of UVB to HaCaT cells to some extent.
8.Effect of Fuyuanbunao Granules on Memory of Experimental Mice

Objective: To probe and reveal the brain tomic and the wise sharpening action of Fuyuanbunao Granules and Its toxicity and side effect. Methods: The effects of Fuyuanbunao Granules on the memory acquisition disturbance induced by pentobarbital sodium or anisodine, the memory consolidation disturbance induced by sodium nitrite, and the memory repetition disturbance induced by alcohol were observed. Meanwhile, the acute toxicity test was done. Results: The granules could obriously improve the memory disturbance induced by the factors obove, and the improvement effect varied with the dose. The effects of larger dose groups were more obvious. There were no death of animals or toxicity and sile effect in test. Conclusion:Fuyuanbunao Granules maybe have the cholinergic acction. It can promote the aerobic metabolism, improve the cerebral anoxia and the inhibited state of center nervous system. It has low toxicity and can be used in clinical application.
9.Experimental study on TCM symptomatological and developmental characteristics of rats with radiation-induced lung injury

Objectives:To explore the possibility of using experimental animal models as a new means for studying the pathogenesis and symptomatologic development of radiation-induced lung injury,and provide theoretical and experimental evidences for early prevention and treatment with the traditional Chinese medicine.Methods:80 female Wistar rats were divided randomly into the model group and control group with 40 rats in each group respectively.The rats in model group were anesthetized and fixed,and a 2cm?3cm area of the right lungs received 6mV X-ray radiation at 3Gy?2/w and a maximal 10/5w.Eight rats randomly selected from each group were sacrificed at the end of weeks of 3,5,8,12 and 26.Diagnostic techniques of TCM were employed to analyze symptomatological and developmental characteristics of TCM in radiation-induced lung injury during exposure by dynamic examination and comparison of living rats,gross lung samples and tissue sections.Results: Signs and symptoms including redness around nose and eyes,yellowish secretion,tachypnea,dry stool,lethargy,sluggishness, inactiveness were observed in model group in early stage of radiation exposure,which consistent with dominant heat in the interior and primordial qi exhaustion syndrome.Swollen lungs,bright redness on surface and red bleeding patches were observed in early stage of radiation;microscopic examination showed interstitial tissues,and readily visible congestion,hemorrhage and edema of pulmonary interstitial tissues,consistent with damaged blood collaterals by heat and blood escaping circulation syndrome.In the later stage of radiation exposure,the lungs showed dark appearance,apparent purple petechia,hard texture and poor elasticity.Microscopic examination showed remote hemorrhage foci,pulmonary capillary stasis or closure,and focal fibrotic clumps,consistent with obstruction of collaterals by blood stasis syndrome.No abnormal changes were observed in the control group.Conclusions:Animal experiments can redeem the clinical inadequacy of traditional inspection,auscultation and olfaction,interrogation and feeling pulse and palpation diagnostic techniques,modern pathological methods can be utilized to study symptomatology of traditional Chinese medicine.Symptomatological and developmental characteristics of'heat toxin impairment in radiation-induced lung injury characterized by heat damaging pulmonary collateral syndrome in early stage and pulmonary collateral stagnation syndrome in the advanced stage'can be revealed at living organism,gross lung sample and microscopic pulmonary tissue levels,thus providing theoretical basis for early prevention and treatment of radiation-induced lung injury using'cooling blood to remove apthogenic heat'.
10.Effect of NGF on the sperm motility of human in vitro

Objective Motility is an important physiological characteristic of a mature sperm.Nerve growth factor(NGF) is a protein essential for the development,maintenance and survival of the peripheral and central nervous systems.NGF and its receptors TrkA and p75 are widely expressed in the testis,accessory reproductive organ,and the epididymal sperms.In the present study,we investigated the role of NGF on human sperm motility.Methods Use 0.1,1 and 10 μmol/L concentrations of NGF,on sperm motility study to investigate the optimal concentration.Use CASA to detect Sperm motility changes every 10 minutes in an hour after 10 μmol/L NGF was added to the semen.Results The parameters of sperm motility increased after NGF incubation had significant difference, in particular,VAP,VSL,VCL,BCF and LIN mean were significantly increased more than 32%.MAD,STR,ALH and WOB mean had no notable difference.Furthermore,NGF promotes the sperm motility in a time- and dose- dependent manner.In addition,the enhancement of NGF on sperm motility was more stronger than those of sperm culture medium.Conclusion Our findings suggest that NGF plays a promoted role in human sperm motility.
