1.Early hemorrhelogical changes of oleic acid-induced RDS in dogs

The hemorrheological changes were observed in dogs in the first 2 hours after they were inflicted with oleic acid-induced RDS.It was found that hematocrit,absolute viscosity and relative viscosity of whole blood were significantly increased but plasma viscosity showed no marked changes.These facts imply that in the early stage of oleic acid-induced RDS,the hemorrheological changes were characterized by hemoconcentration and decrease of the trans-formability of erythrocytes,which might be one of causes of the functional failure of pulmonary microcirculation.
2.Blood vessel dynamic mechanism of hepatic carcinoma with color Doppler ultrasonography

Objective To study the regularity of blood type and Doppler parameters at the interior and surrounding of the hepatic carcinoma.Methods Seventy-five masses with blood flow signals were selected.The size,mean color density (MCD),flood flow type and Doppler parameters including velocity of peak systolic (PS) and resistance index (RI) were studied.Results ①In the interior of masses,the PS of branch and insert types was the highest,and the RI of branch and net types was the highest (P 0.05 ).Conclusions The parameter is associated with the blood type and grade closely in hepatic carcinoma.
3.Progress in treatment of Iead poisoning by antioxidants

Lead poisoning has become a big threat to public health in China. Traditional treatments usually use chelators to accelerate the excretion of lead by forming complex compounds with it. However, chelators exhibit side effects and have little therapeutic effect on lead-induced impairment so that better drugs are needed. As antioxidants are effective for the treatment of lead poisoning, with lasting effect and little side effect, they have been the focus of increasing studies. This review provides a detailed account of updates on the effects of antioxidant drugs in the therapy of lead poisoning, and of the progress in antioxidant activity of puerarin, quercetin, curcumin, allicin and melatonin in treatment of lead poisoning. To some extent, these five types of drugs can reduce lead poisoning by accelerating lead excretion and antioxidant, increase the body′s antioxidant enzyme activity, reduce oxidative stress and repair damage, which promises some clinical value.
4.Effects of tert-butyl hydroperoxide and hypoxia in transient ischemia-like attack in mice

Objective To explore the effects and relationship of tert-butyl hydroperoxide (t-BHP) and hypoxia in transient ischemia-like attack (TIA-like) in Kunming mice.Methods Fifty-six mice were randomly divided into normal saline group (group A), t-BHP group (group B), t-BHP plus hypoxia group (group C) and model group (group D). Fourteen mice in group D were injected t-BHP (0.11 mol/L, 10 ml/kg) through tail vein with an interval of 24 hrs to induce TIA-like attack. The average time of TIA-like was ( 3.7? 1.1) days, and this time was used as experimental evidence to collect blood sample in other groups. Objective To elucidate whether down-regulation of homo sapiens eukaryotic translation elongation factor 1 alpha 2 (EEF1A2) expression induces apoptosis in pancreatic cancer cells and its possible mechanisms. Methods Two siRNAs targeting human EEF1A2 were synthesized and the siRNA/liposome complexes were transfected into the pancreatic cancer cell line BxPC-3. RTPCR and Western blot were used to analyze the change of EEF1A2 expression and the apoptosis rate of BxPC-3 cells was studied using Annexin-V/PI assay. To identify the mechanisms involved, the apoptosis associated proteins such as caspase-3, caspase-8, caspase-9, PARP, cytochrome C and Bid were detected by Western blotting. Results Both EEF1A2-targeting siRNAs reduced the EEF1A2expression, and the No. 2 siRNA inhibited EEF1A2 expression to less than 25 % in mRNA and protein levels. Down-regulation of EEF1A2 expression in BxPC-3 cells enhanced cell apoptosis (15.28% ±3.65%) at a greater level than negative siRNA-expressing cells (10. 11% ± 3. 05%) or mock cells (9.41 % ±4.14 %). Furthermore, reduction of EEF1A2 activated the pro-caspase-8, pro-caspase-3,pro-caspase-9,PARP and Bid to their active forms, and increased the expression of cytochrome C.Conclusions These data suggest that EEF1A2 down-regulation could significantly induce apoptosis of pancreatic cancer cell line BxPC-3, which is likely mediated by the death receptor and mitochondrial apoptotic pathways.

7.Establishment of Experimental Diabetic Nephropathy Rats Model: Optimum Selection Ⅱ

OBJECTIVE: To compare the effects of different factor combinations on diabetic nephropathy(DN)rats model and to determine the preferred plan for the establishment of DN rats model.METHODS:A total of 30 SD rats were randomly divided into 5 group(6 rats in each group):group A was assigned to receive placebo(normal control),group B to receive high-lipids and high-sucrose feeding,group C to receive high-lipids and high-sucrose feeding plus streptozotocin(STZ),group D to receive high-lipids and high-sucrose feeding plus doxorubicin hydrochloride and STZ,group E to receive high-lipids and high-sucrose feeding plus mononephroctomy and STZ 12 weeks later the model rats were put to death for the determination of serum parameters including blood lipid,glycosylated hemoglobin,renal function,aldose reductose(AR),glomerular filtration rate(GFR),SOD and MDA .RESULTS:The rats in group E showed significant increase in blood lipid,in particular the increase of AR,and which also showed significant differences in GFR and oxygen free radical indices as compared with normal group and the other model groups.CONCLUSIONS:The rats model that fed with high-lipids and high-sucrose feeding for 4wk prior to mononephroctomy followed by the postoperative intraperitoneal injection of low dose of STZ 2 weeks later proves to be the optimal one.

9.The effect of silent homo sapiens eukaryotic translation elongation factor 1 alpha 2 gene on the growth of pancreatic cancer xenograft in nude mice

Objective To explore the effect and the possible mechanisms of silent homo sapiens eukaryotic translation elongation factor 1 alpha 2(EEF1A2)gene on the growth of pancreatic cancer cell in vivo.Methods The pancreatic cancer xenograft models in mice were established.The mice were equally divided into control group,negative control group and EEF1A2 group,which were injected with PBS,negative control siRNA and EEF1A2 siRNA into xenograft tumors respectively.The size and weight of tumors in each group were measured.The expression of EEF1A2 and PCNA in tumor tissue of each group was detected by immunohistochemistry.The cell apoptosis rate in tumor tissue of each group was determined by TUNEL.Results In xenograft nude mice models,since the 17th day of injection,the growth of tumor size in EEF1A2 group was obviously slower than that of negative control group and control group(all P<0.05).By the end of the treatment,the tumors were cut off and weighted.The weight of tumors in EEF1A2 group(0.27g± 0.06g)were significantly lower than those of control group and negative control group(0.39g± 0.08g and 0.43g± 0.07g,P<0.05).EEF1A2 mostly expressed in cytoplasm of pancreatic cancer cell.In negative control group and control group,the positive cells distributed densely and the positive rate was(72.58 ± 25.47)% and (76.75±23.19)% respectively.The distribution of positive cells in EEF1A2 group was scattered and the positive rate was(34.78±21.36)%,the difference was statisically significant(P<0.01).The expression of PCNA at protein level in EEF1A2 group was significantly lower those that of control group and negative control group(P< 0.01).The result of TUNEL test indicated that the cell apoptosis rate in EEF1A2 group was higher than those of control group and negative control group (P<0.01).Conclusions The EEF1A2 gene can be effectively silented in vivo,which significantly inhibits the growth of pancreatic cancer cell.It may be related with inhibition of cell proliferation and promotion cell apoptosis.
10.Liver regeneration after transplantation of microencapsulated hepatecytes in rats with acute liver failure

Objective To investigate liver regeneration after transplantation of microencapsulated hepatocytes in rats with acute liver failure (ALF). Methods ALF rat model was established by intraperitoneal injection of D-galactosamine (D-GalN). After 18 h, rats were randomized into control group ( Ⅰ ), free hepatoeyte transplantation group ( Ⅱ ) and the microencapsulated hepatecyte transplantation group (Ⅲ). Six rats for each group were randomly selected and sacrificed at 6, 12, 24, 36, 48, 72, 120, 168 and 240 h after ALF induced and blood samples from inferior vena cava were collected. Liver functions were tested in blood samples, and the expression of proliferating cell nuclear antigen (PCNA) was detected by immunohistochemistry. Results Ten-day survival rates of 3 groups were 26.7% (4/15), 40.0% (6/15) and 73. 3% (11/15), respectively (x2 = 9. 349,P = 0. 009). Survival rate of group Ⅲ was significantly higher than that of group Ⅰ and Ⅱ. Levels of ALT and AST in each group increased significantly at 6 h after ALF induced, and peaked between 48 ~ 72 h. Levels of ALT and AST in group Ⅱ and Ⅲ declined from 36 h, which was more significant in group Ⅲ. Tbil levels in group Ⅰ gradually increased after ALF induced and peaked at 72 h. Tbil in group Ⅱ and Ⅲ declined from 48 h, which was more markedly in group Ⅲ. In normal rats, the expression of PCNA protein was almost negative, but it was strongly expressed in ALF rats and peaked at 48 h. The number of positive cells in group Ⅲ was higher than that in group Ⅰ and Ⅱ, and the differences were of statistical signifieance. Conclusion The transplantation of microencapsulated hepatocytes can promote the regeneration of liver, and it can improve the liver function and prognosis in rats with ALF.
