1.Animal models and evaluation methods of ischemic brain injury

The experimental study of cerebral ischemia plays an important role for understanding the pathogenesis of ischemic brain injury, but its correlation with the clinical therapeutic strategies has certain limitations. One of its main reasons is that the experimental models and methods can not or only partially repeat the pathophysiological processes of natural cerebral ischemia. In order to promote the understanding and interpretation of the experimental data, we review the commonly used experimental animal models and modeling methods and mainly elaborate the methods of current different in vivo and in vitro clinical evaluation. Based on these studies, we believe that the standardized clinical evaluations are hugely important for assessing the experimental results and clinical transformation.
2.Study on TCM Syndrome Distribution and Blood Pressure Variability in 443 Patients with Primary Hypertension

Objective To study the TCM syndrome distribution and the laws of blood pressure variability in patients with primary hypertension.Methods Totally 443 patients with primary hypertension were selected; four diagnostic data were collected to conduct syndrome differentiation (main syndrome was chosen for compound syndrome); 24 h ambulatory blood pressure was monitored and blood pressure variability of each syndrome was compared.Results Among 443 cases of patients, liver-kidney yin deficiency syndrome (157 cases) > qi yin deficiency syndrome (83 cases) > yang deficiency syndrome (76 cases) > phlegm dampness syndrome (65 cases) > liver yang hyperactivity syndrome (62 cases). There was statistical significance in ages of different syndromes, but there was no statistical significance in gender, family history and course of disease (P>0.05). Compared with liver-kidney yin deficiency syndrome, 24 h systolic blood pressure and 24 h mean blood pressure of phlegm dampness syndrome were significantly different (P<0.05); circadian rhythm abnormality rates of liver-kidney yin deficiency syndrome, liver yang hyperactivity syndrome, qi yin deficiency syndrome, phlegm dampness syndrome and yang deficiency syndrome were 80.85% (38/47), 66.67% (14/21) ,81.48% (22/27), 86.96% (20/23), and 78.79% (26/33), respectively, without statistical significance in different syndromes (χ2=3.031,P=0.553).Conclusion Among the 443 hypertensive patients, patients with liver-kidney yin deficiency syndrome were the most, and patients with liveryang hyperactivity syndrome were the least. Phlegm dampness syndrome has the highest blood pressure, while liveryang hyperactivity syndrome has the lowest blood pressure. Patients with liver-kidney yin deficiency syndrome are with the highest blood pressure variability, while patients with qi yin deficiency syndrome are with the lowest blood pressure variability.
3.Significance of fluctuated level of endogenous bactericidal/permeability-increasing protein in patients after major abdominal surgery

Objective To investigate the fluctuated pattern of endogenous bactericidal/permeability-increasing protein(BPI)levels,and to elucidate its significance in endotoxin response after major abdominal surgery.Methods Eleven patients undergoing major abdominal surgery were involved in present study.The levels of plasma BPI,lipopolysaccharide-binding protein(LBP)and interleukin-6(IL-6)were measured preoperatively and on postoperative days 1,3,5,7,and 14 by ELISA.Plasma endotoxin concentration was determined with the chromogenic limulus amebocyte lysate(LAL),which was modified by perchloric acid(PCA)pretreatment of samples.The polymorphonuclear neutrophil count was also done at each time point.Results In comparison with normal controls,plasma BPI/LBP ratios [(1.058?0.074)?10-3,P=0.004] were significantly increased on postoperative day 1,maintained at same level on day 3,and declined on day 5.No plasma IL-6 was detected from the blood of all the 11 patients prior to operation,but plasma IL-6 level rose rapidly on day 1 after operation(27.440?12.144pg/ml,P=0.04),maintaining a high level(11.530?5.312pg/ml)on postoperative day 3,and disappeared on day 5.Conclusions Plasma BPI/LBP ratios were markedly elevated during 3 days after a major abdominal surgery,and endogenous elevated BPI may play an important role in prevention from the development of endotoxemia in surgical patients.

The purpose of this study was to investigate the effect of high mobility group 1 protein (HMG 1) on TNF ? expression as well as release in the kidney, and their potential role in the pathogenesis of acute renal injury associated with sepsis. Using a sepsis model by cecal ligation puncture (CLP), 50 male Wistar rats were randomly divided into normal control group ( n =10), CLP group ( n =20), and sodium butyrate treatment group ( n =20). At serial time points animals in each group were sacrificed, and blood as well as kidney tissue samples were harvested to determine HMG 1/TNF ? mRNA expression, tissue TNF ? protein level, and serum creatinine content. ObjectiveTo investigate the relationship between sepsis induced high mobility group 1 (HMG 1) gene expression and endotoxemia in rats. MethodsSepsis was induced by cecal ligation puncture (CLP). One hundred Wistar model rats were randomly divided into four groups: normal control group ( n =10), sham operation (SO) group ( n =10), CLP group (further subdivided into 2?h, 6?h, 12?h, 24?h, 48?h and 72?h post CLP, n =10 in each subgroup), and bactericidal/permeability increasing protein(rBPI 21 ) treatment group (further subdivided into 12?h, 24?h post CLP, n =10 in each subgroup). Fourteen kinds of ethyl ester of fatty acids form Hai Haozi(Sargassum pallidum) are analysed by GC methods.They are identified by comparision of their rentention time with those of known standards and quantitated by using an internal standard.It shows that this drug contains lots of saturated and unsaturated fatty acids.The analytical resulits may provide material basis for futher research of Hai Haozi.
7.Effects of Jianpi Bushen Huoxue recipe on hematopoietic cell apoptosis in a mouse model of aplastic anemia

OBJECTIVE: Adopting methods of cell culture to explore the effects and mechanisms of Jianpi Bushen Huoxue Prescription (JPBSHXP), a traditional Chinese compound herbal medicine for strengthening spleen, reinforcing kidney and activating blood circulation, in inhibiting hematopoietic cells apoptosis in a mouse model of aplastic anemia (AA). METHODS: Blood serum of AA mice was made from an AA mouse model. Blood serums containing different traditional Chinese compound herbal medicine were made from rats after intragastric administration of JPBSHXP and its related decoctions, respectively. Bone marrow cells of normal mice were incubated by these blood serums for 24 hours, respectively. The apoptosis of the bone marrow cells were assayed by flow cytometry and transmission electron microscopy (TEM). RESULTS: It was indicated that the bone marrow cells of normal mice incubated with blood serum of AA mice displayed typical apoptosis. The apoptosis rates of bone marrow cells of the AA mice incubated by blood serum containing different traditional Chinese herbal medicine were decreased. The effect of Bushen (reinforcing kidey) Recipe was better than Jianpi (strengthening spleen) Recipe and Huoxue (activating blood circulation) Recipe, while the effect of JPBSHXP was the best. TEM results showed that the effect of Bushen Recipe was better than that of the Jianpi Recipe and the Huoxue Recipe, while the effect of JPBSHXP was the best. CONCLUSION: JPBSHXP and its related decoctions can significantly decrease the apoptosis rate of bone marrow mononuclear cells of the AA mice. It is inferred that JPBSHXP can promote bone marrow hematogenesis.
8.Diagnostic significance of the relative band power of in acute focal cerebral ischemia

Objective To investigate the diagnostic significance of the relative band power(RBP) of EEG in acute focal cerebral ischemia (AFCI). Methods EEG monitoring was performed in 20 patients with AFCI (51 lesions) in neurological intensive care unit (NICU) and 20 patients with normal EEG (control group) in NICU. The changes of bilateral RBP were observed and analyzed comparatively. RBP in the infarction group was compared with that in the control group. The diagnostic capabilities of all wave-band RBPs were assessed with the receiver operating characteristic (ROC) analysis. Results AFCI shoved asymmetry on all the wave-band RBPs. The RBP of of δwave on the lesion side was significantly higher than that on the normal side (P <0.01). RBPs of α, β and θ waves were decreased significantly (P <0.05). RBP of δ wave in the infarction group was increased significantly compared to the control group (P < 0.01 ). RBPs of α and β waves were decreased significantly (P <0.01). The diagnostic accuracy of the RBP of α was the highest, and both β and δ waves also had significance for the diagnosis of AFCI. Conclusions RBP had an important significance for the early diagnosis of AFCI. Conclusions RBP had an important significance for the early diagnosis of AFCI.
9.Changes in arterial blood gas and pulmonary pathology of experimental pulmonary emphysema following mesenchymal stem cell transplantation in rats

BACKGROUND: Pathological changes of pulmonary emphysema are not reversible according to the existent pathogenesis of pulmonary emphysema. Research over many years report that injury of pulmonary blood capillary may take part in new pathogenesis of pulmonary emphysema based on lung volume reduction operation and bronchial lumen occlusion. Mesenchymal stem cells (MSCs) have multi-directional differentiation potencies, such as the differentiation into vascular endothelial cells. Therefore, MSCs may promote pulmonary vascularization and repair pulmonary tissue.OBJECTIVE: To observe the effect of MSCs transplantation on pathological changes of arterial blood gas and pulmonary tissue in model rats with pulmonary emphysema, and investigate the therapeutic effects on MSCs on pulmonary emphysema and the pathogenesis of pulmonary emphysema.DESIGN: Randomized controlled animal study.SETTING: The Second Hospital of Shanxi Medical University.MATERIALS: Thirty healthy Wistar rats, 6 weeks old, of either gender, weighing 180-200 g. They were provided by Physiological Experiment Animal Center, Shanxi Medical University. All rats were randomly divided into MSCs treatment group, model group and control group with 10 rats each.METHODS: The experiment was carried out in the Physiological Laboratory of Shanxi Medical University from April 2005 to April 2006. Rats in the MSCs treatment group and in the model group were anesthetized and intratracheally perfused with 250 U/kg Porcine pancreatic elastase (PPE) to establish pulmonary emphysema models; while, rats in the control group were perfused with saline. The models were successfully established 4 weeks later. All rats were anesthetized and then femur and tibia were obtained to separate and culture MSCs in vitro. Immunocytochemistry was used to detect the expression of CD71 in order to evaluate MSCs. Bromium azacytidine-labeled MSCs were inserted along caudal vein into rats in the MSCs treatment group; while, rats in the model group and control group were inserted with the same volume of PBS solution.MAIN OUTCOME MEASURES: ① Changes of arterial blood gas in the three groups; ② Pulmonary tissue was used for pathological sections in order to calculate mean alveolar number, mean alveolar area and mean linear intercept; ③Immunocytochemical staining was used to measure numbers of CD34+ cells so as to determine proliferation of alveolar blood capillary.RESULTS: Three rats in all died during the model establishment, while another 3 rats were supplied. Therefore, an overall number of 30 rats were involved in the final analysis. ① Culture and evaluation of MSCs: At 3 days after inoculation, MSCs were generally adherent to walls and fusiformly shaped. In the third generation, the expression of CD71 was observed on the surface of MSCs.② Comparisons of arterial blood gas in the three groups: There were no significant differences in pH value, PO2, PCO2 and SaO2 in the three groups (P > 0.05). ③ Pathological changes of pulmonary tissue: Pathological changes in the MSCs treatment group were milder than those in the model group;meanwhile, mean alveolar number in the MSCs treatment group was more than that in the model group, and there was significant difference between them (F=80.201, P< 0.05). While mean alveolar area and mean linear intercept in the MSCs treatment group were smaller than those in the model group, and there were significant differences (F =26.755,26.875, P < 0.05). ④ Comparisons of CD34+ expression in pulmonary tissue: Relative positive area of CD34+ in the MSCs treatment group and model group was smaller than that in the control group (F =20.411, P < 0.05), but that in the MSCs treatment group was larger than that in the model group, and there was significant difference between them (F=20.411, P< 0.05).CONCLUSION: MSCs can reverse the pathological changes of pulmonary emphysema; on the other hand, the decrease of the number of pulmonary capillary maybe one of the important pathogeneses of pulmonary emphysema.
10.The Application of MR Diffusion-weighted Imaging in Discriminating Benign and Malignant Vertebral Fractures

Objective To investigate the value in distinguishing benign and malignant vertebral fractures with MR diffusion-weighted imaging(DWI).Methods 31 patients with 58 vertebral fractures were underwent routine MRI scanning and echo-planar DWI(EPI/DWI).The materials were divided into benign group(n=32,20 cases),malignant group(n=18,8cases),tuberculosis group(n=8,3cases) and normal vertebral body group(n=31). Signal intensities on DWI were compared and apparent diffusion coeffient(ADC) values of normal and abnormal vertebral bodies were calculated. ADC values among the benign,malignant,tuberculosis vertebral bodies and normal ones were quantitatively analyzed and compared.Results The ADC values of benign,malingnant,tuberculosis and normal vertebral bodies were (1.44?0.41)?10 -3 mm2/s,(0.94?0.17)?10 -3 mm2/s,(0.98?0.13)?10 -3 mm2/s and(0.42?0.16)?10 -3 mm2/s ,respectively. The ADC values of the three abnormal groups were higher than normal group ( all P0.05). The sensitivity and specificity of ADC for the diagnosis of malignant lesion were 72.2% and 90.6% if the upper bound of 95% confidence interval was set as a differential level.Conclusion DWI provides an important supplement to the routine MRI in the differentiation of benign with malignant vertebral fractures.
