1.Construction of information purchase platform for scientific research materials

The ideas for constructing information purchase platform for scientific research materials were put forward and the targets for its step-by step implementation were worked out according to the analysis of problems existed in purchasing scientific research materials with chemical agents as an example.The construction of information pur-chase platform for scientific research materials plays an important role in enhancing the cost-effectiveness of outlay for scientific research, assuring the safety of founds for scientific research, and improving the service of purchase administrative offices.
2.Molecular cloning and characterization of four small GTPase genes from medicinal fungus Polyporus umbellatus.

Four small GTPase genes which may be relative to sclerotial development were firstly cloned from medicinal fungus Polyporus umbellatus using rapid amplification of cDNA end PCR (RACE) method. The results showed that full-length cDNA of PuRhoA was 698 bp contained 585 bp ORF, which was predicted to encode a 194 amino acid protein with a molecular weight of 21.75 kD with an isoelectric point (pI) of 6.44; the full length cDNA of PuRhoA2 was 837 bp in length and encoded a 194 amino acid protein with a molecular weight of 21.75 kD and an isoelectric point (pI) of 6.33; the full length cDNA of Puypt1 was 896 bp in length and encoded a 204-aa protein with a molecular weight of 22.556 kD and an isoelectric point (pI) of 5.75; the full length cDNA of PuRas was 803 bp in length and encoded a 212-aa protein with a molecular weight of 23.821 kD and an isoelectric point (pI) of 5.2. There are fani acyl transferase enzyme catalytic site and myrcene-transferase enzyme catalytic site in PuRhoA1 while the PuRhoA2 only possess myrcene-transferase enzyme catalytic site. Puypt1 contains the Rab1-Ypt1 conserved domain of small GTPase family and PuRas contains the fani acyl transferase enzyme catalytic site. According to the phylogenetic analysis all these four small GTPase clustered with basidiomycete group. Quantitative real-time PCR analysis revealed that Puypt1, PuRas and PuRhoA1 transcripts were significantly higher in the beginning of sclerotial formation than that in the mycelia, whereas the transcripts levels of PuRhoA2 gene were particularly lower in sclerotia than that in mycelia, suggesting that these four genes might be involved in P umbellatus selerotial development.
3.Expression of recombinant human kallistatin in Pichia pastoris by high density cell culture, and its purification and characterization.

Kallistatin (Kal) is a negative acute phase endogenous protein which can inhibit tumor angiogenesis, growth and metastasis effectively. To express and purify recombinant human kallistatin (rHKal), and characterize its biological activity, P. pastoris was transformed with pPIC9-Kal/GS115 (His4) to express rHKal. The fermentation was carried out in a 7.5 L bioreactor with high density cell culture. 1%-2% methanol was added to the medium to induce the expression of rHKal. The secretion was purified with phenyl sepharose, G-25 sepharose, heparin sepharose and Sephacryl S-100 chromatography. The biological activity of purified bulk rHKal on HUVEC was evaluated with MTT and tube formation assays. The final expression of rHKal in the supernatant reached 50 mg x L(-1), the purity of bulk rHKal after purification was above 98%. A dose-dependent inhibition of rHKal on HUVEC proliferation was observed, however, a U-shaped dose-response curve of rHKal on capillary formation of HUVEC was revealed. The described protocol provides an effective means for preparing rHKal that could be used for anti-angiogenesis therapy in the future.
4.M cell in vitro model and its application in oral delivery of macromolecular drugs.

The oral administration of bioactive macromolecular drugs such as proteins, peptides and nucleic acids represents unprecedented challenges from the drug delivery point of view. One key consideration is how to overcome the gastrointestinal tract absorption barrier. Recent studies suggest that microfold cell (M cell), a kind of specialized antigen-sampling epithelial cell which is characterized by a high endocytic rate and low degradation ability, may play an important role in macromolecule oral absorption. The development of an in vitro M cell coculture system and its modified models greatly advanced the study of M cells and the development of oral delivery system for macromolecular drugs. The special structure, function and formation characteristics, and biomarkers of M cell are summarized in this review. The applications of in vitro M cell models in developing oral delivery system ofbioactive macromolecular drugs are discussed.
5.Effects of Naotaifang on Expression of MMP-9, NF-κB and TIMP-1 after Focal Cerebral Ischemia in Rats

Objective To observe effects of Naotaifang on MMP-9, NF-κB and TIMP-1 after focal cerebral ischemia in rats. Methods The rats were randomly divided into sham operation group, model group, and Naotaifang low- (3 g/kg), medium- (9 g/kg), high- dose (27 g/kg) group. After 3 days of corresponding therapy by intragastric administration once a day, the regional cerebral ischemia model was made by middle cerebral artery occlusion (MCAO) with suture method. Following 3 days, the rats were treated with previous method. On the third day, hippocampal C2 region of ischemic tissue was detected by HE dyeing. And the contents of MMP-9, NF-κB and TIMP1 proteins in hippocampal C2 region were measured by immunohistochemical method. Results The number of normal brain cells in high dose group of Naotaifang was more than that of the model group, and only a few cells appeared nucleus pycnosis. The MMP-9 expression of all dose groups of Naotaifang were significantly decreased than model group (P<0.05). The NF-κB expression of high and medium dose groups of Naotaifang were significantly decreased (P<0.05). The TIMP1 expression of all dose groups of Naotaifang were significantly increased compared with sham operation group (P<0.05). Conclusion The mechanism of Naotaifang protecting blood brain barrier against injury of cerebral ischemia may be involved in ameliorating MMP, NF-κB and increasing TIMP1 expression.

7.Influence of hemoglobin level(Hb) on survival and prognosis of elderly hip fracture at admission.

OBJECTIVE:
To analyze the relationship between hemoglobin(Hb) level on admission and survival prognosis of patients with hip fracture.
METHODS:
From February 2016 to October 2018, 249 elderly patients with hip fracture were surgically treated including 62 males and 187 females;the age ranged from 60 to 91(73.67±10.52) years;the time from injury to operation was (6.79±2.27) d. The clinical and laboratory examination results were collected. The Hb level at admission and the mortality at 30, 90, 180 and 360 days after operation were observed. According to the Hb level at admission, the patients were divided into Hb<120 g/L and Hb≥120 g/L groups. The survival conditions of the two groups at 30, 90, 180 and 360 days after operation were compared and analyzed. Logistic regression was used to analyze the effect of Hb level on death 30, 90, 180 and 360 days after operation.
RESULTS:
The mortality rates at 30, 90, 180 and 360 days after operation were 5.22%, 9.24%, 16.87% and 20.48% respectively. The level of Hb at admission was a risk factor for prognosis and death 30, 90, 180 and 360 days after operation(P<0.05). The OR(95% CI) were 2.431(1.475-4.006), 2.625(1.468-4.695), 2.276(1.320-3.925) and 2.082(1.221-3.551) respectively.
CONCLUSION
The level of Hb at admission can affect the survival and prognosis of elderly patients with hip fracture. We should further study how to manage the level of Hb before operation.

8.Antimicrobial activity of endophytic fungi isolated from Dendrobium species in southwestern China.

OBJECTIVETo isolate and characterize endophytic fungi from seven Dendrobium species, and detect their antimicrobial activities.
METHODFungal endophytes were isolated by strictly sterile sample preparation and fungal identification methods were based on their ITS ribosomal DNA (ITS rDNA gene) sequences. The agar well diffusion method was then employed to evaluate the antimicrobial activity against six pathogenic organisms and the phylogenetic tree of active isolates was constructed by the MEGA.
RESULTNinety-eight endophytic fungi obtained from seven Dendrobium spp., and among them twenty-four isolates, representing 11 genera and 14 species, displayed anti-microbial activities. The phylogenetic assay based on ITS-rDNA showed that 24 active isolates were sorted to 7 taxonomic orders: Hypocreales, Sordariales, Capnodiales, Eurotiales, Botryosphaeriales, Xylariales and Mucorales. The results of antimicrobial activity assay revealed that 1.02%, 10.2%, 18.4%, 1.02%, 1.02% and 10.2% of fermentation broths of 98 isolates displayed significant antimicrobial activities against E. coli, B. subtilis, S. aureus, C. albicans, C. neoformans and A. fumigatus, respectively. Four strains DL-R-3, DL-S-6, DG-R-10 and DN-S-1 displayed strong and broad antimicrobial spectrum.
CONCLUSIONEndophytic fungi associated with Dendrobium species have fungal diversity, and possess diverse antimicrobial activity.

9.Clinical value of three-dimensional ultrasound imaging in the localization and shaping of intrauterine contraceptive device

Objective:To evaluate the clinical value of three-dimensional ultrasound imaging in the localization and shaping of intrauterine contraceptive device.Methods:A total of 46 patients with abnormal intrauterine contraceptive device positions who admitted to Xin'an International Hospital from May 2018 to May 2019 were selected.All patients were examined by two-dimensional ultrasound and three-dimensional ultrasound, and the results were compared with the surgical results of the patients.The application value of three-dimensional ultrasound in the localization and shaping of intrauterine contraceptive device was analyzed.Results:By surgery pathology diagnosed with synthetic in palace in 2 cases, synthetic down in 29 cases, synthetic part or security in 15 cases.The two-dimensional ultrasonic diagnosis of patients with synthetic in the palace in 2 cases, synthetic down in 30 cases, synthetic part or security embedded in 11 cases, abnormal suspicious uncertain in 3 cases, 3 cases of missed diagnosis, 1 case of misdiagnosis.The three-dimensional ultrasound diagnosis in patients with synthetic in the palace in 2 cases, synthetic down in 29 cases, synthetic part or security embedded in 14 cases, abnormal suspicious uncertain in 1 case, no misdiagnosis, missed diagnosis in 1 case, pure synthetic down patients can be removed under hysteroscopy.Patients with IUD outside the uterus were removed laparoscopically without IUD rupture or residual fragments.And synthetic guided by hysteroscopy in patients with incarcerated during extraction, 3 cases of synthetic fracture or residual fragments occurring and compared with synthetic down the difference was statistically significant (χ 2=4.982, P<0.05), compared with the synthetic located outside the uterus, the difference was statistically significant (χ 2=4.634, P<0.05), the incidence of completely bypassed economizer fracture or residual fragments was higher than partial incarceration patients (χ 2=4.845, P<0.05). Conclusion:Three-dimensional ultrasound has higher accuracy in positioning and shaping the intrauterine contraceptive device of patients, which can guide clinicians to choose the treatment program.
10.Application value of 3D energy Doppler ultrasound in clinical diagnosis of endometrial polyps

Objective:To investigate the application value of three-dimensional power Doppler ultrasound in the clinical diagnosis of endometrial polyps.Methods:Seventy-three patients with suspected endometrial lesions admitted to Xin'an International Hospital of Zhejiang Province from June 2018 to June 2019 were enrolled.Three-dimensional power Doppler ultrasound and hysteroscopy were performed on the patients.The results of hysteroscopy and three-dimensional power Doppler ultrasound diagnosis and ROC curve were analyzed.Results:In 73 patients, the pathological diagnosis showed that there were 35 cases in the non-polyp group and 38 cases in the polyp group.The area and standard error under the three-dimensional Doppler ultrasound curve were 0.93 and 0.04, respectively.The area under the hysteroscopic curve and the standard error were 0.92.0.03, respectively, there was no statistically significant difference in the area of ROC curve between the two diagnostic methods ( P=0.051). Conclusion:Three-dimensional power Doppler ultrasound in the diagnosis of endometrial polyps is highly accurate and similar to hysteroscopy.In the clinical examination of endometrial polyps, three-dimensional energy Doppler ultrasonography is worthy of promotion and application.
