Objective:To observe the clinical effect of ulinastatin combined with somatostatin in the treatment of acute severe pan-creatitis. Methods:Totally 130 cases of severe acute pancreatitis patients were randomly divided into two groups with 65 cases in each. The control group was given the routine treatment and 3mg somatostatin in 250ml sodium chloride injections, ivd, bid, and the treatment group was given 10ku ulinastatin in 250ml sodium chloride injections additionally, ivd, bid. After the 10-day treatment, the clinical symptom disappearance time, the recovery time of respiratory and heart rate, length of stay, blood and urine amylase, and clin-ical total effective rate were compared between the two groups. Results:The symptom disappearance time, recovery time of respiratory and heart rate and length of stay in the treatment group were all shorter than those in the control group (P<0. 05). The recovery time of serum and urine amylase in the two groups was without significant difference (P>0. 05). The total effective rate of the treatment group was 95. 38%, while that of the control group was 73. 84% with significant difference (P<0. 05). Conclusion:The application of ulinastatin in the treatment of severe acute pancreatitis shows obviously curative effect, which can effectively improve the clinical symptoms and shorten the course of disease, and is valuable in clinical use.

Objective To investigate the association among daytime sleepiness,serum tumor necrosis factor(TNF)-α levels and microarousal in patients with obstructive sleep apnea hypopnea syndrome (OSAHS).Methods Forty male patients with OSAHS conformed by apnea hypopnea index(AHI)during sleep monitoring of polysomnography were selected as OSAHS group,15 healthy subjects were selected as control group.The level of serum TNF-α was measured by ELISA.The Epworth sleepiness(EP)scale was used in two groups.Results The level of serum TNF-α in OSAHS group[(18.42±6.23)ng/L]was significantly higher than that in control group[(9.75±3.12)ng/L],P＜0.01.The EP scale and micmarousal index were signifieand yhigher in OSAHS group than those in control group[(16.9±4.7)acores vs(4.5±2.3)dcores,(33.6±16.9)times/h vs(11.3±7.3)times/h,P＜0.01].The EP scale in OSAHS group was positively correlated with the levels of serum TNF-α(r=0.461,P＜0.01),AHI(r=0.443,P＜0.01)and microarousal index(r=0.751,P＜0.01)respectively.The levels of serum'INF-α in patients with OSAHS were also positively correlated with microarousal index(r=0.373,P＜0.01).Conclusions The levels of serum TNF-α and mieroarousal index are increased in patients with 0SAHS.The microarousal related to OSAHS plays an important role on the daytime sleepiness,the unusual levels of serum TNF-α maybe lead to interferen of sleepiness.

Objective To study the association of human leukocyte antigen classⅡ(HLA-Ⅱ) alleles with genetic susceptibility and resistance to advanced hepatosplenic schistosomiasis japonica. Methods The allelic types of HLA-DRB1, DPA1, DQA1 and DQB1 were detected by polymerase chain reaction with sequence-specific primers (PCR-SSP) technique in 46 patients with advanced hepatosplenic schistosomiasis, characterized with extensive liver fibrosis. Another 43 subjects with chronic schistosomiasis were used as control. The statistical significance of differences in allelic frequencies was determined by ? 2 test. Results The frequencies of HLA-DRB1*04, DPA1*0103, DQA1*0601, DQB1*0201 in advanced patients were markedly higher than those in control group, while the frequencies of HLA-DQA1*0501 and DQB1*0601 in control group were higher than those in advanced patients. BACKGROUND:With the deepen understanding on the biological function of Rho/ROCK pathway, new ROCK inhibitors continue to be discovered, and ROCK inhibitors show good promoting effects on the survival, proliferation and migration of keratocytes. Research on ROCK inhibitors wil provide more donor materials or seed cels for regenerative medicine and clinical cel transplantation. OBJECTIVE:To summarize and explore the progress in the treatment and application of corneal disease using the ROCK inhibitors Y-27632 and Y-39983. METHODS:The PubMed database and CNKI database were retrieved by computer to search the relevant literature published between 2008 to 2015 using the key words of “corneal endothelial cel, corneal epithelial cel, ROCK inhibitor, Y-39983, Y-27632” in English and Chinese, respectively. Relevant articles in line with the theme were screened and analyzed. RESULTS AND CONCLUSION:Totaly 264 papers were initialy searched. At last, 45 papers were selected. Currently there are two main ROCK inhibitors: Y-27632 and Y-39983, but both of which are stil in basic research stage and clinical testing stage. Y-27632 promotes the proliferation and activity of corneal epithelial stem cel after resuscitation; Y-39983 as a novel ROCK inhibitor can be better to inhibit Rho kinases activity than Y-27632, thereby more effectively promoting the healing of the corneal endothelium. There are many studies on the application of ROCK inhibitors in corneal treatment, but not a stable method established to obtain seed cels. Each method has its own advantages and disadvantages, and how to overcome these disadvantages and to find fast and stable access to seed cels is the future direction of development.

P2Y12 receptor antagonists and morphine are often recommended in patients with acute myocardial infarction.P2Y12 receptor antagonists can rapidly and potently reduce the platelet activity and prevent future thrombotic events.Meanwhile,combined morphine is used to relieve symptoms of angina.A number of studies have confirmed that morphine can decrease plasma concentrations of clopidogrel and impair its anti-platelet activity,which may lead to poor response in clopidogrel-treated patients with acute coronary syndrome.The randomized trials in healthy volunteers and patients with acute myocardial infarction also confirmed the similar drug-drug interaction between morphine and ticagrelor or prasugrel.Although the P2Y12 receptor antagonists combined with morphine are still used for myocardial infarction patients,there are few report on the objective evaluation of this drug interaction.This review,based on the findings of experimental as well as observational and randomized clinical studies,summarizes completely the drug interactions between oral P2Y12 receptor antagonists and morphine.

Objective To construct and express recombinant plasmid containing the structural gene encoding {Mr 31 000} antigen of Trichinella spiralis muscle larvae. MethodsThe target gene TspE1 was amplified by RT_PCR, cloned into pUC18 vector, and sub_cloned into the eukaryotic expression vector pcDNA3. BALB/c mice were immunized with the purified recombinant plasmid pcDNA3_TspE1 by gene_gun bombardment. The expression of recombinant plasmid in the skin tissue was observed by HE staining and immunohistochemical staining. Results and Conclusion The recombinant plasmid pcDNA3_TspE1 was successfully constructed and expressed in the BALB/c mice.

BACKGROUND:Previous studies have demonstrated that total saponins of panax notoginseng can inhibit the ethanol-induced adipogenic differentiation of rabbit bone marrow stromal cells and confirmed that total saponins of panax notoginseng promoted the proliferation and differentiation of rabbit osteoblasts, and improved the osteoprotegerin mRNA relative expression in osteoblasts so as to inhibit receptor activator of nuclear factor kappa B ligand mRNA expression.
OBJECTIVE:To observe effects of total saponins of panax notoginseng on the ultrastructure of the rabbit models of alcoholic avascular necrosis of the femoral head.
METHODS:New Zealand rabbit models of alcohol-induced avascular necrosis of the femoral head were established by gavage of spirit. Successful rabbit models were separately injected with saline, compound bone peptide and total saponins of panax notoginseng group, 0.1 mL/kg, once a day, for 4 consecutive weeks. The ultrastructure of each group were observed by transmission electron microscope.
RESULTS AND CONCLUSON:Transmission electron microscopy showed that osteocytes after alcoholic avascular necrosis of the femoral head presented mitochondrial swel ing and fuzzy crista structure, and degranulation of polysomes on rough endoplasmic reticulum. Lipid droplets were seen in osteocytes. Compared with saline group, mitochondria swel ing subsided, cristae appeared, the number of polysomes increased on rough endoplasmic reticulum, but the number of lipid droplet decreased in total saponins of panax notoginseng group. Morphological changes in ultrastructure were similar between compound bone peptide group and total saponins of panax notoginseng group. Morphological changes in ultrastructure were more significant in the total saponins of panax notoginseng and compound bone peptide groups compared with saline group (P<0.05). Results verified that total saponins of panax notoginseng could effectively restore ultrastructure of osteocytes of rabbit models of alcoholic avascular necrosis of the femoral head in the early stage.

Objective To investigate the expression of CXCR4 and CXCL12 in the labial gland of patients with primary Sj?gren's syndrome (pSS), and to explore their role in the pathogenesis of pSS. Methods The expression of CXCR4 and CXCL12 in labial gland tissues was detected by immunohistochemistry in 32 cases of newly diagnosed pSS and 30 cases of oral mucosa cysts or trauma patients. The expression level, intensity and location were analyzed and compared statistically. χ2 test and Spearman correlation analysis were used for statistical analysis. Results The positive rate of CXCR4 in the test group was 91%(n=29), which was significantly higher than that in the control group (33%, n=10, χ2=21.77, P=0.001). The positive rate of CXCL12 in the test group was 97%(n=31), which was significantly higher than that in the control group 40%(n=12), and the difference was statistically significant ( χ2=23.57, P=0.001). Conclusion CXCR4 and CXCL12 are highly expressed in labial gland of pSS patients, this suggests that they participate in the pathological process of pSS local inflammatory response and play an important role in pSS pathogenesis.

ObjectiveTo observe the effect of 99Tc-methylene diphosphate (99Tc-MDP, Yunke) combined with rehabilitation on ankylosing spondylitis (AS).Methods118 AS patients were randomly divided into the treatment group (n=58) and control group (n=60). The treatment group was treated with meloxicam, sulfasalazine and vein injecting Yunke combined rehabilitation. The control group was treated only with meloxicam and sulfasalazine. The changes of both clinical and experimental indexes of two groups before and after treatment were analyzed.ResultsAfter two therapeutic courses, a much better result was obtained in the treatment group compared with the control group at the respects of releasing arthralgia, shortening morning stiffness, improving range of spinal motion, and decreasing the levels of ESR, CRP, PLT and IgM, ect (P<0.05～0.01) without obvious side effects.ConclusionThe effect of Yunke combined with rehabilitation on AS is evident.

Objective To investigate the inhibitory effect of Xiaoaiping injection (XAP) combined with paclitaxel (PTX) on human ovarian cancer SK-OV-3 cells .Methods In vitro anti-proliferation activity study of XAP combined with PTX on hu-man ovarian cancer SK-OV-3 cells was performed using optical microscope and MTT assay .Human ovarian cancer SK-OV-3 cells were treated with PTX ,XAP ,PTX combined XAP or vehicle control .Each group of cells was treated with drugs for 24 h or 48 h .SK-OV-3 cell morphology was observed with optical microscope .MTT assay was used to detect the A value and cell vi-ability was calculated .In vivo effect of XAP combined with PTX on the growth of SK-OV-3 cells was determined in nude mice . In our study ,thirty-six mice were randomly divided into six groups :G1 (NS) ,G2 (PTX ,10 mg/kg) ,G3 (XAP ,20 ml/kg) , G4 (XAP ,50 ml/kg) ,G5 (PTX 10 mg/kg+XAP 20 ml/kg) and G6 (PTX 10 mg/kg+XAP 50 ml/kg) .Animals were trea-ted for 18 days .Body weight ,tumor volume and tumor inhibition rate were recorded and calculated .The results were analyzed by the SPSS 19 .0 software .Results In vitro study showed that SK-OV-3 cell viability decreased significantly in PTX com-bined XAP group compared to PTX group or XAP group ,in a time and dose-dependent manner .In vivo study showed that the combination of PTX and XAP resulted in decreased tumor weight significantly compared to the control or the PTX alone . Conclusion The combination of XAP and paclitaxel exhibited a synergistic effect both in vitro and in vivo in nude mouse tumor xenograft model .
