Objective To observe the effects of herbal compound in reinforcing the kidney for activating blood and arousing consciousness(HCRAA) on the ABR threshold and the expression of neurotrophic factor 3(NT-3) of the cochlea of gentamicin (GM)-induced ototoxic mice.Methods 40 mice were randomly divided into normal group,model group,and high,middle and low concentration HCRAA groups.Normal group received no treatment.The model group and the HCRAA groups were intraperitoneally injected with GM 100mg/kg per day for consecutive 15 days.At the same time,the model group and the HCRAA groups respectively receiveded normal saline and high,middle and low concentration Chinese medical formula decoction at the same dosage by garage for consecutive 20 days.After the experiment,the mice were tested ABR.The expression of NT-3 of the cochlea in mice was detected by western blot.Results HCRAA at high and middle concentration reduced the GM elevated ABR threshold(P<0.01),and increased the expression of NT-3 in the cochlea(P<0.01).Conclusion HCRAA may effectively reduce the elevated ABR threshold induced by gentamycin by protecting GM damaged cochlear hair cell and neurons and increasing the expression of NT-3 in the cochlea.
2.Correlation between body mass index and physical qualities and functions in 2 056 adults from Guangzhou

AIM: The auxology-related indexes are important for tissue construction. In this article, we investigated the incidence of overweight and obesity and their effects on physical functions and qualities of Guangzhou adults in 2005. METHODS: 2 056 adults including 1 027 males and 1 029 females were randomly selected from Guangzhou in 2005 for physical examination. The anthropometric data such as body mass and height were collected to calculate body mass index (BMI). The subjects were divided according to the BMI criteria recommend by Group of China Obesity Task Force, and their physical functions and qualities were analyzed. RESULTS: In 2005, the detection rate of overweight and obesity in adult males were 28.7% and 5.2%, and in females were 18.6% and 5.3%. The values of diastole pressure and systolic pressure were increased with BMI, but the change tendency of step index and vital capacity/body mass was decreased with BMI. The rate of hypertension was increased with BMI, and male subjects had higher rate of hypertension than that of female subjects in all groups. Obese adult had the poorest values in vertical jump and sit-and-reach test. Those people with normal BMI had better balance ability than that of overweight and obese people. CONCLUSION: The rates of overweight and obesity in Guangzhou adult show a high level. The physical functions and qualities may be improved when the body mass increases or decreases in a certain range, but obesity has impact to those values.
3.Expression of antibacterial peptide gloverin in vitro

AIM: To construct a recombinant vector containing gloverin and express it in vitro by Rapid Translation System(RTS500). METHODS: The gloverin cDNA was amplified by PCR and inserted into the prokaryotic expression vector pIVEX2.3. The recombinant product was identified by PCR and enzyme digestion. The positive reconstructed expression plasmid pIVEX2.3-G was expressed by RTS500 in vitro. The expressed protein was identified by SDS-PAGE and western blotting. RESULTS: Positive recombinant plasmid pIVEX2.3-G was successfully constructed. Target protein of 13.8 Kda with 6-his tag was detected by SDS-PAGE and western blotting. CONCLUSION: Gloverin polypeptide is successfully expressed in inactive E.coli in vitro.

5.The analysis of effect about different therapies on brain metastases from non-small cell lung cancer

Objective To evaluate the prognostic factors of brain metastasis from non-small cell lung cancer and suggest a individualized treatment method proposal with prognostic estimation. Methods From Dec. 2003 to Jan.2007, 183 patients received whole brain radiation therapy (WBRT) were retrospectively analyzed. Kaplan-Meier method was used to analyze the survival. Logrank test was used to evaluate the difference between the groups. Multivariate survival analysis was conducted using a Cox proportional hazard regression model with a backward stepwise procedure. Results The overall l-, 2- and 3-year survival rate was 40.6%, 16.6% and 11.3%, respectively, but with a median survival time of 10.0 months (95% CI 8.6-11.4 months). In multivariate analysis, RAP grouping, weight loss, LDH in blood serum and treatment method were independent prognostic factors. The median survival time of WBRT alone, WBRT with chemotherapy, surgery with chemoradiotherapy and WBRT with Gefitinib was 9.0, 9.0, 22.0 and 13.0 months, respectively, but their difference were statistical significant (X2 = 10.37, P = 0.016). Conclusions The main prognostic factors of brain metastasis from non-small cell lung cancer are RAP grouping, weight loss, LDH in blood serum and treatment method. The survival time is prolonged by proper multidiseiplinary management than WBRT alone. The effect of combined treatment of surgery with chemoradiotherapy is favorable for the patients operated with single region of metastasis.
6.Prognosis analysis of esophageal carcinoma patients with tumor loco-regional recurrence after curative surgery

Objective To evaluate the outcome of radiotherapy for recurrent esophageal cancer after surgery and to determine the prognostic factors. Methods From Jan 2004 to Dec 2009, 93 patients of esophageal carcinoma with loco-regional recurrence as the first site of failure after surgery were retrospectively reviewed. Kaplan-Meier method was used to analyze the survival. Logrank test was used to evaluate the difference between the groups. Multivariate survival analysis was conducted using a Cox proportional hazard regression model with a backward stepwise procedure. Results The overall survival rates at 1, 2 and 3 years were 40. 9% , 10. 1% and 6. 7% ,respectively,but with a median survival time of 11.0 months(95% CI 9. 4-12. 6) . In univariate analysis, age,PS, radiation dose and retreatment methods were independent prognostic factors. In multivariate analysis, only radiation dose and retreatment methods were independent prognostic factors for overall survival. Conclusions The prognosis of patients with postoperative loco-regional recurrence of esophageal cancer is poor. However, a long-term survival maybe expected by definitive chemoradiotherapy.
7.Dosimetric evaluation of different IMRT treatment plans for non-small cell lung cancer

Objective To investigate the optimal treatment planning of intensity modulated radiotherapy (IMRT) for non-small cell lung cancer (NSCLC) .Methods Two types of treatment plans were designed for 11 patients with inoperable NSCLC disease.In the first plan(PTV60 plan) ,60 Gy was prescribed to the planning target volume(PTV) which was created using CTV(GTV +6-8 mm) plus the margin for organ motion and setup uncertainties.In the second plan(PTV70 plan) ,70 Gy was prescribed to the PTV which was created using GTV plus the margin for organ motion and setup uncertainties.The dose-volume histogram,the planning target volume coverage,and other dosimetric parameters of normal structures were compared between the two plans.Results These two plans were not significantly different in the dose heterogeneity,but commpared with the PTV60 plan,the PTV70 plan significantly improved 60 Gy volume with regard to PTV coverage.The values of V_(20) and V_5 of lung were reduced in average by (1.69 ±0.42) % ,and (1.29 ±1.09) % (t = 0.047,0.264,P = 0.002) ,respectively.Conclusions The planning of 70 Gy to the PTV using GTV plus the margin for organ motion and setup uncertainties might be better than that of 60 Gy to the PTV using CTV(GTV +6-8 mm) plus the margin for organ motion and setup uncertainties.
8.Impact of Exercise Rehabilitation on Cardiac Function in Coronary Artery Disease Patients After Percutaneous Coronary Intervention

Objective: To explore the impact of exercise rehabilitation on cardiac function in coronary artery disease (CAD) patients after percutaneous coronary intervention (PCI). Methods: A total of 130 CAD patients received primary PCI in our hospital from 2014-01 to 2015-09 were enrolled. All patients received conventional drug therapy and post-PCI knowledge education, then were randomly divided into 2 groups: Conventional group and Rehabilitation group, in which the patients received cardiac rehabilitation exercise for 3 months at different stage and intensity. n=65 in each group. The cardiac function including left ventricular ejection fraction (LVEF), left ventricular end-diastolic diameter (LVEDD), 6 min walking distance (6MWD) and NYHA classification were compared between 2 groups at 1, 3 and 6 months after the operation. Results: LVEF, LVEDD, 6MWD and NYHA classification were similar between 2 groups at enrollment, P>0.05. ① At 1 month post-operation: 6MWD was different between 2 groups, P<0.05, while LVEF, LVEDD, NYHA classification and the recurrence rate of angina pectoris were similar between 2 groups, P>0.05. In Rehabilitation group, 6MWD and NYHA classification were different from the enrollment condition, P<0.05, while other parameters were similar, P>0.05; in Conventional group, 6MWD was different from the enrollment condition, P<0.05, while other parameters was similar, P>0.05. ② At 3 and 6 months post-operation: all parameters were different between 2 groups, P>0.05. In Rehabilitation group, all parameters were different from the enrollment condition, P<0.05. ③ In Conventional group, at 3 months post-operation: LVEDD and 6MWD were different from the enrollment condition, P<0.05; at 6 months post-operation: 6MWD was different from the enrollment condition, P<0.05. Conclusion: Exercise rehabilitation may improve the cardiac function, therefore enhance the endurance capacity and quality of life in CAD patients after PCI.
9.Prokaryotic expression, purification and identification of anti-bacterial peptide gloverin

Objective To express anti-bacterial peptide gloverin in prokaryotic system and investigate its bioactivity. Methods E.coli BL21 was used to express gloverin. Western blot was used to identify the production and Tachypleus Amebocyte Lysate (TAL) was used to detect the bioactivity of the production. Results The production was identified as gloverin peptide by Western blotting, and the TAL data indicated that the production could neutralize LPS. Conclusion Anti-bacterial peptide gloverin is successfully expressed in E.coli BL21.
