1.The influence of Gardiner bacteria infection on premature birth and changes of endocrine hormone levels

Objective To investigate the effect of Gardiner bacteria infection on preterm delivery and changes of endocrine hor-mone levels.Methods Application of Gardiner bacteria fluorescent marker detection kit in 120 cases of suspected Gardiner bacteria infection pregnant women with immunofluorescence assay,and the levels of serum follicle stimulating hormone (FSH),luteinizing hormone (LH),estradiol (E2),progesterone (P)were measured at the same time,90 cases of non maternal Gardiner bacteria infec-tion were enrolled as control.Results After the treatment,98 cases of pregnant women with Gardiner bacteria infection should ex-perienced premature delivery (24-37 week),accounted for 81.67%,compared with the normal pregnant women were statistically significant (P <0.05);and 22 cases of Gardiner bacteria infection pregnant women after treatment no premature birth,accounted for 18.33%,and to all pregnant women infected Gardiner bacteria and experiencing preterm delivery and the control group levels of endocrine hormone were determined,compared the results of FSH,LH,found no significant differences(P >0.05),E2 increased,P decreased,changed significantly (P <0.05 ).Conclusion The probability of premature delivery in women infected with Gardiner bacteria is significantly higher than that of normal pregnant women,at the same time the secretion of hormone changes,so dealing with pre pregnancy and early pregnancy should carry owt Gardiner bacteria examination.
2.Effect of gastric signet ring cell cancer on T,N,M stages

Objective To evaluate the effect of the gastric signet ring cell cancer(SIG)on the T,N,M stages. Methods Three hundred and fifty-one patients undergoing D2 or greater gastrectomy for gastric cancer were ana lyzed retrospectively.The T,N,M stages of 59 patients with SIG and 161 patients with tubular adenocarcmonla (TUB)were compared using univariate and multivariate analysis.Results The chi-square test showed that there were not significant differences in T,N,M stages between TUB and SIG.Multivariate Logistic regression analysis indicated that SIG Was not a independent factor influencing the T,N.M stage.Conclusion Gastric cancer with SIG has not higher T,N,M stages than that with TUB.
3.Analysis of gene expression profile of multidrug resistant MCF/DOX cell line after benflumetol derivative LY980503 treatment

AIM: To investigate the effect of LY980503(a benflumetol derivative)on multidrug resistance of tumor cell line using DNA microarray. METHODS: Total RNA was extracted from multidrug resistant MCF/DOX cell line. cDNA microarray containing 320 cDNAs was used to detect the gene expression profile. RESULTS: 9 down-regulated genes and 1 up-regulated gene were identified after multidrug resistant MCF/DOX cells were treated with LY980503. CONCLUSION: LY980503 can effectively reverse the resistance of MCF/DOX to DOX in vitro by adjusting the expression of multi-genes.
4.Relationship between angiotensin Ⅱ and pancreatic islet β cell secretion function in patients with type 2 diabetes mellitus

Objective To investigate the relationship between angiotensin Ⅱ and pancreatic islet β cell secretion function under different glucose tolerance statuses. Method Forty-two patients with newly diagnosed type 2diabetes mellitus ( DM group), 38 subjects with impaired fasting glucose/impaired glucose tolerance ( IFG/IGTgroup) ,and 40 normal control subjects (NGT group) underwent intravenous glucose tolerance test. Fasting plasma angiotensin Ⅱ ( Ang Ⅱ ) and adiponectin were assayed by ELISA. Acute insulin response from 3 to 10 min( AIR3-10 ),the area under the curve( AUCⅠ ) and the peak concentration of the first-phase ( 0-10 min) insulin secretion, the area under the curve of the second-phase( 10-120 min) insulin secretion( AUCⅡ), homeostasis model assessment for β cell function index(HOMA-β) and homeostasis model assessment for insulin resistance index(HOMA-IR) were calculated to explore the relationship with Ang Ⅱ. Result ( 1 ) The levels of Ang Ⅱ in DM group and IFG/IGT group were significantly higher than that in NGT group( P<0.05 ). The AIR3-10, AUCⅠ and peak concentration, AUCⅡ ,adiponectin in DM group and IFG/IGT group were significantly lower than those in the NGT group ( P<0. 05), and these results were more significantly reduced in DM group compared with those in IFG/IGT group. (2) Ang Ⅱ was negatively correlated with AIR3-10, AUCⅠ and the peak concentration, AUCⅡ, adiponectin, HOMA-β ( P<0. 01 ), and positively correlated with fasting blood glucose,2 h blood glucose after glucose loading, fasting insulin, HOMA-IR (P<0. 05 ). (3)Multiple stepwise regression analysis showed that Ang Ⅱ was independently associated with AUCⅠ and AUCⅡ.Conclusion Ang Ⅱ was an independent factor that affected the insulin secretion function of pancreatic islet βcells. Ruling out the effect of blood pressure, body position, drugs, and other factors, high levels of Ang Ⅱ could predict the dysfunction of pancreatic islet β cell as well as insulin resistance in patients with type 2 diabetes.
5.Relationship between oxidative stress and the first-phase of glucose-stimulated insulin secretion from pancreatic β cell in individuals with different glucose tolerance

The relationship between oxidative stress and the first-phase of pancreatic β cell insulin secretion in subjects with different statuses of glucose tolerance was explored. Fasting adiponectin, 8-hydroxydeoxyguanosin ( 8-OHdG), malondialdehyde ( MDA ), superoxide dismutase ( SOD ), insulin area under the curve ( AUC ) from 0 to 10 min, and AIR3-5 were measured. The levels of oxidative stress-related markers were elevated, the activities of superoxide dismutase,adiponectin, and first-phase of insulin secretion were reduced with the disease progression. MDA and 8-OHdG were negatively correlated with adiponectin, homeostasis model assessment β cell function index ( HOMA-β ),AUC ,and AIR3-5. SOD was positively correlated with adiponectin, HOMA-β, AUC, AIR3-5. The plasma 8-OHdG and SOD were independently associated with AIR3-5. Oxidative stress exerts a significant effect on the first phase of pancreatic β cell insulin secretion, which may contribute to the pathogenesis of type 2 diabetes.
6.Impact of anti-hypertension therapy on seasonal variability of blood pressure, urinary 8-OHdG levels in essential hypertension

Objective To investigate anti-hypertension threapy on seasonal variability of blood pressure,urinary 8-OHdG levels in essential hypertension in order to provide a basis for seasonal antihypertensive treatment.Methods Fifty hypertensive patients admitted the hospital of Erdos Clinical Medical College of Inner Mongolia Medical University at summer 2013 were selected as our subjects.The final subjects was 42 cases due to 8 lost cases.The patients were randomly divided into two groups,including 30 cases in renin angiotensin aldosterone system inhibitors(RASI) group and 12 cases in Ca2+ channel blocker(CCB) group.Epidata 3.1 software was applied to perform statistic analysis.Urinary 8-OHdG concentration was measured by enzyme-linked immunosorbent assay (ELISA).Blood pressure was measured in spring,summer,autumn and winter.Results Systolic blood pressure(SBP) in RASI group and CCB group at winter periods were (158±20) mmHg,(158 ± 20) mmHg,higher than that in summer periods ((145 ± 12) mmHg,(141 ± 9) mmHg;P< 0.05).Diastolic blood pressure(DPB) in RASI group and CCB group at winter periods were (101 ± 13)mmHg and (100±4)mmHg,significant high than that in summer periods ((93 ±7) mmHg,(90±7) mmHg;P<0.05).8-OhdG levels in RASI group at summer and autumn periods were (243.20±30.94) ng/L and (240.40±47.99) ng/L,significantly higher than that in winter and spring periods((190.80± 15.56) ng/L and (189.06± 13.56) ng/L),and the differences were significant(P<0.001).No significant differences were seen in CCB group among 4 seasons in terms of 8-OhdG levels (P > 0.05).Conclusion Blood pressure change still occur among 4 seasons in hypertensive patients after a single CCB containing RASI-based drug antihypertensive therapy.And blood pressure in winter periods is higher than that in summer,which indicates that therapy medication based on RASI might reduce the level of oxidative stress at winter periods.
7.Impact of Exercise Rehabilitation on Cardiac Function in Coronary Artery Disease Patients After Percutaneous Coronary Intervention

Objective: To explore the impact of exercise rehabilitation on cardiac function in coronary artery disease (CAD) patients after percutaneous coronary intervention (PCI). Methods: A total of 130 CAD patients received primary PCI in our hospital from 2014-01 to 2015-09 were enrolled. All patients received conventional drug therapy and post-PCI knowledge education, then were randomly divided into 2 groups: Conventional group and Rehabilitation group, in which the patients received cardiac rehabilitation exercise for 3 months at different stage and intensity. n=65 in each group. The cardiac function including left ventricular ejection fraction (LVEF), left ventricular end-diastolic diameter (LVEDD), 6 min walking distance (6MWD) and NYHA classification were compared between 2 groups at 1, 3 and 6 months after the operation. Results: LVEF, LVEDD, 6MWD and NYHA classification were similar between 2 groups at enrollment, P>0.05. ① At 1 month post-operation: 6MWD was different between 2 groups, P<0.05, while LVEF, LVEDD, NYHA classification and the recurrence rate of angina pectoris were similar between 2 groups, P>0.05. In Rehabilitation group, 6MWD and NYHA classification were different from the enrollment condition, P<0.05, while other parameters were similar, P>0.05; in Conventional group, 6MWD was different from the enrollment condition, P<0.05, while other parameters was similar, P>0.05. ② At 3 and 6 months post-operation: all parameters were different between 2 groups, P>0.05. In Rehabilitation group, all parameters were different from the enrollment condition, P<0.05. ③ In Conventional group, at 3 months post-operation: LVEDD and 6MWD were different from the enrollment condition, P<0.05; at 6 months post-operation: 6MWD was different from the enrollment condition, P<0.05. Conclusion: Exercise rehabilitation may improve the cardiac function, therefore enhance the endurance capacity and quality of life in CAD patients after PCI.
8.Chromosome microarray analysis of patients with 18q deletion syndrome.

OBJECTIVETo analyze the correlation between the genotype and phenotype of 18q deletion syndrome with chromosome microarray analysis (CMA).
METHODSEight cases with 18q deletion syndrome were selected, including two affected fetuses and six children patients. DNA was extracted and hybridized with Affymetrix CytoScan TM 750K arrays following the manufacturer's standard protocol. The data was analyzed with a special software package.
RESULTSCMA analysis identified pathogenic copy number variations (CNVs) on 18q in all cases, which ranged from 6.612 Mb to 22.973 Mb. NFATC1, GALR1, MBP, SALL3 and TSHZ1 are likely to be causative genes for congenital heart disease, psychological, growth retardation, and cleft palate.
CONCLUSIONCMA can precisely locate the breakpoints of 18q and facilitate definition of the genotype-phenotype correlations, which is useful for prognosis.

9.Failure mode and long-term survival after neoadjuvant therapy for locally advanced esophageal squamous cell carcinoma

Objective:To analyze the fail mode of neoadjuvant therapy combined with surgery for locally advanced esophageal squamous cell carcinoma (ESCC) after long-term follow-up.Methods:Clinical data of consecutive 238 patients with locally advanced resectable ESCC who underwent neoadjuvant therapy combined with surgery in Zhejiang Cancer Hospital from September 2012 to October 2019 were retrospectively analyzed. The failure mode in the whole cohort was analyzed after long-term follow-up. The overall survival (OS) and disease free survival (DFS) rates were analyzed by Kaplan-Meier method. Survival differences were determined by log-rank test.Results:The pathological complete response (pCR) rate was 42.0% in 238 patients. After a median follow-up of 46.1 months, tumor progression occurred in 96 patients (40.3%), including 25 patients (10.5%) with local recurrence, 61 patients (25.6%) with distant metastases, and 10 patients (4.2%) with simultaneous local recurrence and distant metastases. The median OS and DFS were 64.7 months and 49.9 months. And the 3-, 5-, and 7-year OS and DFS rates were 70.0%, 52.8%, 36.4% and 63.5%, 42.5%, and 30.0%, respectively. The 3-, 5-, and 7-year locoregional recurrence-free survival rates and distant metastasis-free survival rates were 86.0%, 71.4%, 61.2% and 70.6%, 55.9%, 43.0%. Compared with non-pCR patients, the overall progression rate and distant metastasis rate of pCR patients were lower (26.0% vs. 50.7%, 16.0% vs. 32.6%, both P<0.05). And the 3-, 5-, and 7-year OS (83.0% vs. 60.2%, 69.7% vs. 41.7%, 50.4% vs. 27.7%, all P<0.001) and DFS rates (80.4% vs. 51.4%, 63.9% vs. 31.2%, 45.9% vs. 20.3%, all P<0.001) were significantly better in pCR patients. Conclusions:Distant metastasis is the main failure mode of patients with locally advanced ESCC after neoadjuvant therapy. Patients with postoperative pCR can achieve better long-term survival.
10. Screening of pathogenic genes in a Chinese familial dilated cardiomyopathy pedigree from Inner Mongolia

Objective:
Screen the pathogenic genes of a pedigree with clinical manifestation of familial dilated cardiomyopathy in Inner Mongolia.
Methods:
A total of 3 patients with dilated cardiomyopathy and 20 family members from the same family were examined in Ordos Central Hospital in Inner Mongolia from October, 2003 to August, 2017. Data on medical history, physical examinations, electrocardiograms, and echocardiography were obtained. 5 ml peripheral blood was sampled for per person. Chip Capture Sequencing technology was used to capture all the exons and splice sites of the genes that associated with hereditary cardiomyopathy and hereditary arrhythmia. The mutations in these genes were detected by high-throughput sequencing. All suspected pathogenic loci identified by high-throughput sequencing were verified by Sanger sequencing used for mutation detection. One hundred and fifty gender, age and race matched healthy people were included as the control group.
Results:
Pathogenic gene variations were detected in 3 symptomatic family members and 1 carrier from the pedigree. Five pathogenic gene variations were identified in the proband (Ⅱ1), a pSer236Gly and a pArg215Cys variation in the MYBPC3 gene, a pGln90Arg variation in the DSP gene, and pAsn2912Asp and pGlu2910Val variation in the DMD gene. One pathogenic variation was detected in Ⅲ3, which was a pArg215Cys variation in the MYBPC3 gene. Two pathogenic variations were detected in Ⅲ7, a pSer236Gly variation in the MYBPC3 gene and a pGln90Arg variation in the DSP gene. Two pathogenic variations were detected in the Ⅳ7, a pSer236Gly variation in the MYBPC3 gene and a pGln90Arg variation in the DSP gene. No gene variation loci were detected in the other family members and the control group.
Conclusion
MYBPC3 gene, DSP gene and DMD gene variations are present in the familial dilated cardiomyopathy pedigree from Inner Mongolia, and these variations may be related with familial dilated cardiomyopathy.
