The present study were to identify a dose-response relationship and study the histologic correlates. Group A : The rabbit's iliac segments of normal control,were performed in radiofrequency balloon angioplasty. Group B: The iliac segments of rabbits, which had been manually separated in to intime- media and media-adventitia layes, were performed in radiofrequency balloon angioplasty. Radiofrequency dose delivered from a metal electrode balloon produced dose-dependent thermal energy with compression. It produces fusion of separated wall layers. 2.Effect of Paroxetine in Treatment of Depressive Patients with Grave's Disease

Objective: To study depression of patients with Grave's disease and the therapeutic effect of Paroxetine (antidepressant). Method: 82 patients with first onset Grave's disease were collected and 52 of them had depression. The depressive patients were divided into Paroxetine and control group. All cases had the same anti-hyperthyroidism treatment. Result: 63.4% (52/82) patients with first onset Grave's disease had depression before Paroxetine treatment. After 4 weeks and 8 weeks treatment, Paroxetine group had greater decrease in FT3 and FT4, and lower scores of SDS and SAS than control group (p<0.01). Conclusion: Paroxetine does enhance the therapeutic effect of anti-hyperthyroidism, as well as improving depression of patients with first-onset Grave's disease.

5.A comparative study of platelet serotonin in patients after traumatic brain injury with obsessive compulsive disorder

Objective To explore the role of serotonin(5-HT) in obsessive compulsive disorder(OCD) and the difference in platelet 5-HT content between OCD and healthy controls, the obsession and the compulsion subgroup. Methods The concentration of serotonin (5-HT) in twenty-seven patients with OCD and twenty-seven patients without OCD were detected in the study. Results Platelet serotonin level in patients with OCD ( ( 139 ±172 ) μg/L) was lower than that in patients without OCD ( ( 248 ± 215 ) μg/L), and the differences were significant (P<0.05). Conclusion The present results support the hypothesis that 5-HT hypofunctionality contribute to OCD. And the differences between the obsession and the compulsion subgroup in the role of 5-HT are significant.
6.Effect of lentivirus-mediated RNA interference silencing IL-1 type Ⅰ receptor on signal transduction in a rat model of osteoarthritis

Objective To observe the effects of lentivirus-mediated RNA interference silencing IL-1 type Ⅰ receptor on the expression of mitogen-activated protein kinase (MAPK) in a rat model of osteoarthritis (OA), and to explore the role of IL-1 and MAPK in OA. Methods Forty SD rats were randomly divided into four groups(A-D, each group: n = 10). The rats (A-C group) were underwent unilateral anterior cruciate ligament transaction and partial excision of the medial meniscus. At day 7 and 9 after operation, the rats of group A were respectively received 40 μL of lentivirus-mediated RNAi silencing IL-1 type Ⅰ receptor by intra-articular injection, while the rats of group B were received lentivirus-mediated RNAi non-silencing IL-1 type Ⅰ receptor, and the rats of group C were received saline. The rats of group D were taken for normal control. All rats were sacrificed four weeks after the surgery. The knees were harvested to observe macropathologic changes of the joint cartilage. ERK1/2, JNK1/2 and p38 proteins in the cartilage were detected by Western blot. Results Cartilage degradation in group A was milder than that in group B and C (P<0.05),but was worse compared to group D (P<0.05). Level of p38 expression in cartilage in group A was lower than that in group B and C (P<0. 05), but had no significant difference compared to group D (P>0.05). Levels of ERK1/2 and JNK1/2 expressions in cartilage in group A were lower than that in group B and C(P<0.05),but was significantly increased compared to group D(P<0.05). Conclusions Lentivirus-mediated RNAi silencing IL-1 type Ⅰ receptor can inhibit the expression of MAPK, in particular,p38 protein was strongly inhibited.
7.Effects of new derivatives of 1H-isoindole-1,3(2H)-dione on angiogenesis in chick chorioallantoic membranes

Aim To investigate the effects of new derivatives of 1H-isoindole-1,3(2H)-dione on angiogenesis in chick chorioallantoic membranes(CAM). Methods The blood vessels of CAM were calculated to determine the antiangiogenic effects of different new compounds. Results Among the 18 new derivatives of 1H-isoindole-1,3(2H)-dione , compound 1,2,3,4,11 and 13 showed significant inhibition on angiogenesis of CAM.Conculsion The 6 new derivatives of 1H-isoindole-1,3(2H)-dione screened out in this experiment may be novel angiogenesis inhibitors.

9.The cDNA cloning of human granulysin Mr 15000 and Mr 9000 active segments from CTL activated by allogenic antigen

Objective; To clone, sequence and analyze the coding sequences of the Mr 15000 and Mr 9000 active segments of natural granulysin derived from human CTLs activated by allogenic antigen ,in order to establish the basis for further purifying and investigating the immune - impairing mechanism of granulysin. Methods; The coding sequences of the Mr 15000 and Mr 9000 granulysin gene were amplified from the total RNA of activated CTLs of healthy person after reverse transcription by Nested - PCR, inserted into pET32a ( + ) vectors and then transformed into E. Coli TOP10, respectively. The recons were identified by PCR, endonuclease digestion and sequencing. Results: The whole coding sequences of the Mr 15000 and Mr 9000 active segments were successfully cloned as expected. The accurate pET32a - Mr 15000 and pET32a - Mr 9000 recons were obtained through Colony - PCR, endonuclease digestion and sequencing. There lied polymorphism on the 119 th amino acid of the product encoded by GLS gene. Conclusion; The coding sequences of the Mr 15000 and Mr 9000 active segments of human granulysin can be obtained and cloned by the methods mentioned above and can be used for subsequent research.
10.Bactericidal activity of human granulysin expressed in macrophages against intracellular Mycobacterium tuberculosis

Objective To construct eukaryotic expression recombinant plasmid containing human granulysin(GLS) and investigate the effect of GLS expression in macrophage RAW264.7 cells on the bactericidal activity against intracellular Mycobacterium tuberculosis.Methods GLS gene was amplified by nested-polymerase chain reaction(PCR) from human cytotoxicity T lymphocyte(CTL) activated by allogenic antigen,and inserted into pBudCE4.1 vector to construct recombinant plasmid.Subsequently,the plasmid was transfccted into RAW264.7 cells which were infected with Mycobacterium tuberculosis H37Rv.The expression of GLS was detected by nested-PCR and immunocytochemistry method.The RAW264.7 cells were lysed after transfected for 96 h,then acidfast stained,cultivated and colony count were done to determine the intraeellular bactericidal activity of GLS.The data were analyzed by t or t' test.Results The pBudCE4.1/GLS eukaryotic expression recombinant plasmid was successfully constructed.The transcriptional and translational expressions of target gene GLS were detected in RAW264.7 cells which were infected with Mycobacterium tuberculosis H37Rv.The bacterial load in macrophages of phorbol myristate acetate(PMA)+pBudCE4.1/GLS group,PMA+pBudCEA.1 group and non-activated group were 1.44±1.25,3.16±0.20 and 3.59±0.21,respectively.The differences between groups were all significant (t=2.403,t=2.854,both P<0.05).Conclusion Eukaryotic expression recombinant plasmid carrying human GLS gene expressed in macrophages has strong bactericidal activity against intracellular mycobacteria,which provide information for the further study on therapeutic vaccine against Mycobacterium tuberculosis.
