1.Determination of Baicalin in Muxiangliqi Tablet by HPLC

Objective To develop a method for the determination of Baicalin in Muxiangliqi tablet. Methods To determine the quantity of Baicalin in the Muxiangliqi tablet by HPLC. The mobile phase was MeOH-H2O-Glacial acetic acid (42∶55∶3). The velocity of flow was 1.0 mL/min. The detectionavelength was 280 nm. The column temperature was room temperature. Results The curve of baicalin was lindar at the range of 0.201~4.02 ?g. The average recovery rate was 96.61%, RSD=1.10%. Conclusion The method was simple, sensitive and stable, and can be used as the method of quality control for Muxiangliqi tablet.
2.Effect of Ginsenosides on Memory Retention of Rats in Animal Models of Schizophrenia

Objective:To explore the effect of ginsenodels on the memory retention of rats of animal models of schizophrenia.Methods:The rats in experiment were divided into experimental group and control group, the rats in the experimental group were orally given Ginsenosides, and rats in control group were given normal saline. Both groups were treated for 5 days, then given injection of MK801 to establish animal models of schizophrenia. Before (d0), during (d1,d3) and after (d5) MK801. 3.Patient-controlled epidural analgesia with ropivacaine combined with fentanyl for labor

Objective To compare two methods of patient controlled epidural analgesia (PCEA) with 0 2% ropivacaine plus fentanyl 2?g?ml -1 with or without background infusion for labor Methods Ninety ASA Ⅰ Ⅱ full term primigravidae in active labor who had a single fetus with vertex presentation and were expected to have vaginal delivery were randomly divided into three groups of 30 each: group A received PCEA without background infusion; group B received PCEA with background infusion and group C received no analgesia of any kind and served as control PCEA included a bolus of 4 ml with a 15 minute lock out When the primigravida was in first stage of labor, an intravenous line was established and 5% glucose normal saline 500 1000 ml was being infused When the external cervical os was dilated to 3 cm, epidural catheter was placed at L 2 3 and a test dose of 4 ml was given 5 min later when no signs of subarachnoid injection was evident, block height was tested by pinprick and another 6 ml was given 30 min later in group B background infusion of 0 2% ropivacaine + fentanyl 2?g?ml -1 was started at a rate of 4 ml?h -1 until the second stage of labor began Maternal vital signs (BP, ECG, SpO 2, P ET CO 2), VAS scores, degree of motor block, drug consumption, side effects of PCEA, gas analysis of umbilical venous blood, progress of labor, and Apgar scores were noted Venous blood samples were taken before PCEA and at the end of first stage of labor for determination of serum epinephrine and norepinephrine levels Results There were no significant differences in Apgar scores, blood gas of umbilical venous blood and the durations of first and second stage of labor among the three groups There were no differences in VAS scores, degree of sensory and motor block, serum concentrations of epinephrine and norepinephrine and percentage of cesarean section between group A and B The percentage of cesarean section was significantly higher in control group than that in group A and B Plasma NE and E concentrations at the end of the first stage of labor were significantly higher in control group than those in group A and B The ropivacaine and fentanyl consumption was less and the incidence of itching and percentage of instrumental delivery were lower in group A than those in group B Conclusions PCEA with 0 2% ropivacaine and fentanyl 2?g?ml -1 was safe and effective It reduces the percentage of cesarean section PCEA without background infusion provides the same level of analgesia as PCEA with background infusion with less drugs and side effects

6.Observation of the effect of three-dimensional conformal radiation dose fractionation treatment of advanced non-small cell lung cancer

Objective To explore the effect of three-dimensional conformal radiation dose fractionation treatment of advanced non-small cell lung cancer(NSCLC).Methods 75 NSCLC patients were treated with hypofractionated 3DCRT( observation group),while 73 cases were treated with conventional fractionated radiotherapy (control group).The efficacy and adverse reactions were observed;Survival after treatment were followed up in 1,2,3 years.Lung function was detected before and after radiotherapy treatment,including FVC,FEV1 and CLCO.Results The total effective cases of control group were 47 cases patients ( 64.4% ),observation group's was 60 cases ( 80.0% ),total effective rate had statistically significant difference ( x2 =4.50,P < 0.05 ).Survival of control group after treatment in 1,2,3year were 50.7%,24.7%,8.2%,the median survival was 13 months,observation group's were 73.3%,45.3%,20.0%,and 19 months.The survival of these two groups was statistically different (x2 =8.07,6,94,4.22,all P < 0.05 ).The patients blood system side effects of observation group were significantly lower than the control group ( x2 =4.73,P <0.05 ) ;acute radiation pneumonia,esophagitis of these two groups had no significant difference in the incidence( P > 0.05 ).Conclusion Hypofractionated 3DCRT treatment of advanced NSCLC had good effect,and its adverse reactions was low,and it was worthy of clinical application.
7.Clinlcal study of hyper-fractionated three-dimensional conformal radiotherapy combined with transcatheter arterial chemoembolization for treating patients with inoperable primary livercancer

Objective To investigate the clinical effect of hyper-fractionated three-dimensional conformal radiotherapy (3DCRT) combined with transcatheter arterial chemeembolization (TACE) for heating patients with inoperable primary liver cancer.Methods One hundred and two patients with inoperable advanced primary liver cancer were divided by random digits table method into control group with 49 cases treated with TACE and observation group with 53 cases treated with hyper-fractionated 3DCRT combined with TACE.The short-term efficacy,adverse reaction and survival rate of the 1st,2nd and 3rd year and serum alpha-fetoprotein (AFP) expression after treatment in two groups were observed.Results The complete remission rate,progress rate and total efficiency were 17.0% (9/53),11.3% (6/53),73.6% ( 39/53 ) in observation group,2.0%( 1/49 ),28.6% (14/49),51.0%(25/49 ) in control group.There were significant differences between two groups (P < 0.05).There was no statistical significance in adverse reactions between two groups ( P > 0.05 ).The survival rates of the 1 st,2nd and 3rd year in control group were 51.0% ( 25/49 ),28.6%(14/49),14.3%(7/49),the median survival time was 15 months;while those in observation groupwere 71.7%(38/53),52.8%(28/53),39.6%(21/53) and 20 months.The survival rates of the 2nd and 3rd year in observation group were significantly higher than those in control group respectively (P <0.05 or <0.01 ).The rate of patients with no obvious serum AFP protein change between observation group [ 9.8%(4/41) ] and control group [ 31.6%(12/38) ] had statistical significance (P < 0.05).Conclusion Hyper-fractionated 3DCRT combined with TACE for treating patients with inoperable primary liver cancer has good clinical efficacy,it is worthy of clinical practice.
8.Relationship between antitumor effect of valdecoxib and COX-2 expression

Objective To evaluate the inhibitory effect of valdecoxib on the growth of the cancer cell lines and involvement of COX-2 in this inhibition. Methods Western blotting and immunocytochemistry were used to detect the expression of COX-2. MTT assay was used to determine inhibitory effect of the drugs on the cell growth. The content of PGE_ 2 in cell medium was determined with PGE_ 2 ELISA kit. Results ①Clone 26 cells expressed high levels of COX-2, whereas BGC-823,HGC-27 and SK-OV-3 cell had no COX-2 expression. ②Valdecoxib inhibited the growth of BGC-823, HGC-27, SK-OV-3 and clone 26 cells, with a IC_ 50 of 110.7, 99.2, 113.3, 117.6 ?mol/L, respectively. ③The inhibitory effect of these drugs on BGC-823 and clone 26 cell was in the descending order of valdecoxib, SC-560 and indomethacin. ④PGE_ 2 did not antagonize the effect of valdecoxib, SC-560 and indomethacin on BGC-823 and clone 26 cells. ⑤The inhibitory effect of valdecoxib and indomethacin on the growth of clone 26 cells was not compatible with that on PGE_ 2 . Conclusion The inhibitory effect of valdecoxib on cell growth is not related to its effect on COX-2.
9.The influences on the structure of rats′aorta with long-term high fat forages diet

Aim To investigate the influences on the aortal structure of rats with long-term high fat forages diet.Methods 14 SD rats were divided into two groups:the control group and the test group. The test rats were fed with high fat forages.12 weeks later, the aortas of the rats were observed with a light microscope, transmission electron microscope(TEM) and scanning electron microscope (SEM).Results In the test group, the aortic tunica intima thickening, endotheliocyte injury and monocyte adhesion were found with a light microscope; the elastic lamina being broken and the smooth muscle cells proliferated. Under TEM, the endothelial cell membrane of the aorta in the test rats was destroyed and appeared to be worm-eclipsed shape.Mitochondria exhibited swelling,vacuole degeneration and its cristae was dissolved, broken or some disappeared.Rough endoplasmic reticula (RER) expanded. The endothelial cell spaces were enlarged and the cell junctions deformed. Monocytes adhering to the endothelial cell stretched out pseudopodia and intruded into the endothelial crevice and the subendothelial layer. Some basement membranes completely sloughed following with endothelial cell. ERE and ribosomes increased in smooth muscle cells. SEM observation showed that the endothelial cells became swelling and the surface of endothelial cell was worm-eclipsed or crater shape. There were deeper crevices between endothelial cells.Conclusions Long-term high fat forages diet can induce injury of the endothelium and elastic lamina, adhesion of the monocytes and its intrusion into endothelial layer and subendothelial layer, proliferation of the subendothelial layer and smooth muscle in the aorta of rats.
10.Apoptosis was involved in the antitumor effect of valdecoxib on Lewis cancer

Aim To evaluate the effect and the mechanism of valdecoxib on Lewis cancer. Methods Western blot was used to detect the expression of VEGF, MMP-2, Bax, Bcl-2 and Caspase-3 in tumor tissue. Flow cytometry was used to observe the effect of valdecoxib on proliferation, apoptosis and the cell cycle distribution. MTT assay was used to observe the lymphocyte transformation rate. Results ① Valdecoxib inhibited the growth of the tumor and increased the survival rate. ② 10~40 mg?kg -1?d -1 Valdecoxib increased the apoptosis rate from 19.1% in control group to 23.1%~29.1%, and did not affect the distribution of cell cycle. ③ The expression level of Bcl-2 was decreased and expression levels of Bax, COX-2, MMP-2, Caspase-3 and VEGF were not affected. ④ Valdecoxib did not affect the weight, the lymphocyte transformation rate, spleen index, and thyrus index of the mice. Conclusion The inhibitory effect of valdecoxib on Lewis tumor is associated with apoptosis.
