Chinese Journal of Anesthesiology 1994;0(06):-
Objective The study consisted of two parts: in part Ⅰ the effects of propofol or/and procaine on CD18, CD62L expression and superoxide anion (SOA) release of phorbol 12-myristal 13-acetate(PMA) stimulated human neutrophils (PMNs) were studied in vitro; in part Ⅱ the effects of propofol or/and procaine on IL-6 and TNF-?production in endotoxin-stimulated human whole blood were studied. Methods PMNs were separated from the whole blood obtained from healthy 20-40yr old subjects. Part Ⅰ consisted of 9 groups: in group 1 (control) phosphate buffer solution was added to PMNs; in group 2 PMNs were stimulated by PMA 100mg?ml-1 ; in group 3-5 different concentrations of propofol (0.4, 4.0,40?g?ml-1) were added to PMA stimulated PMNs; in group 6-8 different concentrations of procaine (1.5,15,150?g?ml-1 ) were added to PMA stimulated PMNs; in group 9 propofol 2?g?ml-1 and procaine 8?g?ml-1 were added to PMA stimulated PMNs. Part II also consisted of 9 groups: in groupl whole blood was mixed with normal saline; in group2 lipopolysaccharide (LPS) l?g?ml-1 was added to whole blood; in group 3-5 different concentrations of propofol (0.4,4.0,40?g?ml-1) were added to LPS stimulated whole blood; in group 6-8 different concentrations of procaine (1.5, 15, 150?g?ml-1 ) were added to LPS stimulated whole blood; in group 9 propofol 2?g?ml-1 and procaine 8?g?ml-1 were added to LPS stimulated whole blood. CD18, CD62L expressions were measured by flow cytometry. SOA release was determined by cytochrome C reduction in the presence or absence of superoxide dismutase (SOD) . IL-6 and TNF-? production were measured by using radioimmunoassay. Results Propofol or/and procaine depressed CD18 up-regulation, CD62L shedding and SOA release of PMA-stimulated PMNs and propofol was more effective than procaine. Propofol enhanced but procaine inhibited the increased production of TNF-? and IL-6 in the LPS stimulated whole blood but when propofol and procaine were used in combination. The effectof procaine predominated. Conclusions Propofol or/and procaine can attenuate tissue damage induced by neutrophils by inhibiting the function of neutrophils.

Chinese Journal of Nephrology 2005;0(08):-
Objective To explore the effects of Sinomenine (Sin) on phenotype transdifferentiation in renal tubular epithelial cells. Methods Mice primary tubular epithelial cells and cell line (MCT) were cultured in vitro. Model group cells were stimulated by IL-1?(10 ng/ml). The cells of Sin group were co-incubated with Sin(10, 100 ,500?mol/L) simultaneously. The cells without IL-1?and Sin treatment were regarded as control group. The protein expression of?-smooth muscle actin (SMA) and Vimentin was assessed by immunohistochemistry and Western blotting. The mRNA expression of?-SMA and Vimentin was examined by RT-PCR. Secretion of matrix metalloproteinases was determined by zymography. Results The mRNA expression and protein level of?-SMA and Vimentin, and the enzyme activity of MMP-2 and MMP-9 in tubular epithelial cells stimulated with IL-1?were significantly higher than those in the controls. Sinomenine significantly reduced the expression levels of protein and mRNA of?-SMA and Vimentin. At the same time, the MMP-2 and MMP-9 activities were reduced markedly. Conclusion Sinomenine can decrease the transdifferentiation in renal tubular epithelial cells stimulated by inflammatory factor IL-1?and prevent renal interstitial fibrosis.

Acta Nutrimenta Sinica 1956;0(04):-
Objective To investigate the effect of taurine on iron bioavailability in rats and its possible mechanism. Method SD rats were randomly divided into three groups:normal group (NG),taurine treated group (TG),experimental control group (EG). TG and EG were induced to be anemic experimentally for 4w before the study. Then the rats were fed iron supplements for 25 d,meanwhile TG was treated with taurine following hemoglobin(Hb) repletion bioassay. Regression analysis of TG and AG were measured following Hb repletion bioassay using iron supplements. And the relative biological value of TG following Hb repletion were calculated. The serum iron, unsaturated iron-binding capacity(UIBC),total iron-binding capacity(TIBC),transferin saturation(TS)and the expression of hepcidin were detected. Results The iron bioavailability of TG was higher than EG,and the RBV of TG was 126%. Other hematological variables of TG were close to NG,and better than EG. The expression of hepcidin was increased compared with NG,but decreased compared with EG. Conclusion Taurine can improve iron bioavailability in rats via suppressing hepcidin expression in rats.

Chinese Journal of Immunology 1986;0(04):-
Objective:To investigate the effects of Tripterygium Wlilfordii on renal interstitial fibrosis and the expression of inflammatory factor in mice kidney with the Unilateral Ureteral Obstruction(UUO).Methods:Mice were randomly assigned to shame operation group(shame proup), UUO group, Tripterygium Wlilfordii and ACEI treatment group after UUO. The protein expression of ?-SMA,ICAM-1 were assessed by immunohistochemistry or Western blot; MCP-1 mRNA were detected by reverse transcription-polymerase chain reaction analyses(RT-PCR). And the change of renal pathology was observed at the same time. Results:In comparison with the shame group, the protein levels of ?-SMA, ICAM-1 and the mRNA expression of MCP-1 in UUO mice and treatment groups increased significantly, meanwhile Tripterygium Wlilfordii and ACEI could significantly reduce the expression of ?-SMA, ICAM-and MCP-1 on levels of protein and gene. The effects of Tripterygium Wlilfordii on ICAM-1 and MCP-1 were more significant than ACEI.Conclusion:Tripterygium Wlilfordii could reduce the transdifferentiation of renal interstitial cells and prevent renal interstitial inflammatory injury.
