1.Fluorescence enhancement of flavoxate hydrochloride in alkali solution and its application in pharmaceutical analysis.

Fluorescence enhancement reaction of flavoxate hydrochloride (FX) in strong alkali solution was studied, the mechanism of the reaction was investigated, and a novel fluorimetric method for analysis of FX in drug sample was established. FX has no intrinsic fluorescence, but it can slowly produce fluorescence in strong alkali solution. Heating can promote the fluorescence enhancement reaction. In 3D fluorescence spectra of the decomposition product of FX, two fluorescence peaks, located respectively at excitation wavelengths λex/ emission wavelength λem =223/410 nm, and 302/410 nm, were observed. Using quinine sulfate as a reference, fluorescence quantum yield of the decomposition product was measured to be 0.50. The structural characteriza- tion and spectral analysis of the decomposition product reveal that ester bond hydrolysis reaction of FX is firstly occurred during heating process, forming 3-methylflavone-8-carboxylic acid (MFA), then a cleavage reaction of the γ-pyrone ring of MFA occurred, producing α, β-unsaturated ketone. This product includes adjacent hydroxyl benzoic acid group in its molecule, which can form intramolecular hydrogen bond under alkaline condition, so that increase the conjugate degree and enhance the rigidity of the molecule, and thereby cause fluorescence enhancement. Based on this fluorescence enhancement reaction, a fluorimetric method was proposed for the determination of FX. A linear calibration curve covered the concentration range 0.020 3-0.487 µg · mL. The regression equation was I(F) = 23.9 + 5357.3 c, with correlation coefficient r = 0.999 7 (n = 8), detection limit D = 1.1 ng · mL(-1). The method was applied to the analysis of FX tablets, with a spiked recovery rate of 100.2%. The reliability of the method was verified by a UV-spectrophotometric method.
2.Image diagnosis of AIDS related toxoplasma encephalitis in AIDS patients

Objective To assess the diagnostic value of CT and MRI findings of toxoplasma encephalitis among AIDS patients.Methods CT and MRI findings of toxoplasma encephalitis were retrospectively studied in 17 AIDS cases.Results The lesions were bilateral and multiple,involving basal nuclei in 12 cases,thalamus in 3 cases,bilateral cerebral hemispheres near corticomedullary junction in 1 case,and cerebellum and brain stem in 1 case.The lesions showed weak hypodensity on CT,and long T 1 and long T2 signal intensity on MRI,with marked peripheral edema effect.After contrast administration,the small ring or twist and target enhancement was seen in 12 cases,large ring enhancement-in 1 case.multiple focus in 15 cases and single focus in 2 cases;MRI was more sensitive in detecting a largest number of cerebral lesions than CT.Conclusion More small ring and twist,nodular,target enhancement are highly suggestive of toxoplasma encephalitis in the basal nuclei.Both CT and MRI are effective in diagnosing toxoplasma encephalitis,but MRI imaging without and with gadolinium is more sensitive than CT in the detection of toxoplasma encephalitis;MRI imaging may reveal a greater number of lesion when it is positive and so the detection rate of MRI is higher than that of CT.
3.Study on the diagnostic value of adiponectin/leptin ratio in metabolic syndrome in elderly men

Objective To explore the correlation between adiponectin/leptin(A/L) ratio and metabolic syndrome diagnosis in elderly men. Methods A total of 256 elderly men (≥60 years) were enrolled and divided into metabolic syndrome group(n= 109) and non metabolic syndrome group (n= 147). Serum levels of adiponectin and leptin were measured by radioimmunoassay (RIA) and the A/L ratio was calculated. Metabolic syndrome diagnosis is based on the definition provided by China Diabetes Society (CDS) in 2004. Results (1) In metabolic syndrome group versus non metabolic syndrome group, the serum levels of leptin and adiponectin were (10. 3±7.0) vs (6.8±4.9)μg/L and(7.8±5.6)g/L vs (9.5±5.9)g/L, and A/L ratio was 0. 94±0. 78 vs 2.15±2.13 respectively. (2) A/L ratio was positively correlated with high density lipoprotein cholesterol (HDL-C) and negatively correlated with body mass index (BMI), triacylglycerol (TG) and serum uric acid(P< 0. 001). (3) The higher value of A/L ratio, the lower possibility of metabolic syndrome. When the A/L ratio was more than 5, the incidence of metabolic syndrome was decreased to O(X2 =34. 891 ,P< 0.001). (4) The more components of metabolic abnormality, the lower value of A/L ratio(F= 10. 876,P<0. 001). Conclusions The A/L ratio may be useful in evaluating the extent of metabolic disorder and diagnosing metabolic syndrome in elderly men.
4.Change of adiponectin to leptin ratio in elderly male non-metabolic syndrome patients with visceral adipose deposit

Objective To observe the serum adiponectin, leptin level, and adiponectin/leptin ratio (A/L ratio) in elderly male patients with normal BMI and visceral adipose deposit but without metabolic syndrome. Methods A total of 109 elderly males (≥60 years old) were enrolled whose BMI were all less than 25 kg m and without metabolic syndrome. They were divided into non visceral adipose deposit group (n = 67) and visceral adipose deposit group (n = 42 ). Serum levels of adiponectin and leptin were measured by radiological immunological assay (RIA) and the A/L ratio was calculated. Visceral adipose deposit was defined as visceral adipose area more than 100 cm2.Metabolic syndrome diagnosis was according to the definition provided by China Diabetes Society (CDS) in 2004. Results (1)The visceral adipose area [(135.56±31.72)cm2 vs. (68. 65±22.64)cm2 , P< 0. 001] and BMI [(22.94 ± 1.35 ) kg/m2 vs. ( 21.38 ± 2.55 ) kg/m2 , P < 0.001] were significantly higher in visceral adipose deposit group than in non visceral adipose deposit group. The A/L ratio was significantly lower in visceral adipose deposit group than in non visceral adipose deposit group ( 2. 17 ± 1.77 vs. 4.54 ± 7.00, P = 0. 034 ) while there was no difference in serum adiponectin or leptin level between those two groups. (2)The A/L ratio was negatively correlated with BMI (r=-0.552, P<0.001), waist circumstances (r=-0.390, P<0.001) and abdominal visceral fat area (r=-0.341, P<0. 001 ). Conclusions The A/L ratio decreases in elderly men with normal BMI and visceral adipose deposit. A/L ratio could be useful in scanning those patients with visceral adipose deposit whose BMI is normal among elderly men.
5.The effect of wet hyperthermia combined with microwave ablation on the proportion and function of CD 4 +CD25 +regulatory T cell in animal model of mouse mammary carcinoma

Objective To investigate the effects of wet hyperthermia combined with microwave ablation on the pro-portion and function of CD 4 +CD25 +regulatory T cell in animal model of mouse mammary carcinoma and explore its significance .Methods An experimental model of mouse mammary carcinoma was established and then treated with microwave ablation on day 14.Local wet hyperthermia was also performed .The proportion of CD4 +CD25 +regula-tory T cells (Tregs) in splenocytes was analyzed by FACS.The relative expression of Foxp3, as well as the expres-sion of CTLA-4 and GITR, on Tregs was determined by real-time PCR assay and FACS respectively .Moreover, the secretion of TGF-βand IL-10 from Tregs also were detected by ELISA .In addition , the suppressive capacity of Tregs on CD4 +CD25 -T cell was also examined by MTT assay .Results The proportion of Tregs in wet hyperther-mia combined with microwave ablation treated group decreased significantly compared with that in wet hyperthermia or microwave ablation treated alone group ( P<0.05 ) .Moreover , the expression level of Foxp 3 on Tregs decreased significantly (P<0.05).Furthermore, the expression of CTLA4 and GITR, as well as the secretion of TGF-βand IL-10, on Tregs also obviously decreased (P<0.05).Finally, the suppressive capacity of Tregs on the proliferation of CD4+CD25-T cells were also significantly impaired ( P<0.05) .Conclusions Wet hyperthermia combined with microwave ablation os more effective than single wet hyperthermia or microwave ablation as a mono -therapy alone on the function of Tregs , which may be closely related to the enhanced suppression of tumor growth .
6.The effect of microwave ablation combined with dendritic cell tumor vaccine treatment on mouse mammary carcinoma and its immunological mechanism

Objective To investigate the therapeutic effects of microwave ablation combined with dendritic cell (DC) tumor vaccine on a murine breast cancer model and to explore its immunological mechanism.Methods Forty-eight Balb/c mice were used to establish models of mouse mammary carcinoma,and then randomly divided into a sham-operation group,a microwave ablation treated group,a DC tumor vaccinated group and a microwave ablation plus DC tumor vaccinated group.Microwave ablation was conducted with the microwave ablation group on day 14 after establishing the model.DC tumor vaccination was performed on day 7 in the vaccinated group.In the microwave ablation plus DC tumor vaccinated group,DC tumor vaccination and microwave ablation were consecutively performed on days 7 and 14,respectively.Tumor growth,lung metastasis and survival were observed.The proportion of CD3+ and CD4+ T cells in splenocytes were analyzed by fluorescence Activated Cell Sorter (FACS) on the 14th day after microwave ablation.Serum levels of interferon-γ (IFN-γ) and interleukin-4 (IL-4) were also detected using ELISA.Results The survival time (56.4 ± 4.3 days),tumor size (67.3 ± 6.1 mm2) and lung metastasis index (0.6 ± 0.4) in the microwave ablation plus DC tumor vaccinated group were all significantly lower than the corresponding averages in the microwave ablation and DC tumor vaccinated group.The proportions of splenic CD3 + T cells (27.8 ±10.3 %) and CD4+ T cells (16.7 ± 8.3 %) in the microwave ablation plus DC tumor vaccinated group were significantly larger than in the control group.Average serum levels of IFN-γ (41.8 ± 22.9 pg/ml) in the microwave ablation plus DC tumor vaccinated group were also significantly larger than in the control group,while the average serum level of IL-4 (15.4± 6.4 pg/ml) was significantly lower.Conclusions Microwave ablation and DC tumor vaccination together could have synergistic effects in treating experimental breast cancer,which might be closely related to the enhanced immune response.
7.Study on the relationship between self-supporting personality and career maturity of medical students

ObjectiveTo investigate the developmental status and their relationship of self-supporting personality and career maturity of medical students.Methods450 medical students were surveyed by self-supporting personality scale and career maturity questionnaire.Descriptive statistical analysis,t-test,correlation analysis and regression analysis were used to do data analysis.Results ( 1 ) Most dimensions of self-supporting personality such as personal initiative,interpersonal initiative,interpersonal flexibility and personal responsibility were positively related to the career maturity ( r=0.53,P<0.01 ; r=0.49,P<0.01; r=0.46,P<0.01 ; r=0.45,P< 0.01 ).(2)Career maturity of medical students can be positively predicted by several dimensions of self-supporting personality ( R2 =0.49,F=60.10,P< 0.01 ).ConclusionThe career maturity of medical students is closely associated with their self-supporting personality.

9.Relationships among posttraumatic growth, social support, and coping style in women with infertility

Objective To explore the relationships among posttraumatic growth (PTG),social support,and coping style in women with infertility.Methods 182 women with infertility were recruited in one public hospital department of assisted reproduction technology.Measures included Posttraumatic Growth Inventory (PTGI),Perceived Social Support Scale (PSSS),and Simplified Coping Style Questionnaire (SCQ).Results ① The PTG score of women with infertility was 42.55 ± 16.83,lower than the average level.② Compared with the lower PTG group,the higher PTG group scored significantly higher in social support,positive coping,and negative coping (t =3.867,P< 0.01 ; t =8.452,P< 0.01 ; t =2.817,P< 0.01).③ There were significantly positive correlations among PTG,social support and positive coping (r =0.295-0.515,P < 0.01).④ Positive coping served as a total mediator in the relationship between social support and PTG.Conclusion There are closely correlations among PTG,social support,and positive coping style.Positive coping style significantly mediate the relationship between social support and PTG.
10.Psychological stress level and features of recruits in training camp

Objective To analyze psychological stress level and features of recruits in training camp.Methods Psychological stress self-evaluation test (PSET) was applied to 1 600 recruits in training camp.Results ①There were 64 recruits(64/1600,4%) who had high level of psychological stress(T score ≥70).②The psychological stress symptoms of recruits in training camp mainly manifested as increased alertness (87.7%),sleep deprivation (57.6%),mental and physical fatigue (48.7%) and nervous irritability (45.2%).③Psychological stress levels of recruits with different ages and different levels of self-feeling in camp had significant differences(P<0.05).Conclusion Recruits in training camp have partly excessive psychological stress responses,targeted emergency decompression trainings should be carried out in recruits.
