1.Preparation of components of defensins from human and rabbit neutrophils by continuous acid-urea-polyacrylamide gel electrophoresis (CAU-PAGE)

Objectives:To develop a preparative method of continuous acid-urea-polyacrylamide gel electrophoresis (CAU-PAGE) to purify the defensin components from human and rabbit neutrophils (HNP-1~3 and RNP-1~5).Methods:Neutrophils were isolated from human peripheral blood or rabbit artificial aseptic ascites.The components of defensin were extracted by 5% acetic acid directly followed by elution of CAU-PAGE,and purified HNP-1~3 or RNP-1~5 were harvested.Polyacrylamide gel filtration was used as the method for comparison.Results:For each elution circa 1.50 mg of purified HNP or 2.50 mg of RNP were obtained from 5×108 of neutrophils,and their natural biological activity was preserved.Conclusions:CAU-PAGE is a relatively simple,rapid,economical method for purifying cationic peptides as the components of defensin.

Hypertensive vascular remodeling has always been a hot program in clinic and basic research.Renin-angiotensin system is an importent regulatory mechanism of human physiological function,which plays a fundamental role in vascular remodel- ing.RAS consists of two axes, ACE-AngII-AT_1 receptor axis and ACE2-Ang(1- 7)-Mas receptor axis.The former affects vasoconstriction and promotes vascular re-modeling through its key member,AngII;the latter offsets the effect of the mentioned AngII with the help of seven-peptide Ang-(1-7).A deep knowledge of the relation between RAS and vascular remodeling would provide more remedies for reducing cardiovascular risk.
3.Protective effects of methylflavonolamine on myocardial injury induced by adriamycin in mice

AIM: To investigate the protective effects and mechanisms of methylflavonolamine (MFA) on myocardial injury induced by adriamycin in mice. METHODS: The myocardial injury was induced by adriamycin (ADR) 1.5 mg?kg -1 ip once every two days for ten days in mice. All mice were taken the electrocardiogram examination before given drugs. The mice with abnormal electrocardiogram were excluded prior to the experiment. The degree of J point elevation, the prolonged degree of the QRS complex duration and the Q T interval, the change of contents of serum creatine phosphokinase (CK), serum lactate dehydrogenase (LDH), myocardial malondialdehyde (MDA), and the activity of myocardial superoxide dismutase (SOD) were observed in control and treated groups. The contents of serum CK and LDH were measured by spectrophotometry, and the content of myocardial MDA was measured by TBA method and the activity of myocardial SOD by hydroxylamine method. RESULTS: The J point was elevated, the Q T interval and the duration of QRS complex were prolonged and the contents of serum CK and LDH were increased in mice with acute myocardial injury induced by ADR, suggesting that a widespread and severe myocardial cell injury occurred in the prepared models. While all these injury indices were reversed by MFA treatment. The content of myocardial MDA was increased and the activity of myocardial SOD was decreased in mice with myocardial injury, and MFA decreased the MDA content and increased the SOD activity, indicating that it possesses the actions of scavenging free oxygen radicals and anti lipoperoxidation. CONCLUSIONS: MFA significantly alleviates the degree of the acute myocardial injury in mice induced by ADR. Its mechanism may be associated with reducing oxygen free radical production and anti lipoperoxidation.
4.Intravascular Stent Insertion for Treatment of Inferior Vera Cava Obstruction

8 cases of inferior vena cava obstruction were treated with intravascular stent insertion,including 3 membranacous obstructions and 5 segmental obstructions.All 8 cases obtained successful results without Severe complications.4 cases were checked up half a year later(2 of which were to checked after one year)with stents still in position and pa- tency of all.inferior vena cavas.In present studies the insertion technique and prevention of complication were discussed.
5.The clinical study on dinoprostone in premature rupture of the membranes at term cervical ripening

Objective To explore clinical effect of dinoprostone in premature rupture of the membranes at term cervical ripening.Methods 42 cases with premature rupture of the membranes at term cervical ripening were selected.All patients were randomly divided into two groups.The dinoprostone group had 21 cases,which applied the treatment of dinoprostone belt the posterior fornix of vagina medicine induced abortion.The oxytocin group had 21 cases,which applied the treatment of intravenous drip oxytocin.The time of induction labor,6h,12h cervical Bishop score after using the medication,drugs acting time,labor conditions and perinatal situation,pregnancy outcome and some other index were observed.Results It taken (6.18 ± 4.48 ) h until parturient and ( 12.62 ± 8.03 ) h until the cervical mouth full open in the dinoprostone group,which were shorter than the oxytocin group,that taken( 10.21 ±5.42) h,( 18.87 ± 9.14) h.( t =2.62,2.35,all P < 0.05 ).There was no significant difference about cervical Bishop score before using the medication.It's(7.52 ± 2.44 ) point 6h after using the medication and( 9.03 ± 1.96 ) point 12h after in the dinoprostone group.They were better than the oxytocin group,which were(5.97 ± 1.95 )point,(7.13 ±2.12 )point.It was significantly different( t =2.27,3.02,all P <0.05 ).The average incubation period of dinoprostone group wss (9.91 ± 1.73 )h.It's longger than the oxytocin group,which was( 8.72 ± 1.34)h.The active period of dinoprostone group was(4.36 ± 0.66 ) h.It's shortter than the oxytocin group,which was( 5.84 ± 1.02 ) h.The difference was significant( t =2.49,5.58,all P < 0.05 ).There was no significant difference in the second and third stage of laber between the two groups( t =0.56,0.33,all P > 0.05 ).There was also no significant difference about intrauterine distress,amniotic fluid contamination,cesarean section,intestinal bleeding,the Apgar score of newborns and some other index in the two groups ( x2 =0.00,0.00,0.52,t =0.84,1.04,all P > 0.05 ).Conclusion Dinoprostone had good promoting effect on cervical ripening for preterm premature rupture of membranes pregnant woman.Its effect was better than oxytocin and it was safe to use.
6.Value of multichannel epidural somatosensory evoked potentials in cortical mapping of primary sensorimotor areas

Objective To assess the value of muhichannel epidural somatosensory evoked potential (SEP) in cortical mapping of primary sensorimotor areas for electrode implantation of motor cortex electrical stimulation.Methods Intraoperative multichannel epidural SEP was recorded in 13 patients with intractable neuropathic pain.SEP mapping was used for determining the location of sensorimotor areas.Results The typical phase reversals of N20 and P20 were recorded and the location of the central sulcus and the primary sensorimotor area were accurately determined in 11 patients.The location of the primary sensorimotor failed to be determined in the other 2 patients because of the atypical SEP.Conclusion The intraoperative muhichannel epidural SEP is reliable in the cortical mapping.
7.The effects of intracarotid transplantation of bone marrow stem cells on learning memory abilities in the brain of VD rats

Objective To observe the survival and migration characteristics after intracarotid transplantation of bone marrow stem cells(BMSCs) and its effect on learning-memory abilities across the blood-brain barrier(BBB) in the vascular dementia(VD) rats .Methods Bone mononuclear cells(BMNCs) were isolated from bone marrow in vitro by standard Ficoll-Hypaque technique , then cells were enriched and expanded by using bone marrow adherent culture .72 Wistar rats were meanly divided into control group ,model group and treatment group .The VD rat model was established by modified pulsinellis 4-vessel occlusion(4 VO) .The treatment group received intracarotid infusion of 0 .5 mL 1 .2 × 107/mL BMSCs which were labeled with BrdU in vitro after opera-tion .Their survival ,migration and the learning-memory abilities were observed at 4th and 8th week .Results BMSCs transplanted by intracarotid transplantation survived and had been found throughout the brain tissue .They major migrated and localized in the is-chemic zone of injury such as hippocampus and cerebral cortex .Compared with the control group ,active avoidance response(AAR) ratio in the model group(42 .1 ± 4 .5) ,group(43 .6 ± 3 .6)showed significantly decrease compared with the control group (90 .0 ± 4 .3) ,(92 .5 ± 5 .0)(P<0 .01) ,and the treatment group(69 .2 ± 4 .7) ,(70 .8 ± 4 .7)was significant higher than the model group(P<0 .01) .Conclusion Intracarotid transplantation of BMSCs could enter the VD rats cerebra parenchyma via BBB ,migrate into the damaged brain tissue to gather and survive .Learning-memory abilities can be improved significantly by transplanted BMSCs .

10.A study on antiviral activity of defensins from neutrophil of human and rabbit against SARS coronavirus in vitro

Objective To determine the anti-SARS coronavirus (SARS-CoV) activity of defensins from human and rabbit, and explore the effects of defensin during the onset and progression of SARS. Methods Defensins were purified from neutrophil of human and rabbit respectively, namely HNP (human neutrophil protein) 1-3 and RNP (rabbit neutrophil protein) 1-5, by acid-urea-polyacrylamide gel electrophoresis (CAU-PAGE). The effect of defensin on Vero-E6 cell infected by SARS CoV BJ01 strain as well as the replication of virus were observed by micro-culture technique. Rusults The HNP 1-3 and RNP 1-5 separated by CAU-PAGE presented a single band with molecular weight of approximately 4000 dalton on 15% SDS-PAGE. HNP 1-3 was identified and validated by quantitative ELISA. The activity of anti-escherichia coli (ATCC25922 strain) of HNP 1-3 and RNP 1-5 was tested by MIC determination method, and the IC50 values obtained were 25~50?g/ml and 5~10?g/ml, respectively. In the antiviral experiment, HNP 1-3 and RNP 1-5 in the concentration from 25 to 100?g/ml were not able to prohibit cytopathic effect (CPE) of SARS-CoV and the replication of virus in Vero-E6. Conclution The natural defensins (HNP 1-3 and RNP 1-5) purified from neutrophils do not play any prohibitory or eliminative effects on SARS-CoV, indicating that that human and rabbit do not have natural immunity against SARS-CoV.
