1.Effects of cognitive function and neuron morphous in hippocampus of the chronic stress depressed rats with gamma-aminobutyric acid and sertraline

Objective To investigate the effects of cognitive function and neuron morphous in hippocampus of the chronic stress depressed rats with gamma-aminobutyric acid and sertraline.Methods Using the chronic mild unpredictable stress to make the model of the chronic stress depressed rats.Sample group,sertraline group,GABA group and research group were gave by saline,sertraline,GABA,GABA and sertraline i.p.respectively for 21 days.Control group reeepted no any stress and drugs.Using open-field test and electric maze test to detect the behavior and memory function of the rats.Using Nissl staining to observe the pyramidal neuron morphous in CA1 and CA3 region of the hippocampus.Results After stress,the squares crossing,rears,grooming movement of the four groups accecepted stress were decreased compared with the control (P < 0.05).After treatment,the squares crossing(respective for 57.25±18.79,39.00±13.69),rears(respective for 23.25±7.01,16.75±4.71),grooming movement(respective for 2.25±0.46,2.25±1.04),number of correct reaction (respective for 17.00±1.93,16.88±2.64) and latency time (respective for 172.88±26.91,206.00±33.54) of the research group and sertraline group were better than sample group(P < 0.05 or 0.01).GABA group showed no significant improvemerit except for the reduced latency time compared with the sample group(P< 0.05).Compared with the control,the morphous,numbers and alignment of pyramidal neuron in CA1 and CA3 in hippocampus of the sertraline group,GABA group and research group were improve with different level,all the better in research group.Conclusion GABA with sertraline can improve the cognitive function of the chronic stress depressed rats by protect the neuron in hippocampus.
2.A method of medical image registration based on frequency domain and time domain

To resolve the problems of relativity translation variant and relativity rotation variant existed in the two medical images, combining the removing characteristics and rotating characteristics in image signal Fourier transform, a method of medical image registration combining with frequency domain and time domain is put forward based on the well-known Fourier property. The method decouples the estimation of rotation and translation. It determines rotation parameters based on amplitude spectrum correlation in frequency domain and estimation translation parameters based on intensity value correlation in time domain. The experimental results of nuclear medical image registration and multi-modality medical image registration show that this method is a very robust and fast registration method with high accuracy.
3.A study on expression changes of a tumor suppressor WWOX in human lung adenocarcinoma cell line A549

Purpose:To detect the abnormalities of WWOX(WW domain containing oxidoreductase) gene in human lung adenocarcinoma cell line A549. Methods:Deletion of WWOX exons 6-8 transcript was analyzed by reverse transcriptase-PCR technology; loss of heterozygosity (LOH) of WWOX gene was analyzed by PCR-based assays for dinucleotide repeat polymorphisms technology. Aberrant expression of WWOX protein was analyzed by western blot. Results:A549 cells samples showed loss of WWOX exons 6-8 transcript.This deletion was not detected in normal primary cultured human bronchial epithelial cells samples.Three microsatellites(D16S3029、D16S3096、D16S504)did not have LOH in the normal primary cultured human bronchial epithelial cells samples, but D16S2029 and D16S3096 were all found to have LOH in A549 Cells samples. We further observed that expression of WWOX protein was significantly lower in A549 cell samples compared to the normal primary cultured human bronchial epithelial cells samples. Conclusions:WWOX gene may be important during tumorigenesis in lung adenocarcinoma cancer.Deletion of exons 6-8,LOH and aberrant expression of protein are all modes of WWOX gene inactivity in lung adenocarcinoma cancer.

Objective To explore the antigen presentation of CT26.WT via intra-peritoneal injection. Methods The intra-peritoneal injection model was made via injecting cell suspensions in mice. The spleen was isolated from BALB/c mice toco-culture with CT26.WT to detect tumor-killed ability. Phenotype identification methods and CCK8 massy were used to measure the ability of antigen presentation and stimulate T lymphocyte proliferation. IHC was used to detect the expression of B7H4 in normal and tumor tissues. Results Along with the extension of intra-peritoneal injection, the surviving number of cells was increased, contrary to the apoptosis. DC cells failed in maturation and impaired in stimulating T lymphocyte proliferation. B7H4 was higher in tumor tissues. Conclusions With the extension of intra-peritoneal injection, the mature DC cells were scared in number, resulting in the impairement of antigen-presentation. Moreover, the higher B7H4 expression in tumor tissues led to the lack of second signals which may stimulate T cells. Consequently, the ability of T cells in killing tumor cells was decreased so that they escape immunosurveillance.

6.Long serial analysis of gene expression in the yeast phase of Candida albicans

Objective To construct a library of long serial analysis of gene expression (LongSAGE) and investigate the characteristics of gene expression in the yeast phase of C. albicans. Methods A LongSAGE library was constructed with long serial analysis of gene expression (LongSAGE). Results A total of 17 773 tags, representing specific 7 333 transcripts, were extracted from 1 000 sequence files in the yeast phase of C. albicans. Tags matching single gene accounted for 16.65%, tags matching multiple genes accounted for 6.59%, tags for hypothetical protein accounted for 16.27%, tags for genome and cosmid accounted for 1.64%, and novel tags accounted for 58.86%. Eleven tags in 33 tags whose expression frequencies exceeded 35 may be new expression sequences. Conclusion LongSAGE is not only a powerful method in investigating the gene expression profiling, allowing the analysis of the expression of thousands of transcripts in a quantitative manner without prior sequence information, but also in finding new genes.

Objective To explore the relationship between smoking, alcohol drinking and gallstone disease in general male population in Guangdong. Methods Each subject received a questionnaire and took an upper abdominal ultrasound examination in a cross-sectional study. 9.Deletion and mutation of WWOX exons 6-8 in human non-small cell lung cancer.

To examine the deletion and point mutation of WWOX (WW domain containing oxidoreductase) exons 6-8 in human non-small cell lung cancer and their possible relationship with pathological stages, tumor tissues and the corresponding normal tissues were obtained from 44 Chinese patients who had undergone surgery for non-small cell lung cancer. RNA was extracted from each sample and deletion and mutation of WWOX exons 6-8 were analyzed by RT-PCR and DNA sequencing. Our results showed that 28 of 44 (63.6%) lung cancer samples showed loss of WWOX exons 6-8 transcript and the deletion was detected in only 3 of 44 (6.8%) corresponding adjacent normal tissues (P < 0.05). The transcript sequencing analyses of the 16 lung cancer samples without transcript loss of WWOX exons 6-8 revealed no difference from the sequence of GenBank. Moreover, the deletion of WWOX exons 6-8 was significantly higher in the smokers when compared with the non-smokers. It is also higher in the men and squamous carcinomas than in women and adenocarcinomas (P < 0. 05). The deletion, however, was not found to be associated with pathological stages of the tumors. Our study documented a high incidence of deletion of WWOX exons 6-8 in non-small cell lung cancer in Chinese patients and suggested that the frequent loss of WWOX exons 6-8 might play an important role in the tumorigenesis of non-small cell lung cancer in Chinese. WWOX exons 6-8 may serves as a candidate molecular target of smoking carcinogenesis, and point mutation is not a predominant way of alteration of WWOX exons 6-8.

10.Efficacy of Hydrochloride Phenazopyridine on Postoperative Pain of Cystoscopy

Objective To explore the clinical effect of hydrochloride phenazopyridine on relieving the pain after cystoscopy. Methods 60 male patients undergoing cystoscopy were divided into observation group and control group according to the block randomization method with 30 patients in each group. 1 h after cystoscopy, the observation group was given orally hydrochloride phenazopyridine 0.2 g, 3 times a day for 0.6 g totally. The control group was not treated with oral medication for pain. The pain degree was evaluated with Numerical Rating Scale (NRS), and the side effects were recorded. Results There was no significant difference in NRS score immediately after cystoscopy between 2 groups (P=0.725). The NRS score was lower in the observation group than in the control group 24 h after cystoscopy (P=0.002).Conclusion Hydrochloride phenazopyridine can effectively relieve the postoperative pain of cystoscopy.
