1.Surgical techniques of orthotopic liver transplantation in rats by a single operator under direct vision

BACKGROUND: Rat model of orthotopic liver transplantation is a very valuable model for experimental study in liver transplantation including organ preservation, tissue ischemia-reperfusion injury, allograft rejection and immune tolerance mechanism. Stable liver transplantation animal model is the basis of the related experimental studies. However, its experimental operation is long and boring, especially performed by a single operator under direct vision. OBJECTIVE: To investigate the operation techniques to establish a stable rat model of orthotopic liver transplantation by a single operator under direct vision. METHODS: The orthotopic liver transplantation was performed using two-cuff method in 50 pairs of rats. We exposed the abdominal cavity fully, perfused the donor liver through abdominal aorta without flipping donor liver; suprahepatic inferior vena cava was in vivo cut down using one-step method, without diaphragm ring; the suprahepatic inferior vena cava was anastomosed with single-row suture, and the cuff of portal vein was installed by fixing the blood vessel forceps on rubber. Hepatic artery was not reconstructed. Fluid replacement was administered to maintain hemodynamic stability in rats after operation. RESULTS AND CONCLUSION: The donor operative time was (36.2± 2.5) minutes, donor liver trimming time was (12.2± 1.5) minutes, receptor operative time was (45.6 ± 3.5) minutes, suprahepatic inferior vena cava anastomosis time was (10.1 ± 2.1) minutes, portal vein cuff time was (1.5 ±0.9) minutes, infrahepatic inferior vena cave cuff time was (1.1 ± 0.6) minutes, anhepatic phase was (15.1 ± 2.2) minutes. The success ratio of the operation was 100% and the survival rates within 1 week and 1 month were all 100%. It is indicated that the key factors of a successful model were stable anesthesia, good donor liver perfusion, adequate exposure, skilled microsurgical technology and vascular anastomosis technique.
2.Efficacy of compound oxymatrine injection combination with postoperative chemotherapy for advanced ovarian carcinoma

ObjectiveTo evaluate the effect of compound oxymatrine injection combination with chemotherapy in patients with advanced ovarian carcinoma.MethodsEighty-six patients with advanced ovarian carcinoma were divided by random digits table method into observation group(43 cases) receiving compound oxymatrine injection plus chemotherapy(paclitaxel plus carboplatin) and control group (43cases) treated with chemotherapy(p aclitaxel plus carboplatin) alone.The change of white blood cell,adverse reaction and effect of the patients were observed.ResultsThe white blood cell of observation group after treatment was obviously higher than that of control group [ (4.7 ± 1.4) × 109/L vs.(3.2 ± 1.6) × 109/L,P < 0.05 ].The rates of nausea and vomiting,transaminase elevating and kidney damage of observation group were obviously lower than those of control group[ 16.3% (7/43),14.0% (6/43),20.9% (9/43) vs.34.9%(15/43),32.6% (14/43),44.2% ( 19/43 ),P < 0.05 ].The complete remission rate of observation group was significantly higher than that of control group[55.8%(24/43) vs.27.9%(12/43),P < 0.05].Conclusion Addition of compound oxymatrine injection to chemotherapy for patients with advanced ovarian carcinoma can effectively protect patients against the decreasing of the white blood cell and attenuate the side effects brought by chemotherapy.
3.Evaluation of the effect of humanized nursing on improvement of treatment compliance and negative emotional state in patients with ovarian cancer

ObjectiveTo investigate the effect of humanized nursing in ovarian cancer patients to improve treatment compliance and negative emotional state. Methods120 cases of ovarian cancer patients from October 2009 to October 2010 in our hospital were chosen as the research object.They were divided into the control group and the observation group according to randomization methods with 60 cases in each group.The control group patients were taken routine care.The patients in the observation group were taken humanized nursing on the basis of routine care.The treatment compliances during treatment were compared.The anxiety,depression scores for the two groups of patients before and after treatment were compared.Based on the three indicators,the effect of humanized nursing in ovarian cancer patients to improve treatment compliance and negative emotional state was evaluated. ResultsThe treatment compliance during treatment for the two groups was compared,the full compliance of patients of the observation group was significantly higher than the control group.The difference between the two groups was statistically significant.The anxiety,depression scores before treatment for the two groups were compared.There was no significant difference between the two groups.The anxiety and depression scores showed significant improvement after the treatment,but improvement level of the observation group was significantly higher than the control group.The difference between the two groups was statistically significant. ConclusionsHumanized nursing can improve the treatment compliance and negative mood of ovarian cancer patients effectively,it is conducive to the rehabilitation of patients,and is worthy of clinical application.
4.LOW RESPONSIVENESS OF CULTURED HUMAN INTESTINAL EPITHELIAL CELLS TO LPS STIMULATION AND ITS POSSIBLE MECHANISM

To investigate the mechanism of low responsiveness of cultured human intestinal epithelial cells (IECs) to LPS stimulation, IL-8 secretion level was detected by ELISA in supernatants harvested after the IECs were treated with LPS for 18h, and TLR4, TLR2, CD14 and MD2 mRNA were detected by reverse transcriptase-polymerase chain reaction(RT-PCR) and RNase protection assay(RPA). It was found that IL-8 was not detectable after IECs were treated with LPS and the expression levels of TLR4, TLR2, CD14 and MD2 mRNA of IECs were low and after exposed to LPS, the expressions of TLR4, CD14 and MD2 mRNA of IECs became further lower, whereas the expression of TLR2 mRNA didn't change obviously. The result suggests that low expressions of LPS signal transduction-associated receptors on IECs may be responsible for low response of IECs to LPS, so it confirms further that TLR4, CD14 and MD2 play important roles in transmembrane LPS signal transduction.
5.Simulation of acoustic response of microvessel containing microbubble in ultrasound field based on finite element analysis and lumped parameter model

BACKGROUND:Exploration on nonlinear acoustic response of the contrast agent microbubble contained in microvessel under ultrasound excitation is of great significance to maximizing ultrasonic energy deposition, promoting the development of quantitative imaging algorithm, revealing the damage mechanism or evaluating the targeted therapy, and overcoming the limitations of the traditional methods that are mainly used in large-size vessels, and measuring microvessel elasticity.
OBJECTIVE:To build a microvessel containing an ultrasound microbubble, revealing the internal mechanism among ultrasound, microbubble, blood flow and microvessel.
METHODS:Based on the finite element analysis and the lumped parameter model, three-dimensional microvessel containing microbubble model was built and simulated on Comsol Multiphysics 4.4 platform.
RESULTS AND CONCLUSION:Microbubble exhibited slower radial motion compared with axial motion due to vascular wal limitation, but maximum displacement and stress were found near the microbubble center because of the oscil ation coupling of the microbubble with the vascular wal . Under the same ultrasound pressure, the excitation frequency increased, accompanied by decreased and stabilized microvessl constriction and dilation;under the same frequency, with the enhancement of ultrasound pressure, the local microbubble oscil ation lasted longer. With the increase of Young’s modulus of the microvessel wal , the frequency of microbubble oscil ation was reduced, while the amplitude increased. Al these findings indicate that the frequency of microbubble oscil ation increased with the reduction of microvessel size, while its amplitude decreased. The frequency of microbubble oscil ation increased with the enhancement of ultrasound excitation, while the amplitude decreased. On the contrary, ultrasound pressure affected the dynamic characteristics of microbubble and microvessel. In particular, it was the first to demonstrate that the elasticity of microvessel has approximate linear positive correlation with the amplitude of microbubble oscil ation, which reveals the relationship between microvessel elasticity and microbubble response so as to provide theoretical basis for indirect measurement of microvessel elasticity.
6.Clinical detection on anti-Toxoplasma gondii antibody,antisperm antibody and interferon-? in infertile females

Objective To determine the levels of anti-Toxoplasma gondii antibody (ATAb) and antisperm antibody (AsAb) in serum samples and interferon-?(IFN-?) in supernatants of peripheral blood cell culture in females with infertility, and to analyze their correlation. Methods ELISA was applied to determine and compare the ATAb and AsAb in serum samples of 182 women with infertility and those of 94 women with normal pregnant history. The level of IFN-? in supernatants of peripheral blood cell culture was also measured and compared by ELISA. Results The positive rates of serum ATAb and AsAb in the cases of infertility were 15.38% and 18.13% respectively, which were significantly higher than those of women with normal pregnant history( 6.38% and 5.32%). 7.p38 MAPK regulates Th2 cytokines release in PBMCs in allergic rhinitis rats.

Th2 cytokines play a pivotal role in the pathogenesis of allergic rhinitis. To investigate the effect of p38 mitogen-activated protein kinase (MAPK) on the production of Th2 cytokines such as IL-4 and IL-5 in allergic rhinitis, a model of allergic rhinitis was established in SD rats. The expression level of p38 MAPK mRNA in PBMCs was detected by means of real time quantitative RT-PCR. The p38 MAPK activity in PBMCs was detected by Western blotting. PBMCs were cultured with various concentrations of p38 MAPK inhibitor SB 239063 or without the treatment, and then IL-4, IL-5 levels of the supernatant were determined by using sandwich ELISA. The results showed that mRNA expression and activity of p38 MAPK in PBMCs were significantly higher in allergic rhinitis rats than in control rats (P<0.05). The p38 MAPK inhibitor SB 239063 decreased the production of IL-4 and IL-5 in a dose-dependent manner. It is concluded that p38 MAPK plays an important role in the pathogenesis of allergic rhinitis which is associated with Th2 cytokines release.
8.Different concentrations of glycyrrhizin on hepatic stellate cell proliferation and its control mechanism

Objective To explore the different concentrations of glycyrrhizin on hepatic stellate cell proliferation and its regulation mechanism.MethodsHepatic stellate cell proliferation rate of three different concentrations of glycyrrhizin was detected by MTT assay.The expression of cyclin E was detected by immunocytochemistry.ResultsMTT assay was used to detect the cell proliferation inhabitation in different concentration groups,from control group 0.8 mg/L glycyrrhizin group to 1.2 mg/L glycyrrhizin group and 1.6 mg/L glycyrrhizin group.The cell proliferation inhibited rates were 0,2.73%,14.75 % and 25.96%.The difference was significant (P<0.05).The numbers of Cyclin E positive cells in different concentrations of glycyrrhizin group were significantly different compared with control group,and it was concentration dependently decreased (P<0.01).ConclusionsGlycyrrhizin concentration dependently inhibited rat hepatic stellate cells proliferation and it could reduce the expression of cyclin E and inhibit the proliferation of rat hepatic stellate cells.
9.Study on Du moxibustion health care underwear intervention effects on quality of life and immunity of sub-health Yang deficiency

Objective To observe the effect of Du moxibustion health care underwear intervention effects on quality of life and immunity of sub-health Yang deficiency.Methods Sub healthy Yang deficiency subjects were randomly divided into observation group and control group,30 cases in each group.The observation group in Du moxibustion health care underwear for 12 weeks,the control group in ordinary clothes,with quality of life scale (SF-36) to observe the change of their integral quality of life energy,physical function,emotional function and mental health of four indicators,IgA and IgG in blood were detected by scatter turbidity rate method.Flow cytometry was used to detect CD4+ and CD8+ in blood.Results The scores of physiological function,energy,emotional function and mental health in the observation group were significantly higher than those in the control group (P<0.01).The levels of IgA,IgG and CD4+ in bservation group were significantly higher than those before,and CD4+ was lower,and the difference were statistically significant when compared with the control group(P<0.05,P<0.01).Conclusion Du moxibustion health care underwear sub-health Yang deficiency can improve their quality of life and immunity.
10.HYPERLIPIDEMIA IN COPPER DEFICIENCY RATS

In order to investigate the interelationship between copper deficiency and lipidemia, an experiment was done in weanling rats fed normol and copper deficient diet for one month. At the end of experimental period, the growth rate, the contents of copper, zinc, cholesterol and triglyceride in plasma and the activity of monoamine oxidase (MAO) in plasma were measured.
