Journal of Chongqing Medical University 2003;0(06):-
Objective:To study the expression of cytokeratin (CK) 20 in the peripheral blood of patients with transitional cell carcinoma of the bladder and to evaluate its clinical significance.Methods:CK20 mRNA in 63 patients with TCCB,19 non-cancerous patients and 19 healthy volunteers was detected respectively.GAPDH was used as a general marker for normal blood cells.CK20 and GAPDH were amplified by using RT-PCR.Results:Nineteen healthy volunteers and 19 non-cancerous patients showed no CK20 signals in peripheral blood.Sixteen of 63(25.4%) patients with TCCB showed positive CK20 signals in peripheral blood, including 0/31 of stage T 1, 2/16 of stage T 2, 7/9 of stage T 3,7/7 of stage T 4.The CK20 expression difference between T 1/T 2 group and T 3-4 groups was significant. Two patients with positive CK20 signals in peripheral blood had distant metastases during a 6-month follow-up period.Conclusion:CK20 is applicable to detect cancer cells in the peripheral blood of transitional cell carcinoma of the bladder and may play an important role in the evaluation of the prognosis and postoperative management of TCCB.

Objective To study the efficacy and safety of reboxetine and citalopram in treating retardant depression. Methods A total of 59 patients with retardant depression were randomly divided into reboxetine group(n=29)and citalopram group(n=30)for the treatment of 6 weeks. The patients were assessed with the Hamilton depression scale(HAMD)and treatment emergent symptom scale(TESS).Results At the end of 6 weeks, the total scores of HAMD in reboxetine(18.48±6.38)group and citalopram(11.60±4.46)group both significantly decreased(P<0.001).Between two groups, the efficacy(82.8% and 56.7%)had significant differences(P<0.05)but side effects had no statistical significance. Conclusion Reboxetine has significant curative efficacy, milder adverse effects and higher safety for treating patients with retardant depression.

Objective:To investigate the eye movement anormalities of Obsessive-Compulsive Disorder (OCD) and their possible clinical significances. Methods: Exploratory eye movement in 57 patients with OCD according to DSM-IV criteria (27 medicated, 30 un-medicated) and 50 normal controls matched with sex, age and education were examined respectively. Results: Compared with normal controls, patients with OCD demonstrated low scores in most of the indices of exploratory eye movement. The rates of positive discrimiant score in OCD and normal controls were 37%(21/57) and 8% (4/50), respectively (X2 = 12.27, P

Objective: To examine the correlation of symptomatic feature and exploratory eye movements in schizophrenia. Methods: The eye movements of 60 patients with schizophrenia and 30 healthy control subjects were measured by the eye-mark recorder (model IV, Japan) and symptom profiles of patients were assessed using the Positive and Negative Syndrome Scale (PANSS) . Pearson correlation analysis was performed to examine the relationship between five-factor scores of PANSS, positive symptoms, negative symptoms and exploratory eye movements respectively. Results: The differences of all indices of eye movements between schizophrenic groups and normal control group had statistical significance ( P

MRI system is composed of many independent subsystems, in which RF, Gradient, Coil, MRI magnet subsystem are the most important and complex subsystems. Every subsystem consists of different components. Based on the analysis of MRI eliminable faults in CBMDisc (1995 ~ 2006), the types and occurrence frequency of MRI eliminable faults are expatiated so as to provide reliable data for reference.

Based on the analysis on "China biomedical medical literature database"(CBMdisc) in the field of MRI Maintenance composed by army writers from May,1994 to 2007,the following results can be obtained: the total number of writers amounts to 75,106 person-time,27 of them are the first writers, and 11 are main writers;among the total 48 pieces of work,33 pieces are written by more then one writer;the unit with the largest number of published writers is Jinan Military Area Command with Nanjing Military Area Command the second,Lanzhou Military Area Command the third;one main writer has a maximum of 7 pieces. The research makes a close study of the army writers in field of MRI maintenance, indicating the lack of intellectual personnel in the domain of Medicine Engineering in military system.

Objective To explore the effects of ligustrazine (TMP) on acute drug -induced free radical for-mation in guinea pig cochlear ,and to explore possible mechanisms of TMP on gentamycin(GM)-induced ototoxici-ty .Methods Sixty guinea pigs were randomly divided into four groups ,control group ,GM group ,TMP group and TMP+GM group .GM group were injected with GM 120 mg/kg per day .TMP group were injected with TMP 40 mg/kg per day .TMP+GM group were injected with GM and TMP at the same dosage .Control group were injected with normal saline .All groups were injected for consecutive 10 days .Before injection and one day after the last injec-tion ,ABR thresholds were measured .Biochemical assays of Superoxide Dismutase (SOD1 ,SOD2 ,T -SOD) activity and ma1ondia1dehyde(MDA) content in guinea pig cochlear were detected by TBA .Results After the injection , ABR thresholds in GM group were significantly increased compared with those of in control group (P<0 .01) ,with significant difference between GM +TMP and GM group(P<0 .01) .Before and after the experiment ,ABR thresholds in control group were almost unchanged(P>0 .05) .SOD activity was significantly decreased while MDA content was increased in cochlear tissues after GM injection (P<0 .05) .Co -treatment with TMP evidently enhanced SOD activity and decreased MDA content (P<0 .05) .Conclusion TMP may enhance SOD activity and prevent lipid per-oxidation ,thus alleviate GM ototoxicity ,and improve auditory function .

OBJECTIVE: To investigate the anti-tumor effect of bufalin and its regulation on Bcl-2 and Bax proteins in orthotopically transplanted tumor of human hepatocellular carcinoma in nude mice. METHODS: Orthotopically transplanted tumor of human hepatocellular carcinoma was established in nude mice. The mice were randomly divided into five groups: high-dose bufalin-treated group (1.5 mg/kg), medium-dose bufalin-treated group (1 mg/kg), low-dose bufalin-treated group (0.5 mg/kg), adriamycin-treated group (8.0 mg/kg), and normal saline-treated group. After 25 days, mice were sacrificed. The tumor volume was measured, and the pathological changes of tumor tissues were detected by HE staining to observe the tumor necrosis degree. Cell morphological changes were also observed by an electron microscopy. Label index of tumor cell apoptosis was assessed by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL), and the expressions of Bcl-2 and Bax proteins were determined by immunohistochemical method. RESULTS: The tumor volume in the bufalin-treated groups was shrunk significantly compared with that in the normal saline-treated group (P<0.01). The survival time of the bufalin-treated groups was prolonged compared with that of the adriamycin-treated group and the normal saline-treated group P<0.05. Apoptotic characteristics could be seen in tumor tissues of the bufalin-treated groups. The label index of tumor cell apoptosis in the bufalin-treated groups (5.87+/-2.13, 8.86+/-2.96 and 10.60+/-3.42 in low-, medium- and high-dose groups respectively) was higher than that in the adriamycin-treated group (3.28+/-0.98) (P<0.05, P<0.01). The expression of Bax was up-regulated, while no changes were detected as to Bcl-2 protein in tumors of the bufalin-treated groups. CONCLUSION: Bufalin has significant anti-tumor effect on the orthotopically transplanted tumor of human hepatocellular carcinoma in nude mice. Its effect might be related to up-regulation of Bax protein and inducement of the tumor cell apoptosis.
