1.C-phycocyanin induces Heme oxygenase-1 expression to protect acute lung injury in septic rats

Objective To observe the protective effect and molecular mechanism of C-phycocyanin (CPC) on acute lung injury (ALI) in septic rats. Method 75 SD rats were randomly divided into control group, model group and CPC group. Cecal ligation and puncture was used to establish a septic acute lung injury rats (model group). For the CPC groups, septic acute lung injury rats were administrated by 20, 40 and 60 mg/kg CPC by intraperitoneal injection. 72 h after the operation, serum and lung tissue were obtained, the wet to dry weight ratio, the content of TNF-α、IL-6 and IL-10 in bronchoalveolar lavage fluid, the activity of myeloperoxidase (MPO) was analyzed. Expression of heme oxygenase (HO)-1,activation of nuclear factor erythroid 2-related factor 2 (Nrf 2) and nuclear factor-kappa B (NF-B) were detected by Western blot. Superoxide and Nitrite/Nitrate Level production in Lungs and bronchoalveolar lavage fluid were measured by chemiluminescence and reduction method, respectively. Results Treatment with CPC significantly inhibited septic-induced inflammatory responses including elevation of superoxide formation, myeloperoxidase activity, leucocytes and protein infiltration in lung tissues, and production of proinflammatory cytokine, and nitrite/nitrate in bronchoalveolar lavage fluid (P<0.05). In addition, CPC could activate Nrf 2 and induce HO-1 expression, and inhibit NF-B activation in ALI rats. However, blocking HO-1 activity by tin protoporphyrin IX (SnPP), an HO-1 inhibitor, markedly abolished these beneficial effects of CPC in septic-induced ALI. Conclusion The protection mechanism of CPC may be through HO-1 induction and suppressing of NF-kB-mediated inflammatory responses.
2.Resistant Characteristic and Mechanisms of Imipenem Resistance in Acinetobacter baumannii

OBJECTIVE To investigate the resistance of Acinetobacter baumannii(ABA),and the mechanism of imipenem resistance in A.baumannii.METHODS All specimens were identified by VITEK-60 and the drug resistance was detected by Kirby-Bauer test.Three dimensional test was used to detect ESBLs and AmpC.PCR and DNA sequencing were performed to determine VIM,IMP,OXA-23 and OXA-24 ?-lactamases.Outer membrane protein was analyzed by SDS-PAGE,Reserpine synergistic inhibition test was used to study the active efflux mechanism.RESULTS Totally 120 strains were isolated from sputum(76.9%),and 16 strains from secretion(10.3%).ICU was the main infected department(51 strains,32.7%).The resistance to sulperazone was the lowest(20.5%),and to imipenem accounted for 38.5%,Of the 20 imipenem resistant strains,10 strains were ESBLs positive(50%),and 20 strains were AmpC positive(100%).VIM,IMP and OXA-24 ?-lactamases were not detected out,19 strains(95%) produced OXA-23.Compared to the imipenem-sensitive strains,the resistant strains lacked the outer membrane protein of 22?103,29?103 and 33?103.The MICs of A.baumannii to imipenem were not decreased by reserpine which demonstrated that excretive mechanism was negative.CONCLUSIONS ICU is the main infected department for ABA.The resistance rate is increasing for longer-term usage of carbopenem;OXA-23 production is the important resistance mechanism in ABA,AmpC production and outer membrane protein lacking show close relation to the drug-resistance in A.baumannii.
3.Comparative analyses of histological and material properties of reticular dermis derived from human, swine and rats

BACKGROUND:Previous studies of our research group have confirmed that the texture of porcine reticular dermis at lateral ventral part is softer and has more extensibility than other parts. Therefore, it may serve as the raw material of xenogenic aceluar dermal matrix. However, its comparison with human and rat reticular dermis has not been reported systematicaly in aspects of histomorphology and material characterization. OBJECTIVE:To compare the reticular dermis from the lateral region of porcine abdomen and rat dorsal part with the reticular dermis from human in histology, biomechanics, molecular structure, thermal stability and other properties. METHODS:The reticular dermis samples were taken from adult human, the lateral region of porcine abdomen, the back of rats, for gross observation. Paraffin sections were observed by hematoxylin and eosin staining and sirius red staining under a light microscopy. The relevant data of micrograph were measured by imagine analysis software. These samples were also vacuum-freezing dried and rehydrated, and then their mechanical properties were tested with a electronic tensile machine to calculate the Young’s modulus. Some vacuum-freezing dried samples were powdered and detected by Fourier transform infrared spectrometer and simultaneous thermal analyzer. RESULTS AND CONCLUSION:There were no differences in colagen fiber bundle diameter of the reticular dermis from adult human and the lateral region of porcine abdomen, but the reticular dermis from the back of rats was thinner than that from adult human (P < 0.01). The gap between the reticular layer of the dermis of the lateral region of porcine abdomen was lower than that from adult human (P < 0.0.1); however, there was no difference in the gap between the reticular layers of the dermis of the rat back and adult human (P=0.17). Colagen fibers of porcine reticular dermis were arranged tightly in disorder; the content of type I colagen and ratio of type I/III colagen were higher than those in the reticular dermis from adult human (P < 0.05), but the content of type III colagen was less than that in the reticular dermis from adult human (P < 0.05). Contents of type I and III colagen and their ratio were similar between the reticular dermis of rats and adult human (P > 0.05). There was no significant difference in the Young’s modulus of the three kinds of reticular dermises. Hydrogen bonds involved in the reticular dermal colagen molecules ranged as folows: rats > swine > human. Rat reticular dermis has better thermal stability than that of swine and adult human.
4.Relationship between maternal red cell index in early pregnancy and gestational diabetes mellitus in Kunming

Objective To investigate the role of hemoglobin (Hb),red blood cell count (RBC) and hematocrit (HCT) during the first trimester in predicting the occurrence of gestational diabetes mellitus (GDM) in the third trimester in nonanemic women in Kunming.Methods A prospective study was carried out.Routine blood tests were performed in 1189 nondiabetic singleton pregnant Chinese women at their initial prenatal healthcare before 14 gestational weeks in the First Affiliated Hospital of Kunming Medical University from January 1,2008 to December 31,2009.They were divided into four groups by quartiles of Hb,RBC and HCT levels respectively.Differences of GDM incidence at the third trimester were compared among groups.Kollmogorov Smirnov test was used to detect normal distribution of Hb,RBC and HCT results.Differences between rates of groups were detected by Chi-square test.Independent risk factors of GDM were screened by Logistic regression analysis.Results The incidence of GDM at the third trimester increased as quartile categories of Hb,RBC and HCT ascended.The GDM incidence (13.1 %,34/259) of ≥75th Hb group was higher than that of < 25th group (5.9 %,18/304,x2 =8.53,P< 0.0 1),25th ~ group (9.2 %,30/326,x2=4.18,P<0.05) and 50th~ group (9.3%,28/300,x2=3.96,P<0.05).The GDM incidence (11.7 %,34/290) of 50th~ RBC group (RBC≥4.44 × 1012/L) was higher than that of <25th group (6.3%,19/304,x2=5.30,P<0.05),25th~ group (7.0%,21/298,x2 =3.93,P<0.05); and the GDM incidence (12.1%,36/297) of ≥75th group was also higher than that of <25th group (6.3%,19/304,x2 =6.49,P<0.05),25th~ group (7.0%,21/298,x2=3.85,P<0.05).The GDM incidence of <25th HCT group (5.3%,16/303) was lower than that of 25th~ group (9.3%,29/311,x2=7.04,P<0.01),50th~ group (10.8%,31/287,x2=3.93,P<0.05) and ≥75th group (11.8%,34/288,x2=6.49,P<0.05).Logistic regression analysis showed that Hb (OR=1.031,95% CI:1.010-1.052),RBC (OR=2.286,95%CI:1.318-3.963) and HCT (OR=1.106,95% CI:1.037-1.179) were independent risk factors of GDM (all P<0.05).Conclusions High maternal Hb,RBC and HCT levels in the first trimester could be used in predicting GDM in pregnant women in Kunming area.
5.Reaction kinetics on renal calculus dissolution by simulating test in vitro

Objective To investigate the reaction kinetics between the stone-dissolving solution and the urinary stone in a model simulating the condition of a kidney for further clinical administration. Methods An artificial upper urinary tract was made by silica gel. Lactic acid prepared in the preliminary study was used to react with artificial stone in the model. The concentration of reaction product in the effluent was measured to identify the reaction velocity. Relationships between the efficiency and dissolution rate or stone surface was investigated. Results The highest utilization rate of dissolution was 100 ~ 150 mL/h. Dissolution efficiency is in positive rela-tion with stone surface. The efficiency correlates with the stone surface and infusion speed in the range of 50~400 mL/h. Conclusions Before dissolution treatment ,the stone should be shattered as deeply as possible to in-crease the surface of reaction. If possible ,the irrigating speed should be as high as possible to eliminate the stone sooner.
6.Comparison of retroperitoneal mini-incision and laparoscopic methods in treatment of varicocele

Objective To evaluate the best mean for the treatment of varicocele. Methods The clini-cal effects of the high ligation of spermatic vein under the small incisal opening of the poritneum meta abdom-inal membrane and the ligation of spermatic vein under the laparoscope for the varicocele were retrospectively reviewed. The operating time, the postoperative days and the cost of hospitalization were compared of the two methods. Results Comparing with the group of operation and the group of laparoscope. Among the 158 vat-icocele patients with unilateral or bilateral varicocele, there was not statistical difference in the operating time between the two methods (P > 0. 05), whi]e there was significant difference in the postoperative days and the cost of hospitalization(P <0. 05). Conclusions When the bilateral varicoceles were simultaneously ligated, the high ligation of spermatic vein under the laparoscope was the best selection; if the unilateral varicocele was highly ligated or the patients were unconditional or the patients were not fit for laparoscopic operation, the high ligation of spermatic vein under the small incisal opening of peritoneum was the best operating method.
7.Epidemiological characteristics of influenza B virus in Guizhou Province in 2013-2016

Objective To understand the epidemic characteristics and regularity of influenza B virus in Guizhou Province, and provide scientific evidence for the control and prevention of influenza.Methods Results of reverse transcription polymerase chain reaction(PT-PCR) of influenza B virus in Guizhou Province from April 1, 2013 to March 31, 2016 were statistically analyzed.Results A total of 1 904 samples were detected influenza B virus by RT-PCR, B/Yamagata (By) lineage and B/Victoria (Bv) lineage were 1 215 and 642 respectively.In April 2013-March 2014 and April 2014-March 2015, the predominant strains of influenza B were both By lineage, in April 2015-March 2016, the predominant strains of influenza B were Bv and By lineages, the epidemic peaks were in winter and spring;there's a higher positive percentage of influenza B in male, accounting for 56.83%;the highest detection rate of influenza B virus was found in population aged <15 years(70.80%),Bv and By lineages were the highest in the 0~ (42.37%) and 5~ age groups (35.56%) respectively;the main pathogen causing mixed infection was By+Bv (67.65%),mixed infection with influenza B virus accounted for 95.59%.Conclusion There are two lineages By and Bv epidemic in Guizhou Province, the epidemic peaks of influenza B are in winter and spring, male cases are higher than female, people under 15 years old are the high-risk group for influenza B, it is of great significance to strengthen the vaccination and surveillance of influenza in low age population.
8.Molecular characteristic analysis of neuraminidase genes of avian influenza virus H9N2 in environments in Weining, Guizhou Province during 2015-2017

Objective To understand the genetic variations of neuraminidase (NA) genes of avian influenza virus H9N2 in Weining,Guizhou Province,and to provide the scientific evidence for the prevention and control of avian influenza virus.Methods Ribonucleic acids (RNA) were extracted and NA genes were amplified and sequenced from 13 randomly selected H9N2 positive samples from the live poultry market (LPM)environments in north of Weining Yi and Hui and Miao autonomous county (Weining),Guizhou Province during 2015 to 2017.Then the homology,genetic evolution,and sites of stalk deletion areas,potential N-glycosylation,receptor binding regions and drug resistance of H9N2 subtype avian influenza viruses were analyzed by a series of bioinformation software.Results Homology analysis revealed that there were 93.0%-100.0% and 92.1%-100.0% similarity among 13 strains H9N2 avian influenza viruses in nucleotide and amino acid of the NA gene,respectively.All strains belonged to DK/HK/Y280/97 sub-lineage,but their genetic sources were complex and diverse.Thirteen strains had a stalk deletion of 3 amino acid residues TEI at positions 63-65 and 3 isolates had mutation QN to QK at positions 39-40.The potential N-glycosylation sites at amino acid residues 86,146,200,and 234 of the NA protein of all strains were highly conserved,while other N-glycosylation sites had quantity and site mutations.There were different mutation types at the three sialic acid binding site areas,especially at 399-404 area.All NA protease activity sites and key sites of the 13 strains had no mutations associated with resistance to the neuraminidase inhibitor drugs.Conclusions All 13 strains H9N2 viruses belongs to DK/HK/Y280/97 sub-lineage in Weining,Guizhou Province during 2015-2017,and their genetic sources are complex and diverse.The mutations on sites of stalk areas,potential N-glycosylation and sialic acid binding site areas are presented at different degrees.Hence,enhancing surveillance and controlling H9N2 avian influenza virus is necessary.
9.Characteristics of dermatitis herpetiformis in China: a literature review from 1995 to 2018

Objective:To summarize characteristics of dermatitis herpetiformis (DH) patients in China.Methods:Published literature on DH in Chinese patients was searched in China National Knowledge Infrastructure database, Wanfang data and PubMed database from establishment of these databases to December 2020. Patients were screened according to the inclusion and exclusion criteria, and characteristics of patients meeting the diagnostic criteria were analyzed.Results:A total of 55 patients with DH were involved in this study. Among these patients, the average age of onset was 44.9 ± 18.5 years, the ratio of males to females was about 2.5∶1, and the average period of delayed diagnosis was 4.1 years. Skin lesions mainly manifested as tense blisters on an erythematous base, and were distributed on the buttocks, elbows, back, knees, etc. Histopathological examination of 39 cases showed subepidermal blisters in 37, and neutrophil infiltration in 17. Direct immunofluorescence study of perilesional skin tissues of 33 cases showed granular IgA deposition in 31, and IgA deposits limited to the dermal papilla in 23. Among 39 cases with available data on treatment, 25 were treated with dapsone alone or in combination with a gluten-free diet, most of whom achieved marked therapeutic outcomes with subsidence of skin lesions within 1 month.Conclusions:DH in China mainly clinically manifests as tense blisters on an erythematous base, and is mainly characterized by granular IgA deposition under a direct immunofluorescence microscope. Genetic factors are involved in its etiology, and dapsone is effective for its treatment.
