1.Screening of Phage Display Peptide Libraries Using ComplexBiological Systems as Selector Targets

During recent years, some new methods for phage display panning have been developed by using complex biological systems instead of purified antigens, antibodies or receptors as selector targets. These complex biological systems include living cells, viruses, mouse tissues and tumors etc. The peptides identified by using tumors as selector molecules can selectively home to blood vessels of experimental tumors and may have potential applications in targeted anti-tumor treatment.
2.Purification and characterization of non-fusion recombinant human interleukin-18 expressed in E. coli

Objective To develop the purification method of rhIL 18 expressed in E coli Methods The harvested cells were disrupted using lysozyme and sonication The rhIL 18 inclusion bodies were extracted by centrifugation Washed inclusion bodies were solubilized in 8 mol/L urea The solubilized inclusion bodies were first purified by ion exchange chromatography under denaturing conditions After renaturation, gel filtration chromatography was used for further purification Samples were analyzed by SDS PAGE, HPLC, Western blotting and N terminal amino acid sequencing The biological activity of purified rhIL 18 was also measured Results It was found that rhIL 18 was expressed intracellularly in E coli as insoluble inclusion bodies The purity of rhIL 18 in extracted inclusion bodies was above 80% The final purified rhIL 18 was of high purity and exhibited the mass of molecule 19?10 3 by SDS PAGE The sequence of 15 amino acid residues form NH2 terminus of the purified rhIL 18 was consistent with the predicted sequence The purified rhIL 18 also induced IFN ? production of Con A stimulated human PBMC Conclusion The established method for purifying rhIL 18 is simple and effective and might be useful in the development of the large scaled purification process of rhIL 18
3.An empirical study on the payment effect of hospitalization expense under the urban employee basic medical insurance in Guangzhou

Objective To evaluate the payment effect of hospitalization expense under the urban employee basic medical insurance in Guangzhou.Methods Analyze the differences between the expenses paid by medical insurance with actual hospitalization expenses of the 22 settlement units from 1 5 tertiary hospitals,and the expense settlement data of the insured patients,based on the early,the mid-term,and the recent periods since the urban employee basic medical insurance was implemented since 2002.Results The ratio of good payment effect units reduced to 9.09%(the recent)from 42.86%(the earlier).The ratio of poor payment effect units was 42.86% in the early,to mid-term 18.18%,and sharply increased to 77.27%in the near term.The settlement units which exceeded its flat standard accounted for 52%,50%, and 91%respectively(in the early,the mid-term,and the recent).The medical insurance agency and the hospital shared the overrun costs by 52.88%and 47.12%respectively in 201 1.Conclusion The payment effect of the urban employee medical insurance was greatly influenced by the adjustment of medical insurance policy and the payment ability of the pooling fund.It should improve the payment effect of the medical insurance timely,so as to ensure the hospitals’operation normally.The hospitals should take the effective cost management measures so as to deal with the increasing cost control pressure.
4.Operative treatment of degenerative sympotomatic lumbar scoliosis:an analysis of 37 patients

[Objective]To evaluate the surgical results and value of posterior lumbar decompression and intervertebral bone fusion with pedicle screw interval fixation in degenerative sympotomatic lumbar scoliosis.[Method]From Jan.1999 to Apr.2006,37 cases with degenerative sympotomatic lumbar scoliosis underwent posterior lumbar decompression and intervertebral bone fusion with pedicle screw interval fixation,including 21 male and 16 female patients,aged 62.6 years old averagely.All the patients were assessed by routine X-ray and Bending X-ray.CT and MRI were performed necessarily.According to patients ' symptom,total or semi-laminectomy,intervertebral bone fusion with pedicle screw interval fixation were performed.[Result]The fellow-up period varied from 1 year to 6 years(averaged,2 years and 7 months),clinical symptoms were effectively relieved after operation in all cases.Neurological functions were evaluated using JOA system,the clinical success rate in all 37 patients was 91.9%.The corrective rate was 54.8%.The average solid arthrodsis time was 11.6 weeks.The height of intervertebral disc and corrective rate had no loss.All patients X-ray of the third month and the first year didn 't show the interplantation 's clinch and breakage.[Conclusion]In the treatment of the elderly patients with degenerative sympotomatic lumbar scoliosis,first of all,the particular principle of surgical operation should be applied,the old age and long course of disease are not absolute contraindication.In addition,The technique of posterior lumbar decompression and intervertebral bone fusion with pedicle screw interval fixation can be safely performed in the elderly patients with degenerative sympotomatic lumbar scoliosis.
5.Clinical value of peripheral platelet count in assessing active pulmonary tuberculosis

Objective To understand the role of increasing peripheral blood platelet count in the diagnosis and prognosis of patients with active pulmonary tuberculosis.Methods This analysis included 80 inpatients with active pulmonary tuberculosis evidenced by positive sputum smear,43 patients in recovery and 89 healthy controls during the period from January to June in 2012.Peripheral white blood cell,platelet count,and neutrophil percentage were assayed.Erythrocyte sedimentation rate (ESR)and serum C-reactive protein (CRP)were measured and compared.The data were analyzed by using SPSS 13.0 software.Results ESR and CRP values were significantly higher in the patients with active pulmonary tuberculosis than in the patients in recovery (P=0.006 5 and P=0.007 3,respectively).The peripheral blood platelet count exceeded normal range in 41.94% of the patients with active pulmonary tuberculosis,which was significantly higher than that in the patients in recovery (P=0.001 4)and controls (P=0.000 0).Platelet count was positively correlated with CPR (r=0.515,P<0.000 1)and ESR (r=0.398,P<0.001)value.However,peripheral white cell count and neutrophil count were not different from the corresponding values of controls.Conclusions Increasing peripheral platelet count may play a role in assessment of tuberculosis disease activity and the effect of anti-tuberculosis treatment.
6.Experimental study on anti-inflammatory and analgesic effects of Yitieling Paste

OBJECTIVE: To observe the anti-inflammatory and analgesic effects of Yitieling Paste (YTLP). METHODS: YTLP and aspirin (as a control drug) were introduced to treat the edematous ears of mice induced by xylene and swollen toes of mice induced by carrageenin, and to relieve the pain in mice induced by heat and acetic acid. The swelling degree, pain threshold and body distortions were measured. RESULTS: The repression rates of the ear edema in groups of YTLP of high and low dosages and the aspirin group were 67.92%, 52.52% and 58.28%, respectively. The repression rate of the toe swelling in YTLP high dosage group was higher than that of the aspirin group, which was higher than that of the YTLP low dosage group. The results of analgesic effect of the three YTLP-treated groups showed that with the increase of dosage, the pain thresholds were higher and higher, and the body distortions were lower and lower. The pain thresholds of high and medium dosage YTLP groups were near to the aspirin group. CONCLUSION: YTLP possesses a strong anti-inflammatory and analgesic effect.
7.Practice and reflection on enhancing clinical pathology general education in clinical medicine graduates

Because of ignoring the basic knowledge training, the medicine graduates lack ba-sic knowledge of clinical pathology, which has seriously affected their improvement of medical skills. The paper analyzes the status of pathology teaching in clinical medicine graduates and the necessity to strengthen clinical pathology general education. Then it proposes measures about enhancing clinical pathology general education, so as to increase the basic clinical pathology knowledge of medical grad-uates.
8.Production and identification of specific monoclonal antibodies against recombinant human interleukin-18(hIL-18)

Objective:To produce hybridoma cell lines of monoclonal antibodies against recombinant human interleukin 18.Methods:Hybridoma techniques were used to fuse BALB/C mice myeloma cell line SP2/0 with B lymphocytes of spleen of BALB/C mice which were immunostimulated with interleukin 18 antigen through polyethyleneglycol (PEG).Single cell lines which stably produce antibodies against interleukin 18 were picked out and identified by enzyme linked immunosorbent assay.Results:Two hybridoma cell lines were obtained and named 1C7 and 1F5,which produce monoclonal antibodies specificity against interleukin 18.They respectively own 88 and 92 chromosomes, the purities of IgG in ascites are all over 95% through affinity chromatography, the titer degrees of ascies are 1?10 -4 and 1?10 -5 respectively.Two antibodies are against two different antigen binding epitopes on the surface of interleukin 18.It was evaluated by Western blot that the two antibodies can bind to interleukin 18 specifically and are made of only single protein strap.Conclusion:Have prepared two hybridoma cell lines which stably secrete high titer and high specific monoclonal antibodies against interleukin 18,which provide the foundation for further studying the structure and the biological function of interleukin 18.

10.Rapid Identification of Traditional Chinese Medicine Using Electronic Nose Based on RBF-RF Cascade Classifier

This study was aimed to apply the electronic nose (E-nose) in the research of traditional Chinese medicine (TCM). The discussion was made on difficulties of using E-nose. The solution plan was proposed and the discrimination model was established. It provided a simple, rapid and effective analysi method in the identification of TCM. It also provided new ideas for the research and application of gas sensor arrays. E-nose was used in the ex-traction of TCM scent characteristics. Based on ion mobility spectrometry of MOS sensor, the fingerprint of TCM scent was established. The maximum response value of the sensor was used as analysis index. According to the diffi-culties of identification, two solution plans were proposed. Firstly, different detectors were employed to complete the classification. Secondly, radial basis function (RBF) and random forests (RF) were combined and then a cascade classifier was constructed in order to achieve the maximum of information obtained in conditions where the number of measurements, metal oxide semiconductor sensors in E-nose was limited. The results showed that both plans were accurate and practical with relatively high upper correct judge rate and better cross-validation (The highest upper correct judge rates were 95% and 100%, 96% and 80%, respectively). It was concluded that this study firstly ap-plied cascade classifier in the establishment of TCM identification by E-nose. With limited amount of sensors, the maximum information was received through data mining. Using E-nose in the identification of TCM was rapid and accurate. The established pattern recognition method was maneuverable with accurate identification rate and stability compared to conventional sensory identification method. It provided a simple and rapid analysis method for the iden-tification of TCM.
